1Modeling study of regional severe hazes over mid-eastern China in January 2013 and its implications on pollution prevention and control显示文摘The Nested Air Quality Prediction Model System(NAQPMS)was used to investigate the temporal and spatial variations of PM2.5over tropospheric central eastern China in January 2013.The impact of regional transport and its implications on pollution prevention and control were also examined.Comparison between simulated and observed PM2.5showed NAQPMS was able to reproduce the evolution of PM2.5during heavy haze episodes.The results indicated that regional transport of PM2.5played an important role in regional haze episodes in the city cluster including Hebei,Beijing and Tianjin(HBT).The cross-city clusters transport outside HBT and transport among cities inside HBT contributed 20%–35%and 26%–35%of PM2.5as compared with local emission,in HBT respectively.To meet the Air Quality Standards for Grade II,90%,90%and65%of emissions would have to be cut down in Hebei,Tianjin and Beijing,if non-control strategy was taken in the surrounding city clusters of HBT.This implicated that control of emissions in one city cluster is not sufficient to reduce regional haze events,and joint efforts among city clusters are essential.Besides regional transports,two-way feedback between boundary-layer evolution and PM2.5also significantly contributed to the formation of heavy hazes,which contributed 30%of monthly average PM2.5concentration in HBT.WANG ZiFa LI Jie WANG Zhe YANG WenYi TANG Xiao GE BaoZhu YAN PinZhong ZHU LiLi CHEN XueShun CHEN HuanSheng WAND Wei LI JianJun LIU Bing WANG XiaoYan WAND Wei ZHAO YiLin LU Ning SU DeBin 2014Science China Earth Sciences2014,57,1:106
2Management of cytokine release syndrome related to CAR-T cell therapy显示文摘Chimeric antigen receptor T (CAR-T) cell therapy is a novel cellular immunotherapy that is widely used to treat hematological malignancies, including acute leukemia, lymphoma, and multiple myeloma. Despite its remarkable clinical effects, this therapy has side effects that cannot be underestimated. Cytokine release syndrome (CRS) is one of the most clinically important and potentially life-threatening toxicities. This syndrome is a systemic immune storm that involves the mass cytokines releasing by activated immune cells. This phenomenon causes multisystem damages and sometimes even death. In this study, we reported the management of a patient with recurrent and refractory multiple myeloma and three patients with acute lymphocytic leukemia who suffered CRS during CAR-T treatment. The early application of tocilizumab, an anti-IL-6 receptor antibody, according to toxicity grading and clinical manifestation is recommended especially for patients who suffer continuous hyperpyrexia, hypotensive shock, acute respiratory failure, and whose CRS toxicities deteriorated rapidly. Moreover, low doses of dexamethasone (5-10 mg/day) were used for refractory CRS not responding to tocilizumab. 4Design of high efficiency achromatic metalens with large operation bandwidth using bilayer architecture显示文摘Achromatic metalens composed of arrays of subwavelength nanostructures with spatially varying geometries is attractivefor a number of optical applications. However, the limited degree of freedom in the single layer achromatic metasurfacedesign makes it difficult to simultaneously guarantee the sufficient phase dispersion and high diffraction efficiency,which restricts the achromatic bandwidth and efficiency of metalens. Here we propose and demonstrate a high efficiencyachromatic metalens with diffraction-limited focusing capability at the wavelength ranging from 1000 nm to 1700 nm. Themetalens comprises two stacked nanopillar metasurfaces, by which the required focusing phase and dispersion compensationcan be controlled independently. As a result, in addition to the large achromatic bandwidth, the averaged focusingefficiency of the bilayer metalens is higher than 64% at the near-infrared region. 5Deep-seated structure and metallogenic dynamics of the Ailaoshan polymetallic mineralization concentration area, Yunnan Province, China显示文摘The Ailaoshan poly-metallic mineralization concentrated area (MCA) consists of the well known Ailaoshan metallogenic belt and adjacent mineral districts and/or deposits. Located in an area of several complex and intersecting tectonic units, the Ailaoshan poly-metallic MCA is controlled by deep crustal and mantle tectonism. Through interpretation of remote sensing images, we identified a large ring structure system that surrounds the MCA. This ring structure encloses regional deep-crustal faults, ductile shear zones, geothermal anomalies, magmatic rocks, and the major mineral deposits, all of which are the reflections of deep tectonic geodynamics that have been long active in this area. Geophysical data indicate that the crust is comprised of relatively stable two or three layers, with some irregular lower-velocity belts. The Moho in the ring sutures occurs as an area of local uplift. There exists an obvious transitional zone between the crust and mantle boundaries. Asthenopheric mantle shows multi-layer upwelling, which indicates multiple events during different geological epochs. It is believed that these mantle events or pulses were responsible for the formation of the regional shear zones, magmatic rocks, and polymetallic orebodies. 6Method for Six-Legged Robot Stepping on Obstacles by Indirect Force Estimation显示文摘Adaptive gaits for legged robots often requires force sensors installed on foot-tips,however impact,temperature or humidity can affect or even damage those sensors.Efforts have been made to realize indirect force estimation on the legged robots using leg structures based on planar mechanisms.Robot Octopus III is a six-legged robot using spatial parallel mechanism(UP-2UPS) legs.This paper proposed a novel method to realize indirect force estimation on walking robot based on a spatial parallel mechanism.The direct kinematics model and the inverse kinematics model are established.The force Jacobian matrix is derived based on the kinematics model.Thus,the indirect force estimation model is established.Then,the relation between the output torques of the three motors installed on one leg to the external force exerted on the foot tip is described.Furthermore,an adaptive tripod static gait is designed.The robot alters its leg trajectory to step on obstacles by using the proposed adaptive gait.Both the indirect force estimation model and the adaptive gait are implemented and optimized in a real time control system.An experiment is carried out to validate the indirect force estimation model.The adaptive gait is tested in another experiment.Experiment results show that the robot can successfully step on a 0.2 m-high obstacle.This paper proposes a novel method to overcome obstacles for the six-legged robot using spatial parallel mechanism legs and to avoid installing the electric force sensors in harsh environment of the robot's foot tips.XU Yilin GAO Feng PAN Yang CHAI Xun 2016Chinese Journal of Mechanical Engineering2016,29,4:16
7Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 8Unusual high-density and saline aqueous inclusions in ultrahigh pressure metamor- phic rocks from Sulu terrane in eastern China显示文摘Primary high-density fluid inclusions were identified in garnet from ultrahigh pressure eclogite in the southern part of the Sulu terrane. They occur isolatedly or in cluster together with relatively low-density two-phase inclu-sions. The eutectic temperature of the inclusions is as low as ≤ -52℃. A bubble was nucleated in a liquid inclusion dur-ing the specific stage of cyclic cooling-heating runs, and the liquid-gas homogenization temperature was measured to be ≤-12.5℃. The composition of the inclusions modeled by the system CaCl2-NaCl-H2O, yields the fluid density of 1.27g/cm3 that corresponds to a pressure of ca. 2.4 GPa at thetemperature of peak eclogite-facies metamorphism, close tothe ultrahigh pressure metamorphic conditions. 9Fluid/melt in continental deep subduction zones: Compositions and related geochemical fractionations显示文摘Plate subduction is the most magnificent process in the Earth. Subduction zones are important sites for proceeding matter- and energy- transports between the Earth's surface and the interior, continental crust growth, and crust-mantle interactions. Besides, a number of geological processes in subduction zones are closely related to human beings' daily life, such as volcanic eruptions and earthquakes, formation of mineral deposits. Subduction process thus has long been the centric topic of Earth sciences. The finding in 1980 s that continental crust could be subducted to mantle depths is a revolutionary progress in plate tectonic theory. Compared to oceanic crust, continental crust is colder, drier, lighter, and much more geochemically/isotopically heterogeneous. Hence, continental subduction process would affect the structure, compositions and evolutions of the overlying mantle wedge even more. During continental subduction and subsequent exhumation, fluids and melts can be generated in the(de)hydration process and partial melting process, respectively. These melts/fluids play important roles in crust-mantle interactions, elemental migrations, isotopic fractionations, and mantle metasomatism. By summarizing recent research works on subduction zones in this paper, we present a review on the types, physicochemical conditions and compositions of fluids/melts, as well as the migration behaviors of fluid-related characteristic elements(Nb-Ta-V) and the fractionation behaviors of non-traditional stable isotopes(Li-Mg) in subduction zones. 11Bright-field holography:cross-modality deep learning enables snapshot 3D imaging with bright-field contrast using a single hologram显示文摘Digital holographic microscopy enables the 3D reconstruction of volumetric samples from a single-snapshot hologram.However,unlike a conventional bright-field microscopy image,the quality of holographic reconstructions is compromised by interference fringes as a result of twin images and out-of-plane objects.Here,we demonstrate that cross-modality deep learning using a generative adversarial network(GAN)can endow holographic images of a sample volume with bright-field microscopy contrast,combining the volumetric imaging capability of holography with the speckle-and artifact-free image contrast of incoherent bright-field microscopy.We illustrate the performance of this“bright-field holography”method through the snapshot imaging of bioaerosols distributed in 3D,matching the artifact-free image contrast and axial sectioning performance of a high-NA bright-field microscope.This data-driven deep-learning-based imaging method bridges the contrast gap between coherent and incoherent imaging,and enables the snapshot 3D imaging of objects with bright-field contrast from a single hologram,benefiting from the wave-propagation framework of holography.Yichen Wu Yilin Luo Gunvant Chaudhari Yair Rivenson Ayfer Calis Kevin de Haan Aydogan Ozcan 2019Light(Science & Applications)2019,8,1:12

13All-in-one cellulose based hybrid tribo/piezoelectric nanogenerator显示文摘Aybrid tribo/piezoelectric nanogenerators (HTPENG) have been proven to be highly efficient and versatile as far as the collection and conversion of ambient energy are concerned, and the introduction of flexible and green materials is a key step for their potential applications. Here, we developed a HTPENG by using nitrocellulose nanofibril paper as the triboelectric layer and BaTiCVMWCNT@ bacterial cellulose paper as the piezoelectric layer. The output of the triboelelctric paper has considerable performance as fluorinated ethylene propylene, and the output of piezoelectric paper is more than ten times higher than the BTO/polydimethylsiloxane structure. The integrated outputs of the sandwich structured HTPENG are 18 V and 1.6 pA·cm^-2, which are capable of lighting up three LED bulbs and charging a 1 pF capacitor to 2.5 V in 80 s. In addition, the voltage signal generated by the HTPENG in contact-separation mode can be used for dynamic pressure detection. The linear range of dynamic pressure is from 0.5 to 3 N·cm^-2 with a high sensitivity of 8.276 V·cm^-2·N^-1 and a detection limit of 0.2 N·cm^-2. 15A telomere-to-telomere gap-free reference genome of watermelon and its mutation library provide important resources for gene discovery and breeding显示文摘Watermelon,Citrullus lanatus,is the world's third largest fruit crop.Reference genomes with gaps and a narrow genetic base hinder functional genomics and genetic improvement of watermelon.Here,we report the assembly of a telomere-to-telomere gap-free genome of the elite watermelon inbred line G42 by incorporating high-coverage and accurate long-read sequencing data with multiple assembly strategies.All 11 chromosomes have been assembled into single-contig pseudomolecules without gaps,representing the highest completeness and assembly quality to date.The G42 reference genome is 369321829 bp in length and contains 24205 predicted protein-coding genes,with all 22 telomeres and 11 centromeres characterized.Furthermore,we established a pollen-EMS mutagenesis protocol and obtained over 200000M1 seeds from G42.In a sampling pool,48 monogenic phenotypic mutations,selected from 223M1and 78 M2 mutants with morphological changes,were confirmed.The average mutation density was 1 SNP/1.69Mband1 indel/4.55 Mb per M1 plant and 1SNP/1.08Mb and 1 indel/6.25 Mb per M2 plant.Taking advantage of the gap-free G42 genome,8039 mutations from 32 plants sampled from M1 and M2 families were identified with 100%accuracy,whereas only 25% of the randomly selected mutations identified using the 97103v2 reference genome could be confirmed.Using this library and the gap-free genome,two genes responsible for elongated fruit shape and male sterility(CiMs1)were identified,both caused by a single basechange from G to A.The validated gap-free genome and its EMS mutation library provide invaluable resources for functional genomics and genetic improvement of watermelon.Yun Deng Shoucheng Liu Yilin Zhang Jingsheng Tan Xiaopeng Li Xiao Chu Binghua Xu Yao Tian Yudong Sun Bosheng Li YunbiXu Xing Wang Deng Hang He Xingping Zhang 2022Molecular Plant2022,15,8:9

17Neural progenitor cells but not astrocytes respond distally to thoracic spinal cord injury in rat models显示文摘Traumatic spinal cord injury(SCI) is a detrimental condition that causes loss of sensory and motor function in an individual. Many complex secondary injury cascades occur after SCI and they offer great potential for therapeutic targeting. In this study, we investigated the response of endogenous neural progenitor cells,astrocytes, and microglia to a localized thoracic SCI throughout the neuroaxis. Twenty-five adult female Sprague-Dawley rats underwent mild-contusion thoracic SCI(n = 9), sham surgery(n = 8), or no surgery(n = 8). Spinal cord and brain tissues were fixed and cut at six regions of the neuroaxis. Immunohistochemistry showed increased reactivity of neural progenitor cell marker nestin in the central canal at all levels of the spinal cord. Increased reactivity of astrocyte-specific marker glial fibrillary acidic protein was found only at the lesion epicenter. The number of activated microglia was significantly increased at the lesion site,and activated microglia extended to the lumbar enlargement. Phagocytic microglia and macrophages were significantly increased only at the lesion site. There were no changes in nestin, glial fibrillary acidic protein,microglia and macrophage response in the third ventricle of rats subjected to mild-contusion thoracic SCI compared to the sham surgery or no surgery. These findings indicate that neural progenitor cells, astrocytes and microglia respond differently to a localized SCI, presumably due to differences in inflammatory signaling. These different cellular responses may have implications in the way that neural progenitor cells can be manipulated for neuroregeneration after SCI. 18Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. During each month the SIC trends are negative over the Arctic Ocean, wherein the largest(smallest) rate of decline found in September(March) is-0.48%/a(-0.10%/a).The summer(-0.42%/a) and autumn(-0.31%/a) seasons show faster decrease rates than those of winter(-0.12%/a) and spring(-0.20%/a) seasons. Regional variability is large in the annual SIC trend. The largest SIC trends are observed for the Kara(-0.60%/a) and Barents Seas(-0.54%/a), followed by the Chukchi Sea(-0.48%/a), East Siberian Sea(-0.43%/a), Laptev Sea(-0.38%/a), and Beaufort Sea(-0.36%/a). The annual SIC trend for the whole Arctic Ocean is-0.26%/a over the same period. Furthermore, the in?uences and feedbacks between the SIC and three climate indexes and three climatic parameters, including the Arctic Oscillation(AO), North Atlantic Oscillation(NAO), Dipole anomaly(DA), sea surface temperature(SST), surface air temperature(SAT), and surface wind(SW), are investigated. Statistically, sea ice provides memory for the Arctic climate system so that changes in SIC driven by the climate indices(AO, NAO and DA) can be felt during the ensuing seasons. Positive SST trends can cause greater SIC reductions, which is observed in the Greenland and Barents Seas during the autumn and winter. In contrast, the removal of sea ice(i.e., loss of the insulating layer) likely contributes to a colder sea surface(i.e., decreased SST), as is observed in northern Barents Sea. Decreasing SIC trends can lead to an in-phase enhancement of SAT, while SAT variations seem to have a lagged in?uence on SIC trends. 20Attenuation of rock blasting induced ground vibration in rock-soil interface显示文摘Blasting has been widely used in mining and construction industries for rock breaking.This paper presents the results of a series of field tests conducted to investigate the ground wave propagation through mixed geological media.The tests were conducted at a site in the northwestern part of Singapore composed of residual soil and granitic rock.The field test aims to provide measurement data to better understand the stress wave propagation in soil/rock and along their interface.Triaxial accelerometers were used for the free field vibration monitoring.The measured results are presented and discussed,and empirical formulae for predicting peak particle velocity (PPV) attenuation along the ground surface and in soil/rock were derived from the measured data.Also,the ground vibration attenuation across the soil-rock interface was carefully examined,and it was found that the PPV of ground vibration was decreased by 37.2% when it travels from rock to soil in the vertical direction.Bhagya Jayasinghe Zhiye Zhao Anthony Goh Teck Chee Hongyuan Zhou Yilin Gui 2019Journal of Rock Mechanics and Geotechnical Engineering2019,11,4:7
