| 1 | Influence of Ren and Du meridian electro-acupuncture on neural stem cell proliferation and extracellular signal-regulated kinase pathway in a rat model of focal cerebral ischemia injury显示文摘BACKGROUND:Studies have shown that electro-acupuncture at the Ren meridian could improve proliferation of subventricular zone neural stem cells in cerebral-ischemic rats. However,there are few reports on the influence of electro-acupuncture at the Du meridian on neural stem cell proliferation. OBJECTIVE:To observe the influence of electro-acupuncture at Ren and Du meridians on neural stem cell proliferation in the subventricular zone and altered signal transduction in cerebral ischemia rats. DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed at the Laboratory of Human Anatomy,Medical College of Sun Yat-sen University from May 2006 to February 2008. MATERIALS:Mouse anti-rat bromodeoxyuridine (BrdU) monoclonal antibody was provided by Sigma,USA; mouse anti-rat nestin monoclonal antibody and extracellular signal-regulated protein kinase (ERK) specific inhibitor PD98059 were provided by Calbiochem,Germany; acupuncture needle was provided by Suzhou Acupuncture Supplies,China. METHODS:A total of 126 rats were randomly assigned to four groups:model (n = 36),Du meridian (n = 36),Ren/Du meridian (n = 36),and Ren/Du meridian + PD98059 (n = 18). Rats in the Ren /Du meridian + PD98059 group were observed on days 7 (n = 6) and 14 (n = 12) after cerebral ischemia injury. Rats in the model,Du meridian,and Ren/Du meridian groups were observed on days 7,14,and 28 after cerebral ischemia injury,with 12 rats per group at each time point. Thread occlusion was used to establish middle cerebral artery occlusion models. Electro-acupuncture was performed at Renzhong (DU 26) and Baihui (DU 20) acupoints in the Du meridian group,as well as Chengjiang (RN 24),Guanyuan (RN 4),Renzhong,and Baihui acupoints in the Ren/Du meridian and Ren/Du meridian + PD98059 groups 2 days after model establishment. In addition,electro-acupuncture stimulation with disperse-dense waves was performed,with 30 Hz disperse wave,100 Hz dense wave,and 5 V intensity for 20 minutes. Rats in the Ren/Du meridian + PD98059 group were treated with 0.2 μg PD98059 injection into the subventricular zone,2 μL per rat. Rats in the model group were not treated with electro-acupuncture. MAIN OUTCOME MEASURES:BrdU/nestin immunofluorescent staining was used to detect proliferating neural stem cells in the subventricular zone of cerebral ischemia rats; Western blot was used to determine phosphorylated ERK1 and 2 (pERK1/2) expression in the subventricular zone. RESULTS:On days 14 and 28 after cerebral ischemia,there were significantly more BrdU-positive and BrdU/nestin-positive cells in the Ren /Du meridian group compared with the Du meridian group (P < 0.05). PD98059 decreased the number of BrdU-positive and BrdU/nestin-positive cells induced by electro-acupuncture at the Ren and Du meridians (P < 0.05). On days 7,14,and 28 after treatment,pERK1/2 expression was significantly greater in the Du meridian and Ren/Du meridian groups compared with the model group (P < 0.05). The promoting effect of electro-acupuncture at Ren and Du meridians on ERK1/2 phosphorylation was superior to electro-acupuncture at the Du meridian alone on day 14 after model induction (P < 0.05). However,PD98059 completely abolished the promoting effect of electro-acupuncture at Ren/Du meridians on pERK1/2 expression (P < 0.05). | 3 | A novel colloidal gold immunochromatography assay strip for the diagnosis of schistosomiasis japonica in domestic animals显示文摘Background:Schistosomiasis remains a major public health concern in China and an epidemiological survey has revealed that schistosome-infected bovines and goats are the main transmission sources for the disease.Therefore,development of a sensitive technique for the diagnosis of schistosomiasis in domestic animals is necessary.Method:A novel colloidal gold immunochromatography assay(GICA)strip was developed for detecting Schistosoma japonicum in domestic animals.The colloidal gold was conjugated with recombinant streptococcal protein G(rSPG).As the test and control lines,the schistosome soluble egg antigen and rSPG,respectively,were blotted on nitrocellulose membrane.Results:The lowest detectable serum dilution was 1∶640 for schistosome-infected buffaloes.The cross-reaction rate of GICA was 14.29%with Paramphistomum sp.in buffaloes,16.67%with Haemonchus sp.in goats,and 33.33%with Orientobilharzia sp.in goats.These results were slightly lower and similar to those obtained through ELISA.Moreover,the strips for detecting S.japonicum in mice,rabbits,buffaloes,and goats showed high sensitivity(100.00%,100.00%,100.00%,and 100.00%,respectively)and specificity(100.00%,100.00%,94.23%,and 88.64%,respectively).And the sensitivity or specificity of the GICA strips did not present any significant differences after storage for 12 months at room temperature.When compared with ELISA,the GICA strips exhibited similar sensitivity and specificity in the diagnosis of schistosomiasis in mice,rabbits,buffaloes,and goats.Besides,only 5μl of serum are required for the test and the detection can be completed within 5 min.Conclusion:This study is the first to develop a GICA strip using gold-rSPG conjugate for the diagnosing of schistosomiasis in domestic animals,and preliminary results showed that the developed strip may be suitable for large-scale screening of schistosomiasis in endemic areas. | Rui Xu Jintao Feng Yang Hong Chao Lv Dengyun Zhao Jiaojiao Lin Ke Lu Hao Li Jinming Liu Xiaodan Cao Tao Wang Jinli Zai Zhaozhe Wang Bingguang Jia Qian Han Chuangang Zhu | 2017 | Infectious Diseases of Poverty2017,6,1: | 12 |

| 5 | Effect of quercetin on the expression of Bcl-2/Bax apoptotic proteins in endometrial cells of lipopolysaccharide-induced-abortion mice显示文摘OBJECTIVE: To explore the effect of quercetin on the expressions of Bcl-2/Bax apoptotic proteins in endometrial cells in mice with abortion induced by lipopolysaccharide.METHODS: For in vivo experiment, twenty five Kunming mice were randomly divided into five groups at day 4 of pregnancy, with 5 mice per group. The mice were treated with lipopolysaccharide(LPS)through tail vein intravenous injection at day 4 of pregnancy, followed by different concentrations of quercetin by oral gavage consecutively at days 5 to6 of pregnancy. On day 7 of gestation, the mice were sacrificed and the histopathological changes of the uterus tissues were observed. Immunohistochemical staining was applied to the detection of Bcl-2/Bax apoptotic proteins in the endometrial cells. For in vitro experiment, the primary endometrial cells werecultured using a uterus tissue mass culturing method sampled at day 4.5 of pregnancy. The cells were treated with LPS with or without different dosages of quercetin, respectively, for 12 h after 80% confluence. The expression of Bcl-2/Bax apoptotic proteins were detected by western blotting.RESULTS: Both the in vivo and in vitro experiments showed decreased expression of Bcl-2 and enhanced expression of Bax after LPS treatment, leading to a decreased Bcl-2/Bax ratio. | 8 | Could CMIP6 climate models reproduce the early-2000s global warming slowdown?显示文摘The unexpected global warming slowdown during 1998–2013 challenges the existing scientific understanding of global temperature change mechanisms,and thus the simulation and prediction ability of state-of-the-art climate models since most models participating in phase 5 of the Coupled Model Intercomparison Project(CMIP5)cannot simulate it.Here,we examine whether the new-generation climate models in CMIP6 can reproduce the recent global warming slowdown,and further evaluate their capacities for simulating key-scale natural variabilities which are the most likely causes of the slowdown.The results show that although the CMIP6 models present some encouraging improvements when compared with CMIP5,most of them still fail to reproduce the warming slowdown.They considerably overestimate the warming rate observed in 1998–2013,exhibiting an obvious warming acceleration rather than the observed deceleration.This is probably associated with their deficiencies in simulating the distinct temperature change signals from the human-induced long-term warming trend and/or the three crucial natural variabilities at interannual,interdecadal,and multidecadal scales.In contrast,the 4 models that can successfully reproduce the slowdown show relatively high skills in simulating the long-term warming trend and the three keyscale natural variabilities.Our work may provide important insight for the simulation and prediction of near-term climate changes. | Meng WEI Qi SHU Zhenya SONG Yajuan SONG Xiaodan YANG Yongqing GUO Xinfang LI Fangli QIAO | 2021 | Science China Earth Sciences2021,64,6: | 5 |
| 9 | Seasonal Variations of Several Main Water Masses in the Southern Yellow Sea and East China Sea in 2011显示文摘The seasonal variations of several main water masses in the southern Yellow Sea (SYS) and East China Sea (ECS) in 2011 were analyzed using the in-situ data collected on four cruises.There was something special in the observations for the Yellow Sea Warm Current (YSWC) ,the Yellow Sea Cold Water Mass (YSCWM) and the Changjiang Diluted Water (CDW) during that year.The YSWC was confirmed to be a seasonal current and its source was closely associated with the Kuroshio onshore intrusion and the northerly wind.It was also found that the YSCWM in the summer of 2011 occupied a more extensive area in comparison with the climatologically-mean case due to the abnormally powerful wind prevailing in the winter of 2010 and decaying gradually thereafter.Resulting from the reduced Changjiang River discharge,the CDW spreading toward the Cheju Island in the summer of 2011 was weaker than the long-term mean and was confined to flow southward in the other seasons.The other water masses seemed normal without noticeable anomalies in 2011.The Yellow Sea Coastal Current (YSCC) water,driven by the northerly wind,flowed southeastward as a whole except for its northeastward surface layer in summer.The Taiwan Warm Current (TWC) was the strongest in summer and the weakest in winter in its northward movement.The Kuroshio water with an enhanced onshore intrusion in autumn was stable in hydrographic features apart from the seasonal variation of its surface layer. | QUAN Qi MAO Xinyan YANG Xiaodan HU Yingying ZHANG Haiyan JIANG Wensheng | 2013 | Journal of Ocean University of China2013,12,4: | 5 |
| 10 | Transplantation of placenta-derived mesenchymal stem cells in type 2 diabetes:a pilot study显示文摘Mesenchymal stem cells(MSC)have been used in clinical trials for severe diabetes,a chronic disease with high morbidity and mortality.Bone marrow is the traditional source of human MSC,but human term placenta appears to be an alternative and more readily available source.Here,the therapeutic effect of human placenta-derived MSC(PD-MSC)was studied in type 2 diabetes patients with longer duration,islet cell dysfunction,high insulin doses as well as poor glycemic control in order to evaluate the safety,efficacy and feasibility of PD-MSC treatment in type 2 diabetes(T2D).Ten patients with T2D received three intravenous infusions of PDSC,with one month interval of infusion.The total number of PDSC for each patient was(1.22–1.51)×10^(6)/kg,with an average of 1.35×10^(6)/kg.All of the patients were followed up after therapy for at least 3 months.A daily mean dose of insulin used in 10 patients was decreased from 63.7±18.7 to 34.7±13.4 IU(P<0.01),and the C-peptide level was increased from 4.1±3.7 ng/mL to 5.6±3.8 ng/mL(P<0.05)respectively after therapy.In 4 of 10 responders their insulin doses reduced more than 50%after infusion.The mean levels of insulin and C-peptide at each time point in a total of 10 patients was higher after treatment(P<0.05).No fever,chills,liver damage and other side effects were reported.The renal function and cardiac function were improved after infusion.The results obtained from this pilot clinical trial indicate that transplantation of PD-MSC represents a simple,safe and effective therapeutic approach for T2D patients with islet cell dysfunction.Further large-scale,randomized and well-controlled clinical studies will be required to substantiate these observations. | Ranhua JIANG Zhibo HAN Guangsheng ZHUO Xiaodan QU Xue LI Xin WANG Yuankang SHAO Shimin YANG Zhong Chao HAN | 2011 | Frontiers of Medicine2011,5,1: | 5 |

| 12 | Active loading liposomal irinotecan hydrochloride:Preparation,in vitro and in vivo evaluation显示文摘The aim of the present study was to investigate the effect of various transmembrane ammonium salt gradients and different lipid composition on the loading efficiency of liposomal formulations of irinotecan hydrochloride(CPT-11),their behavior in vivo and cytotoxicity.Among ammonium salts studied,ammonium sulfate was successfully used to load CPT-11 into liposomes with the highest encapsulation efficiency.Subsequently,liposomal CPT-11 with different lipid composition was prepared by ammonium sulfate gradient method.CPT-11 can be loaded to a level over 90%into liposomes composed of soybean phospholipids/cholesterol(SPC-L)or hydrogenated natural soybean phospholipids/cholesterol(HSPC-L).In vitro release profiles were also investigated,indicating that HSPC-L had a lower release than that in SPC-L.In vivo,encapsulation of CPT-11 in both liposomal formulations showed higher area under the curve(AUC),a lower rate of clearance(CL)and smaller volume of distribution for CPT-11 than those of irinotecan hydrochloride solution(CPT-11-S).However,CL and AUC of 7-ethyl-10-hydroxycamptothecin(SN-38)were moderately improved in HSPC-L group.Based on the results of comparative pharmacokinetics of liposomal CPT-11 with different lipid composition,the in vitro cytotoxicity of HSPC-L was evaluated with human tumor cell.The result indicated that liposomal CPT-11 showed a great enhancement in vitro cytotoxicity.The results suggested that entrapment of CPT-11 in liposomes especially in those with high phase-transition temperature lipid by ammonium sulfate gradient would be a promising formulation with a better in vivo behavior. | Hongyan Wei Juan Song Hao Li Yang Li Shanshan Zhu Xiaodan Zhou Xiwen Zhang Li Yang | 2013 | Asian Journal of Pharmaceutical Sciences2013,8,5: | 3 |
| 13 | An Active De-anonymizing Attack Against Tor Web Traffic显示文摘Tor is pervasively used to conceal target websites that users are visiting. A de-anonymization technique against Tor, referred to as website fingerprinting attack, aims to infer the websites accessed by Tor clients by passively analyzing the patterns of encrypted traffic at the Tor client side. However, HTTP pipeline and Tor circuit multiplexing techniques can affect the accuracy of the attack by mixing the traffic that carries web objects in a single TCP connection. In this paper, we propose a novel active website fingerprinting attack by identifying and delaying the HTTP requests at the first hop Tor node. Then, we can separate the traffic that carries distinct web objects to derive a more distinguishable traffic pattern. To fulfill this goal, two algorithms based on statistical analysis and objective function optimization are proposed to construct a general packet delay scheme. We evaluate our active attack against Tor in empirical experiments and obtain the highest accuracy of 98.64%, compared with 85.95% of passive attack. We also perform experiments in the open-world scenario. | 18 | Targeting of anti-microbial proteins to the hyphal surface amplifies protection of crop plants against Phytophthora pathogens显示文摘Phytophthora pathogens are a persistent threat to the world's commercially important agricultural crops,including potato and soybean.Current strategies aim at reducing crop losses rely mostly on disease-resistance breeding and chemical pesticides,which can be frequently overcome by the rapid adaptive evolution of pathogens.Transgenic crops with intrinsic disease resista nee offer a promising alternative and con tinue to be developed.Here,we explored Phytophthora-derived PI3P(phosphatidylinositol 3-phosphate)as a novel control target,using proteins that bind this lipid to direct secreted anti-microbial peptides and proteins(AMPs)to the surface of Phytophthora pathogens.In transgenic Nicotiana benthamiana,soybean,and potato plants,significantly enhanced resistance to different pathogen isolates was achieved by expression of two AMPs(GAFP1 or GAFP3 from the Chinese medicinal herb Gastrodia elata)fused with a PI3P-specific binding domain(FYVE).Using the soybean pathogen P.sojae as an example,we demonstrated that the FYVE domain could boost the activities of GAFPs in multiple independent assays,including those performed in vitro,in vivo,and in planta.Mutational analysis of P.sojae PI3K1 and PI3K2 genes of this pathogen confirmed that the enhanced activities of the targeted GAFPs were correlated with PI3P levels in the pathogen.Collectively,our study provides a new strategy that could be used to confer resistance not only to Phytophthora pathogens in many plants but also potentially to many other kinds of plant pathogens with unique targets. | Yang Zhou Kun Yang Qiang Yan Xiaodan Wang Ming Cheng Jierui Si Xue Xue Danyu Shen Maofeng Jing Brett M.Tyler Daolong Dou | 2021 | Molecular Plant2021,14,8: | 2 |
