| 1 | Wearable strain sensing textile based on one-dimensional stretchable and weavable yarn sensors显示文摘Flexible, wearable, and even stretchable sensors are the key components ofsmart electronic textiles. However, most reported flexible and wearable sensorsfor wearable electronics are usually fabricated in two-dimensional (2D) planarstrip configurations, which cannot be properly integrated into textile structuresand thus greatly degrade intrinsic properties such as the softness, flexibility, andair permeability of textiles and the aesthetic feeling of clothing. In this work, a newone-dimensional weavable strain sensing yarn consisting of an elastic polyurethane(PU) core, a conductive Ag-nanopartides/graphene-microsheets composite sheath,and a silicone encapsulation layer was designed and fabricated through aneasily manipulated protocol. Arising from the reasonable structural design andappropriate material selection, the as-fabricated strain sensor not only exhibitedexcellent flexibility, stretchability, and highly repeatable electromechanicalstability (a repeatability error of 1.56%) but also possessed both high sensitivity(a gauge factor of nearly 500) and a relatively wide working range (0-50%applied strain) with good linearity (a correlation coefficient of 0.98). In addition,the facile, nearly all-solution-based fabrication protocol enabled the scalableproduction of long conductive yams. Thus, the proper yarn length and superbmechanical properties endowed the stretchable conductive yarn with goodweavability. The excellent wearability of the stretchable conductive yarn wasderived from the outermost isolating, hydrophobic, and biocompatible siliconeencapsulation layer. | 2 | Early inoculation with caecal fermentation broth alters small intestine morphology,gene expression of tight junction proteins in the ileum,and the caecal metabolomic profiling of broilers显示文摘Background:The establishment of stable microbiota in early life is beneficial to the individual.Changes in the intestinal environment during early life play a crucial role in modulating the gut microbiota.Therefore,early intervention to change the intestinal environment can be regarded as a new regulation strategy for the growth and health of poultry.However,the effects of intestinal environmental changes on host physiology and metabolism are rarely reported.This study was conducted to investigate the effects of early inoculation with caecal fermentation broth on small intestine morphology,gene expression of tight junction proteins in the ileum,and cecum microbial metabolism of broilers.Results:Our data showed that early inoculation with caecal fermentation broth could improve intestine morphology.The small intestine villus height was significantly increased(P<0.05)in the intervened broilers compared to the control group,especially on day 28.A similar result was observed in the ratio of villus height to crypt depth(P<0.05).Meanwhile,we found early inoculation significantly increased(P<0.05)the expression levels of zonula occludens-1(ZO1)on days 14 and 28,claudin-1(CLDN1)on day 28,whereas the gene expression of claudin-2(CLDN2)was significantly decreased(P<0.05)on days 14 and 28.Gas chromatography time-of-flight/mass spectrometry(GC-TOF/MS)technology was further implemented to systematically evaluate the microbial metabolite profiles.Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)displayed a distinct trend towards separation between the fermentation broth group(F group)and the control group(C group).The differentially expressed metabolites were identified,and they were mainly functionally enriched in beta-alanine metabolism and biosynthesis of unsaturated fatty acids.In addition,1,3-diaminopropane was selected as a key biomarker that responded to early inoculation with caecal fermentation broth.Conclusions:These results provide insight into intestinal metabolomics and confirm that early inoculation with caecal fermentation broth can be used as a potential strategy to improve intestinal health of broilers. | Yujie Gong Wenrui Xia Xueting Wen Wentao Lyu Yingping Xiao Hua Yang Xiaoting Zou | 2020 | Journal of Animal Science and Biotechnology2020,11,2: | 3 |

| 4 | Pooled Plasmid Sequencing Reveals the Relationship Between Mobile Genetic Elements and Antimicrobial Resistance Genes in Clinically Isolated Klebsiella pneumoniae显示文摘Plasmids remain important microbial components mediating the horizontal gene transfer(HGT)and dissemination of antimicrobial resistance.To systematically explore the relationship between mobile genetic elements(MGEs)and antimicrobial resistance genes(ARGs),a novel strategy using single-molecule real-time(SMRT)sequencing was developed.This approach was applied to pooled conjugative plasmids from clinically isolated multidrug-resistant(MDR)Klebsiella pneumoniae from a tertiary referral hospital over a 9-month period.The conjugative plasmid pool was obtained from transconjugants that acquired antimicrobial resistance after plasmid conjugation with 53 clinical isolates.The plasmid pool was then subjected to SMRT sequencing,and 82 assembled plasmid fragments were obtained.In total,124 ARGs(responsible for resistance to b-lactam,fluoroquinolone,and aminoglycoside,among others)and 317 MGEs[including transposons(Tns),insertion sequences(ISs),and integrons]were derived from these fragments.Most of these ARGs were linked to MGEs,allowing for the establishment of a relationship network between MGEs and/or ARGs that can be used to describe the dissemination of resistance by mobile elements.Key elements involved in resistance transposition were identified,including IS26,Tn3,IS903 B,ISEcp1,and ISKpn19.As the most predominant IS in the network,a typical IS26-mediated multi-copy composite transposition event was illustrated by tracing its flanking 8-bp target site duplications(TSDs).The landscape of the pooled plasmid sequences highlights the diversity and complexity of the relationship between MGEs and ARGs,underpinning the clinical value of dominant HGT profiles. | Yan Jiang Yanfei Wang Xiaoting Hua Yue Qu Anton Y.Peleg Yunsong Yu | 2020 | Genomics, Proteomics & Bioinformatics2020,18,5: | 1 |
| 5 | Global effect of an RNA polymerase β-subunit mutation on gene expression in the radiation-resistant bacterium Deinococcus radiodurans显示文摘The β-subunit of RNA polymerase, which is involved in rifampin binding, is highly conserved among prokaryotes, and Rif r mutants detected in many bacteria are the result of amino acid changes. Spontaneous rifampin resistance mutations resulting in amino acid replacement (L420R) and deletion (1258-66 9 bp deletion) have been previously isolated in the rpoB gene of Deinococcus radiodurans. In this study, a β-subunit mutation in D. radiodurans resulted in a unique effect on growth rate. We used DNA microarrays and biochemical assays to investigate how the Rif r mutation in the β-subunit led to changes in growth rate via altered regulation of multiple genes. The expression of genes with predicted functions in metabolism, cellular processes and signaling, and information storage and processing were significantly altered in the 9 bp-deletion rpoB mutant. The consensus promoter sequence of up-regulated genes in the 9 bp-deletion rpoB mutant was identified as an AT-rich sequence. Greater levels of reactive oxygen species accumulated in the L420R and 9 bp-deletion rpoB mutants compared with wild type. | 7 | Bautype: Capsule and Lipopolysaccharide Serotype Prediction for Acinetobacter baumannii Genome显示文摘Acinetobacter baumannii has three clusters of genes for polysaccharide synthesis.Capsular polysaccharides and lipopolysaccharides are major virulence determinants in many Gram-negative bacteria,and play key roles in the overall fitness and virulence in A.baumannii,meanwhile poly-beta-1-6-N-acetylglucosamine was critical for biofilm formation.A variety of studies have been conducted which focused on the surveillance of antimicrobial resistance genes and clones in A.baumannii.However,a tool to perform serotype prediction in A.baumannii is still lacking.Here we present Bautype,a tool for whole genome sequencingbased K and O locus typing in A.baumannii.Using Bautype,users can upload and analyze the genome assembled in a web browser,with the results being displayed in a tabular or graphical format.We demonstrated the high accuracy of predicting K and O locus diversity using Bautype when analyzing more than 3000 A.baumannii genomes.Using our software,we were able to analyze K and O locus diversity among 593 genomes belonging to ST208(3 K and 8 O).These diverse loci can serve as epidemiological markers for Multi-drug Resistance(MDR)strain subtyping.We also identified 45 novel O locus types.Our findings highlight the importance in monitoring the capsule type and LOS of A.baumannii strains for which Bautype is a tailored clinical software.Bautype is available at http://gffzzf27235bfe84f4a38hp9qq5ovknfvn69wx.ffgz.tsg.suse.edu.cn. | Xiaoting Hua Qian Liang Li Fang Jintao He Meixia Wang Wenjie Hong Sebastian Leptihn Hengliang Wang Yunsong Yu Huan Chen | 2020 | Infectious Microbes & Diseases2020,2,1: | 0 |
