1Expression of plasma vascular endothelial growth factor in patients with hepatocellular carcinoma and effect of transcatheter arterial chemoembolization therapy on plasma vascular endothelial growth factor level显示文摘AIM: To investigate the expression level of plasma vascularendothelial growth factor (P-VEGF) in patients withhepatocellular carcinoma (HCC) and its relationship withthe clinicopathologic characteristics, and to examine thechanges of P-VEGF in the course of transcatheter arterialchemoembolization (TACE).METHODS: Peripheral blood samples were taken from 45HCC patients before and 1, 3, 7 d, and 1 mo after TACE.Plasma VEGF level was measured with the quantitativesandwich enzyme-linked immunosorbent assay (ELISA).Twenty patients with benign liver lesions and 17 healthycontrol subjects were also included in this study.RESULTS: Plasma VEGF levels in HCC patients weresignificantly elevated as compared to those in patients withbenign liver lesions (P = 0.006) and in the normal controls(P = 0.003). Significant differences were observed whenP-VEGF was categorized by tumor size (P = 0.006), portalvein thrombosis (P= 0.011), distant metastasis (P= 0.017),arterial-portal vein shunting (P = 0.026), and InternationalUnion Against Cancer (UICC) TNM stage (P = 0.044). Therewas no correlation between plasma level of VEGF and thelevel of alpha fetoprotein (^-FP) (r = 0.068, P = 0.658) andweakly correlated with the number of platelets (r = 0.312,P = 0.038). P-VEGF levels increased significantly andreached the peak value on the first day after TACE, and thendecreased gradually. The change rate of P-VEGF concentration(one month post-TACE/pre-TACExl00%) was correlatedwith the retention rate of lipiodol oil (rs = 0.494, P= 0.001)and the tumor volume change (r s = 0.340, P = 0.034).The patients who achieved a partial or complete responseto TACE therapy showed significantly less pre-treatmentP-VEGF than those nonresponders (P = 0.025). A high pre-therapeutic P-VEGF level was associated with poor responseto treatment (P = 0.018).CONCLUSION: A high pre-treatment P-VEGF level is auseful marker for tumor nroeression, esBeciallv for vascularinvasion. 2Risk Identification of Seismic Landslides by Joint Newmark and RockFall Analyst Models: A Case Study of Roads Affected by the Jiuzhaigou Earthquake显示文摘Geological disasters are a great threat to people's lives and property. At present, it is difficult to evaluate quantitatively the cascading effects of regional geological disasters, and the development of new methods for such evaluation is much needed. In this study, the authors have developed a joint procedure that couples the Newmark model and the RockFall Analyst model based on a GIS platform in order to identify the impact of seismic landslides on roads. The new method effectively combines two processes—seismic landslide occurrence probability analysis and mass movement trajectory simulation. The permanent displacement derived from the Newmark model is used to identify potential source areas of landslides.Based on the RockFall Analyst model, the possible impact of mass movement on the roads can be simulated. To verify the reliability of the method, the landslides induced by the 2017 Jiuzhaigou Earthquake were taken as a case study.The results suggest that about 21.37% of the study area is at high risk of seismic landslides, and approximately 3.95 km of road sections are at extremely high risk of large landslides. The simulated area is consistent with the distribution of disasters revealed by post-earthquake remote sensing image interpretation and field investigation in existing studies. 9Anchoring CoP nanoparticles on the octahedral CoO by self-phosphating for enhanced photocatalytic overall water splitting activity under visible light显示文摘Designing an effective and stable composite photocatalyst is of significance for the further realization of practical applications. In this study, a series of CoP/CoO composites are successfully prepared by a straight one-step phosphating method. The reasonable design and controllable preparation of CoP/CoO composite make it exhibit improved photocatalytic performance for overall water splitting and excellent stability under visible light irradiation in comparison with pure CoO, which is derived from the CoP nanoparticles well dispersed on the(111) facets of CoO octahedrons, intimate interface between them and efficiently accelerated of photo-induced electrons from CoO to CoP. 12Integrated genomic and transcriptomic analysis reveals genes associated with plant height of foxtail millet显示文摘Foxtail millet(Setaria italica)is an important C4 model crop;however,due to its high-density planting and high stature,lodging at the filling stage resulted in a serious reduction in yield and quality.Therefore,it is imperative to identify and deploy the genes controlling foxtail millet plant height.In this study,we used a semi-dwarf line 263A and an elite high-stalk breeding variety,Chuang 29 to construct an F2 population to identify dwarf genes.We performed transcriptome analysis(RNA-seq)using internode tissues sampled at three jointing stages of 263A and Chuang 29,as well as bulk segregant analysis(BSA)on their F2 population.A total of 8918 differentially expressed genes(DEGs)were obtained from RNA-seq analysis,and GO analysis showed that DEGs were enriched in functions such as‘‘gibberellin metabolic process”and‘‘oxidoreductase activity”,which have previously been shown to be associated with plant height.A total 593 mutated genes were screened by BSA-seq method.One hundred and seventy-six out of the 593 mutated genes showed differential expression levels between the two parental lines,and seven genes not only showed differential expression in two or three internode tissues but also showed high genomic variation in coding regions,which indicated they play a crucial role in plant height determination.Among them,we found a gibberellin biosynthesis related GA20 oxidase gene(Seita.5G404900),which had a single-base at the third exon,leading to the frameshift mutation at 263A.Cleaved amplified polymorphic sequence assay and association analysis proved the single-base in Seita.5G404900 co-segregated with dwarf phenotype in two independent F2 populations planted in entirely different environments.Taken together,the candidate genes identified in this study will help to elucidate the genetic basis of foxtail millet plant height,and the molecular marker will be useful for marker-assisted dwarf breeding.Mengyuan Zhu Qiang He Mingjie Lyu Tiantian Shi Qian Gao Hui Zhi Huan Wang Guanqing Jia Sha Tang Xiliu Cheng Rui Wanga Andi Xu Haigang Wang Zhijun Qiao Jun Liu Xianmin Diao Ying Gao 2023The Crop Journal2023,11,2:0
13TEAseq-based identification of 35,696 Dissociation insertional mutations facilitates functional genomic studies in maize显示文摘In plants,transposable element(TE)-triggered mutants are important resources for functional genomic studies.However,conventional approaches for genome-wide identification of TE insertion sites are costly and laborious.This study developed a novel,rapid,and high-throughput TE insertion site identification workflow based on next-generation sequencing and named it Transposable Element Amplicon Sequencing(TEAseq).Using TEAseq,we systemically profiled the Dissociation(Ds)insertion sites in 1606 independent Ds insertional mutants in advanced backcross generation using K17 as background.The Ac-containing individuals were excluded for getting rid of the potential somatic insertions.We characterized 35,696 germinal Ds insertions tagging 10,323 genes,representing approximately 23.3%of the total genes in the maize genome.The insertion sites were presented in chromosomal hotspots around the ancestral Ds loci,and insertions occurred preferentially in gene body regions.Furthermore,we mapped a loss-of-function AGL2 gene using bulked segregant RNA-sequencing assay and proved that AGL2 is essential for seed development.We additionally established an open-access database named MEILAM for easy access to Ds insertional mutations.Overall,our results have provided an efficient workflow for TE insertion identification and rich sequence-indexed mutant resources for maize functional genomic studies.Mingjie Lyu Huafeng Liu Joram Kiriga Waititu Ying Sun Huan Wang Junjie Fu Yanhui Chen Jun Liu Lixia Ku Xiliu Cheng 2021Journal of Genetics and Genomics2021,48,11:0
