| 1 | Increased globulin and its association with hemorrhagic transformation in patients receiving intra-arterial thrombolysis therapy显示文摘Previous studies have identified a diverse set of predisposing factors for the occurrence of hemorrhagic transformation(HT), but the independent clinical predictors of HT after intraarterial thrombolysis have not been determined.In this retrospective study, we investigated the characteristics of patients with or without HT who had received intra-arterial thrombolysis therapy, using biochemical analysis, renal function test, routine blood test, blood lipid test, coagulation blood test,liver function test, random blood glucose test, timewindow for intra-arterial thrombolysis, recanalization,National Institutes of Health Stroke Scale(NIHSS)score and systolic blood pressure before intra-arteria thrombolysis. The mortality rates were similar in the HT and non-HT groups(P = 0.944). In the singlefactor analysis, patients with a higher globulin leve(P <0.002), prothrombin time activity percentage(PTA; P = 0.026), and NIHSS score(P = 0.002),had a significantly increased risk of developing HT.In the multifactor logistic regression model involving globulin level, PTA, white blood cell count, and NIHSS score, the globulin level(P <0.001; OR,1.185; 95% confidence interval [CI], 1.090–1.288),PTA(P = 0.018; OR, 1.016; 95% CI, 1.003–1.029),white blood cell count(P = 0.025; OR, 1.097; 95%CI, 1.012–1.190) and NIHSS score(P = 0.003; OR,1.097; 95% CI, 1.031–1.166) were significantlyincreased in the HT group. The increase in globulin level is an independent risk factor for HT in patients receiving intra-arterial thrombolysis. | 2 | A newly discovered Gigantopithecus fauna from Sanhe Cave, Chongzuo, Guangxi, South China显示文摘Among the most important faunas in the Late Cenozoic, the Gigantopithecus faunas have received a good deal of attention. The Gigantopithecus fauna recently discovered in Sanhe Cave consists of more than 80 mammal species, including cf. Hominidae, Pongo sp., Hylobates sp., Sinomastodon yangzien-sis, Stegodon preorientalis, Cervavitus fenqii, Dicoryphochoerus ultimus and Sus xiaozhu. It is the southernmost Gigantopithecus fauna found so far in China. Its geological age is estimated to be Early Pleistocene based on the fauna and stratigraphic correlation. The significant increase in the estimated body sizes of Ailuropoda, Gigantopithecus and Tapirus shows that the Sanhe fauna is middle Early Pleistocene, later than those from Wushan and Liucheng (early Early Pleistocene). Paleomagnetic dating of the fossil-bearing strata in Sanhe Cave gives an age of approximately 1.2 Ma. The fauna is characterized by tropical-subtropical forest types, including Pongo sp., Tupaia sp., Ia sp., Typhlomys intermedius, etc., and it lacks Palaearctic types. It is a typical tropical forest fauna, suggesting an en-vironment with a lush forest and a warm and humid climate. | 3 | Underwater explosion in centrifuge partⅠ:Validation and calibration of scaling laws显示文摘Centrifuge is a promising tool for underwater explosion(UNDEX) research as both Mach and Froude similitudes could be satisfied with hyper-gravity in models, which would result in similarities in both shock wave and bubble oscillation. Scaling laws for UNDEX in centrifuge have been proposed based on dimensional analysis. Two dimensionless numbers, i.e.,π_3 and π_4, are used to characterize shock wave and bubble oscillation, respectively. To validate scaling laws, 17 UNDEX tests are designed by varying accelerations or explosive weights and positions in centrifuge. The tests are classified into different groups to validate scaling laws as well as calibrate coefficients in empirical formulae for both shock wave and bubble oscillation. The results show that changes of gravity acceleration or hydrodynamic pressure almost has no influence on shock wave peak pressure and time constant as long as π3 is constant. The dimensionless bubble period and maximum radius agreed with each other when π4 is constant. | 4 | The Homo sapiens Cave hominin site of Mulan Mountain,Jiangzhou District,Chongzuo,Guangxi with emphasis on its age显示文摘One of the most hotly debated and frontal issues in paleoanthropology focuses on the origins of modern humans. Recently, an incomplete hominin mandible with a distinctly weaker mental protuberance than modern human and a great variety of coexisting fossil mammals were unearthed from the Homo sapiens Cave of Mulan Mountain, Chongzuo, Guangxi. The mammalian fauna from the Homo sapiens Cave characterized by the combination of Elephas kiangnanensis, first occurring Elephas maixmus, and Megatapirus augustus, and strikingly different from the Early Pleistocene Gigantopithecus fauna and the Middle Pleistocene Ailuropoda-Stogodon fauna of South China could be regarded as an early representive of the typical Asian elephant fauna. Faunal analysis, biostratigraphic correlation, and, most importantly, U-series dating all consistently support an estimate of ca. 110 ka for the age of the fossil Homo sapiens and coexisting mammalian fauna, that is, the early Late Pleistocene. The fauna is mainly made up of tropical-subtropical elements, but grassland elements have a much greater variety than forest elements, which probably indicates a drier climate at that time. | 5 | A four-compartment model for the COVID-19 infection—implications on infection kinetics,control measures,and lockdown exit strategies显示文摘Objective:To analyse the impact and repercussions of the surge in healthcare demand in response to the COVID-19 pandemic,assess the potential effectiveness of various infection/disease control measures,and make projections on the best approach to exit from the current lockdown.Design:A four-compartment model was constructed for SARS-CoV-2 infection based on the Wuhan data and validated with data collected in Italy,the UK,and the US.The model captures the effectiveness of various disease suppression measures in three modifiable factors:(a)the per capita contact rate(β)that can be lowered by means of social distancing,(b)infection probability upon contacting infectious individuals that can be lowered by wearing facemasks,personal hygiene,etc.,and(c)the population of infectious individuals in contact with the susceptible population,which can be lowered by quarantine.The model was used to make projections on the best approach to exit from the current lockdown.Results:The model was applied to evaluate the epidemiological data and hospital burden in Italy,the UK,and the US.The control measures were identified as the key drivers for the observed epidemiological data through sensitivity analyses.Analysing the different lockdown exit strategies showed that a lockdown exit strategy with a combination of social separation/general facemask use may work,but this needs to be supported by intense monitoring whichwould allowre-introduction/tightening of the controlmeasures if the number of newinfected subjects increases again.Conclusions and relevance:Governments should act early in a swift and decisive manner for containment policies.Any lockdown exit will need to be monitored closely,with regards to the potential of lockdown reimplementation.This mathematical model provides a framework for major pandemics in the future. | Tianbing Wang Yanqiu Wu Johnson Yiu-Nam Lau Yingqi Yu Liyu Liu Jing Li Kang Zhang Weiwei Tong Baoguo Jiang | 2020 | Precision Clinical Medicine2020,3,2: | 5 |
| 6 | Monascus vinegar alleviates high-fat-diet-induced inflammation in rats by regulating the NF-κB and PI3K/AKT/mTOR pathways显示文摘Monascus vinegar(MV) is a typical fermented food with various health-promoting effects. This study aimed to evaluate the role of MV in alleviating high-fat-diet-induced inflammation in rats with hyperlipidemia and elucidate the possible regulatory mechanisms. In the study, serum lipid profiles, liver pathology and liver inflammatory cytokines were analyzed in hyperlipidemia rats with MV(0.5 mL/kg mb, 2 mL/kg mb). Results showed that the administration of MV alleviated dyslipidemia by decreasing the serum and liver levels of triglyceride and total cholesterol. Increase in hepatic lipase and carnitine palmitoyl transferase 1(CPT-1)levels and decrease in hepatocyte steatosis, nephritis, and intestinal tissue injury in the HD group showed that high-dose MV can significantly suppress hepatic lipid accumulation and steatosis. In addition, compared with the model(MOD) group, the HD group showed significantly down-regulated the level of serum or hepatic alanine aminotransferase(ALT), aspartate aminotransferase(AST), CPT-1, interleukin(IL)-2, IL-6, IL-12,and tumor necrosis factor α(TNF-α). Moreover, the HD group showed repressed hepatic nuclear factor κB(NF-κB) pathway and inactivated phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR) pathway mitigated liver inflammation. Similar results were obtained from cell experiments. | 7 | Exosomes secreted from cardiomyocytes suppress the sensitivity of tumor ferroptosis in ischemic heart failure显示文摘Heart failure(HF)patients in general have a higher risk of developing cancer.Several animal studies have indicated that cardiac remodeling and HF remarkably accelerate tumor progression,highlighting a cause-and-effect relationship between these two disease entities.Targeting ferroptosis,a prevailing form of non-apoptotic cell death,has been considered a promising therapeutic strategy for human cancers.Exosomes critically contribute to proximal and distant organ-organ communications and play crucial roles in regulating diseases in a paracrine manner.However,whether exosomes control the sensitivity of cancer to ferroptosis via regulating the cardiomyocyte-tumor cell crosstalk in ischemic HF has not yet been explored.Here,we demonstrate that myocardial infarction(MI)decreased the sensitivity of cancer cells to the canonical ferroptosis activator erastin or imidazole ketone erastin in a mouse model of xenograft tumor.Post-MI plasma exosomes potently blunted the sensitivity of tumor cells to ferroptosis inducers both in vitro in mouse Lewis lung carcinoma cell line LLC and osteosarcoma cell line K7M2 and in vivo with xenograft tumorigenesis model.The expression of miR-22-3p in cardiomyocytes and plasma-exosomes was significantly upregulated in the failing hearts of mice with chronic MI and of HF patients as well.Incubation of tumor cells with the exosomes isolated from post-MI mouse plasma or overexpression of miR-22-3p alone abrogated erastin-induced ferroptotic cell death in vitro.Cardiomyocyte-enriched miR-22-3p was packaged in exosomes and transferred into tumor cells.Inhibition of cardiomyocyte-specific miR-22-3p by AAV9 sponge increased the sensitivity of cancer cells to ferroptosis.ACSL4,a pro-ferroptotic gene,was experimentally established as a target of miR-22-3p in tumor cells.Taken together,our findings uncovered for the first time that MI suppresses erastin-induced ferroptosis through releasing miR-22-3p-enriched exosomes derived from cardiomyocytes.Therefore,targeting exosome-mediated cardiomyocyte/tumor pathological communication may offer a novel approach for the ferroptosis-based antitumor therapy. | Ye Yuan Zhongting Mei Zhezhe Qu Guanghui Li Shuting Yu Yingqi Liu Kuiwu Liu Zhihua Shen Jiaying Pu Yanquan Wang Changhao Wang Zhiyong Sun Qian Liu Xiaochen Pang Ao Wang Zijing Ren Tong Wang Ying Liu Jinhuan Hong Jiajie Xie Xin Li Zhonghua Wang Weijie Du Baofeng Yang | 2023 | Signal Transduction and Targeted Therapy2023,8,4: | 2 |
