| 1 | FADD and TRADD expression and apoptosis in primary hepatocellular carcinoma显示文摘AIM To investigate the clinical features ofFADD and TRADD expressions in primaryhepatocellular carcinoma(HCC)and todetermine their relationship with hepaticapoptosis.METHODS FADD and TRADD expressions weredetected by immunohistochemistry and hepaticapoptosis were determined by in situ end-labeling(ISEL).RESULTS Ten(25.6%)cases of HCC weredetected to express FADD protein.The positiverate in HCC is lower than that in non-cancerousadjacent liver tissues(62.5%)(P<0.05).Inthose of grade Ⅰ-Ⅱ,8(38.1%)cases wereFADD positive,while only 2/18(11.1%)casesof grade Ⅲ-Ⅳ had detectable FADD protein(P<0.05).No relationship was found betweenFADD expression and other clinical features,such as gender,age,tumor size,differentiationor metastasis.ISEL positive cells can be seen inall cases of HCC.The hepatic apoptosis wasassociated with FADD expression as moreapoptotic cells were detected in those caseswhich had moderately to strongly positiveFADD,as compared with negative or weakpositive FADD cases(P<0.05).No relationshipwas found between FADD expression and hepaticapoptosis in non-cancerous adjacent livertissues.Fifteen of 39(38.5%)cases of HCCwere found positive for TRADD protein,andsimilar positive rate(37.5%)in non-cancerousadjacent liver tissues(P>0.05).The expressionof TRADD is correlated with HCC differentiation, as only 22.2% of moderately to highlydifferentiated HCC showed positive TRADDprotein,while as high as 52.4% of poorlydifferentiated HCC had TRADD(P<0.05),Norelationship was found between TRADDexpression and gender,age,tumor size or gradeor metastasis,although 42.9% of HCC of gradeⅠ/Ⅱ showed positive TRADD which wasslightly higher than that of grade Ⅲ/Ⅳ(33.3%,P>0.05).Hepatic apoptosis was not related toTRADD expression in HCC or non-cancerousadjacent liver tissues.CONCLUSION Loss of FADD expression playsan important role in HCC carcinogenesis,andexpression of TRADD also contributes to HCCdevelopment.The cell apoptosis in HCC isassociated with FADD expression.However,theexpression of TRADD does not correlate wellwith hepatic apoptosis in HCC. | Sun BH Zhao XP Wang BJ Yang DL Hao LJ | 2000 | World Journal of Gastroenterology2000,6,2: | 22 |
| 2 | Cloning and expression of MXR7 gene in human HCC tissue显示文摘AIM To clone and identify the whole cDNA ofMXR7 gene and to find out its expression inhuman HCC,and normal tissues.METHODS The DNA primers were designed andsynthesized according to the whole cDNAsequence of MXR? gene.The cDNA of humanHCC was taken as the template while the cDNA ofMXR7 gene was synthesized by polymerasechain reaction(PCR).Recombinant DNAconforming to reading frame was constructed byconnecting purified PCR product of the cDNA ofMXR? gene with expression vector pGEX-5X-1 offusion protein.The plasmid MXRT/pGEX-5X-1was identified by sequencing.Using 32p labeledMXR? cDNA as probe,MXR7 mRNA expressionwas detected by Northern blot analysis in 12different human normal tissues,7 preoperativelyuntreated non-liver tumor tissues,30preoperatively untreated HCC,theparacancerous liver tissues and 12 normal livertissues samples.RESULTS Restriction enzyme and sequenceanalysis confirmed that the insertion sequence invector pGEX-5X-1 was the same as the cDNAsequence of MXR7 gene.Northern blot analysisshowed no expression of MXR? mRNA in 12 kindsof normal human tissues including liver,7 tumortissues in other sites and 12 normal livertissues,the frequencies of MXR7 mRNA expression in HCC and paracancerous livertissues were 76.6% and 13.3%,respectively.The frequency of MXR7 mRNA expression in HCCwithout elevation of serum AFP and in HCC<5cm was 90%(9/10)and 83.3%(5/6),respectively.CONCLUSION MXR7 mRNA is highly expressedin human HCC,which is specific and occurs at anearly stage of HCC,suggesting MXR7 mRNA canbe a tumor biomarker for HCC.The detection ofMXR7 mRNA expression in the biopsied livertissue is helpful in discovering early subclinicalliver cancer in those with negative serum AFP. | Zhou XP Wang HY Yang GS Chen ZJ Li BA Wu MC | 2000 | World Journal of Gastroenterology2000,6,1: | 23 |
| 3 | Cultivation of human liver cell lines with microcarriers acting as biological materials of bioartificial liver显示文摘AIM To improve the Cultivation efficiency and yield of human liver cell line CL--l.METHODS High--density cultivation of CL--l onmicrocarriers was carried out with periodicobservation of their growth and proliferation.The specific functions of human liver cell werealso determined.RESULTS Cells of CL-1 cell line grew well onmicrocarrier Cytodex--3 and on the 7th day thepeak was reached. The amount of CLI cells was2.13 x 10’ and the total amount of albuminsynthesis reached 71 .23 ig, urea synthesis23 .32 mg and diazepam transformation 619 .7 ugrespectively. The yield of CL-l on microcarrierswas 49.3 times that of conventional cultivation.The amounts of albumin synthesis, ureasynthesis and diazepam transformation were39.8 times, 41 .6 times and 33.3 times those ofconventional Cultivation, respectively.CONCLUSION The human liver cell line CL--1 canbe Cultivated to a high density with Cytodex--3and has better biological functions.
