| 2 | Preclinical evaluation and pilot clinical study of [^(18)F]AlF-labeled FAPI-tracer for PET imaging of cancer associated fibroblasts显示文摘In recent years,fibroblast activation protein(FAP)has emerged as an attractive target for the diagnosis and radiotherapy of cancers using FAP-specific radioligands.Herein,we aimed to design a novel^(18)Flabeled FAP tracer([^(18)F]Al F-P-FAPI)for FAP imaging and evaluated its potential for clinical application.The[^(18)F]Al F-P-FAPI novel tracer was prepared in an automated manner within 42 min with a non-decay corrected radiochemical yield of 32±6%(n=8).Among A549-FAP cells,[^(18)F]Al F-P-FAPI demonstrated specific uptake,rapid internalization,and low cellular efflux.Compared to the patent tracer[^(18)F]FAPI-42,[^(18)F]Al F-P-FAPI exhibited lower levels of cellular efflux in the A549-FAP cells and higher stability in vivo.Micro-PET imaging in the A549-FAP tumor model indicated higher specific tumor uptake of[^(18)F]Al F-P-FAPI(7.0±1.0%ID/g)compared to patent tracers[^(18)F]FAPI-42(3.2±0.6%ID/g)and[68 Ga]Ga-FAPI-04(2.7±0.5%ID/g).Furthermore,in an initial diagnostic application in a patient with nasopharyngeal cancer,[^(18)F]Al F-P-FAPI and[^(18)F]FDG PET/CT showed comparable results for both primary tumors and lymph node metastases.These results suggest that[^(18)F]Al F-P-FAPI can be conveniently prepared,with promising characteristics in the preclinical evaluation.The feasibility of FAP imaging was demonstrated using PET studies. | Kongzhen Hu Junqi Li Lijuan Wang Yong Huang Li Li Shimin Ye Yanjiang Han Shun Huang Hubing Wu Jin Su Ganghua Tang | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 4 |

| 4 | Identification of berbamine dihydrochloride from barberry as an anti-adipogenic agent by high-content imaging assay显示文摘Objective:Lipid droplet(LD)deposition in adipose tissue is a critical factor leading to metabolic dysfunction.Various herbal medicines in traditional Chinese medicine(TCM)are used to treat hyperlipidemia,type 2 diabetes,obesity,and other diseases.The objective of this study was to identify potential anti-adipogenic agents from TCM herbal compounds.Methods:One hundred and twenty compounds were evaluated in terms of their effect on adipocyte differentiation through image-based high content screening.Anti-adipogenic effects of identified hits were further confirmed at various concentrations.In addition,drug-induced liver injury assay was performed with HepG2 cells to test the hepatotoxicity of hit compounds.Results:Berbamine(BBM),a chemical isolated from barberry,and a derivative of BBM,berbamine dihydrochloride(BBMD),reduced LDs formation by more than 50%.Dose-dependent effects were observed and the IC50 values of the two hits,BBM and BBMD,were determined as 1.88 mM and 0.95 mM,respectively.Moreover,BBM induced mild HepG2 cell injury,while its dihydrochloridedBBMD did not exhibit hepatotoxicity within 40 mM.Conclusion:This study demonstrates that BBMD may be a potential therapeutic candidate for disorders associated with elevated LDs accumulation. | Shifeng Wang Qiao Zhang Yuxin Zhang Yanling Zhang Qinghua Wu Shiyou Li Yanjiang Qiao | 2016 | Journal of Traditional Chinese Medical Sciences2016,3,2: | 2 |

| 7 | Chronic hypoperfusion due to intracranial large artery stenosis is not associated with cerebralβ-amyloid deposition and brain atrophy显示文摘Background:Insufficient cerebral perfusion is suggested to play a role in the development of Alzheimer disease(AD).However,there is a lack of direct evidence indicating whether hypoperfusion causes or aggravates AD pathology.We investigated the effect of chronic cerebral hypoperfusion on AD-related pathology in humans.Methods:We enrolled a group of cognitively normal patients(median age:64 years)with unilateral chronic cerebral hypoperfusion.Regions of interest with the most pronounced hypoperfusion changes were chosen in the hypoperfused region and were then mirrored in the contralateral hemisphere to create a control region with normal perfusion.11C-Pittsburgh compound-positron emission tomography standard uptake ratios and brain atrophy indices were calculated from the computed tomography images of each patient.Results:The median age of the 10 participants,consisting of 4 males and 6 females,was 64 years(47-76 years).We found that there were no differences in standard uptake ratios of the cortex(volume of interest[VOI]:P=0.721,region of interest[ROI]:P=0.241)and grey/white ratio(VOI:P=0.333,ROI:P=0.445)and brain atrophy indices(Bicaudate,Bifrontal,Evans,Cella,Cella media,and Ventricular index,P>0.05)between the hypoperfused regions and contralateral normally perfused regions in patients with unilateral chronic cerebral hypoperfusion.Conclusion:Our findings suggest that chronic hypoperfusion due to large vessel stenosis may not directly induce cerebralβ-amyloid deposition and neurodegeneration in humans. | Dongyu Fan Huiyun Li Dongwan Chen Yang Chen Xu Yi Heng Yang Qianqian Shi Fangyang Jiao Yi Tang Qiming Li Fangyang Wang Shunan Wang Rongbing Jin Fan Zeng Yanjiang Wang | 2022 | Chinese Medical Journal2022,,5: | 1 |

| 20 | Effect of high cholesterol diet on spatial learning and memory in a rat model of Alzheimer’s disease显示文摘BACKGROUND: Recently study indicates a potentially important link between cholesterol, Aβ deposit, and clinicopathological manifestation of Alzheimer’s disease (AD). OBJECTIVE: To study the effect of high cholesterol diet on cognitive function and neuronal loss of hippocampal dentate gyrus in AD model rats. DESIGN, TIME AND SETTING: A randomized controlled animal study, which was performed in the Laboratory of Stem Cells, Department of Pathology, Third Military Medical University of Chinese PLA from February 2006 to March 2007. MATERIALS: Twenty healthy, male, Wistar rats, aged 3-4 months and weighing (300 ± 20) g, were selected for this study. Aβ1-40 was provided by Sigma Company, USA. Standard diet and high cholesterol diet mixed with cholesterol (5%), sodium hypocholic acid (1%), lard (10%), and ordinary rat food (84%) were provided by Experimental Animal Center, Institute of Field Surgery, Daping Hospital, Third Military Medical University of Chinese PLA. METHODS: Rats were fed on high cholesterol diet or standard diet for eight successive weeks. Then, rats were randomly divided into cholesterol diet +Aβ, high cholesterol diet + phosphate buffered saline(PBS), standard diet + Aβ, and standard diet + PBS group, with five rats in each group. AD rat models were established by local injection of Aβ1-40 solution (10 μL) into the hippocampal dentate gyrus. Rats in the control group were injected with the same volume of PBS. After injection, rat were fed for two weeks MAIN OUTCOME MEASURES: Neuronal cells in the hippocampal dentate gyrus were detected by Nissl staining; spatial navigation and spatial probe were detected by Morris water maze to reflect learning and memory. RESULTS: Twenty rats were included in the final analysis, without any loss. (1) Neuronal numbers: neuronal loss in the high cholesterol diet + Aβ and standard diet + Aβ groups was significantly higher than in the PBS groups (P < 0.01). In particular, loss of pyramidal cells in the high cholesterol diet + Aβ group was significantly higher than in the standard diet + Aβ group (P < 0.01). (2) Average escape latency: average escape latency was ranked as follows: cholesterol diet + Aβ group > standard diet + Aβ group > high cholesterol diet + PBS group > standard diet + PBS group. Moreover, the number of occasions on which the platform was passed—as well as percentage of swimming distance between two quadrants within two minutes—negatively correlated with average escape latency.
