| 1 | Establishment of cell clones with different metastatic potential from the metastatic hepatocellular carcinoma cell line MHCC97显示文摘ALM To establish clone cells with different metastatic potential for the study of metastasis-related mechanisms. METHODS Cloning procedure was performed on parental hepatocellular carcinoma (HCC) cell line MHCC97. andbiological characteristics of the target clones selected by in vivo screening were studied.``RESULTS Two clones with high MHCC97-H and IowMHCC9--L1 metastatic potential were isolated from theparent cell line. Compared with MHCC97-L. MHCC97-H hadsmaller cell size average cell diameter 43 um vs 50 μmand faster in vitro and in vivo growth rate tumor celldoubling time was 34.2 h vs 60.0 h. The main ranges ofchromosomes were 5.5 58 in MHCC97-H and 57 62 inMHCC97-L. Boyden chamber in vitro invasion assay demonstrated that the number of penetrating cells through the artificial basement membrane was 137.5 - 11 .0) cellsfield for MHC_C99--H vs 17.7 - 6.3) field for MHCC97-L.The proportions of cells in GO Gl phase. | 5 | A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. | 9 | Mesozoic tectono-magmatic activities in South China:Retrospect and prospect显示文摘The South China Block was formed through the collisional orogeny between the Cathaysia Block and the Yangtze Block in the Early Neoproterozoic.The northern,western and southern sides of the South China Block were affected by disappearance of the Paleo-Tethyan Ocean during the Paleozoic.The southern and northern sides of the South China Block were respectively collided with the Indo-China Block and North China Block in the latest Paleozoic to form the basic framework of the Eastern China.The Eastern China has been affected by the westward subduction of the Pacific Plate since the Mesozoic.Therefore,the South China Block was influenced by the three major tectonic systems,leading to a superposed compound tectonics.The comparative study of the Mesozoic geology between the South China Block and its surrounding areas suggests that although the Mesozoic South China Block was adjacent to the subduction zone of the western Pacific,no juvenile arc-type crust has been found in the eastern margin.The main Mesozoic geology in South China is characterized by reworking of ancient continental margins to intracontinental tectonics,lacking oceanic arc basalts and continental arc andesites.Therefore,a key to understanding of the Mesozoic geology in South China is to determine the temporal-spatial distribution and tectonic evolution of Mesozoic magmatic rocks in this region.This paper presents a review on the tectonic evolution of the South China Block through summarizing the magmatic rock records from the compressional to extensional tectonic process with the transition at the three juncture zones and using the deformation and geophysic data from the deep part of the South China continental lithosphere.Our attempt is to promote the study of South China’s geology and to make it as a typical target for development of plate tectonic theory. | MAO JianRen LI ZiLong YE HaiMin | 2014 | Science China Earth Sciences2014,57,12: | 64 |
| 10 | Changes of NF-kB,p53,Bcl-2 and caspase in apoptosis induced by JTE-522 in human gastric adenocarcinoma cell line AGS cells:role of reactive oxygen species显示文摘AIM: To identify whether JTE-522 can induce apoptosis inAGS cells and ROS also involved in the process, and toinvestigate the changes in NF-kB, p53, bcl-2 and caspase in the apoptosis process.METHODS: Cell culture, MTT, Electromicroscopy, agarosegel electrophoresis, lucigenin, Western blot andelectrophoretic mobility shift assay (EMSA) analysis wereemployed to investigate the effect of JTE-522 on cellproliferation and apoptosis in AGS cells and relatedmolecular mechanisms.RESULTS: JTE-522 inhibited the growth of AGS cells andinduced the apoptosis. Lucigenin assay showed the generationof ROS in cells under incubation with JTE-522. The inc eareasedROS generation might contribute to the induction of AGS cellsto apoptosis. EMSA and Westem blot revealed that NF-kBactivity was almost completely inhibited by preventing thedegradation of IkBα. Additionally, by using Western blot weconfirmed that the level of bcl-2 was decreased, whereas p53showed a great increase following JTE-522 treatment. | 11 | Autophagy and multidrug resistance in cancer显示文摘Multidrug resistance(MDR) occurs frequently after long-term chemotherapy, resulting in refractory cancer and tumor recurrence.Therefore, combatting MDR is an important issue. Autophagy, a self-degradative system, universally arises during the treatment of sensitive and MDR cancer. Autophagy can be a double-edged sword for MDR tumors: it participates in the development of MDR and protects cancer cells from chemotherapeutics but can also kill MDR cancer cells in which apoptosis pathways are inactive. Autophagy induced by anticancer drugs could also activate apoptosis signaling pathways in MDR cells, facilitating MDR reversal. Therefore, research on the regulation of autophagy to combat MDR is expanding and is becoming increasingly important. | 18 | L-forms of H. pylori显示文摘AIM: To study the occurrence of L-forms of H. pyloriinfection in patients with peptic ulcers and its association with possible changes of cellular immune function in the patients.METHODS: Endoscopic biopsy specimens of gastric antrum and gastric corpus were taken from 228 patients with peptic ulcers and inoculated into Skirrow selective medium for H.pylorivegetative forms and special medium for H. pylori Lforms, followed by bacterial isolation and identification. And peripheral venous blood of the patients was taken to detect the percentage of CD3+, CD4+ and CD8+ with biotin-streptavidin (BSA) and the level of IL-2, IL-6 and IL-8 with ElISA.RESULTS: (1) The detection rates of H. pyloriL-forms and vegetative forms in the patients were 50.88 % (116/228)and 64.91 % (148/228) respectively, and the co-infection rate of H. pyloriL-forms and vegetative forms was 78.38 % (116/148). To be more exact, the detection rates of H. pylori L-forms in male and female patients were 57.04 % (77/135) and 41.94 % (39/93) respectively, and statistics found significant difference between them (P<0.05). Furthermore, the detection rates of H. pyloriL-forms in patients aged 14 years-, 30 years-, 40 years- and 50 years- were 31.91%(15/47), 42.86 % (24/56), 56.94 % (41/72) and 67.92 %(36/53) respectively, and there was significant difference between them (P<0.011). (2) The percentages of CD3+, CD4+,CD8+, the ratio of CD4+/CD8+, and the level of IL-2, IL-6,IL-8 in H. pylori-positive patients were (52.59±5.44) %,(35.51±5.74) %, (27.77±8.64) %, (1.56±0.51), (2.66±0.47)mg/L, (108.62±5.85) ng/L and (115.79±7.18) ng/Lrespectively, compared with those in H. pylori-negative patients, the percentages of CD3+, CD4+ and the ratio of CD4+/CD8+ decreased, but the level of IL-2, IL-6 increased, and the difference was significant (P<0.001-P<0.011).Moreover, the percentages of CD3+, CD4+, CD8+, the ratio of CD4+/CD8+, and the level of IL-2, IL-6, IL-8 in the patients with mixed infection of bothH. pyloriL-forms and vegetative forms were (51.69±5.28) %, (34.75±5.89) %, (27.15±7.45) %, (1.48±0.47), (2.16±0.38) mg/L, (119.45±5.44) ng/L and (123.64±6.24) ng/L respectively, compared with those in patients with simple infection of H. pylorivegetative forms,the percentage of CD4+, the ratio of CD4+/CD8+ and the level of IL-2 increased, but the level of IL-6 and IL-8decreased, statistical difference was found between them (P<0.001-P<0.05).CONCLUSION: L-forms variation often occurs in patients with peptic ulcers who are infected byH. pylori, which is commonly found in male patients and related to ages. | 19 | SHRIMP zircon U-Pb dating for volcanic rocks of the Dasi Formation in southeast Hubei Province,middle-lower reaches of the Yangtze River and its implications显示文摘The Jinniu Basin in southeast Hubei,located at the westernmost part of middle-lower val-ley of the Yangtze River,is one of the important vol-canic basins in East China. Volcanic rocks in the Jin-niu Basin are distributed mainly in the Majiashan Formation,the Lingxiang Formation and the Dasi Formation,consisting of rhyolite,basalt and basaltic andesite,(trachy)-basalt and basaltic trachy-andesite and (trachy)-andesite and (trachy)-dacite and rhyolite respectively,in which the Dasi volcanism is volumet-rically dominant and widespread. The Dasi volcanic rocks were selected for SHRIMP zircon U-Pb dating to confirm the timing of volcanism. The results indi-cate that there exist a large amount of magmatic zir-cons characterized by high U and Th contents in the volcanic rocks. The concordia ages for 13 points are 128 ± 1Ma (MSWD = 3.0). On account of the shape of zircons and Th/U ratios,this age is considered to represent the crystallization time of the Dasi volcan-ism. The volcanic rocks in the Dasi,Majiashan and Lingxiang Formations share similar trace elementand REE partition patterns as well as Sr-Nd isotopic compositions. In combination with the regional geol-ogy,it is proposed that the southeast Hubei volcanic rocks were formed mainly during the Early Creta-ceous,just like other volcanic basins in middle-lower Yangtze valley. | 20 | Current Trends of the Prevalence of Childhood Asthma in Three Chinese Cities: A Multicenter Epidemiological Survey显示文摘Objective To analyze the prevalence of asthma and asthma related symptoms among children aged 0-14 years in three Chinese cities and to obtain a crude estimation of the trend of childhood asthma prevalence in China. Methods A cross-sectional, population-based survey of prevalence of asthma was conducted in children aged from 0 to 14 years in 3 major cities of China (Beijing, Chongqing, and Guangzhou) with different geographic locations. All the subjects were randomly selected by a multi-stage sampling method. Three to five schools and kindergartens in 2 urban districts in each city were randomly selected for the survey, and a validated questionnaire that included the core questions of the International Study of Asthma and Allergies in Childhood, Phase Ⅲ questionnaire and several additional questions were used. All questionnaires were completed by parents or guardians of the selected children. Children whose parents responded affirmatively to the question' Has your child ever been diagnosed as asthma by a doctor' were recognized as victims of asthma. Results The prevalence of asthma in Beijing, Chongqing, and Guangzhou was 3.15%, 7.45%, and 2.09%, respectively. These values were significantly higher than those obtained 10 years ago in the national epidemiological survey in 2000 which used the same method of investigation and the same diagnotic criteria (χ2 =3.938, P=0.047; χ2 =73.506, P≤0.001; χ2 =11.956, P=0.001, in each city). Of the asthmatic children 57.21%, 69.91%, and 60.00% had their first attack before the age of 3 in Beijing, Chongqing, and Guangzhou, respectively. Wheezing was the primary clinical manifestation for all asthmatic children , followed by persistent cough and repeated respiratory infections. Both the prevalence of asthma and asthma-related symptoms were statistically higher in males than in females.
