| 1 | Autophagy receptor CCDC50 tunes the STING-mediated interferon response in viral infections and autoimmune diseases显示文摘DNA sensing and timely activation of interferon(IFN)-mediated innate immunity are crucial for the defense against DNA virus infections and the clearance of abnormal cells.However,overactivation of immune responses may lead to tissue damage and autoimmune diseases;therefore,these processes must be intricately regulated.STING is the key adaptor protein,which is activated by cyclic GMP-AMP,the second messenger derived from cGAS-mediated DNA sensing.Here,we report that CCDC50,a newly identified autophagy receptor,tunes STING-directed type I IFN signaling activity by delivering K63-polyubiquitinated STING to autolysosomes for degradation.Knockout of CCDC50 significantly increases herpes simplex virus 1(HSV-1)-or DNA ligand-induced production of type I IFN and proinflammatory cytokines.Ccdc50-deficient mice show increased production of IFN,decreased viral replication,reduced cell infiltration,and improved survival rates compared with their wild-type littermates when challenged with HSV-1.Remarkably,the expression of CCDC50 is downregulated in systemic lupus erythematosus(SLE),a chronic autoimmune disease.CCDC50 levels are negatively correlated with IFN signaling pathway activation and disease severity in human SLE patients.CCDC50 deficiency potentiates the cGAS-STING-mediated immune response triggered by SLE serum.Thus,our findings reveal the critical role of CCDC50 in the immune regulation of viral infections and autoimmune diseases and provide insights into the therapeutic implications of CCDC50 manipulation. | Panpan Hou Yuxin Lin Zibo Li Ruiqing Lu Yicheng Wang Tian Tian Penghui Jia Xi Zhang Liu Cao Zhongwei Zhou Chunmei Li Jieruo Gu Deyin Guo | 2021 | Cellular & Molecular Immunology2021,18,10: | 3 |
| 2 | Impact of EV load uncertainty on optimal planning for electric vehicle charging station显示文摘With the development of electric vehicles(EV), there is a huge demand for electric vehicle charging stations(EVCS). The utilization of renewable energy sources(RES) in EVCS can not only decrease the energy fluctuation by participating in peakload reduction of the grid, but also reduce the pollution to the environment by cutting down the use of fossil fuels. In this paper,the optimal planning for grid-connected EVCS with RES is studied by considering EV load uncertainty. Nine scenarios are set based on a different characteristic of EV load to reveal the impact of EV load on net present cost(NPC) and to express the relationship between the optimal capacity and energy flow. Moreover, since electricity price also plays an important role in EVCS planning, an economic comparison between different cases with different electricity prices for peak-valley-flat period is carried out. | 3 | Imaging algorithm of multi-ship motion target based on compressed sensing显示文摘An imaging algorithm based on compressed sensing(CS) for the multi-ship motion target is presented. In order to reduce the quantity of data transmission in searching the ships on a large sea area, both range and azimuth of the moving ship targets are converted into sparse representation under certain signal basis. The signal reconstruction algorithm based on CS at a distant calculation station, and the Keystone and fractional Fourier transform(FRFT) algorithm are used to compensate range migration and obtain Doppler frequency. When the sea ships satisfy the sparsity, the algorithm can obtain higher resolution in both range and azimuth than the conventional imaging algorithm. | 14 | Evaluation of necrosis avidity of radioiodinated 5-hydroxytryptophan and its potential applications in myocardial infarction imaging显示文摘This study aimed to seek necrosis avid agents with high safety from DNA binding agents. The interaction of 5-hydroxytryptophan(5-HTP) with DNA was investigated by a series of spectroscopic studies. Then, 5-HTP was labeled with iodine-131([^(131)I]5-HTP) and the necrosis avidity of [^(131)I]5-HTP was evaluated by in vitro cell binding assays, in vivo biodistribution experiments and blocking experiment. Finally, the potential of [^(131)I]5-HTP to image necrotic myocardium was explored in rat models with myocardial infarction by SPECT/CT imaging. Results showed that 5-HTP bound to DNA in groove binding mode and the binding constant was 1.69×10~4 L/mol. [~(^(131))I]5-HTP showed specific affinity to necrotic A549 cells,which might be related to cell nucleus. Biodistribution and autoradiography results showed preferential accumulation of [^(131)I]5-HTP in necrotic muscle(necrotic/viable muscle ratio: 6.95 ? 0.68 at 3 h postinjection(p.i.)), which could be blocked by 5-HTP with 66.7% decline, indicating that [^(131)I]5-HTP might share the same necrotic targets with 5-HTP. On SPECT/CT images, a hotspot was clearly observed at 3h p.i.in the necrotic myocardium while not in the control myocardium. In conclusion, [^(131) I]5-HTP is a novel necrosis avid agent and can rapidly visualize necrotic myocardium at 3 h p.i.
