2A structural view of the antibiotic degradation enzyme NDM-1 from a superbug显示文摘Gram-negative Enterobacteriaceae with resistance to carbapenem conferred by New Delhi metallo-β-lactamase 1(NDM-1)are a type of newly discovered antibioticresistant bacteria.The rapid pandemic spread of NDM-1 bacteria worldwide(spreading to India,Pakistan,Europe,America,and Chinese Taiwan)in less than 2 months characterizes these microbes as a potentially major global health problem.The drug resistance of NDM-1 bacteria is largely due to plasmids containing the blaNDM-1 gene shuttling through bacterial populations.The NDM-1 enzyme encoded by the blaNDM-1 gene hydrolyzes β-lactam antibiotics,allowing the bacteria to escape the action of antibiotics.Although the biological functions and structural features of NDM-1 have been proposed according to results from functional and structural investigation of its homologues,the precise molecular characteristics and mechanism of action of NDM-1 have not been clarified.Here,we report the threedimensional structure of NDM-1 with two catalytic zinc ions in its active site.Biological and mass spectroscopy results revealed that D-captopril can effectively inhibit the enzymatic activity of NDM-1 by binding to its active site with high binding affinity.The unique features concerning the primary sequence and structural conformation of the active site distinguish NDM-1 from other reported metallo-β-lactamases(MBLs)and implicate its role in wide spectrum drug resistance.We also discuss the molecular mechanism of NDM-1 action and its essential role in the pandemic of drug-resistant NDM-1 bacteria.Our results will provide helpful information for future drug discovery targeting drug resistance caused by NDM-1 and related metallo-β-lactamases.Yu Guo Jing Wang Guojun Niu Wenqing Shui Yuna Sun Honggang Zhou Yaozhou Zhang Cheng Yang Zhiyong Lou Zihe Rao 2011Protein & Cell2011,2,5:16
3Automatic Malware Classification via PRICoLBP显示文摘Creating effective features is a critical issue in malware analysis. It requires a proper tradeoff between discriminative power and invariance. Previous studies have shown that it is fairly effective to design features based on the binary code. However, the current existing binary-based features seldom take into consideration the problem of obfuscation, such as relocated sections, incomplete code and redundant operations. In this paper, we propose a novel Pairwise rotation invariant co-occurrence local binary pattern(PRICo LBP) feature, and further extend it to incorporate the Term frequency-inverse document frequency(TFIDF) transform. Different from other static analysis techniques, our method not only achieves better linear separability, but also appears to be more resilient to obfuscation. In addition, we evaluate PRICo LBPTFIDF comprehensively on three datasets from different perspectives, e.g., classification performance, classifier selection and performance against obfuscation. 4From graphite to porous graphene-like nanosheets for high rate lithium-ion batteries显示文摘Graphene nanosheets possess a promising potential as electrodes in Li-ion batteries (LIBs); consequently, the development of low-cost and high-productivity synthetic approaches is crudal. Herein, porous grapheneqike nanosheets (PGSs) have been synthesized from expandable graphite (EG) by initially intercalating phosphoric acid, and then performing annealing to enlarge the interlayer distance of EG, thus fadlitating the successive intercalation of zinc chloride. Subsequently, the following pyrolysis of zinc chloride in the EG interlayer promoted the formation of the porous PGS structure; meanwhile, the gas produced during the formation of the porous structure could exfoliate the EG to graphene-like nanosheets. The synthetic PGS material used as LIB anode exhibited superior Li+ storage performance, showing a remarkable discharge capacity of 830.4 mAh.g-1 at 100 mA.g-1, excellent rate capadty of 211.6 mAh'g-1 at 20,000 mA-g-1, and excellent cycle performance (near 100% capacity retention after 10,000 cycles). The excellent rate performance is attributed to the Li+ ion rapid transport in porous structures and the high electrical conductivity of graphene-like nanosheets. 5Effect of Post-weld Heat Treatment on Properties of Friction Welded Joint Between TC4 Titanium Alloy and 40Cr Steel Rods显示文摘Dissimilar metal joining of Ti–6Al–4V(TC4) titanium alloy to as-rolled 40 Cr steel rods was conducted with friction welding, and the effect of post-weld heat treatment(PWHT) on the microstructure and mechanical properties of the resultant joints was investigated. The average tensile strength of the as-welded joints reached 766 MPa and failure occurred in 40 Cr steel base metal. However, after PWHT at 600 °C for 0.5, 1, 2 and 3 h, the tensile strength of the joints decreased and fracture happened through the interface with quasi-cleavage features. The bending angle of specimens was improved from 9.6° in as-welded state to 32.5° after PWHT for 2 h. The tensile strength of the joint was enhanced by martensitic transformation near the interface in as-welded state. Sorbite formed near the interface in PWHT state and improved the bending ductility of the joint. Ti C brittle phase formed at the interface after PWHT for 0.5 h and deteriorated the tensile strength and bending ductility of the joint. After PWHT for 2 h, no Ti C phase was detected at the interface. 7The nucleoprotein of severe fever with thrombocytopenia syndrome virus processes a stable hexameric ring to facilitate RNA encapsidation显示文摘Severe fever with thrombocytopenia syndrome virus(SFTSV),a member of the Phlebovirus genus from the Bunyaviridae family endemic to China,is the causative agent of life-threatening severe fever with thrombocyto-penia syndrome(SFTS),which features high fever and hemorrhage.Similar to other negative-sense RNA viruses,SFTSV encodes a nucleocapsid protein(NP)that is essen-tial for viral replication.NP facilitates viral RNA encapsida-tion and is responsible for the formation of ribonucleopro-tein complex.However,recent studies have indicated that NP from Phlebovirus members behaves in inhomogene-ous oligomerization states.In the present study,we report the crystal structure of SFTSV NP at 2.8Åresolution and demonstrate the mechanism by which it processes a ring-shaped hexameric form to accomplish RNA encapsida-tion.Key residues essential for oligomerization are identi-fi ed through mutational analysis and identifi ed to have a signifi cant impact on RNA binding,which suggests that correct formation of highly ordered oligomers is a criti-cal step in RNA encapsidation.The fi ndings of this work provide new insights into the discovery of new antiviral reagents for Phlebovirus infection.Honggang Zhou Yuna Sun Ying Wang Min Liu Chao Liu Wenming Wang Xiang Liu Le Li Fei Deng Hualin Wang Yu Guo Zhiyong Lou 2013Protein & Cell2013,4,6:4
8Optical wafer defect inspection at the 10 nm technology node and beyond显示文摘The growing demand for electronic devices, smart devices, and the Internet of Things constitutes the primary driving force for marching down the path of decreased critical dimension and increased circuit intricacy of integrated circuits. However, as sub-10 nm high-volume manufacturing is becoming the mainstream, there is greater awareness that defects introduced by original equipment manufacturer components impact yield and manufacturing costs. The identification, positioning, and classification of these defects, including random particles and systematic defects, are becoming more and more challenging at the 10 nm node and beyond.Very recently, the combination of conventional optical defect inspection with emerging techniques such as nanophotonics, optical vortices, computational imaging, quantitative phase imaging, and deep learning is giving the field a new possibility. Hence, it is extremely necessary to make a thorough review for disclosing new perspectives and exciting trends, on the foundation of former great reviews in the field of defect inspection methods. In this article, we give a comprehensive review of the emerging topics in the past decade with a focus on three specific areas:(a) the defect detectability evaluation,(b) the diverse optical inspection systems,and(c) the post-processing algorithms. 9WAF-Based Chinese Character Recognition for Spam Image Filtering显示文摘We address the problem of filtering image spam, a kind of rapidly spread spam in which the text is embedded into images to defeat text-based spam filter.Particularly, we focus on image spam with Chinese text as 'spam' which is a more challenging task. A popular way to detect image spam is by Optical character recognition(OCR) system, which detects and recognizes the embedded text, then followed by a text classifier that discriminate spam from ham. However, spammers start to obscure image text to prevent OCR system discovering the spam text. To compensate for the shortcomings of OCR system, a novel method which essentially is a keyword reconstruction algorithm based on Word activation force(WAF) model is proposed. It is effective on discovering keywords, hence is benefit for the later classification stage and notably improve the performance of image spam filtering.
