| 1 | MicroRNA-24 promotes pancreatic beta cells toward dedifferentiation to avoid endoplasmic reticulum stress-induced apoptosis显示文摘Current research indicates that beta cell loss in type 2 diabetes may be attributed to beta cell dedifferentiation rather than apoptosis;however,the mechanisms by which this occurs remain poorly understood.Our previous study demonstrated that elevation of microRNA-24(miR-24)in a diabetic setting caused beta cell dysfunction and replicative deficiency.In this study,we focused on the role of miR-24 in beta cell apoptosis and dedifferentiation under endoplasmic reticulum(ER)stress conditions.We found that miR-24 overabundance protected beta cells from thapsigargin-induced apoptosis at the cost of accelerating the impairment of glucosestimulated insulin secretion(GSIS)and enhancing the presence of dedifferentiation markers.Ingenuity?Pathway Analysis(IPA)revealed that elevation of miR-24 had an inhibitory effect on XBP1 and ATF4,which are downstream effectors of two key branches of ER stress,by inhibiting its direct target,Irela.Notably,elevated miR-24 initiated another pathway that targeted Mafa and decreased GSIS function in surviving beta cells,thus guiding their dedifferentiation under ER stress conditions.Our results demonstrated that the elevated miR-24,to the utmost extent,preserves beta cell mass by inhibiting apoptosis and inducing dedifFerentiation.This study not only provides a novel mechanism by which miR-24 dominates beta cell turnover under persistent metabolic stress but also offers a therapeutic consideration for treating diabetes by inducing dedifferentiated beta cells to re-differentiation. | Yunxia Zhu Yi Sun Yuncai Zhou Yan Zhang Tao Zhang Yating Li Weiyan You Xiaoai Chang Li Yuan Xiao Han | 2019 | Journal of Molecular Cell Biology2019,11,9: | 9 |

| 3 | Antibacterial Effects of Chinese Herbal Medicines against Streptococcus iniae in Oreochromis niloticus显示文摘In this study,the antibacterial effects of Chinese herbal medicines against Streptococcus iniae in Oreochromis niloticus were investigated by drug sensitivity test. The results indicated that Streptococcus iniae was extremely sensitive to Galla Chinensis,Fructus Hippophae and Fructus Mume,highly sensitive to Sappan Lignum,Pericarpium Granati,Folium Eucalypti and Radix Scutellariae,moderately sensitive to Rhizoma Coptidis,Radix et Rhizoma Rhei,Flos Caryophyllata,Cortex phellodendri and Rabdosia serra( Maxim.) Hara,and insensitive to Herba Portulacae,Herba Houttuyniae,Polygonum hydropiper L.,Herba Menthae,Radix Glycyrrhizae,Pericarpium Citri Reticulatae,Herba Andrographitis and Rhizoma Acori Tatarinowii. Chinese medicinal herbs which Streptococcus iniae was extremely sensitive or moderately sensitive to were selected and prepared into three Chinese herbal medicine formulae for medicated bath of Oreochromis niloticus infected artificially with Streptococcus iniae. The results showed that medicated bath elicited therapeutic effects on infected Oreochromis niloticus. | 4 | Targeting EZH1/2 induces cell cycle arrest and inhibits cell proliferation through reactivation of p57CDKN1C and TP53INP1 in mantle cell lymphoma显示文摘Objective: To explore the effect of dysregulation of epigenetic regulator EZH1 and EZH2 on the proliferation in MCL and the underlying mechanisms.Methods: In this study, we elucidated the role of EZH1 and EZH2 overexpression by immunohistochemistry and correlated them to clinical outcome in 41 MCL patients.Quantitative real-time PCR and Western blot were applied to confirm the level of EZH1 and EZH2 in well-characterized MCL cell lines which were compared to those of na?ve B cells.Then we manipulated the expression of EZH1 and EZH2 in MCL cells using CRISPR/Cas9 system to directly investigate their functional roles in MCL.We also evaluated the effect of two small molecule selective inhibitors, EPZ005687 and UNC1999, on MCL cell proliferation, cell cycle distribution and apoptosis in vitro.Finally, we performed RNA-sequencing(RNA-Seq) and Chromatin immunoprecipitation(ChIP) assay to further gain insight into the underlying molecular mechanisms.Results: We found that EZH2 protein is overexpressed in approximately half of this cohort of MCL cases.More importantly, the overexpression of EZH2 is associated with poor OS in the patients.Nevertheless, simple EZH2 depletion in vitro has little impact on the viability of MCL cells, predominantly because of the consequent up-regulation of EZH1.Consistently, UNC1999, a dual EZH1/2 inhibitor, unlike the EZH2 selective inhibitor EPZ005687, exerts a potent inhibitory effect on MCL cells.Furthermore, we discover CDKN1C and TP53 INP1 as the two important cell cycle regulators, the expression of which are repressed by EZH1/2 mediated epigenetic regulation and are restored by EZH1/2 dual inhibition.Conclusions: Our study suggests that EZH2 participates in the pathogenesis of MCL which may serve as a potential biomarker for prognosis prediction.The dual inhibition of EZH1/2 is a promising therapeutic strategy for MCL. | Wei Li Chengfeng Bi Yating Han Tian Tian Xianhuo Wang Huijing Bao Xiaoying Xu Xuhan Zhang Lu Liu Weiwei Zhang Hai Gao Huaqing Wang Huilai Zhang Bin Meng Xi Wang Kai Fu | 2019 | Cancer Biology & Medicine2019,16,3: | 2 |
| 5 | Filling Ti_(3)C_(2)T_(x)nanosheets into melamine foam towards a highly compressible all-in-one supercapacitor显示文摘The development of compressible supercapacitors strongly relies on the design of electrode materials combining superior compressibility,high conductivity with the stable electrochemical cycling performance.In this work,we report a facile yet scalable strategy to construct a highly compressible supercapacitor by integrating the current collector,active materials and the separator into one device.We use the highly compressive melamine foam(MF)as scaffold and the Ti_(3)C_(2)T_(x)nanosheets as the active materials.Filling the few-layer Ti_(3)C_(2)T_(x)nanosheets into the skeleton of MF by capillary force followed by freeze-drying yields the MF/Ti_(3)C_(2)T_(x)composite with superior structural integrity that can be compressed at a large strain of 50%for 100 cycles.The electrochemical performances of the all-in-one supercapacitor were systematically investigated under diverse compression strains.The improved conductivity and reduced ion diffusion length allow the all-in-one supercapacitor to exhibit fast ion and electron kinetics even at high strain of 60%,delivering a maximal volumetric specific energy of 0.37 mWh∙cm^(-3)at power density of 0.42 mW∙cm^(-3)and extraordinary cycling performance during the 2,500 compression cycles. | Rui Guo Xiying Han Peng Yuan Xuexia He Qi Li Jie Sun Liqin Dang Zonghuai Liu Yating Zhang Zhibin Lei | 2022 | Nano Research2022,15,4: | 1 |

| 12 | Determination of Fosthiazate Residue in Ginger by Gas Chromatography显示文摘[Objectives] An analytical method was established for determining fosthiazate residue in ginger by gas chromatography with flame photometric detector(GC-NPD) to evaluate the safety of fosthiazate in ginger. [Methods] The fosthiazate residue in ginger was extracted with dichloromethane, and subjected to sodium chloride salt-out and liquid-liquid extraction with hexane and dichloromethane(the plant and soil samples had no need for the later two steps). The analysis was performed on gas chromatograph equipped with a flame photometric detector(GC-FPD), and the fosthiazate residue was quantitatively determined by calculating the peak area. [Results] The results of degradation dynamics showed that the half-life of fosthiazate was 7.2-8.7 d in ginger plants, and was 9.9-19.8 d in the soil. The results of the final residue test showed that after applying the fosthiazate granules according to the recommended high dose(active component 58.2 g a.i/m^2,WK) and low dose(38.8 g a.i/m^2,WK) once, the final residual amounts of fosthiazate in the ginger and soil were estimated to be <0.02 mg/kg and <0.02-0.103 mg/kg, respectively, while no fosthiazate residue was detected in all control samples(<0.02 mg/kg). The maximum residue limit(MRL) of fosthiazate in ginger was not established in China, and was 0.2 mg/kg in Japan. [Conclusions] The method was proved to meet the basic requirements of pesticide residue analysis on sensitivity, accuracy and precision and have good linearity. | 14 | A unified model of cusp spot, High Latitude Dayside aurora (HiLDA)/(Space Hurricane), and 15MLT-PCA显示文摘Cusp spot,High Latitude Dayside aurora(HiLDA),‘space hurricane’,and 15MLT-PCA(Polar Cap Arc observed around 15:00 Magnetic Local Time)are mesoscale auroral structures observed in the polar cap region.They share many common properties and,at the same time,have notable differences.A 15MLT-PCA is a polar cap arc connected to the auroral oval,but the others are auroral spots detached from the oval.A cusp spot differs from a HiLDA in local time location.A space hurricane differs from a HiLDA in having spiral-arm structures.Until now,relationships among these auroras have not been depicted clearly.Here we propose a unified model,based on lobe reconnection,that encompasses their similarities and differences.We then provide critical supporting evidence for the model.The model suggests that different reconnection sites under different IMF conditions result in these differently-appearing auroral forms.The model explains all the characteristic features of these auroras and has implications for understanding the dawn/dusk and inter-hemispheric asymmetries observed in their occurrence patterns.We anticipate confirmation of the model by observations to be made during the Solar wind-Magnetosphere-Ionosphere Link Explorer(SMILE)mission.Moreover,since the model indicates that these auroras appear on the footprint of the poleward cusp boundary,we expect data from the SMILE mission to make it possible to estimate the approximate shape of the cusp. | DeSheng Han YaTing Xiong Run Shi HuiXuan Qiu HuiTing Feng | 2023 | Earth and Planetary Physics2023,7,4: | 0 |
| 15 | Cholesterol accumulation on dendritic cells reverses chronic hepatitis B virus infection-induced dysfunction显示文摘Chronic hepatitis B(CHB)infection remains a serious public health problem worldwide;however,the relationship between cholesterol levels and CHB remains unclear.We isolated peripheral blood mononuclear cells from healthy blood donors and CHB patients to analyze free cholesterol levels,lipid raft formation,and cholesterol metabolism-related pathways.Hepatitis B virus(HBV)-carrier mice were generated and used to confirm changes in cholesterol metabolism and cell-surface lipid raft formation in dendritic cells(DCs)in the context of CHB.Additionally,HBV-carrier mice were immunized with a recombinant HBV vaccine(rHBVvac)combined with lipophilic statins and evaluated for vaccine efficacy against HBV.Serum samples were analyzed for HBsAg,anti-HBs,and alanine aminotransferase levels,and liver samples were evaluated for HBV DNA and RNA and HBcAg.CHB reduced free cholesterol levels and suppressed lipid raft formation on DCs in patients with CHB and HBV-carrier mice,whereas administration of lipophilic statins promoted free cholesterol accumulation and restored lipid rafts on DCs accompanied by an enhanced antigen-presentation ability in vitro and in vivo.Cholesterol accumulation on DCs improved the rHBVvac-mediated elimination of serum HBV DNA and intrahepatic HBV DNA,HBV RNA,and HBcAg and promoted the rHBVvac-mediated generation and polyfunctionality of HBV-specific CD11a^(hi) CD8α^(lo) cells,induction of the development of memory responses against HBV reinfection,and seroconversion from HBsAg to anti-HBs.The results demonstrated the important role of cholesterol levels in DC dysfunction during CHB,suggesting that strategies to increase cholesterol accumulation on DCs might enhance therapeutic vaccine efficacy against HBV and support development toward clinical CHB treatment. | Huajun Zhao Yating Yu Yucan Wang Lianhui Zhao Ailu Yang Yifei Hu Zhaoyi Pan Zixuan Wang Jiarui Yang Qiuju Han Zhigang Tian Jian Zhang | 2022 | Cellular & Molecular Immunology2022,19,12: | 0 |

| 20 | CircUCP2 promotes the tumor progression of non-small cell lungcancer through the miR-149/UCP2 pathway显示文摘Non-small cell lung cancer(NSCLC)is a highly lethal cancer,and better treatments are urgently needed.Many studies have implicated circular RNAs(circRNAs)in the progression of multiple malignant tumors.Nonetheless,the functions of circRNAs in NSCLC remain unclear.To study new targets for the treatment of NSCLC,circRNA expression profiling was performed on NSCLC tissues and para-carcinoma nonmalignant tissues.RNA was isolated and used for circRNA sequencing.Biological studies were performed in vitro and in vivo to determine the functions of circRNAs in NSCLC,including their functions in cell proliferation and migration.How circRNAs function in NSCLC was explored to clarify the underlying regulatory mechanisms.We found that circUCP2 was upregulated in NSCLC tissues compared with neighboring nonmalignant tissues.circUCP2 promoted the proliferation and metastasis of NSCLC cells.circUCP2 promoted NSCLC progression by sponging miR-149 and upregulating UCP2.The circUCP2/miR-149/UCP2 axis accelerates the progression of NSCLC,and circUCP2 may therefore be a novel diagnostic biomarker for the progression of NSCLC. | WEI DU FANG YIN YATING ZHONG MINJIE LUO ZHEN WANG PENG LIN QING LIU HAN YANG | 2023 | Oncology Research2023,31,6: | 0 |
