| 1 | The HuangZaoSi Maize Genome Provides Insights into Genomic Variation and Improvement History of Maize显示文摘Maize is a globally important crop that was a classic model plant for genetic studies. Here, we report a 2.2 Gb draft genome sequence of an elite maize line, HuangZaoSi (HZS). Hybrids bred from HZS-improved lines (HILs) are planted in more than 60% of maize fields in China. Proteome clustering of six completed sequeneed maize genomes show that 638 proteins fall into 264 HZS-specific gene families with the majority of contributions from tandem duplication events. Resequencing and comparative analysis of 40 HZSrelated lines reveals the breeding history of HILs. More than 60% of identified selective sweeps were clustered in identity.by.descent conserved regions, and yield-related genes/QTLs were enriched in HZS characteristic selected regions. Furthermore, we dem on strated that HZS-specific family genes were not uniformly distributed in the genome but enriched in improvement/function.related genomic regions. | 4 | Multi-Branch Cable Harness Layout Design Based on Genetic Algorithm with Probabilistic Roadmap Method显示文摘Current studies on cable harness layouts have mainly focused on cable harness route planning.However,the topological structure of a cable harness is also extremely complex,and the branch structure of the cable harness can affect the route of the cable harness layout.The topological structure design of the cable harness is a key to such a layout.In this paper,a novel multi-branch cable harness layout design method is presented,which unites the probabilistic roadmap method(PRM)and the genetic algorithm.First,the engineering constraints of the cable harness layout are presented.An obstacle-based PRM used to construct non-interference and near to the surface roadmap is then described.In addition,a new genetic algorithm is proposed,and the algorithm structure of which is redesigned.In addition,the operation probability formula related to fitness is proposed to promote the efficiency of the branch structure design of the cable harness.A prototype system of a cable harness layout design was developed based on the method described in this study,and the method is applied to two scenarios to verify that a quality cable harness layout can be efficiently obtained using the proposed method.In summary,the cable harness layout design method described in this study can be used to quickly design a reasonable topological structure of a cable harness and to search for the corresponding routes of such a harness. | Yingfeng Zhao Jianhua Liu Jiangtao Ma Linlin Wu | 2021 | Chinese Journal of Mechanical Engineering2021,34,2: | 2 |

| 9 | RIPK3 promotes hantaviral replication by restricting JAK-STAT signaling without triggering necroptosis显示文摘Hantaan virus(HTNV)is a rodent-borne virus that causes hemorrhagic fever with renal syndrome(HFRS),resulting in a high mortality rate of 15%.Interferons(IFNs)play a critical role in the anti-hantaviral immune response,and IFN pretreatment efficiently restricts HTNV infection by triggering the expression of a series of IFNstimulated genes(ISGs)through the Janus kinase-signal transducer and activator of transcription 1(JAK-STAT)pathway.However,the tremendous amount of IFNs produced during late infection could not restrain HTNV replication,and the mechanism remains unclear.Here,we demonstrated that receptor-interacting protein kinase 3(RIPK3),a crucial molecule that mediates necroptosis,was activated by HTNV and contributed to hantavirus evasion of IFN responses by inhibiting STAT1 phosphorylation.RNA-seq analysis revealed the upregulation of multiple cell death-related genes after HTNV infection,with RIPK3 identified as a key modulator of viral replication.RIPK3 ablation significantly enhanced ISGs expression and restrained HTNV replication,without affecting the expression of pattern recognition receptors(PRRs)or the production of type I IFNs.Conversely,exogenously expressed RIPK3 compromised the host's antiviral response and facilitated HTNV replication.RIPK3^(-/-)mice also maintained a robust ability to clear HTNV with enhanced innate immune responses.Mechanistically,we found that RIPK3 could bind STAT1 and inhibit STAT1 phosphorylation dependent on the protein kinase domain(PKD)of RIPK3 but not its kinase activity.Overall,these observations demonstrated a noncanonical function of RIPK3 during viral infection and have elucidated a novel host innate immunity evasion strategy utilized by HTNV. | Yue Si Haijun Zhang Ziqing Zhou Xudong Zhu Yongheng Yang He Liu Liang Zhang Linfeng Cheng Kerong Wang Wei Ye Xin Lv Xijing Zhang Wugang Hou Gang Zhao Yingfeng Lei Fanglin Zhang Hongwei Ma | 2023 | Virologica Sinica2023,38,5: | 0 |
| 10 | Development of the expressed immunoglobulinμchain repertoire during maturation of mice B cells显示文摘In the bone marrow and spleen,the developing B cell populations undergo both negative and positive selections to shape their B cell receptor repertoire.To gain insight into the shift of the immunoglobulin heavy(IgH)chain repertoire during B cell development,we undertook large scale Igμchain repertoire analysis of pre-B,immature B and spleen B cell populations.We found that the majority of VH gene segments,VH families,JH and D gene segments,were observed to have significantly different usage frequencies when three B cell populations were compared,but the usage profile of the VH,D,and JH genes between different B cell populations showed high correlations.In both productive and nonproductive rearrangements,the length of CDRH3 shortened significantly on average when B cells entered the periphery.However,the CDRH3 length distribution of nonproductive rearrangements did not follow a Gaussian distribution,but decreased successively in the order 3n–2,3n–1 and 3n,suggesting a direct correlation between mRNA stability and CDRH3 length patterns of nonproductive rearrangements.Further analysis of the individual components comprising CDRH3 of productive rearrangements indicated that the decrease in CDRH3 length was largely due to the reduction of N addition at the 5′and 3′junctions.Moreover,with development,the amino acid content of CDRH3 progressed toward fewer positively charged and nonpolar residues but more polar residues.All these data indicated that the expressed Igμchain repertoire,especially the repertoire of CDRH3,was fine-tuned when B cells passed through several checkpoints of selection during the process of maturation. | Jingwen LIANG Yingfeng LUO Yi SUN Meng LEI Bing ZHANG Songnian HU Yaofeng ZHAO | 2014 | Frontiers of Agricultural Science and Engineering2014,1,3: | 0 |
