1Application of Next-generation Sequencing Technology in Forensic Science显示文摘Next-generation sequencing(NGS) technology, with its high-throughput capacity and low cost, has developed rapidly in recent years and become an important analytical tool for many genomics researchers. New opportunities in the research domain of the forensic studies emerge by harnessing the power of NGS technology, which can be applied to simultaneously analyzing multiple loci of forensic interest in different genetic contexts, such as autosomes, mitochondrial and sex chromosomes. Furthermore, NGS technology can also have potential applications in many other aspects of research. These include DNA database construction, ancestry and phenotypic inference,monozygotic twin studies, body fluid and species identification, and forensic animal, plant and microbiological analyses. 2Characterization of miRNomes in Acute and Chronic Myeloid Leukemia Cell Lines显示文摘Myeloid leukemias are highly diverse diseases and have been shown to be associated with microRNA(miRNA) expression aberrations. The present study involved an in-depth miRNome analysis of two human acute myeloid leukemia(AML) cell lines, HL-60 and THP-1, and one human chronic myeloid leukemia(CML) cell line, K562, via massively parallel signature sequencing. mRNA expression profiles of these cell lines that were established previously in our lab facilitated an integrative analysis of miRNA and mRNA expression patterns. miRNA expression profiling followed by differential expression analysis and target prediction suggested numerous miRNA signatures in AML and CML cell lines. Some miRNAs may act as either tumor suppressors or oncomiRs in AML and CML by targeting key genes in AML and CML pathways. Expression patterns of cell type-specific miRNAs could partially reflect the characteristics of K562, HL-60 and THP-1 cell lines, such as actin filament-based processes, responsiveness to stimulus and phagocytic activity. miRNAs may also regulate myeloid differentiation, since they usually suppress differentiation regulators. Our study provides a resource to further investigate the employment of miRNAs in human leukemia subtyping, leukemogenesis and myeloid development. 3Provenance and tectonic setting transition as recorded in the Neoproterozoic strata, western Jiangnan Orogen:Implications for South China within Rodinia显示文摘The Neoproterozoic Tonian strata (ca.870-725 Ma) in the western Jiangnan Orogen archive the records of sedimentary provenance and tectonic setting which can be used to understand the geological evolution of the South China Continent.These strata are separated into the basement and cover sequences by a regional angular unconformity.The basement sequence can be subdivided into the lower and the upper parts by the widespread interbedded ca.840 Ma basalt with pillow structure.In the present work,234 concordant detrital zircon analyses are obtained from three Tonian sandstone samples in the Fanjingshan district,Guizhou Province.Combined with previous results,a total of 1736 analyses of detrital zircon U-Pb ages derived from 12 formations of Tonian strata in the western Jiangnan Orogen are used to decipher the integrated sedimentary and tectonic histories.The zircons from the lowermost part of the basement sequence (the Yujiagou Formation) show oval morphology and display two Paleoproterozoic age peaks at 2325 Ma and 1845 Ma which are similar with the detrital zircon age peaks from the Late Paleoproterozoic to Early Mesoproterozoic Dongchuan/Dahongshan/Hekou groups,suggesting a passive margin basin in which the sediments were mainly sourced from the southwestern Yangtze Block.However,the zircon age population of the lower part of the basement sequence (the Xiaojiahe,Huixiangping formations and their equivalents) indicates the sedimentary derivation from bidirectional sources (the ca.870 Ma arc materials in the south and the old detritus from the southwestern Yangtze Block) which is consistent with a back arc setting for the deposition of the sediments.Zircons from the upper part of the basement sequence (the Duyantang Formation and its equivalent) show euhedral and subangular morphology and display a unimodal age peak at ca.835 Ma.This sequence was possibly deposited in a convergent setting and the detritus were came from the locally distributed syn-collisional igneous rocks.The lower part of the cover sequence (the Xinzhai and Wuye formations and their equivalents) shows a distinct zircon age peak at 815-809 Ma and two subordinate peaks at 2485 Ma and 2018 Ma,suggesting that the basin had gradually transformed into a continental rift basin and received the detritus from the ca.815 Ma post-collisional magmatic rocks as well as from different Paleoproterozoic source rocks in the northern Yangtze Block.We propose a tectonic evolution model that envisages eruption of ca.840 Ma basalt in a back arc basin that existed during ca.870-835 Ma,an angular unconformity was formed during amalgamation of the Yangtze Block and the Cathaysia Block at ca.835-820 Ma and the rifting of the South China Continent was initiated at ca.800 Ma.Our study concludes that the South China Continent was formed on the periphery of the Rodinia supercontinent.Jiawei Zhang Taiping Ye Yaran Dai Jianshu Chen Hui Zhang Chuangu Dai Guohua Yuan Kaiyuan Jiang 2019Geoscience Frontiers2019,10,5:4
4Preparation of SiC Porous Ceramics by Crystalline Silicon Cutting Waste显示文摘SiC porous ceramics were prepared at 1 400 ℃ for4 h with crystalline silicon cutting waste and activated carbon as main starting materials and NH_4HCO_3 as the pore-forming agent. Effects of NH_4HCO_3 additions( 0,20%,30%,40%,by mass) on the phase composition,microstructure,sintering properties,cold compressive strength and thermal shock resistance of as-prepared Si C porous ceramics were investigated. The results show that:( 1) addition of NH_4HCO_3 remarkably influences the apparent porosity and cold compressive strength of specimens. The apparent porosity achieves its maximum value( 63. 40%) when 40% NH_4HCO_3 is added,while the minimum cold compressive strength is 4. 77 MPa;( 2) the specimen with 40% NH_4HCO_3 has the best thermal shock resistance. 5Thrombin is a therapeutic target for non-small-cell lung cancer to inhibit vasculogenic mimicry formation显示文摘Tumor cells transform into endothelial cells by epithelial-to-mesenchymal transition,which is characterized by vasculogenic mimicry(VM).VM not only accelerates tumor progression but also increases drug-induced resistance.However,very little is currently known about the molecular determinants that enable VM.Targeting VM might bring a new breakthrough in cancer treatment.Thrombin is the key enzyme of the blood coagulation system and could contribute to tumor progression.Nevertheless,the association between thrombin and VM formation remains largely unknown.We found that VM was associated with the overall survival of non-small-cell lung cancer(NSCLC)patients,and that thrombin expression was closely related to VM formation.This research revealed that thrombin induced VM formation via PAR-1-mediated NF-κB signaling cascades.The novel thrombin inhibitors r-hirudin and DTIP inhibited VM formation and spontaneous metastases in subcutaneous tumors.Clinical pathological analysis confirmed that NSCLC patients with thrombin-positive/PAR-1-high expression had the poorest prognosis and were the most likely to form VM.The promotional activity of thrombin in VM formation and tumor metastasis was abolished in PAR-1-deficient NSCLC cells.The EGFR inhibitor gefitinib had no effect on VM and increased VEGF expression in tumors.The combination therapy of DTIP and gefitinib achieved a better therapeutic effect than either agent alone.This study is the first to illustrate that thrombin substantially contributes,together with PAR-1,to VM formation and to illustrate that VM might be a target of r-hirudin and DTIP to suppress tumor progression.The anticoagulants r-hirudin and DTIP could be employed for antitumor therapy.Combination therapy with DTIP with an EGFR inhibitor might achieve superior therapeutic effects.Bing Zhao Mengfang Wu Zhihuang Hu Yixin Ma Wang Qi Yanling Zhang Yaran Li Min Yu Huijie Wang Wei Mo 2020Signal Transduction and Targeted Therapy2020,5,1:2
6Selective adsorption of phosphate in water using lanthanum-based nanomaterials:A critical review显示文摘In recent years,lanthanum-based nanomaterials(La-NMs)are selected as an efficient nano-adsorbent for phosphate removal because La3+has a strong affinity with oxygen-donor atoms from phosphate.Additionally,there are a broad interest and literature base for the effect of different synthesis optimization and environmental parameters on the adsorption performance of La-NMs.A considerable amount of research has also investigated the regeneration and application of La-NMs to real wastewater in a laboratory scale.Based on the literature survey,it was found that La-NMs are often produced via co-precipitation and hydrothermal methods.Moreover,phosphate’s adsorption process and behavior onto La-NMs are described well with the pseudo-second-order model and Langmuir model.The interaction mechanism between phosphate and La-NMs are dominated by ligand exchange,surface complexation and electrostatic attraction.Furthermore,phosphate could easily desorb from La-NMs due to the weak H-bonding interaction between phosphate and the H-bond acceptor groups on the surface of La-NMs.Despite the wealth of literature available in this area,there is a lack of systematic review to evaluate the gaps in the use of La-NMs to eliminate phosphate in water.In this review,we mainly summarize and discuss the role and the effect of the synthesis techniques on the physicochemical properties and the adsorption behavior of La-NMs.The possible adsorption mechanism,regeneration efficiency,and the application of La-NMs to the real environmental samples are also presented and highlighted.Mamitiana Roger Razanajatovo Wenyan Gao Yaran Song Xuan Zhao Qina Sun Qingrui Zhang 2021Chinese Chemical Letters2021,32,9:2

10Effects of Pore-forming Agents on Properties of Silicon Carbide Porous Ceramics Prepared from Crystalline Silicon Cutting Waste显示文摘SiC powder was rapidly synthesized in an induction furnace with crystalline silicon cutting waste and active carbon as raw materials,and then SiC porous ceramics were prepared at 1600 t for 4 h with carbon embedded using the powder as raw material,the starch and the graphite as pore-forming agents.Effects of additions of different pore-forming agents on the phase composition,microstructures,physical properties,and cold crushing strength of the porous ceramics were investigated.The results show that the main crystalline phases of the synthetic powder areα-S iC(6H-SiC)andβ-SiC(3C-SiC).The phase composition of the porous ceramics includesα-S iC(6H-SiC),β-SiC(3C-SiC),FeSi,quartz and Si2N20.The apparent porosity and closed porosity of the porous ceramics prepared by adding starch are higher,and the cold compressive strength of the porous ceramics added with graphite is higher.As increasing the additions of the starch,the apparent porosity,closed porosity and linear shrinkage ratio of the porous ceramics increase,and the bulk density decreases correspondingly.When 20 mass%starch is added,the apparent porosity,closed porosity,linear shrinkage ratio and cold compressive strength are 57.05%,2.03%,5.10%and 10.20 MPa,respectively.MA Beiyue WU Hua GAO Zhi REN Xinming ZHANG Yaran 2019China's Refractories2019,28,4:1
11Effect of microstructure evolution and crystal structure on thermal properties for plasma-sprayed RE_(2)SiO_(5)(RE=Gd,Y,Er)environmental barrier coatings显示文摘In this work,the microstructure evolution,thermal expansion,thermal conductivity,and thermal shock resistance properties of the plasma-sprayed Xl-Gd_(2)SiO_(5),X2-Y_(2)SiO_(5),and X2-Er_(2)SiO_(5)coatings were evaluated and compared by experimental measurement and theoretical exploration.Results showed that significant microstructure evolution such as crystallization of amorphous phase,grain growth,and defects reduction was observed in the RE_(2)SiO_(5)coatings after thermal aging at 1400℃.The Xl-Gd_(2)SiO_(5)coating exhibited higher CTE values than the X2-Y_(2)SiO_(5)and X2-Er_(2)SiO_(5)coatings,which was related to their crystal structure.The thermal conductivity of thermal-aged RE_(2)SiO_(5)coating was much higher than that of the as-sprayed RE_(2)SiO_(5)coating,and thermal conductivity was determined not only by crystal structure but also mainly by the microstructure of the coatings.The X2-Y_(2)SiO_(5)and X2-Er_(2)SiO_(5)coatings with lower thermal mismatch stre s ses presented much better thermal shock resistance than the X1-Gd_(2)SiO_(5)coating.Xin Zhong Tao Zhu Yaran Niu Haijun Zhou Le Zhang Xiangyu Zhang Qilian Li Xuebin Zheng 2021Journal of Materials Science & Technology2021,,26:1

13DNA methylation and alternative splicing modulate FBXW11 gene expression in Holstein bull testis and are correlated with sperm quality显示文摘F-box and WD-40 domain protein 11(FBXW11)is an important component of the E3 ubiquitin-ligase enzyme that plays a key role in the ubiquitin-dependent regulation of spermatogenesis.In our previous research,the mRNA expression of FBXW11 in bull sperm with high motility is significantly higher than that with low motility.In the present study,the protein expression levels of FBXW11 in bull testicular tissues with low-performance sperm quality groups were significantly higher than those in normal performance groups.The immunohistochemistry result demonstrated that FBXW11 protein was located in the periphery of Leydig cells and seminiferous tubules.Three splice variants of the FBXW11 gene,namely,FBXW11-tv1,FBXW11-tv2,and FBXW11-tv3,were identified in testicular tissues.The splicing patterns of the three variants are exon skipping.The transcript FBXW11-tv2 expressions were the highest in each sample.The low-performance groups displayed higher FBXW11-tv1 and FBXW11-tv2 transcript expressions than the normal performance groups.Two CpG islands were located within the 5’UTR and exon 1-2 region of the FBXW11 gene.Bisulfite sequencing PCR results demonstrated that the methylation levels of 11 methylation sites in the CpG island 2 from−99 to−43 in the normal performance groups were significantly lower than those in the low-performance groups.Pearson correlation analysis suggested that the CpG island 2 methylation level was negatively correlated with sperm motility and the transcript FBXW11-tv2 expression level.Our data revealed that alternative splicing and DNA methylation jointly regulated FBXW11 gene expression and were correlated with sperm quality traits during spermatogenesis in Holsteins.YONG LIU ZHIHUA JU QIANG JIANG WENHAO LIU CHUNHONG YANG YARAN ZHANG XIUGE WANG YAPING GAO XIAOCHAO WEI YAN SUN JINPENG WANG MINGHAI HOU LING YANG JINMING HUANG 2021BIOCELL2021,45,1:1
