| 1 | Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. | 6 | Expert consensus on the evaluation and diagnosis of combat injuries of the Chinese People's Liberation Army显示文摘The accurate assessment and diagnosis of combat injuries are the basis for triage and treatment of combat casualties. A consensus on the assessment and diagnosis of combat injuries was made and discussed at the second annual meeting of the Professional Committee on Disaster Medicine of the Chinese People's Liberation Army(PLA). In this consensus agreement, the massive hemorrhage, airway, respiration, circulation and hypothermia(MARCH) algorithm, which is a simple triage and rapid treatment and field triage score, was recommended to assess combat casualties during the first-aid stage, whereas the abbreviated scoring method for combat casualty and the MARCH algorithm were recommended to assess combat casualties in level Ⅱ facilities. In level Ⅲ facilities, combined measures, including a history inquiry, thorough physical examination, laboratory examination, X-ray, and ultrasound examination, were recommended for the diagnosis of combat casualties. | 7 | Finite element model validation of bridge based on structural health monitoring——Part I: Response surface-based finite element model updating显示文摘In the engineering practice, merging statistical analysis into structural evaluation and assessment is a tendency in the future. As a combination of mathematical and statistical techniques, response surface(RS) methodology has been successfully applied to design optimization, response prediction and model validation. With the aid of RS methodology,these two serial papers present a finite element(FE) model updating and validation method for bridge structures based on structural health monitoring. The key issues to implement such a model updating are discussed in this paper, such as design of experiment,parameter screening, construction of high-order polynomial response surface model,optimization methods and precision inspection of RS model. The proposed procedure is illustrated by a prestressed concrete continuous rigid-frame bridge monitored under operational conditions. The results from the updated FE model have been compared with those obtained from online health monitoring system. | 10 | Radial propagation of magnetospheric substorm-injected energetic electrons observed using a BD-IES instrument and Van Allen Probes显示文摘In cases where substorm injections can be observed simultaneously by multiple spacecraft,they can help elucidate the potential mechanisms of particle transport and energization,of great importance to understanding and modeling the magnetosphere.In this paper,using data returned from the BeiDa-IES(BD-IES) instrument onboard a satellite in an inclined(55°) geosynchronous orbit(IGSO),in combination with two geo-transfer orbiting(GTO) satellite Van Allen Probes(A and B),we analyze a substorm injection event that occurred on the 16 th of October 2015.During this substorm injection,the IGSO onboard BD-IES was outbound,while both Van Allen Probe satellites(A and B) were inbound,a configuration of multiple trajectories that provides a unique opportunity to simultaneously investigate both the inward and outward radial propagation of substorm injection.Indicated by AE/AL indices,this substorm was closely related to an IMF/solar wind discontinuity that showed a sharp change in IMF Bz direction to the north.The innermost signature of this substorm injection was detected by Van Allen Probes A and B at L-3.7,while the outermost signature was observed by the onboard BD-IES instrument at L~\0.These data indicate that the substorm had a global,rather than just local,effect.Finally,we suggest that electric fields carried by fast-mode compressional waves around the substorm injection are the most likely candidate mechanism for the electron injection signatures observed in the inner- and outermost inner magnetosphere. | ZONG QiuGang HAO YiXin ZOU Hong FU SuiYan ZHOU XuZhi REN Jie WANG LingHua YUAN ChongJing LIU ZhiYang JIA XiangHong QUAN Lin LIU Ying WANG YongFu | 2016 | Science China Earth Sciences2016,59,7: | 8 |
| 11 | Hierarchical activation of compartmentalized pools of AMPK depends on severity of nutrient or energy stress显示文摘AMPK, a master regulator of metabolic homeostasis, is activated by both AMP-dependent and AMP-independent mechanisms. The conditions under which these different mechanisms operate, and their biological implications are unclear. Here, we show that, depending on the degree of elevation of cellular AMP, distinct compartmentalized pools of AMPK are activated, phosphorylating different sets of targets. Low glucose activates AMPK exclusively through the AMP-independent, AXIN-based pathway in lysosomes to phosphorylate targets such as ACC1 and SREBP1c, exerting early anti-anabolic and pro-catabolic roles. Moderate increases in AMP expand this to activate cytosolic AMPK also in an AXIN-dependent manner. In contrast, high concentrations of AMP, arising from severe nutrient stress, activate all pools of AMPK independently of AXIN. Surprisingly, mitochondrion-localized AMPK is activated to phosphorylate ACC2 and mitochondrial fission factor (MFF) only during severe nutrient stress. | 13 | Oscillation analysis of a DFIG-based wind farm interfaced with LCC-HVDC显示文摘With the increasing capacity of wind farm, HVDC technology has become a promising transmission scheme for long distance transportation of large-scale wind power. However oscillation caused by this system will have a great influence on the security and stability of power system operation. In this paper, the oscillation of a doubly-fed induction generator(DFIG)-based wind farm interfaced with line commutated converter(LCC) based HVDC is discussed. Low-frequency oscillation and subsynchronous oscillation(SSO) are studied since these two oscillations are the particularly concerned oscillations in the stability study of power system in recent years. The model of a DFIG-based wind farm interconnected with LCC-HVDC is developed. The impact of drive train model's structure and parameters on the oscillation characteristics is analyzed. Eigenvalue and participation factor analysis are used to identify the three main modes, which include controller mode, electromechanical mode, and shaft mode. The effects of DFIG controller's parameters, wind speed and operating conditions of HVDC on those modes are studied. | 17 | Leaf photosynthesis and yield components of mung bean under fully open-air elevated[CO_2]显示文摘Mung bean(Vigna radiata L.) has the potential to establish symbiosis with rhizobia,and symbiotic association of soil micro flora may facilitate the photosynthesis and plant growth response to elevated[CO_2].Mung bean was grown at either ambient CO_2 400 μmol mol^(-1) or[CO_2]((550+17) μmol mol^(-1)) under free air carbon dioxide enrichment(FACE) experimental facility in North China.Elevated[CO_2]increased net photosynthetic rate(P_n),water use efficiency(WUE) and the non-photochemical quenching(NPQ) of upper most fully-expanded leaves,but decreased stomatal conductance(G_s),intrinsic efficiency of PSII(F_v '/F_m'),quantum yield of PSII(φ_(PSll)) and proportion of open PSII reaction centers(q_p).At elevated[CO_2],the decrease of F_v'/F_m',φ_(PSII),q_p at the bloom stage were smaller than that at the pod stage.On the other hand,P_n was increased at elevated[CO_2]by 18.7 and 7.4%at full bloom(R2) and pod maturity stages(R4),respectively.From these findings,we concluded that as a legume despite greater nutrient supply to the carbon assimilation at elevated[CO_2],photosynthetic capacity of mung bean was still suppressed under elevated[CO_2]particularly at pod maturity stage but plant biomass and yield was increased by 11.6 and 14.2%,respectively.Further,these findings suggest that even under higher nutrient acquisition systems such as legumes,nutrient assimilation does not match carbon assimilation under elevated[CO_2]and leads photosynthesis down-regulation to elevated[CO_2]. | GAO Ji HAN Xue Saman Seneweera LI Ping ZONG Yu-zheng DONG Qi LIN Er-da HAO Xing-yu | 2015 | Journal of Integrative Agriculture2015,14,5: | 6 |

Methods We conducted T2-Weighted Magnetic Resonance Imaging(MRI) and 1H-MRS with a 3.0-Tesla animal MRI system to investigate the early microstructural and metabolic profiles in vivo in the striatum of rats following carbon monoxide(CO) poisoning. Results Compared to baseline, we found significant cortical surface deformation, cerebral edema changes, which were indicated by the unclear gray/white matter border, and lateral ventricular volume changes in the brain. A significant reduction in the metabolite to total creatine(Cr) ratios of N-acetylaspartate(NAA) was observed as early as 1 h after the last CO administration, while the lactate(Lac) levels increased marginally. Both the Lac/Cr and NAA/Cr ratios leveled off at 6 h and showed no subsequent significant changes. In addition, compared to the control, the choline(Cho)/Cr ratio was slightly reduced in the early stages and significantly increased after 6 h. In addition, a pathological examination revealed mild cerebral edema on cessation of the insult and more severe cerebral injury after additional CO poisoning. Conclusion The present study demonstrated that 1H-MRS of the brain identified early metabolic changes after CO poisoning. | 19 | cAMP elevators inhibit LPS-induced IL-12 p40 expression by interfering with phosphorylation of p38 MAPK in Murine Peritoneal Macrophages显示文摘cAMP mediated signaling may play a suppressive role in immune response. We previously found that the cAMP-elevators (CTx and 8-Br-cAMP) inhibited IL-12, IL-la, IL-6 gene expression, but increased the transcriptional levels of IL-10 and IL-1Ra in LPS-treated murine peritoneal macrophages. The present study examined a possible molecular mechanism involved in cAMP elevators-induced inhibition of IL-12 p40 expression in response to LPS. Our data demonstrated that cAMP elevators downregulated IL-12 p40 mRNA expression and IL-12 pT0 production in murine peritoneal macrophages. Subsequent studies revealed that cAMP-elevators blocked phosphorylation of p38 MAPK, but did not affect the activity of NF-κB binding to IL-12 promoter (-136/-112). | 20 | Chinese expert consensus on the treatment of modern combat-related spinal injuries显示文摘The battlefield treatments of spinal and spinal cord injury vary from civilian settings. However, there is no unified battlefield treatment guidelines for spine trauma in PLA. An expert consensus is reached, based on spine trauma epidemiology and the concepts of battlefield treatment combined with the existing levels of military medical care in modern warfare. Since the specialized treatment for spine trauma are no significant difference between civilian settings and modern war, the first aid, emergency treatment and early treatment of spine trauma are introduced separately in three levels in this consensus. In Level I facilities, the fast and accurate evaluation of spine trauma followed by fixation and stabilization are recommended during the first-aid stage. Re-evaluation, further treatment for possible hemorrhagic shock, dyspnea and infection are recommended at Level II facilities. At Level III facilities, it is recommended to strengthen the intensive care and the prevention of urinary system and lung infection for the wounded with severe spinal injury, however, spinal surgery is not recommended in a battlefield hospital.
