| 1 | Hacking the optical diffraction limit: Review on recent developments of fluorescence nanoscopy显示文摘Subject to the diffraction limit, the resolution of conventional optical microscopy is constrained to about 200 and 500 nm in the lateral and axial planes, respectively. The advantage of optical microscopy in the life sciences over electronic microscopy, especially fluorescence microscopy, drives scientists to develop novel 'hacks' to reach nanoscale resolutions by optical means. In this review, three aspects of the techniques are discussed: (1) lateral super-resolution; (2) axial super-resolution; (3) super-resolution in three dimensions. The principles of how the methods achieve the cross-barrier resolution are discussed, and recent advances in current techniques are described. | 2 | Diurnal and Seasonal Variations of Thermal Stratification and Vertical Mixing in a Shallow Fresh Water Lake显示文摘Among several influential factors, the geographical position and depth of a lake determine its thermal structure. In temperate zones, shallow lakes show significant differences in thermal stratification compared to deep lakes. Here,the variation in thermal stratification in Lake Taihu, a shallow fresh water lake, is studied systematically. Lake Taihu is a warm polymictic lake whose thermal stratification varies in short cycles of one day to a few days. The thermal stratification in Lake Taihu has shallow depths in the upper region and a large amplitude in the temperature gradient,the maximum of which exceeds 5°C m–1. The water temperature in the entire layer changes in a relatively consistent manner. Therefore, compared to a deep lake at similar latitude, the thermal stratification in Lake Taihu exhibits small seasonal differences, but the wide variation in the short term becomes important. Shallow polymictic lakes share the characteristic of diurnal mixing. Prominent differences on the duration and frequency of long-lasting thermal stratification are found in these lakes, which may result from the differences of local climate, lake depth, and fetch. A prominent response of thermal stratification to weather conditions is found, being controlled by the stratifying effect of solar radiation and the mixing effect of wind disturbance. Other than the diurnal stratification and convection, the representative responses of thermal stratification to these two factors with contrary effects are also discussed. When solar radiation increases, stronger wind is required to prevent the lake from becoming stratified. A daily average wind speed greater than 6 m s–1 can maintain the mixed state in Lake Taihu. Moreover, wind-induced convection is detected during thermal stratification. Due to lack of solar radiation, convection occurs more easily in nighttime than in daytime. | 3 | Analysis of oral microorganism diversity in healthy individuals before and after chewing areca nuts using PCR-denatured gradient gel electrophoresis显示文摘To analyze oral microbial diversity in the saliva of 8 healthy individuals before and after chewing areca nuts. Saliva samples were collected before chewing areca nuts, after chewing areca nuts for 5 min and after chewing areca nuts for 30 min. DNA was extracted, and microbial diversity was examined using PCR-denaturing gradient gel electrophoresis(PCR-DGGE). When examining DGGE profiles collectively,the bands associated with Streptococcus and Veillonella were the most intense, making them the most prevalent bacteria. Furthermore, the band intensities did not decrease after chewing areca nuts for 5 or30 min; thus, these bacteria were unaffected. However, when examining some individuals, the band intensities for Streptococcus and Veillonella became more intense after 5 min of chewing and then returned to the pre-chewing level. This difference may be attributed to the mechanical movements of the oral cavity or individual differences. | 5 | Towards win-win: weighted-Voronoi-diagram based channel quantization for security enhancement in downlink cloud-RAN with limited CSI feedback显示文摘Physical layer(PHY) security is recently proved to enable improving the security of wireless communication networks. In downlink frequency division duplex(FDD) cloud radio access network(C-RAN), the performance of PHY security highly relies on the channel state information(CSI) which is usually acquired through the codebook-quantization-based technique at the transceiver. However, the conventional quantization method aggravates the leakage of privacy information in C-RAN under the eavesdropping environment. In this paper, a novel channel quantization method is investigated to improve the secrecy-rate performance of C-RAN by exploiting the high-dimension space geometry. Based on this method, it is proved that when the statistical distribution of the channel matrices of both the legitimate user and the eavesdropper is exploited, a win-win situation can be created where secrecy-rate gains are improved without sacrificing beamforming gains from the point of view of ergodic rate. Particularly, a secrecy-oriented criterion is devised to implement the proposed method for generating codebooks. Then a weighted Voronoi diagram(WVD) is formulated on the complex Grassmann manifold and finally, a vector quantization based algorithm is proposed to build up novel quantization codebooks iteratively. | 7 | Preparation,model construction and efficacy lipid-lowering evaluation of kiwifruit juice fermented by probiotics显示文摘In this study,kiwi juice was fermented with acid-resistant and bile-resistant probiotics to screen out lactic acid bacteria that can lower cholesterol and were suitable for kiwi fermentation.A non-linear fitting equation was used to establish a kinetic model of probiotics fermenting kiwi juice(R^(2)>0.90),and then a fermented juice with special effects of lowering fat was prepared.We have studied the lipid-lowering effect of fermented fruit juice through in vivo validation experiments in mice,and determined the lipid-lowering effect of fermented fruit juice according to the mouse serum indicators and the distribution of intestinal flora.The results suggested that the juice fermented with Bifidobacterium bifidum 6169 and Lactobacillus plantarum 21805 were more effective in cholesterol degradation,and the cholesterol degradation rates were 67.57%and 64.22%,respectively.The number of viable bacteria after fermentation was significantly higher than other groups(p<0.05),and the sensory score was also higher.In addition,the serum indicators were significantly ameliorated and the number of beneficial microorganisms in the intestinal flora increased after the high-fat mice were treated with fermented kiwifruit juice.The research indicates that the fermented products have a regulatory effect on cholesterol levels in mice with hyperlipidemia. | Yu Wang Hongcai Li Yichen Ren Yaqin Wang Ren Yaopeng Wang Xiaowei Yue Tianli Wang Zhouli Gao Zhenpeng | 2022 | Food Bioscience2022,47,3: | 1 |

| 10 | TNFR2 is a potent prognostic biomarker for post-transplant lung metastasis in patients with hepatocellular carcinoma显示文摘Objective:Lung metastasis is a common and fatal complication of liver transplantation for hepatocellular carcinoma(HCC).The precise prediction of post-transplant lung metastasis in the early phase is of great value.Methods:The mRNA profiles of primary and paired lung metastatic lesions were analyzed to determine key signaling pathways.We enrolled 241 HCC patients who underwent liver transplantation from three centers.Tissue microarrays were used to evaluate the prognostic capacity of tumor necrosis factor(TNF),tumor necrosis factor receptor 1(TNFR1),and TNFR2,particularly for post-transplant lung metastasis.Results:Comparison of primary and lung metastatic lesions revealed that the TNF-dependent signaling pathway was related to lung metastasis of HCC.The expression of TNF was degraded in comparison to that in para-tumor tissues(P<0.001).The expression of key receptors in the TNF-dependent signaling pathway,TNFR1 and TNFR2,was higher in HCC tissues than in para-tumor tissues(P<0.001).TNF and TNFR1 showed no relationship with patients’outcomes,whereas elevated TNFR2 in tumor tissue was significantly associated with worse overall survival(OS)and increased recurrence risk(5-year OS rate:31.9%vs.62.5%,P<0.001).Notably,elevated TNFR2 levels were also associated with an increased risk of post-transplant lung metastasis(hazard ratio:1.146;P<0.001).Cox regression analysis revealed that TNFR2,Hangzhou criteria,age,and hepatitis B surface antigen were independent risk factors for post-transplant lung metastasis,and a novel nomogram was established accordingly.The nomogram achieved excellent prognostic efficiency(area under time-dependent receiver operating characteristic=0.755,concordance-index=0.779)and was superior to conventional models,such as the Milan criteria.Conclusions:TNFR2 is a potent prognostic biomarker for predicting post-transplant lung metastasis in patients with HCC.A nomogram incorporating TNFR2 deserves to be a helpful prognostic tool in liver transplantation for HCC. | Huigang Li Zuyuan Lin Jianyong Zhuo Modan Yang Wei Shen Zhihang Hu Yichen Ding Hao Chen Chiyu He Xinyu Yang Siyi Dong Xuyong Wei Beicheng Sun Shusen Zheng Ren Lang Di Lu Xiao Xu | 2023 | Chinese Journal of Cancer Research2023,35,1: | 0 |
