1Reprogramming of phytopathogen transcriptome by a non-bactericidal pesticide residue alleviates its virulence in rice显示文摘Bacteria equipped with virulence systems based on highly bioactive small molecules can circumvent their host's defense mechanisms.Pathogens employing this strategy are currently threatening global rice production.In the present study,variations in the virulence of the highly destructive Barkholderiaplantarii were observed in different rice-producing regions.The environment-linked variation was not attributable to any known host-related or external factors.Co-occurrence analyses indicated a connection between reduced virulence and 5-Amino-l,3,4-thiadiazole-2-thiol(ATT),a non-bactericidal organic compound.ATT,which accumulates in rice plants during metabolization of specific agrochemicals,was found to reduce virulence factor secretion by B.plantarii up to 88.8%and inhibit pathogen virulence by hijacking an upstream signaling cascade.Detailed assessment of the newly discovered virulence inhibitor resulted in mechanistic insights into positive effects of ATT accumulation in plant tissues.Mechanisms of virulence alleviation were deciphered by integrating high-throughput data,gene knockout mutants,and molecular interaction assays.TroK,a histidine protein kinase in a two-component system that regulates virulence factor secretion,is likely the molecular target antagonized by ATT.Our findings provide novel insights into virulence modulation in an important plant-pathogen system that relies on the host's metabolic activity and subsequent signaling interference.Haruna Matsumoto Yuan Qian Xiaoyan Fan Sunlu Chen Yanxia Nie Kun Qiao Dandan Xiang Xinzhong Zhang Meng Li Bo Guo Peilin Shen Qiangwei Wang Yunlong Yu Tomislav Cemava Mengcen Wang 2022Fundamental Research2022,2,2:2
2Overhauling the assessment of agrochemical-driven interferences with microbial communities for improved global ecosystem integrity显示文摘Recent studies have shown that various agrochemicals can substantially affect microbial communities;especially those that are associated with cultivated plants.Under certain circumstances,up to 50% of the naturally occurring microorganisms can be negatively affected by common agricultural practices such as seed coating with fungicide-based matrices.Nevertheless,the off-target effects of commonly applied agrochemicals are still understudied in terms of their interferences with microbial communities.At the same time,agrochemical inputs are steadily increasing due to the intensification of agriculture and the increasing pathogen pressure that is currently observed worldwide.In this article,we briefly reflect on the current knowledge related to pesticide interference with microbial communities and discuss negative implications for the plant holobiont as well as such that are spanning beyond local system borders.Cumulative effects of pesticide inputs that cause alterations in microbial functioning likely have unforeseen implications on geochemical cycles that should be addressed with a high priority in ongoing research.A holistic assessment of such implications will allow us to objectively select the most suitable means for food production under the scenario of a growing global population and aggravating climatic conditions.We present three hypothetical solutions that might facilitate a more sustainable and less damaging application of pesticides in the future.Mengcen Wang Tomislav Cernava 2020Environmental Science and Ecotechnology2020,,4:2
3Bacterial-fungal interactions under agricultural settings:from physical to chemical interactions显示文摘Bacteria and fungi are dominant members of environmental microbiomes.Various bacterial-fungal interactions(BFIs)and their mutual regulation are important factors for ecosystem functioning and health.Such interactions can be highly dynamic,and often require spatiotemporally resolved assessments to understand the interplay which ranges from antagonism to mutualism.Many of these interactions are still poorly understood,especially in terms of the underlying chemical and molecular interplay,which is crucial for inter-kingdom communication and interference.BFIs are highly relevant under agricultural settings;they can be determinative for crop health.Advancing our knowledge related to mechanisms underpinning the interactions between bacteria and fungi will provide an extended basis for biological control of pests and pathogens in agriculture.Moreover,it will facilitate a better understanding of complex microbial community networks that commonly occur in nature.This will allow us to determine factors that are crucial for community assembly under different environmental conditions and pave the way for constructing synthetic communities for various biotechnological applications.Here,we summarize the current advances in the field of BFIs with an emphasis on agriculture.Yaqi Zhou Hongkai Wang Sunde Xu Kai Liu Hao Qi Mengcen Wang Xiaoyulong Chen Gabriele Berg Zhonghua Ma Tomislav Cernava Yun Chen 2022Stress Biology2022,2,1:1
4Characterization of the chemical fungicides-responsive and bacterial pathogen-preventing Bacillus licheniformis in rice spikelet显示文摘Seed-borne bacterial pathogens cause severe yield loss and biotoxin contamination in rice,leading to increasing concern on the global food supply and environmental safety.Plant native microbes play an important role in defending against diseases,but their actions are often influ-enced by the chemical fungicides applied in the field.Here,Bacillus licheniformis mmj was isolated from rice spikelet,which uniquely showed not only fungicide-responsiveness but also broad-spectrum antimicrobial activity against major rice bacterial pathogens including Xanthomonas oryzae pv.oryzae,Burkholderia plantari and Burkholderia glumae.To understand the hallmark underlying the environmental adaptation and anti-microbial activity of B.licheniformis mmj,the genome sequence was determined by SMRT and subjected to bioinformatics analysis.Genome sequence analysis enabled the identification of a set of antimicrobial-resistance and antibacterial activity genes together with an array of harsh environment-adaptive genes.Moreover,B.licheniformis mmj metabolites were analyzed with gas chromatography coupled to triple quadrupole mass spectrometry,and the volatile components that were linked with the antimicrobial activity were preliminarily profiled.Collectively,the present findings reveal the genomic and metabolic landscapes underlying fungicide-responsive B.licheniformis,which offers a new opportunity to design harsh environment-adaptive biopesticides to cope with prevalent bacterial phytopathogens.Chengfang Zhan Mengchen Wu Hongda Fang Xiaoyu Liu Jiuyue Pan Xiaoyan Fan Mengcen Wang Haruna Matsumoto 2023Food Quality and Safety2023,7,2:1
5Predicting rice diseases using advanced technologies at different scales: present status and future perspectives显示文摘The past few years have witnessed significant progress in emerging disease detection techniques foraccurately and rapidly tracking rice diseases and predicting potential solutions. In this review we focuson image processing techniques using machine learning (ML) and deep learning (DL) models related tomulti-scale rice diseases. Furthermore, we summarize applications of different detection techniques,including genomic, physiological, and biochemical approaches. In addition, we also present the state-ofthe-art in contemporary optical sensing applications of pathogen–plant interaction phenotypes. 6Gut microbiota dysbiosis involves in host non-alcoholic fatty liver disease upon pyrethroid pesticide exposure显示文摘A growing body of evidence has demonstrated the significance of the gut microbiota in host health,while the association between gut microbiota dysbiosis and multiple diseases is yet elusive in the scenario of exposure to widely used pesticides.Here,we show that gut microbiota dysbiosis involves in host's abnormal lipid metabolism and consequently the non-alcoholic fatty liver disease in Xenopus laevis upon exposure to cis-bifenthrin,one of the most prevalent pyrethroid insecticides in the world.With the guidance of gut microbiota analysis,we found that cis-bifenthrin exposure significantly perturbed the gut microbial community,and the specific taxa that served as biomarkers were identified.Metabolomics profiling and association analysis further showed that a significant change of intestinal metabolites involved in lipid metabolic pathways were induced along with the microbiota dysbiosis upon exposure to cis-bifenthrin.Detailed investigation showed an altered functional regulation of lipids in the liver after cis-bifenthrin exposure and the accumulation of lipid droplets in hepatocytes.Specifically,a change in deoxycholic acid alters bile acid hepatoenteral circulation,which affects lipid metabolism in the liver and ultimately causes the development of fatty liver disease.Collectively,these findings provide novel insight into the gut microbiota dysbiosis upon pesticide exposure and their potential implication in the development of chronic host diseases related to liver metabolic syndrome.Meng Li Tingting Liu Teng Yang Jiaping Zhu Yunqian Zhou Mengcen Wang Qiangwei Wang 2022Environmental Science and Ecotechnology2022,,3:0
