2Rapid, automated, and reliable antimicrobial susceptibility test from positive blood culture by CAST‐R显示文摘Antimicrobial susceptibility tests(ASTs)are pivotal in combating multidrug resistant pathogens,yet they can be time‐consuming,labor‐intensive,and unstable.Using the AST of tigecycline for sepsis as the main model,here we establish an automated system of Clinical Antimicrobials Susceptibility Test Ramanometry(CAST‐R),based on D2O‐probed Raman microspectroscopy.Featuring a liquid robot for sample pretreatment and a machine learning‐based control scheme for data acquisition and quality control,the 3‐h,automated CAST‐R process accelerates AST by>10‐fold,processes 96 paralleled antibiotic‐exposure reactions,and produces high‐quality Raman spectra.The Expedited Minimal Inhibitory Concentration via Metabolic Activity is proposed as a quantitative and broadly applicable parameter for metabolism‐based AST,which shows 99%essential agreement and 93%categorical agreement with the broth microdilution method(BMD)when tested on 100 Acinetobacter baumannii isolates.Further tests on 26 clinically positive blood samples for eight antimicrobials,including tigecycline,meropenem,ceftazidime,ampicillin/sulbactam,oxacillin,clindamycin,vancomycin,and levofloxacin reveal 93%categorical agreement with BMD‐based results.The automation,speed,reliability,and general applicability of CAST‐R suggest its potential utility for guiding the clinical administration of antimicrobials.Pengfei Zhu Lihui Ren Ying Zhu Jing Dai Huijie Liu Yuli Mao Yuandong Li Yuehui He Xiaoshan Zheng Rongze Chen Xiaoting Fu Lili Zhang Lijun Sun Yuanqi Zhu Yuetong Ji Bo Ma Yingchun Xu Jian Xu Qiwen Yang 2022mLife2022,1,3:3

6Differential gene expression profile of Buyanghuanwu decoction in rats with ventricular remodeling post-myocardial infarction显示文摘OBJECTIVE: To investigate the effect of Buyanghuanwu decoction(BYHWD) on gene expression in ventricular remodeling post-myocardial infarction in rats.METHODS: Animal models of myocardial infarction were established by permanent ligation of the left anterior descending coronary artery. Echocardiography measurements were performed after the treatment of BYHWD(18 g·kg-1 collagen was observ·d-1) for 90 days.Myocardialed by mallory trichrome staining. Capillary density was quantified by using Factor rentially expⅧre immunohistochemical staining.Diffessed genes were explored by a short-read sequencing technology combined with a tag-based digital gene expression profiling(DGE)system. Real-time quantitative polymerase chain reaction detecting system(q PCR) was used to validate the sequencing results. After assembling the gene information from Sham, model and BYHWD groups, we constructed three DGE libraries based on each group. The sequencing of three libraries generated 66 000-73 000 unique tags, which were mapped to reference sequences for annotation of expressed genes.RESULTS: Among them, 511 and 352 differentially expressed genes were found in comparison with sham/model and model/BYHWD, respectively. Fifty-five genes exhibited reversed direction of gene expression differences between Sham/Model and Model/BYHWD groups. We found that transforming growth factor beta receptor-1, junctophilin-2,monocyte chemotactic protein 1, neuropeptide Y,arachidonate 5-Lipoxygenase, arachidonate 15-Lipoxygenase were significantly modulated, which suggested the involvement of these genes in BYHWD treatment.CONCLUSION: The DGE profiling data provide comprehensive gene expression information at the transcriptional level that could facilitate our understanding of the pharmacological mechanisms of BYHWD in ventricular remodeling post-myocardial infarction.Zhang Tao Hua Yue Luo Hao Chen Hongmei Shao Meng Fu Xiuqiong John Man-Tak Chu Huang Guiqiong Liu Bin Zhou Yingchun 2017Journal of Traditional Chinese Medicine2017,37,3:1

10A new DRIE cut-off material in SOG MEMS process显示文摘The silicon on glasses process is a common preparation method of micro-electro-mechanical system inertial devices,which can realize the processing of thick silicon structures.This paper proposes that indium tin oxides(ITO)film can serve as a deep silicon etching cut-off layer because ITO is less damaged under the attack of fluoride ions.ITO has good electrical conductivity and can absorb fluoride ions for silicon etching and reduce the reflection of fluoride ions,thus reducing the foot effect.The removal and release of ITO use an acidic solution,which does not damage the silicon structure.Therefore,the selection of the sacrificial layer has an excellent effect in maintaining the shape of the MEMS structure.This method is used in the preparation of MEMS accelerometers with a structure thickness of 100μm and a feature size of 4μm.The over-etching of the bottom of the silicon structure caused by the foot effect is negligible.The difference between the simulated value and the designed value of the device characteristic frequency is less than 5%.This indicates that ITO is an excellent deep silicon etch stopper material.Chaowei Si Yingchun Fu Guowei Han Yongmei Zhao Jin Ning Zhenyu Wei Fuhua Yang 2023Journal of Semiconductors2023,44,4:0
11Double heterozygous pathogenic mutations in KIF3C and ZNF513 cause hereditary gingival fibromatosis显示文摘Hereditary gingival fibromatosis(HGF)is a rare inherited condition with fibromatoid hyperplasia of the gingival tissue that exhibits great genetic heterogeneity.Five distinct loci related to non-syndromic HGF have been identified;however,only two diseasecausing genes,SOS1 and REST,inducing HGF have been identified at two loci,GINGF1 and GINGF5,respectively.Here,based on a family pedigree with 26 members,including nine patients with HGF,we identified double heterozygous pathogenic mutations in the ZNF513(c.C748T,p.R250W)and KIF3C(c.G1229A,p.R410H)genes within the GINGF3 locus related to HGF.Functional studies demonstrated that the ZNF513 p.R250W and KIF3C p.R410H variants significantly increased the expression of ZNF513 and KIF3C in vitro and in vivo.ZNF513,a transcription factor,binds to KIF3C exon 1 and participates in the positive regulation of KIF3C expression in gingival fibroblasts.Furthermore,a knock-in mouse model confirmed that heterozygous or homozygous mutations within Zfp513(p.R250W)or Kif3c(p.R412H)alone do not led to clear phenotypes with gingival fibromatosis,whereas the double mutations led to gingival hyperplasia phenotypes.In addition,we found that ZNF513 binds to the SOS1 promoter and plays an important positive role in regulating the expression of SOS1.Moreover,the KIF3C p.R410H mutation could activate the PI3K and KCNQ1 potassium channels.ZNF513 combined with KIF3C regulates gingival fibroblast proliferation,migration,and fibrosis response via the PI3K/AKT/mTOR and Ras/Raf/MEK/ERK pathways.In summary,these results demonstrate ZNF513+KIF3C as an important genetic combination in HGF manifestation and suggest that ZNF513 mutation may be a major risk factor for HGF.Jianfan Chen Xueqing Xu Song Chen Ting Lu Yingchun Zheng Zhongzhi Gan Zongrui Shen Shunfei Ma Duocai Wang Leyi Su Fei He Xuan Shang Huiyong Xu Dong Chen Leitao Zhang Fu Xiong 2023International Journal of Oral Science2023,15,3:0
12Diverse O-methyltransferases catalyze the biosynthesis of floral benzenoids that repel aphids from the flowers of waterlily Nymphaea prolifera显示文摘Nymphaea is a key genus of the ANA grade(Amborellales,Nymphaeales,and Austrobaileyales)of basal flowering plants,which serve as a key model to study the early evolution of floral traits.In this study,we comprehensively investigated the emission,biosynthesis,and biological function of the floral scent in a night-blossoming waterlily Nymphaea prolifera.The headspace volatile collection combined with GC-MS analysis showed that the floral scent of N.prolifera is predominately comprised by methylated benzenoids including anisole,veratrole,guaiacol,and methoxyanisole.Moreover,the emission of these floral benzenoids in N.prolifera exhibited temporal and spatial pattern with circadian rhythm and tissue specificity.By creating and mining transcriptomes of N.prolifera flowers,12 oxygen methyltransferases(NpOMTs)were functionally identified.By in vitro enzymatic assay,NpOMT3,6,and 7 could produce anisole and NpOMT5,7,9,produce guaiacol,whereas NpOMT3,6,9,11 catalyzed the formation of veratrole.Methoxyanisole was identified as the universal product of all NpOMTs.Expression patterns of NpOMTs provided implication for their roles in the production of the respective benzenoids.Phylogenetic analysis of OMTs suggested a Nymphaea-specific expansion of the OMT family,indicating the evolution of lineage-specific functions.In bioassays,anisole,veratrole,and guaiacol in the floral benzenoids were revealed to play the critical role in repelling waterlily aphids.Overall,this study indicates that the basal flowering plant N.prolifera has evolved a diversity and complexity of OMT genes for the biosynthesis of methylated benzenoids that can repel insects from feeding the flowers.These findings provide new insights into the evolutional mechanism and ecological significance of the floral scent from early-diverged flowering plants.Guanhua Liu Jianyu Fu Lingyun Wang Mingya Fang Wanbo Zhang Mei Yang Xuemin Yang Yingchun Xu Lin Shi Xiaoying Ma Qian Wang Hui Chen Cuiwei Yu Dongbei Yu Feng Chen Yifan Jiang 2023Horticulture Research2023,10,12:0
13Neurofilament 200 expression in a rat model of complete spinal cord injury following growth-associated protein-43 treatment显示文摘BACKGROUND:Growth-associated protein-43(GAP-43) expression in the nervous system has been demonstrated to promote neural regeneration,neuronal growth and development,as well as synaptic reconstruction.Neurofilament 200(NF200) expression could reflect degree of injury and repair in injured spinal axons. OBJECTIVE:To observe NF200 expression changes in a rat model of complete spinal cord injury following GAP-43 treatment and to explore the effects of GAP-43 following spinal cord injury. DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed at the Laboratory of Histology and Embryology of Kunming Medical University between March 2007 and October 2008. MATERIALS:GAP-43 and GAP-43 antibody were provided by Beijing Boao Biology,China;mouse anti-rat NF200 antibody was purchased from Chemicon,USA. METHODS:Female,8-week-old,Sprague Dawley rats were randomly assigned into three groups following complete spinal cord injury,with 20 animals in each group:GAP-43 antibody,GAP-43,and model groups.In addition,each group was subdivided into four subgroups according to sampling time after modeling,i.e.,3-,5-,9-,and 15-day groups,with 5 rats in each group.GAP-43 antibody or GAP-43 was injected into injury sites of the spinal cord,5μg/0.2 mL,respectively,twice daily for three consecutive days,followed by three additional days of injection,once daily.The model group did not receive any treatment following injury. MAIN OUTCOME MEASURES:NF200 expression in the damaged spinal area at different stages was detected by immunohistochemistry;lower limb motion function following injury was evaluated using the Basso,Beattie and Bresnahan(BBB) locomotor rating scale. RESULTS:NF200 expression was significantly reduced in the GAP-43 antibody group,compared with GAP-43 and model groups,at 3 and 5 days after spinal cord injury(P<0.05).In addition,the model group expressed significantly less NF200 than the GAP-43 group(P<0.05).BBB scores from the GAP-43 antibody and model groups were remarkably less than the GAP-43 group(P<0.05).At 9 and 15 days of injury after drug withdrawal,NF200 expression was increased in the GAP-43 antibody group,and NF200 expression and BBB scores in the GAP-43 antibody and GAP-43 groups were significantly greater than in the model group(P<0.05).In particular,the GAP-43 group exhibited greater BBB scores than the GAP-43 antibody group at day 9(P<0.05). 15Lightweight design of an electric bus body structure with analytical target cascading显示文摘Lightweight designs of new-energy vehicles can reduce energy consumption,thereby improving driving mileage.In this study,a lightweight design of a newly developed multi-material electric bus body structure is examined in combination with analytical target cascading(ATC).By proposing an ATC-based two-level optimization strategy,the original lightweight design problem is decomposed into the system level and three subsystem levels.The system-level optimization model is related to mass minimization with all the structural modal frequency constraints,while each subsystem-level optimization model is related to the sub-structural performance objective with sub-structure mass constraints.To enhance the interaction between two-level systems,each subsystem-level objective is reformulated as a penalty-based function coordinated with the system-level objective.To guarantee the accuracy of the model-based analysis,a finite element model is validated through experimental modal test.A sequential quadratic programming algorithm is used to address the defined optimization problem for effective convergence.Compared with the initial design,the total mass is reduced by 49 kg,and the torsional stiffness is increased by 17.5%.In addition,the obtained design is also validated through strength analysis.Puyi WANG Yingchun BAI Chuanliang FU Cheng LIN 2023Frontiers of Mechanical Engineering2023,18,1:0
