1Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 2Highly Stretchable,Elastic,and Sensitive MXene-Based Hydrogel for Flexible Strain and Pressure Sensors显示文摘Electronic skin is driving the next generation of cutting-edge wearable electronic products due to its good wearability and high accuracy of information acquisition.However,it remains a challenge to fulfill the requirements on detecting full-range human activities with existing flexible strain sensors.Herein,highly stretchable,sensitive,and multifunctional flexible strain sensors based on MXene-(Ti_(3)C_(2)T_(x)-)composited poly(vinyl alcohol)/polyvinyl pyrrolidone double-network hydrogels were prepared.The uniformly distributed hydrophilic MXene nanosheets formed a three-dimensional conductive network throughout the hydrogel,endowing the flexible sensor with high sensitivity.The strong interaction between the double-network hydrogel matrix and MXene greatly improved the mechanical properties of the hydrogels.The resulting nanocomposited hydrogels featured great tensile performance(2400%),toughness,and resilience.Particularly,the as-prepared flexible pressure sensor revealed ultrahigh sensitivity(10.75 kPa^(-1))with a wide response range(0-61.5 kPa),fast response(33.5 ms),and low limit of detection(0.87 Pa).Moreover,the hydrogel-based flexible sensors,with high sensitivity and durability,could be employed to monitor fullrange human motions and assembled into some aligned devices for subtle pressure detection,providing enormous potential in facial expression and phonation recognition,handwriting verification,healthy diagnosis,and wearable electronics.Yao Lu Xinyu Qu Wen Zhao Yanfang Ren Weili Si Wenjun Wang Qian Wang Wei Huang Xiaochen Dong 2020Research2020,,1:14

4Embryonic endothelial evolution towards first hematopoietic stem cells revealed by single-cell transcriptomic and functional analyses显示文摘Hematopoietic stem cells(HSCs)in adults are believed to be born from hemogenic endothelial cells(HECs)in mid-gestational embryos.Due to the rare and transient nature,the HSC-competent HECs have never been stringently identified and accurately captured,let alone their genuine vascular precursors.Here,we first used high-precision single-cell transcriptomics to unbiasedly examine the relevant EC populations at continuous developmental stages with intervals of 0.5 days from embryonic day(E)9.5 to E11.0.As a consequence,we transcriptomically identified two molecularly different arterial EC populations and putative HSCprimed HECs,whose number peaked at El 0.0 and sharply decreased thereafter,in the dorsal aorta of the aorta-gonadmesonephros(AGM)region.Combining computational prediction and in vivo functional validation,we precisely captured HSCcompetent HECs by the newly constructed Neurl3-EGFP reporter mouse model,and realized the enrichment further by a combination of surface markers(Procr+Kit+CD44+,PK44).Surprisingly,the endothelial-hematopoietic dual potential was rarely but reliably witnessed in the cultures of single HECs.Noteworthy,primitive vascular ECs from E8.0 experienced two-step fate choices to become HSC-primed HECs,namely an Initial arterial fate choice followed by a hemogenic fate conversion.This finding resolves several previously observed contradictions.Taken together,comprehensive understanding of endothelial evolutions and molecular programs underlying HSC-primed HEC specification in vivo will facilitate future investigations directing HSC production in vitro.Siyuan Hou Zongcheng Li Xiaona Zheng Yun Gao Ji Dong Yanli Ni Xiaobo Wang Yunqiao Li Xiaochen Ding Zhilin Chang Shuaili Li Yuqiong Hu Xiaoying Fan Yu Hou Lu Wen Bing Liu Fuchou Tang Yu Lan 2020Cell Research2020,30,5:8

6Pyrolysis mechanism of glucose and mannose: The formation of 5-hydroxymethyl furfural and furfural显示文摘Fast pyrolysis of biomass will produce various furan derivatives, among which 5-hydroxymethyl furfural(5-HMF) and furfural(FF) are usually the two most important compounds derived from holocellulose. In this study, density functional theory(DFT) calculations are utilized to reveal the formation mechanisms and pathways of 5-HMF and FF from two hexose units of holocellulose, i.e., glucose and mannose. In addition, fast pyrolysis experiments of glucose and mannose are conducted to substantiate the computational results, and the orientation of 5-HMF and FF is determined by ^(13)C-labeled glucoses. Experimental results indicate that C1 provides the aldehyde group in both 5-HMF and FF, and FF is mainly derived from C1 to C5 segment. According to the computational results, glucose and mannose have similar reaction pathways to form 5-HMF and FF with d-fructose(DF) and 3-deoxy-glucosone(3-DG) as the key intermediates. 5-HMF and FF are formed via competing pathways. The formation of 5-HMF is more competitive than that of FF, leading to higher yield of 5-HMF than FF from both hexoses. In addition, compared with glucose,mannose can form 5-HMF and FF via extra pathways because of the epimerization at C2 position. 7MXene-composited highly stretchable,sensitive and durable hydrogel for flexible strain sensors显示文摘The flourishing development in flexible electronics has provoked intensive research in flexible strain sensors to realize accurate perception acquisition under different external stimuli.However,building hydrogel-based strain sensors with high stretchability and sensitivity remains a great challenge.Herein,MXene nanosheets were composited into polyacrylamide-sodium alginate matrix to construct mechanical robust and sensitive double networked hydrogel strain sensor.The hydrophilic MXene nanosheets formed strong interactions with the polymer matrix and endowed the hydrogel with excellent tensile properties(3150%),compliant mechanical strength(2.03 kPa^(-1)in Young’s Module)and long-lasting stability and fatigue resistance(1000 dynamic cycles under 1,600%strain).Due to the highly oriented MXene-based three dimensional conductive networks,the hydrogel sensor achieved extremely high tensile sensitivity(18.15 in gauge factor)and compression sensitivity(0.38 kPa^(-1)below 3 kPa).MXene hydrogel-based strain sensors also displayed negligible hysteresis in electromechanical performance,typical frequent-independent feature and rapid response time to external stimuli.Moreover,the sensor exhibited accurate response to different scales of human movements,providing potential application in speech recognition,expression recognition and handwriting verification.Wei Yuan Xinyu Qu Yao Lu Wen Zhao Yanfang Ren Qian Wang Wenjun Wang Xiaochen Dong 2021Chinese Chemical Letters2021,32,6:3
8Ultradurable,freeze-resistant,and healable MXene-based ionic gels for multi-functional electronic skin显示文摘Hydrogel is a potential matrix material of electronic-skins(E-skins)because of its excellent ductility,tunability,and biocompatibility.However,hydrogel-based E-Skins will inevitably lose their sensing performance in practical applications for water loss,physical damage,and ambient interferences.It remains a challenge to manufacture highly durable gel-based E-skins.Herein,an E-Skin is fabricated by introducing ionic liquids(ILs)into MXene-composited binary polymer network.The obtained ionic gel shows excellent mechanical properties,strong adhesion,and superior tolerance to harsh environments.The E-skin exhibits high sensitivity to both strain and pressure in a wide range of deformations,which enables a monitoring function for various human motions and physiological activities.Importantly,the E-skin shows consistent electrical response after being stored in the open air for 30 days and can be quickly healed by irradiation with 808 nm near-infrared light,originating from the photo-thermal effect induced self-healing acceleration.It is noteworthy that the E-skin also reveals a highly sensitive perception of temperature and near-infrared light,displaying the promising potential applications in the multifunctional flexible sensor.Yao Lu Xinyu Qu Siying Wang Ye Zhao Yanfang Ren Wenli Zhao Qian Wang Chencheng Sun Wenjun Wang Xiaochen Dong 2022Nano Research2022,15,5:2

11Arginine methylation of ribose-5-phosphate isomerase A senses glucose to promote human colorectal cancer cell survival显示文摘Cancer cells remodel their metabolic network to adapt to variable nutrient availability. Pentose phosphate pathway(PPP) plays protective and biosynthetic roles by oxidizing glucose to generate reducing power and ribose. How cancer cells modulate PPP activity in response to glucose supply remains unclear. Here we show that ribose-5-phosphate isomerase A(RPIA), an enzyme in PPP, directly interacts with co-activator associated arginine methyltransferase 1(CARM1) and is methylated at arginine 42(R42). R42 methylation up-regulates the catalytic activity of RPIA. Furthermore, glucose deprivation strengthens the binding of CARM1 with RPIA to induce R42 hypermethylation. Insufficient glucose supply links to RPIA hypermethylation at R42, which increases oxidative PPP flux. RPIA methylation supports ROS clearance by enhancing NADPH production and fuels nucleic acid synthesis by increasing ribose supply. Importantly, RPIA methylation at R42 significantly potentiates colorectal cancer cell survival under glucose starvation.
