| 1 | Brain cell apoptosis and enhancement of nervous excitability in pregnant rats with high plasma levels of homocysteine显示文摘Hyperhomocysteinemia is an important risk factor for preeclampsia-eclampsia. This study established a pregnant rat model of hyperhomocysteinemia, in which blood plasma homocysteine concentrations were twice or three times greater than that of normal pregnant rats. TUNEL revealed an increase in the number of apoptotic cells in the frontal cortex of pregnant rats with hyperhomocysteinemia. In addition,immunohistochemical staining detected activated nuclear factor-κB-positve cells in the frontal cortex.Reverse transcription-PCR detected that mRNA expression of the anti-apoptotic gene bcl-2 diminished in the frontal cortex. In situ hybridization and western blotting revealed that N-methyl-D-aspartate receptor 1 mRNA and protein expression was upregulated in the frontal cortex and hippocampus. | 3 | Abnormal expression of VEGF and its gene transcription status as diagnostic indicators in patients with non-small cell lung cancer显示文摘Objective Angiogenesis is known to be essential for the survival,growth,invasion,and metastasis of lung cancer cells. Vascular endothelial growth factor(VEGF) is an important factor regulating angiogenesis of non-small cell lung cancer(NSCLC); however,its pathologic features and significance are unclear. In this study,the tissue VEGF expression levels and its gene transcriptional status,as well as circulating VEGF levels,were investigated in patients with lung disease. Methods VEGF protein and m RNA expression levels in 38 lung tissue samples were analyzed by immunohistochemistry and reverse transcription-polymerase chain reaction(RT-PCR),respectively. Circulating VEGF levels were detected quantitatively by an enzyme linked immuno-sorbent assay. Results The level of VEGF expression was significantly higher in lung cancer tissue than in the corresponding paracancerous or non-cancerous tissues. The average level of VEGF-positive staining was 76% in tissue samples from NSCLC patients; the levels were 89% in tissue samples from stage III patients and 92% in stage IV patients. High VEGF expression was also evident in cases with lymph node metastasis(84%),distant metastasis(90%),and lower differentiation degree(89%). VEGF m RNA in cancerous tissues was represented predominantly by the VEGF121 and VEGF165 isoforms. Circulating VEGF levels were significantly higher in NSCLC patients [(840 ± 324) pg/m L] than in patients with benign lung diseases [(308 ± 96) pg/m L] or in healthy individuals serving as controls [(252 ± 108) pg/m L]. | 10 | Clinicopathological features of hypoxia-inducible factor-1α and vascular endothelial growth factor expression in patients with lung cancer显示文摘Objective The aim of the study was to investigate the clinicopathological characteristics of hypoxiainducible factor-1α(HIF-1α) and vascular endothelial growth factor(VEGF) expression in patients with lung cancer.Methods Cancerous and noncancerous tissues were collected post-operation from 115 patients with lung cancers by the self-control method. Total RNA was extracted from the lung tissues. The status of tissue HIF-1α expression and intercellular distribution was observed by immunochemistry using a tissue microarray. The expression levels of circulating HIF-1α and VEGF were detected by enzyme-linked immunosorbent assay(ELISA).Results The expression of serum HIF-1α [(138.3 ± 28.8) μg/L] in the group of patients with lung cancer was significantly higher(P < 0.01) than that in the group of patients with pneumonia [(58.8 ± 14.5) μg/L] and the control group of patients ((24.1 ± 3.3) μg/L)There was a strong positive correlation of serum HIF-1α levels(r = 0.937, P < 0.01) with serum VEGF levels. The specific concentration of total RNA [(1.52 ± 1.14) μg/mg wet lung tissues] in the cancerous tissues was significantly higher(t = 8.494, P < 0.001) than that in the noncancerous tissues ((0.58 ± 0.33) μg/mg)The clinicopathological features of HIF-1α expression in lung cancer tissues revealed a significant relationship between positive HIF-1α expression and patient sex(χ~2 = 4.494, P = 0.034), tumor size(χ~2 = 4.679, P = 0.031), differentiation degree(χ~2= 8.846, P = 0.012), and presence of lymphatic node metastasis(χ~2= 6.604, P = 0.037).Conclusion Abnormal HIF-1α expression in lung cancer is closely related with nucleic acid metabolism and angiogenesis, and it may be helpful in the diagnosis and identification of lung cancer. | Xuli Yang Li Wang Wenli Sai Yin Cai Juanjuan Gu Xin Chen Dengfu Yao | 2016 | Oncology and Translational Medicine2016,2,6: | 1 |

| 13 | Soil C, N and P stocks and stoichiometry under different vegetation on the surface of the Leshan Giant Buddha显示文摘The accumulation of soil organic matter and nutrients is an important pathway in effectivelyunderstanding the mechanisms of plant settlement and rock weathering, while the characteristics ofsoil organic carbon (C), nitrogen (N) and phosphorus (P) under different vegetation remain unclear.In this study, the stocks and stoichiometry of soil organic C, N and P were determined in differentpositions and types of vegetation on the surface of the Leshan Giant Buddha. We found that the totalstocks of soil organic C, N and P were 1689.77, 134.6 and 29.48 kg, respectively, for the Buddha.The stocks of soil organic C, N and P under vascular plants were higher than those under othervegetation, with highest values observed under herb. Higher stocks per unit area (m2) of soil organicC, N and P were found on the left and right arms, shoulders, and two platforms. | 16 | Differential bone metabolism and protein expression in mice fed a high-fat diet versus Daurian ground squirrels following natural pre-hibernation fattening显示文摘This study compared the effects on bone metabolism and morphology of pathological obesity induced by excessive fat intake in a non-hibernator(mice)versus healthy obesity due to pre-hibernation fattening in a hibernator(ground squirrels).Kunming mice were fed a high-fat diet to provide a model of pathological obesity(OB group).Daurian ground squirrels fattened naturally in their pre-hibernation season(PRE group)were used as a healthy obesity model.Micro-computed tomography(micro-CT)and three-point bending tests were used to determine the microstructure and mechanical properties of bone.Western blots were used to analyze protein expression levels related to bone metabolism(Runt-related transcription factor 2(RunX2),osteocalcin(OCN),alkaline phosphatase(ALP),osteoprotegerin(OPG),receptor activator of nuclear factor-κB ligand(RANKL),cathepsin K,matrix metallopeptidase 9(MMP9),patched protein homolog 1(Ptch1),phosphorylatedβ-catenin(P-β-catenin),and glycogen synthase kinase-3β(GSK-3β)).Compared with controls,there was no obvious bone loss in the OB mice,and the stiffness of the femur was increased significantly.Compared with summer active squirrels,bone formation was enhanced but the mechanical properties did not change in the PRE group squirrels.In OB mice,western blots showed significantly increased expression levels of all proteins except RunX2,OPG,and Ptch1.PRE ground squirrels showed significantly increased expression of most proteins except OCN and Ptch1,which decreased significantly,and P-β-catenin and OPG,which did not change.In conclusion,for non-hibernating mice,moderate obesity had a certain protective effect on bones,demonstrating two-way regulation,increasing both bone loss and bone formation.For pre-hibernating ground squirrels,the healthy obesity acquired before hibernation had a positive effect on the microstructure of bones,and also enhanced the expression levels of proteins related to bone formation,bone resorption,and Wnt signaling. | Xuli GAO Shenyang SHEN Qiaohua NIU Weilan MIAO Yuting HAN Ziwei HAO Ning AN Yingyu YANG Yu ZHANG Han ZHANG Kenneth BSTOREY Hui CHANG | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,12: | 0 |
| 17 | Identification of potential anti-pneumonia pharmacological components of Glycyrrhizae Radix et Rhizoma after the treatment with Gan An He Ji oral liquid显示文摘Glycyrrhizae Radix et Rhizoma,a traditional Chinese medicine also known as Gan Cao(GC),is frequently included in clinical prescriptions for the treatment of pneumonia.However,the pharmacological components of GC for pneumonia treatment are rarely explored.Gan An He Ji oral liquid(GAHJ)has a simple composition and contains GC liquid extracts and paregoric,and has been used clinically for many years.Therefore,GAHJ was selected as a compound preparation for the study of GC in the treatment of pneumonia.We conducted an in vivo study of patients with pneumonia undergoing GAHJ treatments for three days.Using the intelligent mass spectrometry data-processing technologies to analyze the metabolism of GC in vivo,we obtained 168 related components of GC in humans,consisting of 24 prototype components and 144 metabolites,with 135 compounds screened in plasma and 82 in urine.After analysis of the metabolic transformation relationship and relative exposure,six components(liquiritin,liquiritigenin,glycyrrhizin,glycyrrhetinic acid,daidzin,and formononetin)were selected as potential effective components.The experimental results based on two animal pneumonia models and the inflammatory cell model showed that the mixture of these six components was effective in the treatment of pneumonia and lung injury and could effectively downregulate the level of inducible nitric oxide synthase(iNOS).Interestingly,glycyrrhetinic acid exhibited the strongest inhibition on iNOS and the highest exposure in vivo.The following molecular dynamic simulations indicated a strong bond between glycyrrhetinic acid and iNOS.Thus,the current study provides a pharmaceutical basis for GC and reveals the possible corresponding mechanisms in pneumonia treatment. | Xiaojuan Jiang Yihua Lin Yunlong Wu Caixia Yuan Xuli Lang Jiayun Chen Chunyan Zhu Xinyi Yang Yu Huang Hao Wang Caisheng Wu | 2022 | Journal of Pharmaceutical Analysis2022,12,6: | 0 |
| 18 | Programmable dual responsive system reconstructing nerve interaction with small-diameter tissue-engineered vascular grafts and inhibiting intimal hyperplasia in diabetes显示文摘Small-diameter tissue-engineered vascular grafts(sdTEVGs)with hyperglycemia resistance have not been constructed.The intimal hyperplasia caused by hyperglycemia remains problem to hinder the patency of sdTEVGs.Here,inspired by bionic regulation of nerve on vascular,we found the released neural exosomes could inhibit the abnormal phenotype transformation of vascular smooth muscle cells(VSMCs).The transformation was a prime culprit causing the intimal hyperplasia of sdTEVGs.To address this concern,sdTEVGs were modified with an on-demand programmable dual-responsive system of ultrathin hydrogels.An external primary Reactive Oxygen Species(ROS)-responsive Netrin-1 system was initially triggered by local inflammation to induce nerve remolding of the sdTEVGs overcoming the difficulty of nerve regeneration under hyperglycemia.Then,the internal secondary ATP-responsive DENND1A(guanine nucleotide exchange factor)system was turned on by the neurotransmitter ATP from the immigrated nerve fibers to stimulate effective release of neural exosomes.The results showed nerve fibers grow into the sdTEVGs in diabetic rats 30 days after transplantation.At day 90,the abnormal VSMCs phenotype was not detected in the sdTEVGs,which maintained long-time patency without intima hyperplasia.Our study provides new insights to construct vascular grafts resisting hyperglycemia damage. | Yanzhao Li Yeqin Wang Fangchao Xue Xuli Feng Zhaojing Ba Junjie Chen Zhenhua Zhou Yanhong Wang Ge Guan Guanyuan Yang Ziwei Xi Hao Tian Yong Liu Ju Tan Gang Li Xiewan Chen Mingcan Yang Wen Chen Chuhong Zhu Wen Zeng | 2022 | Bioactive Materials2022,7,1: | 0 |
| 19 | Analysis of Biological Activity and Immunogenicity of Extracellular Products from Streptococcus iniae显示文摘[Objectives] This study aimed to analyze the biological activity and immunogenicity of extracellular products from Streptococcus iniae. [Methods] S.iniae was incubated with brain heart infusion agar medium( BHIA + 4% calf serum) for 60 h. The bacterial liquid was rinsed with PBS,centrifuged,and filtered through microporous filtering film to collect extracellular products( ECPs). [Results]Extracellular proteinase( ECPase) of S. iniae exhibited amylase,protease,lecitinase,gelatinase,lipase activities and hemolytic activity but had no urease activity. EDTA,DTT and PMSF could reduce ECPase activity to 72. 4%,77. 6%and 72. 4%,respectively. Cu^(2+),Ca^(2+),K+and Mg^(2+)exhibited an inhibitory effect on ECPase activity,whereas Fe^(3+),Co^(2+)and Mn^(2+)could activate ECPase activity. ECPs had good heat stability and exhibited relatively high activities under alkaline conditions. The optimal temperature for ECPs was 55 ℃. Two-dimensional electrophoresis was performed to analyze the main protein of ECPs. The results indicated that there are 12 main bands of ECPs,and the molecular weights mainly ranged between 28-68 kD a. About 120 protein spots were detected,and the molecular weights mainly ranged between 26-95 kD a. The mouse anti-S. iniae was used for Western-blot analysis of ECPs,and the results showed that there were four proteins,with molecular weights of 26,37,95,and 97 kD a,respectively. The pathogenicity assay indicated that ECPs of S. iniae were highly pathogenic to tilapia. The mortality rate of tilapia was enhanced as the concentration of ECPs increased.
