Large noncoding RNAs(lnc RNAs) are an important class of gene regulators that play essential roles in a variety of physiologic and pathologic processes. Dozens of lnc RNAs are now identified to control ESC self-renewal and differentiation. Research on lnc RNAs may provide novel insights into manipulating the cell fate or reprogramming somatic cells into induced pluripotent stem cells(i PSCs). | 5 | Thermal evolution and applications of aromatic hydrocarbons in highly mature coal-bearing source rocks of the Upper Triassic Xujiahe Formation in the northern Sichuan Basin显示文摘Based on the GC-MS analytical data of aromatic fractions of over forty highly mature coal-bearing source rock samples collected from the Upper Triassic Xujiahe Formation in the northern Sichuan Basin, the thermal evolution of aromatic hydrocarbons during late-mature to over-mature stage(Ro=1.13%–2.85%) was characterized, and aromatic indicators suitable for recognizing the organic source and sedimentary environment of high maturity source rocks were discussed. The results indicated that the concentrations of low carbon-cycle naphthalene as well phenanthrene series reduce gradually with increasing Ro at the highly mature levels. However, some high-cyclic components such as chrysene, benzofluoranthene, and benzo[e]pyrene are relatively enriched, in companying an enhancement of parent aromatic compounds. The variations are attributed to thermal cracking and polymerization reactions due to continuous dehydrogenation under enhanced burial temperature. As thermal maturity rises, MPI1(Methylphenanthrene Index) values display a two-modal varying trend, namely, increasing when Ro is below 1.80% and decreasing above 1.8% Ro. The relationships between Ro and MPI1 are Ro=0.98MPI1+0.37 for Ro<1.80% and Ro=?0.90MPI1+3.02 at Ro>1.8%, being different from the previous research. The amount of dibenzofurans declines sharply at Ro higher than 1.1%, leading to a significant change of relative composition among dibenzothiophenes, dibenzofurans and fluorenes(referred as three-fluorenes series composition). Thus, this parameter appears to be unsuitable for identifying the sedimentary environment of the highly matured source rocks. 4-/1-MDBT(methyldibenzothiophene) ratio could be served as an effective indicator for organic facies, and can distinguish coals from mudstones at over-maturity in this case. | 10 | Urban-rural disparities in mild cognitive impairment and its functional subtypes among community- dwelling older residents in central China显示文摘background Substantial variations in the prevalence of mild cognitive impairment(MCI)and its subtypes have been reported,although mostly in geographically defined developed countries and regions.Less is known about MCI and its subtypes in rural areas of less developed central China.Aims The study aimed to compare the prevalence of MCI and its subtypes in residents aged 65 years or older in urban and rural areas of Hubei Province,China.Methods Participants aged 65 years or older were recruited between 2018 and 2019.Inperson structured interviews and clinical and neuropsychological assessments were performed at city health community centres and township hospitals.results Among 2644 participants without dementia,735 had MCI,resulting in a prevalence of 27.8%for total MCI,20.9%for amnestic MCI(aMCI)and 6.9%for non-amnestic MCI(naMCI).The prevalence of MCI in urban and rural areas was 20.2%and 44.1%,respectively.After adjusting for demographic factors,the prevalence of total MCI,aMCI and naMCI differed significantly between rural and urban areas(adjusted odds ratio(OR)2.10,1.44 and 3.76,respectively).Subgroup analysis revealed an association between rural socioeconomic and lifestyle disadvantage and MCI and its subtypes.Conclusions Our findings suggest that the prevalence of MCI among urban residents in central China is consistent with that in other metropolis areas,such as Shanghai,but the prevalence in rural areas is twice that in urban areas.Prospective studies and dementia prevention in China should focus on rural areas. | Dan Liu Lin Li Lina An Guirong Cheng Cong Chen Mingjun Zou Bo Zhang Xuguang Gan Lang Xu Yangming Ou Qingming Wu Ru Wang Yan Zeng | 2021 | General Psychiatry2021,34,5: | 3 |

| 13 | Development of the novel ACLY inhibitor 326E as a promising treatment for hypercholesterolemia显示文摘Hepatic cholesterol accumulation is an important contributor to hypercholesterolemia,which results in atherosclerosis and cardiovascular disease(CVD).ATP-citrate lyase(ACLY)is a key lipogenic enzyme that converts cytosolic citrate derived from tricarboxylic acid cycle(TCA cycle)to acetyl-CoA in the cytoplasm.Therefore,ACLY represents a link between mitochondria oxidative phosphorylation and cytosolic de novo lipogenesis.In this study,we developed the small molecule 326E with an enedioic acid structural moiety as a novel ACLY inhibitor,and its CoA-conjugated form 326E-CoA inhibited ACLY activity with an IC_(50)=5.31±1.2μmol/L in vitro.326E treatment reduced de novo lipogenesis,and increased cholesterol efflux in vitro and in vivo.326E was rapidly absorbed after oral administration,exhibited a higher blood exposure than that of the approved ACLY inhibitor bempedoic acid(BA)used for hypercholesterolemia.Chronic 326E treatment in hamsters and rhesus monkeys resulted in remarkable improvement of hyperlipidemia.Once daily oral administration of 326E for 24 weeks prevented the occurrence of atherosclerosis in ApoE^(-/-)mice to a greater extent than that of BA treatment.Taken together,our data suggest that inhibition of ACLY by 326E represents a promising strategy for the treatment of hypercholesterolemia. | Zhifu Xie Mei Zhang Qian Song Long Cheng Xinwen Zhang Gaolei Song Xinyu Sun Min Gu Chendong Zhou Yangming Zhang Kexin Zhu Jianpeng Yin Xiaoyan Chen Jingya Li Fajun Nan | 2023 | Acta Pharmaceutica Sinica B2023,13,2: | 1 |

| 15 | Glycosylation and an amino acid insertion in the head of hemagglutinin independently affect the antigenic properties of H5N1 avian influenza viruses显示文摘Antigenic drift forces us to frequently update influenza vaccines; however, the genetic basis for antigenic variation remains largely unknown. In this study, we used clade 7.2 H5 viruses as models to explore the molecular determinants of influenza virus antigenic variation. We generated eight monoclonal antibodies(MAbs) targeted to the hemagglutinin(HA) protein of the index virus A/chicken/Shanxi/2/2006 and found that two representative antigenically drifted clade 7.2 viruses did not react with six of the eight MAbs. The E131 N mutation and insertion of leucine at position 134 in the HA protein of the antigenically drifted strains eliminated the reactivity of the virus with the MAbs. We also found that the amino acid N131 in the H5 HA protein is glycosylated.
