| 1 | Characteristics and Origin of Tight Oil Accumulations in the Upper Triassic Yanchang Formation of the Ordos Basin,North-Central China显示文摘The Upper Triassic oil accumulations in the Ordos Basin is the most successful tight oil play in China,with average porosity values of less than 10% and permeability values below 1.0 mD.This study investigated the geological characteristics and origin of the tight oil accumulations in the Chang 6 member of the Upper Triassic Yanchang Formation in the Shanbei area based on over 50,000 petrological,source-rock analysis,well logging and production data.The tight oil accumulation of the Chang 6 member is distributed continuously in the basin slope and the centre of the basin.The oilwater relationships are complex.Laumontite dissolution pores are the most important storage spaces,constituting 30%-60% of total porosity and showing a strong positive relationship with oil production.The pore-throat diameter is less than 1 μm,and the calculated critical height of the oil column is much larger than the tight sand thickness,suggesting that the buoyancy was probably of limited importance for oil migration.The pressure difference between the source rocks and sandstone reservoirs is inferred to have provided driving force for hydrocarbon migration.Two factors of source-reservoir configuration and laumontite dissolution contributed to the formation of the Chang 6 tight oil accumulations.Intense hydrocarbon generation and continuous sand bodies close to the hydrocarbon kitchen are the foundation for the large-scale oil distribution.Dissolution of feldspar-laumontite during the process of organic matter evolution generated abundant secondary pores and improved the reservoir quality. | WU Songtao ZOU Caineng ZHU Rukai YAO Jingli TAO Shizhen YANG Zhi ZHAI Xiufen CUI Jingwei LIN Senhu | 2016 | Acta Geologica Sinica(English Edition)2016,90,5: | 13 |
| 2 | Identifying disease modules and components of viral infections based on multi-layer networks显示文摘With the emergence of multi-dimensional data, we can more comprehensively analyze the pathogenic mechanisms of complex diseases, and thereby improve the diagnosis, treatment and prevention of these diseases.This study presents a novel multi-layer network-based strategy that integrates multi-dimensional data, and identifies disease-related modules and components of viral infections. We first propose a systematic method that constructs a virus-host interaction network with three layers: a viral protein layer, a host protein layer and a host gene layer. This method combines the data of high-throughput gene expression, viral protein interactions,virus-host interactions, protein-protein interactions and transcriptional regulatory relationships. To extract the underlying mechanisms of viral infections from the multi-layer networks, we identify the intra-layer and crosslayer modules, and investigate the conserved modules across multiple datasets. The essential components in the multi-layer networks are detected by singular-value decomposition. The identified conserved modules and essential components are combined into a functional enrichment analysis that reveals their contributions to influenza virus replication. By this analysis, we elucidate the common and specific mechanisms of the replication cycles of two influenza strains. | 15 | Dynamic Host Immune and Transcriptomic Responses to Respiratory Syncytial Virus Infection in a Vaccination-Challenge Mouse Model显示文摘Respiratory syncytial virus(RSV) is the major cause of lower respiratory tract infections in children. Inactivated RSV vaccine was developed in the late 1960’s, but the vaccine-enhanced disease(VED) occurred to vaccinated infants upon subsequent natural RSV infection. The excessive inflammatory immunopathology in the lungs might be involved in the VED, but the underlying mechanisms remain not fully understood. In this study, we utilized UV-inactivated RSV in the prime/boost approach followed by RSV challenge in BALB/c mice to mimic RSV VED. The dynamic virus load,cytokines, histology and transcriptome profiles in lung tissues of mice were investigated from day 1 to day 6 post-infection.Compared to PBS-treated mice, UV-RSV vaccination leads to a Th2 type inflammatory response characterized by enhanced histopathology, reduced Treg cells and increased IL4^(+)CD4 T cells in the lung. Enhanced production of several Th2 type cytokines(IL-4, IL-5, IL-10) and TGF-b, reduction of IL-6 and IL-17 were observed in UV-RSV vaccinated mice. A total of 5582 differentially expressed(DE) genes between PBS-treated or vaccinated mice and na?ve mice were identified by RNA-Seq. Eleven conserved high-influential modules(HMs) were recognized, majorly grouped into regulatory networks related to cell cycle and cell metabolism, signal transduction, immune and inflammatory responses. At an early time post-infection, the vaccinated mice showed obvious decreased expression patterns of DE genes in 11 HMs compared to PBS-treated mice. The extracellular matrix(HM5) and immune responses(HM8) revealed tremendous differences in expression and regulation characteristics of transcripts between PBS-treated and vaccinated mice at both early and late time points.
