2Betaine affects muscle lipid metabolism via regulating the fatty acid uptake and oxidation in finishing pig显示文摘Background: Betaine affects fat metabolism in animals, but the specific mechanism is still not clear. The purpose of this study was to investigate possible mechanisms of betaine in altering lipid metabolism in muscle tissue in finishing pigs.Methods: A total of 120 crossbred gilts(Landrace × Yorkshire × Duroc) with an average initial body weight of 70.1 kg were randomly allotted to three dietary treatments. The treatments included a corn–soybean meal basal diet supplemented with 0, 1250 or 2500 mg/kg betaine. The feeding experiment lasted 42 d.Results: Betaine addition to the diet significantly increased the concentration of free fatty acids(FFA) in muscle(P < 0.05). Furthermore, the levels of serum cholesterol and high-density lipoprotein cholesterol were decreased(P < 0.05) and total cholesterol content was increased in muscle(P < 0.05) of betaine fed pigs. Experiments on genes involved in fatty acid transport showed that betaine increased expression of lipoprotein lipase(LPL), fatty acid translocase/cluster of differentiation(FAT/CD36), fatty acid binding protein(FABP3) and fatty acid transport protein(FATP1)(P < 0.05). The abundance of fatty acid transport protein and fatty acid binding protein were also increased by betaine(P < 0.05). As for the key factors involved in fatty acid oxidation, although betaine supplementation didn't affect the level of carnitine and malonyl-CoA, betaine increased mR NA and protein abundance of carnitine palmitransferase-1(CPT1)and phosphorylated-AMPK(P < 0.05).Conclusions: The results suggested that betaine may promoted muscle fatty acid uptake via up-regulating the genes related to fatty acid transporter including FAT/CD36, FATP1 and FABP3. On the other hand, betaine activated AMPK and up-regulated genes related to fatty acid oxidation including PPARα and CPT1. 4A Fully Homomorphic Encryption Scheme with Better Key Size显示文摘Fully homomorphic encryption is faced with two problems now.One is candidate fully homomorphic encryption schemes are few.Another is that the efficiency of fully homomorphic encryption is a big question.In this paper,we propose a fully homomorphic encryption scheme based on LWE,which has better key size.Our main contributions are:(1) According to the binary-LWE recently,we choose secret key from binary set and modify the basic encryption scheme proposed in Linder and Peikert in 2010.We propose a fully homomorphic encryption scheme based on the new basic encryption scheme.We analyze the correctness and give the proof of the security of our scheme.The public key,evaluation keys and tensored ciphertext have better size in our scheme.(2) Estimating parameters for fully homomorphic encryption scheme is an important work.We estimate the concert parameters for our scheme.We compare these parameters between our scheme and Bra 12 scheme.Our scheme have public key and private key that smaller by a factor of about logq than in Bra12 scheme.Tensored ciphertext in our scheme is smaller by a factor of about log2 q than in Bra 12 scheme.Key switching matrix in our scheme is smaller by a factor of about log3 q than in Bra 12 scheme.CHEN Zhigang WANG Jian ZHANG ZengNian SONG Xinxia 2014China Communications2014,11,9:5
5An implantable microelectrode array for simultaneous L-glutamate and electrophysiological recordings in vivo显示文摘L-glutamate,the most common excitatory neurotransmitter in the mammalian central nervous system(CNS),is associated with a wide range of neurological diseases.Because neurons in CNS communicate with each other both electrically and chemically,dualmode(electric and chemical)analytical techniques with high spatiotemporal resolution are required to better understand glutamate function in vivo.In the present study,a silicon-based implantable microelectrode array(MEA)composed of both platinum electrochemical and electrophysiological microelectrodes was fabricated using micro-electromechanical system.In the MEA probe,the electrophysiological electrodes have a low impedance of 0.018 MΩat 1 kHz,and the electrochemical electrodes show a sensitivity of 56 pAμM^(−1) to glutamate and have a detection limit of 0.5μM.The MEA probe was used to monitor extracellular glutamate levels,spikes and local field potentials(LFPs)in the striatum of anaesthetised rats.To explore the potential of the MEA probe,the rats were administered to KCl via intraperitoneal injection.K+significantly increases extracellular glutamate levels,LFP low-beta range(12–18 Hz)power and spike firing rates with a similar temporal profile,indicating that the MEA probe is capable of detecting dual-mode neuronal signals.It was concluded that the MEA probe can help reveal mechanisms of neural physiology and pathology in vivo.Wenjing Wei Yilin Song Li Wang Song Zhang Jinping Luo Shengwei Xu Xinxia Cai 2015Microsystems & Nanoengineering2015,1,1:4
6Small-volume solution current-time behavior study for application in reverse iontophoresis-based non-invasive blood glucose monitoring显示文摘The reverse iontophoresis-based glucose monitoring circumstance is similar to the small-volume solution in which mass diffusion controls the current response of the electrochemical biosensors.In this study,the law of mass transfer in this type of solution was analyzed and a mathematic model was established to depict the current-time behavior of the fabricated planar electrode used in the non-invasive meter designed by ourselves.A small-volume glucose solution was directly constructed on the electrode to simulate the reverse iontophoresis-based sensing condition.The correctness of the model was demonstrated by chronoamperometry.Animal assay was subsequently carried out to verify the practicality of the model in determination of blood glucose.The results processed by the new method accurately traced the authentic value,confirming the advantage of the new method and the potential in clinical analysis.CHANG LingQian LIU ChunXiu HE YinZeng XIAO HongHui CAI XinXia 2011Science China Chemistry2011,54,1:4
7CBP-mediated Slug acetylation stabilizes Slug and promotes EMT and migration of breast cancer cells显示文摘Slug,a member of the Snail family of transcriptional repressors,plays a key role in cancer progression,cellular plasticity,and epithelial to mesenchymal transition(EMT).Slug is a fast-turnover protein and its stability is controlled by post-translational modifications.Here,we identified that Slug is acetylated by acetyltransferase CREB-binding protein(CBP)in breast cancer cells.CBP directly interacts with the C-terminal domain of Slug through its catalytic histone acetyltransferase(HAT)domain,leading to acetylation of Slug at lysines 166 and 211.Analysis with acetylation-specific antibodies revealed that Slug is highly acetylated in metastatic breast cancer cells.Notably,Slug acetylation,mediated by CBP at lysines 166 and 211,doubles its halflife and increases its stability.Further,acetylated Slug downregulates the expression of E-cadherin,the epithelial marker,and upregulates the expression of N-cadherin and vimentin,thereby promoting breast cancer cell migration.In conclusion,the present study demonstrates that CBP-mediated Slug acetylation increases its stability,promoting EMT and migration of breast cancer cells.Xiaoyan Dai Yanli Xin Weizhi Xu Xinxia Tian Xiaofan Wei Hongquan Zhang 2021Science China(Life Sciences)2021,64,4:4
8Selective electrochemical production of hydrogen peroxide at zigzag edges of exfoliated molybdenum telluride nanoflakes显示文摘The two-electron reduction of molecular oxygen represents an effective strategy to enable the green,mild and on-demand synthesis of hydrogen peroxide.Its practical viability,however,hinges on the development of advanced electrocatalysts,preferably composed of non-precious elements,to selectively expedite this reaction,particularly in acidic medium.Our study here introduces 2H-MoTe2 for the first time as the efficient non-precious-metal-based electrocatalyst for the electrochemical production ofhydrogen peroxide in acids.We show that exfoliated 2H-MoTe2 nanoflakes have high activity(onset overpotential〜140 mV and large mass activity of 27 A g^-1 at 0.4 V versus reversible hydrogen electrode),great selectivity(H2O2 percentage up to 93%)and decent stability in 0.5 M H2SO4.Theoretical simulations evidence that the high activity and selectivity of 2H-MoTe2 arise from the proper binding energies of HOO*and O*at its zigzag edges that jointly favor the two-electron reduction instead of the four-electron reduction of molecular oxygen.Xuan Zhao Yu Wang Yunli Da Xinxia Wang Tingting Wang Mingquan Xu Xiaoyun He Wu Zhou Yafei Li Jonathan NColeman Yanguang Li 2020National Science Review2020,7,8:3
9Low sample volume origami-paper-based graphene-modified aptasensors for label-free electrochemical detection of cancer biomarker-EGFR显示文摘In this work,an electrochemical paper-based aptasensor was fabricated for label-free and ultrasensitive detection of epidermal growth factor receptor(EGFR)by employing anti-EGFR aptamers as the bio-recognition element.The device used the concept of paper-folding,or origami,to serve as a valve between sample introduction and detection,so reducing sampling volumes and improving operation convenience.Amino-functionalized graphene(NH 2-GO)/thionine(THI)/gold particle(AuNP)nanocomposites were used to modify the working electrode not only to generate the electrochemical signals,but also to provide an environment conducive to aptamer immobilization.Electrochemical characterization revealed that the formation of an insulating aptamer–antigen immunocomplex would hinder electron transfer from the sample medium to the working electrode,thus resulting in a lower signal.The experimental results showed that the proposed aptasensor exhibited a linear range from 0.05 to 200 ngmL^(−1)(R^(2)=0.989)and a detection limit of 5pgmL^(−1) for EGFR.The analytical reliability of the proposed paper-based aptasensor was further investigated by analyzing serum samples,showing good agreement with the gold-standard enzyme-linked immunosorbent assay.Yang Wang Shuai Sun Jinping Luo Ying Xiong Tao Ming Juntao Liu Yuanyuan Ma Shi Yan Yue Yang Zhugen Yang Julien Reboud Huabing Yin Jonathan MCooper Xinxia Cai 2020Microsystems & Nanoengineering2020,6,1:2
10Significantly influenced photocatalytic performance for H_(2)O_(2)generation over ultrathin g-C_(3)N_(4)through regulating the migration orientation of photogenerated charge carriers显示文摘H_(2)O_(2)has been widely applied in the fields of chemical synthesis,medical sterilization,pollutant removal,etc.,due to its strong oxidizing property and the avoidable secondary pollution.Despite of the enhanced performance for H_(2)O_(2)generation over g-C_(3)N_(4)semiconductors through promoting the separation of photo-generated charge carriers,the effect of migration orientation of charge carriers is still ambiguous.For this emotion,surface modification of g-C_(3)N_(4)was employed to adjust the migration orientation of charge carriers,in order to investigate systematically its effect on the performance of H_(2)O_(2)generation.It was found that ultrathin g-C_(3)N_(4)(UCN)modified by boron nitride(BN),as an effective hole-attract agent,demonstrated a significantly enhanced performance.Particularly,for the optimum UCN/BN-40%catalyst,4.0-fold higher yield of H_(2)O_(2)was obtained in comparison with the pristine UCN.As comparison,UCN modified by carbon dust demonstrated a completely opposite tendency.The remarkably improved performance over UCN/BN was ascribed to the fact that more photo-generated electrons were remained inside of triazine structure of g-C_(3)N_(4),leading to the formation of larger amount of 1,4-endoxide.It is anticipated that our work could provide new insights for the design of photocatalyst with significantly improved performance for H_(2)O_(2)generation.Xinxia He Hongyan Shang Chuan Wang Le Chen Zehan Gong Jun Wang Shilin Zhao Jun Ma 2021Chinese Chemical Letters2021,32,11:2
11Real-time simultaneous recording of electrophysiological activities and dopamine overflow in the deep brain nuclei of a non-human primate with Parkinson’s disease using nano-based microelectrode arrays显示文摘Parkinson’s disease(PD)is characterized by a progressive degeneration of nigrostriatal dopaminergic neurons.The precise mechanisms are still unknown.Since the neuronal communications are inherently electrical and chemical in nature,dual-mode detection of PD-related neuroelectrical and neurochemical information is essential for PD research.Subthalamic nucleus(STN)highfrequency stimulation(HFS)can improve most symptoms of PD patients and decrease the dosage of antiparkinsonian drugs.The mechanism of STN-HFS for PD still remains elusive.In this study,a silicon-based dual-mode microelectrode array(MEA)probe was designed and fabricated,and systematic dual-mode detection methods were established.The recording sites were modified using Pt nanoparticles and Nafion to improve the signal-to-noise(SNR)ratio.To evaluate its applicability to PD research,in vivo electrophysiological and electrochemical detection was performed in normal and hemiparkinsonian models,respectively.Through comparison of the dual-mode signals,we demonstrated the following in a PD monkey:(1)the maximum dopamine concentration in the striatum decreased by 90%;(2)the spike firing frequency increased significantly,especially in the region of the cortex;(3)the spectrogram analysis showed that much power existed in the 0–10 Hz frequency band;and(4)following repeated subthalamic nucleus high-frequency stimulation trials,the level of DA in the striatum increased by 16.5μM,which led to a better elucidation of the mechanism of HFS.The dual-mode MEA probe was demonstrated to be an effective tool for the study of neurological disorders.Song Zhang Yilin Song Mixia Wang Guihua Xiao Fei Gao Ziyue Li Guoxian Tao Ping Zhuang Feng Yue Piu Chan Xinxia Cai 2018Microsystems & Nanoengineering2018,4,1:2
12Species-specific aminoacylation of Oryza sativa mitochondrial tRNA^(Trp)显示文摘Abstract The details of species- spe- cific aminoacylation in Oryza sativa mitochondrial tRNATrp by bacterial and eukaryotic (cytoplasm) tryptophanyl-tRNA synthetases (TrpRS) were inves- tigated. Seven single or multiple mutations of three bases (G73, U72, A 68) were made in O. sativa mi- tochondrial tRNATrp to the corresponding nucleotides present in human tRNATrp. In vitro transcripts of these mutant genes were tryptophanylated by Bacil- lus subtilis and human tryptophanyl-tRNA syntheta- ses (TrpRS), and the kinetic parameters were deter- mined. The results showed that the aminoacylation of seven mutant transcripts by B. subtilis TrpRS was 53.33%―99.79% less efficient than that by wild-type O. sativa mitochondrial tRNATrp, but was 4―330 times more efficient than that by human TrpRS. The mutant MPH7 (G73, U72 and C68 in O. sativa mito- chondrial tRNA were all replaced by the counterpart residues from human tRNATrp and showed a great change in aminoacylation efficiency. Our results in- dicate that the species-specific identity elements of O. sativa mitochondrial tRNATrp are similar to bacterial and eukaryotic (cytoplasm). They are mainly located at the discriminator base, the first and the fifth pairs of bases, the discriminator base G73, two bases in the acceptor stem G1/U72 and U5/A68. 13Rapid detection of Cyfra 21-1 by optical-biosensing based on chemiluminescence immunoassay using bio-functionlized magnetic nanocomposites显示文摘This letter reports a chemiluminescene immunoassay method combined with immunomagnetic separation to rapidly detect Cyfra 21-1, in which bio-functionlized magnetic nanocomposites were used as mobile substrate for capturing and isolating the cyfra 21-1 proteins. After the captured Cyfra 21-1 further reacted with horseradish peroxidase-conjugated anti-Cyfra 21-1 antibody to form a sandwich immunocomplex, the chemiluminescence would be produced as a result of addition of the chemiluminescent substrate. A home-made optical biosensor was designed to detect the chemiluminescence instead of other large instruments. There is a good linear response between the chemiluminescence intensity and the concentration of Cyfra 21-1 in the range from 0.2 to 50 ng/mL. The whole detection process including incubation, washing and detection could be performed within 45 min.
