| 1 | Antidiabetic Effects of Gegen Qinlian Decoction via the Gut Microbiota Are Attributable to Its Key Ingredient Berberine显示文摘Gegen Qinlian Decoction(GQD),a traditional Chinese medicine(TCM)formula,has long been used for the treatment of common metabolic diseases,including type 2 diabetes mellitus.However,the main limitation of its wider application is ingredient complexity of this formula.Thus,it is critically important to identify the major active ingredients of GQD and to illustrate mechanisms underlying its action.Here,we compared the effects of GQD and berberine,a hypothetical key active pharmaceutical ingredient of GQD,on a diabetic rat model by comprehensive analyses of gut microbiota,short-chain fatty acids,proinflammatory cytokines,and ileum transcriptomics.Our results show that berberine and GQD had similar effects on lowering blood glucose levels,modulating gut microbiota,inducing ileal gene expression,as well as relieving systemic and local inflammation.As expected,both berberine and GQD treatment significantly altered the overall gut microbiota structure and enriched many butyrate-producing bacteria,including Faecalibacterium and Roseburia,thereby attenuating intestinal inflammation and lowering glucose.Levels of short-chain fatty acids in rat feces were also significantly elevated after treatment with berberine or GQD.Moreover,concentration of serum proinflammatory cytokines and expression of immune-related genes,including Nfkb1,Stat1,and Ifnrg1,in pancreatic islets were significantly reduced after treatment.Our study demonstrates that the main effects of GQD can be attributed to berberine via modulating gut microbiota.The strategy employed would facilitate further standardization and widespread application of TCM in many diseases. | Xizhan Xu Zezheng Gao Fuquan Yang Yingying Yang Liang Chen Lin Han Na Zhao Jiayue Xu Xinmiao Wang Yue Ma Lian Shu Xiaoxi Hu Na Lyu Yuanlong Pan Baoli Zhu Linhua Zhao Xiaolin Tong Jun Wang | 2020 | Genomics, Proteomics & Bioinformatics2020,18,6: | 29 |

| 3 | Computed tomography-based radiomics for prediction of neoadjuvant chemotherapy outcomes in locally advanced gastric cancer: A pilot study显示文摘Objective: The standard treatment for patients with locally advanced gastric cancer has relied on perioperative radio-chemotherapy or chemotherapy and surgery. The aim of this study was to investigate the wealth of radiomics for pre-treatment computed tomography(CT) in the prediction of the pathological response of locally advanced gastric cancer with preoperative chemotherapy.Methods: Thirty consecutive patients with CT-staged II/III gastric cancer receiving neoadjuvant chemotherapy were enrolled in this study between December 2014 and March 2017. All patients underwent upper abdominal CT during the unenhanced, late arterial phase(AP) and portal venous phase(PP) before the administration of neoadjuvant chemotherapy. In total, 19,985 radiomics features were extracted in the AP and PP for each patient.Four methods were adopted during feature selection and eight methods were used in the process of building the classifier model. Thirty-two combinations of feature selection and classification methods were examined. Receiver operating characteristic(ROC) curves were used to evaluate the capability of each combination of feature selection and classification method to predict a non-good response(non-GR) based on tumor regression grade(TRG).Results: The mean area under the curve(AUC) ranged from 0.194 to 0.621 in the AP, and from 0.455 to 0.722 in the PP, according to different combinations of feature selection and the classification methods. There was only one cross-combination machine-learning method indicating a relatively higher AUC(>0.600) in the AP, while 12 cross-combination machine-learning methods presented relatively higher AUCs(all >0.600) in the PP. This kind of deposits have simple mineralogy dominated by galena and sphalerite commonly associated with calcite and other gangue minerals. Both galena and sphalerite have low and high variable Re concentrations and thus Re-Os dating of these minerals have been less promising. In addition, the recovery of Re is extremely low for galena when conventional method was applied, lending additional difficulty in precisely dating galena. In this study, we investigate the recovery of Re using different media for anion exchange separation and reporte a revised preparation method for Re-Os dating of galena and sphalerite. | 5 | Thermodynamic Analysis of a Mixed Refrigerant Ejector Refrigeration Cycle Operating with Two Vapor-liquid Separators显示文摘A mixed refrigerant ejector refrigeration cycle operating with two-stage vapor-liquid separators(MRERC2) is proposed to obtain refrigeration temperature at-40℃. The thermodynamic investigations on performance of MRERC2 using zeotropic mixture refrigerant R23/R134 a are performed, and the comparisons of cycle performance between MRERC2 and MRERC1(MRERC with one-stage vapor-liquid separator) are conducted. The results show that MRERC2 can achieve refrigeration temperature varying between-23.9℃ and-42.0℃ when ejector pressure ratio ranges from 1.6 to 2.3 at the generation temperature of 57.3-84.9℃. The parametric analysis indicates that increasing condensing temperature decreases coefficient of performance(COP) of MRERC2, and increasing ejector pressure ratio and mass fraction of the low boiling point component in the mixed refrigerant can improve COP of MRERC2. | 12 | Effects of Traditional Chinese Medicine herbs for tonifying Qi and kidney, and replenishing spleen on intermittent asthma in children aged 2 to 5 years old显示文摘OBJECTIVE: To evaluate the efficacy and safety of Traditional Chinese Medicine(TCM) herbs for tonifying Qi and kidney, and replenishing spleen on intermittent asthma in children aged 2 and 5 years.METHODS: A randomized, single-blind, placebo controlled trial was conducted. Children with intermittent asthma were enrolled and their baseline conditions were measured using a questionnaire. A total of 60 participants, aged 2 to 5, were randomized into either the treatment group(n = 40) or the placebo group(n = 20). The treatment group was treated with granules of TCM herbs for tonifying Qi and kidney, and replenishing spleen, and the placebo group was given placebo granules for 3 months.The number of asthma attacks was counted and TCM syndrome scores were measured at 1, 2, 3, 6, 9,and 12 months. The airway resistance and levels of eosinophil cationic protein for the two groups were observed before and after 3 months. The results were statistically analyzed.RESULTS: Compared with the placebo group, the number of asthma attacks significantly decreased in the treatment group(P < 0.05). For the treatment group, the TCM syndrome scores decreased after 1 and 2 months; there was also a significant difference in scores between the two groups(P < 0.05).The difference remained after the medicine was stopped for 9 months(P < 0.05). After the 3-month treatment, compared with the placebo group, the airway resistance decreased in the treatment group(P < 0.05). Why and how to develop cloud manufacturing platform(CMP), however, remains the key concern of both platform operators and users. A microscopic model is proposed to investigate advantages and diffusion forces of CMP through exploration of its diffusion process and mechanism. Specifically, a three-stage basic evolution process of CMP is innovatively proposed. Then, based on this basic process, a more complex CMP evolution model has been established in virtue of complex network theory, with five diffusion forces identified. Thereafter, simulations on CMP diffusion have been conducted. The results indicate that, CMP possesses better resource utilization,user satisfaction, and enterprise utility. Results of simulation on impacts of different diffusion forces show that both the time required for CMP to reach an equilibrium state and the final network size are affected simultaneously by the five diffusion forces. | 17 | Efficacy of Lidan Tang on high-fat-diet induced gallstone in mice and possible mechanism显示文摘OBJECTIVE:To investigate efficacy of Lidan Tang(LDT)on gallstone induced by high fat diet in mice,and to study its underlying mechanism.METHODS:Mice were fed with high fat diet every day and treated with LDT(9.01 times of human clinic dosage).Mice were randomly divided into 6 groups as control group,gallstone model group(high-fat diet),positive control ursodeoxycholic acid(UDCA)group(80 mg·kg^-1·d^-1,i.g.),LDT low dose group(6 kg/d,i.g.),LDT middle dose group(12 kg/d,i.g.),and LDT high dose group(24 kg/d,i.g.).The whole experiment was lasted for 4 weeks.The levels of ALT,AST,LDH,CHO,HDL-C and LDL-C in serum were measured,the pathological sections were observed by hematoxylin-eosin staining,the activities of antioxidant enzymes were measured by kits,and the proteins related to oxidative stress and lipid transport were detected by Western blot analysis.RESULTS:LDT could significantly reduce the contents of ALT and AST in serum and improve the pathological tissue of liver.LDT could significantly reduce the content of MDA and LPO,and increase the level of GSH and GSH-PX in liver tissue.The data of Western blot showed that LDT had antioxidant effect promoting Keap1/Nrf2 pathway and regulated the process of lipid transport,which was statistically significant.In addition,LDT treatment inhibited the expression of ATP-binding cassette transports ABCG5/8 in liver,and reduced cholesterol transport from the hepatocytes to the gallbladder.CONCLUSION:LDT has protective effect on gallstones induced by high fat diet in mice,which might be based on the protective effect on liver,including enhancing the antioxidant capacity of liver and reducing the production of lipid peroxides. | Gao Yan Liu Caipin Li Juntong Zhai Yingying Lin Meiyu Wu Qinglin Chu Shifeng Zhang Zhao Li Jianping Zhou Xin Li Yueting Chen Naihong | 2020 | Journal of Traditional Chinese Medicine2020,40,4: | 4 |

| 20 | Targeting slug-mediated non-canonical activation of c-Met to overcome chemo-resistance in metastatic ovarian cancer cells显示文摘Metastasis-associated drug resistance accounts for high mortality in ovarian cancer and remains to be a major barrier for effective treatment. In this study, SKOV3/T4, a metastatic subpopulation of ovarian cancer SKOV3 cells, was enriched to explore potential interventions against metastaticassociated drug resistance. Quantitative genomic and functional analyses were performed and found that slug was significantly increased in the SKOV3/T4 subpopulation and contributed to the high resistance of SKOV3/T4. Further studies showed that slug activated c-Met in a ligand-independent manner due to elevated levels of fibronectin and provoked integrin α V function, which was confirmed by the significant correlation of slug and p-Met levels in 121 ovarian cancer patient samples. Intriguingly,c-Met inhibitor(s) exhibited greatly enhanced anti-cancer effects in slug-positive ovarian cancer models both in vitro and in vivo. Additionally, IHC analyses revealed that slug levels were highly correlated with reduced survival of ovarian cancer patients.
