1Survey of nutrients and quality assessment of crab paste by 1~H NMR spectroscopy and multivariate data analysis显示文摘Crab paste is a traditional and popular food for people in the coastal area of China.Visual inspection is currently the only method of quality assessment.We developed a new analytical method of a comprehensive survey of nutrients and quality assessment for crab paste based on a combination of 1 H NMR spectroscopy with multivariate data analysis.The aqueous extract of crab paste was dominated by amino acids,sugars,carboxylic acids,nucleotides and amines(including 19 first-time-reported compounds such as choline,uracil and guanosine).Two grades of crab paste had significant compositional differences in terms of amino acids,lactate,N-acetylglutamate,choline,dimethylamine,uridine,1-methylnicotinamide and 2-pyridienmethanol.These results provided important information on the grade-dependence of crab-paste composition,and demonstrated that NMR-MDA was effective not only for the comprehensive survey of nutrients,but also for quality assessment of crab paste.YE YangFang ZHANG LiMin TANG HuiRu YAN XiaoJun 2012Chinese Science Bulletin2012,57,25:9

10Dynamic metabolite alterations of Portunus trituberculatus during larval development显示文摘A mass mortality often occurs from molting to the megalopa stage during the larval development of the swimming crab Portunus trituberculatus. Larvae with insuffcient nutrient accumulation during the zoeal stages are probably unable to develop into juvenile swimming crabs. However, the nutritional information such as the primary metabolites is scarce for P. trituberculatus larvae. The aim of this work is to obtain an insight into the metabolite traits of P. trituberculatus at early developmental stages. 1 H nuclear magnetic resonance spectroscopy coupled with multivariate data analysis was used to determine how the metabolite pro?les shift during larval development in P. trituberculatus. Our results show that the trend of total metabolites exhibited a rise from zoea 1 to zoea 3, followed by a drop from zoea 4 to megalopa and recovery during the ?rst juvenile stage. A large-scale depletion of total metabolites in the zoea 4 and megalopa stages suggests a deep depression of metabolic activity, which may be linked to the mass mortality from molting to the megalopa stage. 11NMR-based metabolomic responses of freshwater gastropod Bellamya aeruginosa to MC-producing and non MC-producing Microcystis aeruginosa显示文摘Molluscan metabolomic analysis is essential for the understanding of the regulatory mechanism of aquatic invertebrate in response to hepatotoxic microcystins(MCs)stress.To understand the system responses of the gastropod to MC exposure,metabolomic alterations caused by two strains(MC-producing and non MC-producing)of Microcystis aeruginosa were characterized indiff erent biological matrices(hepatopancreas and muscle)of Bellamya aeruginosa(Gastropoda)using 1 H nuclear magnetic resonance(NMR)spectroscopy combined with MCs detections after exposure for 1,7,and 14 d.Although ELISA analysis showed that no MCs was detected in both tissues after non MC-producing M.aeruginosa exposure,MCs concentrations were increasing in the hepatopancreas(from 1.29±0.48μg/g to 3.17±0.11μg/g)and foot muscle(from 0.07±0.02μg/g to 0.21±0.08μg/g)after 14-d exposure of MC-producing M.aeruginosa.Meanwhile,we observed that MC induced signifi cant increase in creatine,a variety of amino acids(leucine,isoleucine,valine,threonine,alanine,methionine,glutamate,aspartate,and lysine),carboxylic acids(lactate,acetate,and D-3-hydroxybutyrate),and choline and its derivatives(phosphocholine and glycerophosphocholine)but decreased the energy substance(lipids,glucose,and glycogen)in the hepatopancreas.However,no signifi cant metabolite diff erences were observed in the muscle between MC-producing and non MC-producing cyanobacteria treated groups.These results suggest that MC exposure may cause hepatic energy expenditure accompanied with various metabolic disorders that involve lipid metabolism,protein catabolism,osmoregulation,glycolysis,glycogenolysis,and tricarboxylic acid(TCA)cycle.Moreover,metabolic perturbation was aggravated as the level of accumulated MCs raised over time in the MC-producing cyanobacteria treatment.These fi ndings indicated that MCs accumulation might lead to oxidative-stress-mediated damage of mitochondria functions.Wen YANG Yangfang YE Kaihong LU Zhongming ZHENG Jinyong ZHU 2022Journal of Oceanology and Limnology2022,40,1:0
12IgE Reactivity of Potential Allergens from the Swimming Crab Portunus trituberculatus and the Research of ELISA Reagent for Detecting Crab Food Anaphylaxis显示文摘Crustacean is one of the major allergic foods.It is of great significance to identify more crab allergens and research the detection methods for crab food anaphylaxis.In this study,IgE reactivity to three recombinant proteins from Portunus trituberculatus,including tropomyosin(rPtTM),myosin light chain(rPtMLC),and pancreatic lipase(rPtPL),were detected by enzyme-linked im-munosorbent assay(ELISA).The expressions of tropomyosin(TM)in various tissues of P.trituberculatus were detected by Western blot(WB).Furthermore,microplates were coated with rPtTM and the ELISA conditions were optimized.The cut-off value was deter-mined by the receiver operating characteristic(ROC)curve.Among 51 crab-allergic sera,21(41.2%)showed positive IgE to rPtTM.Other 70 crab-allergic sera more frequently recognized rPtPL(9/70;12.9%),followed by rPtMLC(1/70;1.4%).WB results showed that TM was mainly expressed in the muscle,followed by the heart and a small amount in gills and the testis.The optimal results showed that the coating condition of rPtTM was 50 ng per well with coating for 3 h at 37℃.The optimal blocking condition was 1.2%BSA with blocking for 3 h at 37℃,and the optimal dilution of the second antibody was 1:1500.The ROC curve showed that the ELISA reagent had high sensitivity(83.52%)and specificity(98.00%)when the cut-off value was 0.45.All results indicated that tropomyosin is the major allergen of P.trituberculatus,and myosin light chain and pancreatic lipase are the potential allergens.Addi-tionally,the ELISA reagent developed with the rPtTM was feasible for laboratory detection of crab anaphylaxis.QIU Xueni ZHOU Qiongyan ZHU Xiaoxia LIN Wei MU Changkao LI Ronghua YE Yangfang SONG Weiwei SHI Ce LIU Lei WANG Huan WANG Chunlin XU Suling 2022Journal of Ocean University of China2022,21,2:0
13Phospholipid Compositions in Portunus trituberculatus Larvae at Different Developmental Stages显示文摘Phospholipids are used to improve the growth and survival of Portunus trituberculatus,a widely cultured crab species in China.However,only total phospholipids or several classes are applied in crab diets.In this study,we employed a targeted lipidomic method to investigate the comprehensive phospholipid composition in P.trituberculatus larvae and reveal the changing phospholipid profile over the larval development.Results showed that P.trituberculatus larvae contain 112 phospholipid species belonging to 10 phospholipid classes,in which phosphatidylcholine(PC)and phosphatidylethanolamine(PE)species are the most abundant,and PC,PE,phosphatidic acid,and phosphatidylserine(PS)are with high concentrations.The levels of all phospholipids significantly changed with larval development,which was highlighted by the downward parabolic changes in PE,phosphatidylglycerol,phosphatidylino-sitol,PS,lysophosphatidic acid,and sphingomyelin levels.In addition,nearly all phospholipid species were depleted at the M stage,which probably contributed to the mass mortality of crab larvae.These findings on the composition and alterations of phospholipids in P.trituberculatus larvae provide novel perspectives for the targeted supplementation of phospholipids in crab diets.Our work also highlights the use of targeted UHPLC-MS lipidomics in understanding the changes of phospholipids during crab development.LU Zhibin SHI Ce LIU Lei MU Changkao YE Yangfang WANG Chunlin 2022Journal of Ocean University of China2022,21,1:0
14Music stimulus has a positive eff ect on survival and development of the larvae in swimming crab Portunus trituberculatus显示文摘The sound stimulus has a signifi cant impact on marine animals.However,many studies have focused on the negative impacts of sound on marine vertebrates.The positive impacts of sound on marine animals,especially on marine invertebrates,were rarely studied.In the present study,we evaluated the eff ects of music on the survival and development of an economically important marine invertebrate,swimming crab Portunus trituberculatus.Three pieces of classical music and a control group,i.e.,Eine Kleine Nachtmusik by Mozart(EK),Bach-Violin Concerto No.1(BV),Anonymous Romanza by Anonymous(AR),and environmental sound(EN)were used.The sound pressure received were 163-dB root mean square(RMS)re 1μPa for EK,166-dB RMS re 1μPa for BV,165-dB RMS re 1μPa for AR,44-dB RMS re 1μPa for EN,respectively.The larvae were subject to 3-h classical music transmission every day during the entire larval development period.The result showed that BV and AR signifi cantly elevated the cumulative survival rate of the larva from 3.42%±1.77%(EN)to 10.65%±1.69%(BV)and 7.12%±1.37%(AR)respectively(P<0.05).Though music did not aff ect the cumulative development time of larvae from ZoeaⅠto ZoeaⅣ,it signifi cantly increased the molting rate of megalopa from 24.00%±11.78%(EN)to 44.00%±13.06%(AR)and 46.67%±10.50%(BV)(P<0.05).Musical exposure during critical growth intervals may contribute to the survival and development of larval crustaceans.From an application point of view,music intervention is much economically feasible.The observed music eff ects imply that music could further enhance growth,welfare,and production.Zhiming REN Jiechi WANG Chunlin WANG Changkao MU Yangfang YE Ce SHI 2022Journal of Oceanology and Limnology2022,40,3:0
15Using iTRAQ-Based Quantitative Proteomics Analysis to Identify Differentially Expressed Proteins Related to Larval Development of Portunus trituberculatus显示文摘The swimming crab Portunus trituberculatus is an economically important marine crustacean species.Here isobaric tags for relative and absolute quantitation(iTRAQ)analysis were used to identify proteins that are differentially expressed during larval development of P.trituberculatus to elucidate the underlying mechanisms.In comparison with the first zoea stage(Z1),3980 proteins were identified from 32789 peptides,which were matched with 115497 spectrums.A total of 241 proteins were screened with significantly differential expressions in all development stages.These 241 proteins are involved in various biological processes,such as cytoskeleton organization,protein synthesis,energy production and substance metabolism,physiological activities,and transport.Cluster analyses of the 241 differentially expressed proteins led to the generation of four protein clusters based on the overall similarity in protein expression patterns.Exactly 54,70,36,and 45 proteins clustered in profiles 10(0,0,0,−1,0,0),15(0,1,0,1,0,1),18(0,1,2,2,1,0),and 19(0,1,2,3,4,5),respectively.Muscle development and exoskeleton renewal were important processes throughout the development stages.In addition,protein synthesis,degradation,and digestion actively occurred,especially at the Z4 stage.These results provide novel insights into the mechanisms underlying larval development of swimming crab and can assist in larval rearing.CHEN Xiayue MU Changkao LI Ronghua YE Yangfang SONG Weiwei SHI Ce LIU Lei WANG Huan WANG Chunlin 2021Journal of Ocean University of China2021,20,4:0
16Postmortem Metabolite Profile Changes of Mud Crab(Scylla paramamosain)Under Different Storage Conditions显示文摘Mud crab(Scylla paramamosain)is one of the most consumed crabs in China.Generally,it is believed that dead mud crabs are not edible.Herein,we investigated the postmortem metabolite profile changes of the muscle and hepatopancreas of mud crab associated with storage method using nuclear magnetic resonance-based metabolomics coupled with multivariate data analysis.Our results showed that postmortem changes of trimethylamine,organic acids(lactate,formate,and succinate),as well as ATP and its breakdown products are closely affected by the storage method.These metabolites can be used as indicators for monitoring loss of crab freshness.Furthermore,dead mud crabs preserved an edible quality at least for 4 h at room temperature,12h on ice,and 16 d at−20℃,strongly suggesting that dead crab consumption quality should be determined by freshness.They shouldn’t be discarded without considering the effects of different storage conditions on their freshness.ZHANG Limin GUO Liping MU Changkao YE Yangfang WANG Chunlin 2021Journal of Ocean University of China2021,20,3:0
