1SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li 2019Horticulture Research2019,6,1:39

3Case-based learning in education of Traditional Chinese Medicine: a systematic review显示文摘OBJECTIVE:To assess the effect of case-based learning(CBL)in the education of Traditional Chinese Medicine(TCM).METHODS:The studies concerning TCM courses designed with CBL were included by searching the databases of EBSCO,Pubmed,Science Citation Index,China National Knowledge Infrastructure,Chongqing VIP database.The valid data was extracted in accordance with the included criteria.The quality of the studies was assessed with Gemma Flores-Masteo.RESULTS:A total of 22 articles were retrieved that met the selection criteria:one was of high quality;two were of low quality;the rest were categorized as moderate quality.The majority of the studiesdemonstrated the better effect produced by CBL,while a few studies showed no difference,compared with the didactic format.All included studies confirmed the favorable effect on learners'attitude,skills and ability.CONCLUSION:CBL showed the desirable results in achieving the goal of learning.Compared to didactic approach,it played a more active role in promoting students'competency.Since the quality of the articles on which the study was based was not so high,the findings still need further research to become substantiated.Ji Chen Ying Li Yong Tang Fang Zeng Xi Wu Fanrong Liang 2013Journal of Traditional Chinese Medicine2013,33,5:30
4Behaviors and kinetics of toluene adsorption-desorption on activated carbons with varying pore structure显示文摘This work was undertaken to investigate the behaviors and kinetics of toluene adsorption and desorption on activated carbons with varying pore structure. Five kinds of activated carbon from different raw materials were selected. Adsorption isotherms and breakthrough curves for toluene were measured. Langmuir and Freundlich equations were fitted to the equilibrium data, and the Freundlich equation was more suitable for simulating toluene adsorption. The process consisted of monolayer, multilayer and partial active site adsorption types. The effect of the pore structure of the activated carbons on toluene adsorption capacity was investigated. The quasi-first-order model was more suitable for describing the process than the quasi-second-order model. The adsorption data was also modeled by the internal particle diffusion model and it was found that the adsorption process could be divided into three stages. In the external surface adsorption process, the rate depended on the specific surface area. During the particle diffusion stage, pore structure and volume were the main factors affecting adsorption rate. In the final equilibrium stage, the rate was determined by the ratio of meso-and macro-pores to total pore volume. The rate over the whole adsorption process was dominated by the toluene concentration. The desorption behavior of toluene on activated carbons was investigated,and the process was divided into heat and mass transfer parts corresponding to emission and diffusion mechanisms, respectively. 6Effect of severe acute pancreatitis on pharmacokinetics of Da-Cheng-Qi Decoction components显示文摘AIM:To investigate the effect of severe acute pan- creatitis(SAP)on pharmacokinetics of Da-Cheng-Qi Decoction(DCQD)components in rats. METHODS:Rats were divided into SAP group and sham-operation group as a control group(n=6). Rhein,chrysophanol,rheochrysidin,magnolol,hesperidin and naringin in DCQD were quantified in rat serum by high performance liquid chromatography tandem mass spectrometry for studying their pharmacokinetics. RESULTS:Early absorption of each DCQD component was tended to degrade in SAP group after treatment with DCQD by gavage.The Cmax(chrysophanol,P= 0.0059;rheochrysidin,P=0.0288;magnolol,P= 0.0487;hesperidin,P=0.0277;naringin,P=0.0023) and AUC(rhein,P=0.0186;chrysophanol,P=0.0013; magnolol,P=0.001;hesperidin,P=0.0081;naringin, P=0.0272)of DCQD component were obviously lower in SAP group than in control group.The T1/2α of chrysophanol and rheochrysidin(P=0.0467 and 0.0005,respectively)and Tmax of chrysophanol and rheochrysidin(P=0.0101 and 0.0037,respectively) lasted longer in SAP group than in control group. 10High HIV incidence epidemic among men who have sex with men in china: results from a multi-site cross-sectional study显示文摘Background:Recent upsurge of new HIV infections among men who have sex with men(MSM)is a major concern in China.Paucity of national-level information regarding the burden and predictors of this progressive epidemic of new infections called for a multi-centric,timely and comprehensive investigation.Methods:Mixed methods were used to recruit MSM from seven cities in China between 2012 and 2013.Recent and established HIV infections were estimated by Western Blot and BED HIV-1 capture enzyme immunoassay.Syphilis and herpes simplex virus-2(HSV-2)were also tested.Results:A total of 4496 eligible MSM were recruited.The majority was aged≤35 years(77.5%),migrants(60.3%),never married(69.8%),and played receptive role in anal sex(70.5%).The HIV prevalence was 9.9%,and 41.9%were recently infected,with sensitivity/specificity adjusted HIV incidence of 8.9(95%CI:7.6-10.2)/100 Person-Years.The prevalence of history HSV-2 and syphilis were 12.5%and 8.5%,respectively.Recent HIV infection was associated with having multiple male partners(aOR=1.4,95%CI 1.1-1.9),recreational drug use(aOR=2.2,95%CI 1.6-3.0),anal bleeding(aOR=2.1,95%CI 1.4-3.0),syphilis infection(aOR=2.8,95%CI 1.9-4.3)and history HSV-2 infection(aOR=2.3,95%CI 1.5-3.3).Conclusion:High rate of recent HIV infection is potentially resulting in progressive deterioration of the overall HIV epidemic among MSM in China.Targeted interventions to address high-risk MSM including those having multiple partners,history of recreational drug use and syphilis or HSV-2 infection seemed to be the need of the hour.Jun-Jie Xu Wei-Ming Tang Hua-Chun Zou Tanmay Mahapatra Qing-Hai Hu Geng-Feng Fu Zhe Wang Lin Lu Ming-Hua Zhuang Xi Chen Ji-Hua Fu Yan-Qiu Yu Jin-Xin Lu Yong-Jun Jiang Wen-Qing Geng Xiao-Xu Han Hong Shang 2016Infectious Diseases of Poverty2016,5,1:14
11Evaluation on the accuracy of digital elevation models显示文摘There is a growing interest in investigating the accuracy of digital elevation model (DEM). However people usually have an unbalanced view on DEM errors. They emphasize DEM sampling errors, but ignore the impact of DEM resolution and terrain roughness on the accuracy of terrain representation. This research puts forward the concept of DEM terrain representation error (Et) and then investigates the generation, factors, measurement and simulation of DEM terrain representation errors. A multi-resolution and multi-relief comparative approach is used as the major methodology in this research. The experiment reveals a quantitative relationship between the error and the variation of resolution and terrain roughness at a global level. Root mean square error (RMS Et) is regressed against surface profile curvature (V) and DEM resolution (R) at 10 resolution levels. It is found that the RMS Et may be expressed as RMS Et = (0.0061 ( V+ 0.0052) ( R - 0.022 ( V + 0.2415. 12A multicenter hospital-based diagnosis study of automated breast ultrasound system in detecting breast cancer among Chinese women显示文摘Objective: The automated breast ultrasound system(ABUS) is a potential method for breast cancer detection;however, its diagnostic performance remains unclear. We conducted a hospital-based multicenter diagnostic study to evaluate the clinical performance of the ABUS for breast cancer detection by comparing it to handheld ultrasound(HHUS) and mammography(MG).Methods: Eligible participants underwent HHUS and ABUS testing; women aged 40–69 years additionally underwent MG. Images were interpreted using the Breast Imaging Reporting and Data System(BI-RADS).Women in the BI-RADS categories 1–2 were considered negative. Women classified as BI-RADS 3 underwent magnetic resonance imaging to distinguish true-and false-negative results. Core aspiration or surgical biopsy was performed in women classified as BI-RADS 4–5, followed by a pathological diagnosis. Kappa values and agreement rates were calculated between ABUS, HHUS and MG.Results: A total of 1,973 women were included in the final analysis. Of these, 1,353(68.6%) and 620(31.4%)were classified as BI-RADS categories 1–3 and 4–5, respectively. In the older age group, the agreement rate and Kappa value between the ABUS and HHUS were 94.0% and 0.860(P<0.001), respectively; they were 89.2% and0.735(P<0.001) between the ABUS and MG, respectively. Regarding consistency between imaging and pathology results, 78.6% of women classified as BI-RADS 4–5 based on the ABUS were diagnosed with precancerous lesions or cancer; which was 7.2% higher than that of women based on HHUS. For BI-RADS 1–2, the false-negative rates of the ABUS and HHUS were almost identical and were much lower than those of MG.Conclusions: We observed a good diagnostic reliability for the ABUS. 13Saline soil enzyme activities of four plant communities in Sangong River basin of Xinjiang,China显示文摘Soil enzyme activity plays an important role in the conversion of soil organic carbon into inorganic carbon,which is significant for the global carbon cycle.In this study,we investigated the soil enzyme activities of two ligninolytic enzymes(peroxidase and polyphenol oxidase)and five non-ligninolytic enzymes(α-1,4-glucosidase(AG);β-1,4-glucosidase(BG);N-acetyl-β-glucosaminidase(NAG);β-D-cellobiosidase(CBH);andβ-xylosidase(BXYL))in four plant communities of the Sangong River basin in Fukang,North Xinjiang,China.The four typical plant communities were dominated by Haloxylon ammodendron,Reaumuria soongonica,Salsola passerina,and Tamarix rarmosissima,respectively,with saline soils of varied alkalinity.The results showed that the soil peroxidase activity decreased seasonally.The activities of the five non-ligninolytic enzymes decreased with increasing soil depths,while those of the two ligninolytic enzymes did not show such a trend.In the four plant communities,BG had the highest activity among the five non-ligninolytic enzymes,and the activities of the two ligninolytic enzymes were higher than those of the four non-ligninolytic ones(AG,NAG,CBH,and BXYL).The community of H.ammodendron displayed the highest activity with respect to the two ligninolytic enzymes in most cases,but no significant differences were found among the four plant communities.The geometric mean of soil enzyme activities of the four plant communities was validated through an independently performed principal component analysis(PCA),which indicated that different plant communities had different soil enzyme activities.The correlation analysis showed that soil polyphenol oxidase activity was significantly positively correlated with the activities of the five non-ligninolytic enzymes.The soil pH value was positively correlated with the activities of all soil enzymes except peroxidase.Soil microbial carbon content also showed a significant positive correlation(P<0.01)with the activities of all soil enzymes except polyphenol oxidase.The results suggested that the H.ammodendron community has the highest ability to utilize soil organic carbon,and glucoside could be the most extensively utilized non-ligninolytic carbon source in the saline soil of arid areas in Xinjiang.ZhengJun GUAN Qian LUO Xi CHEN XianWei FENG ZhiXi TANG Wei WEI YuanRun ZHENG 2014Journal of Arid Land2014,6,2:11
14Outcomes in refractory diffuse large B-cell lymphoma:results fromamulticenter real-world study in China显示文摘Background:Diffuse large B-cell lymphoma(DLBCL)patients refractory to rituximab-based immunochemotherapy have a dismal prognosis.However,the definition of refractory DLBCL remains inconsistent and no large cohort study data is available from Asian countries.To validate the definition and outcomes of refractory DLBCL in China,we conducted a multicenter,retrospective cohort study.Methods:The REtrospective AnaLysis of Treatment REspoNse of refractory DLBCL(REAL-TREND)study was performed using real-world data from 8 centers in China.DLBCL patients with curative intent were included in the REAL-TREND dataset.Overall survival(OS)was estimated using the Kaplan-Meier method and compared by the log-rank test.Due to heterogeneity in response rates among different centers,the response rates of refractory patients were pooled using random-effect models.Multivariate survival analysis was performed using the Cox regression model.Results:A total of 2778 DLBCL patients diagnosed between January,2010 and December,2015 were enrolled to this study.After validating previous definitions,the SCHOLAR-1 study was most suitable to define refractory DLBCL.The estimated 5-year cumulative incidence of refractory patients was 20%(95% confidence Interval[CI]=18%-22%).After the determination of refractory disease,overall response rate and complete remission rate were 30%(95%CI=22%-38%)and 9%(95%CI=4%-15%),respectively.Patients with either no response to immunochemotherapy or relapse within 12 months after stem-cell transplantation had inferior survival with a median OS of 5.9 months(95%CI=5.5-7.1 months)and 2-year OS rate of 16%(95%CI=12%-20%).International prognostic index score 4-5(hazard ratio[HR]=2.22;95%CI=1.47-3.35),central nervous systemrelapse(HR=1.43;95%CI=1.04-1.97),and best response status(HR=2.68;95%CI=1.42-5.03 for partial remission.HR=5.97,95%CI=3.21-11.11 for stable disease/progressive disease)were independent unfavorable prognostic factors.Conclusions:This is the first large-scale Asian cohort study focusing on outcomes of refractory DLBCL.The definition of the SCHOLAR-1 study identifies patients with homogenously inferior survival,thus is appropriate to select refractory DLBCL.Due to poor clinical outcomes in the rituximab era,patients with refractory DLBCL may be potential candidates for novel treatment modalities.Shuo Wang Li Wang Jianda Hu Wenbin Qian Xi Zhang Yu Hu Qi Zhu Bobin Chen Depei Wu Chung-Chou H.Chang Pengpeng Xu Xiaoyun Zheng Juying Wei Yao Liu Guohui Cui Yong Tang Yan Ma Haiwen Huang Hongmei Yi Weili Zhao 2021Cancer Communications2021,41,3:10
15Extraction, identification, and antioxidant property evaluation of limonin from pummelo seeds显示文摘Limonin, the main bioactive phytochemical constituent of limonoids with multi-functions, is enriched in citrus fruits and often found at a high concentration in citrus seeds. The present study was attempted to introduce a new and efficient extraction method to isolate limonoids from pummelo seeds, and to evaluate the antioxidant property of the main constituent limonin in Hep G2 cells. Three key single factors were identified for the extraction of limonoids from pummelo seeds using the Box-Behnken experiment design of response surface methodology(RSM), and the optimized extraction parameters were treatment with 89.68 m L of anhydrous acetone for 4.62 h at 78.94C, while the yield of limonoids was 11.52 mg/g. The structure of isolated main constituent of the limonoids was further identified as limonin by Fourier transform infrared(FT-IR) spectrometer and nuclear magnetic resonance(NMR)spectrum. Moreover, the molecular data in Hep G2 cells revealed that limonin exerted its anti-oxidant property mainly by the activation of nuclear factor(erythroid-2)-like 2(Nrf2)/kelch-like ECHassociated protein 1(Keap1)-antioxidant response element(ARE) pathway in the form of transcriptional regulation of Nrf2 m RNA and posttranscriptional regulation of Nrf2/Keap1 system. 18Plasma metabolomic and lipidomic alterations associated with COVID-19显示文摘The pandemic of the coronavirus disease 2019(COVID-19)has become a global public health crisis.The symptoms of COVID-19 range from mild to severe,but the physiological changes associated with COVID-19 are barely understood.In this study,we performed targeted metabolomic and lipidomic analyses of plasma from a cohort of patients with COVID-19 who had experienced different symptoms.We found that metabolite and lipid alterations exhibit apparent correlation with the course of disease in these patients,indicating that the development of COVID-19 affected their whole-body metabolism.In particular,malic acid of the TCA cycle and carbamoyl phosphate of the urea cycle result in altered energy metabolism and hepatic dysfunction,respectively.It should be noted that carbamoyl phosphate is profoundly down-regulated in patients who died compared with patients with mild symptoms.And,more importantly,guanosine monophosphate(GMP),which is mediated not only by GMP synthase but also by CD39 and CD73,is significantly changed between healthy subjects and patients with COVID-19,as well as between the mild and fatal cases.In addition,dyslipidemia was observed in patients with COVID-19.Overall,the disturbed metabolic patterns have been found to align with the progress and severity of COVID-19.This work provides valuable knowledge about plasma biomarkers associated with COVID-19 and potential therapeutic targets,as well as an important resource for further studies of the pathogenesis of COVID-19.Di Wu Ting Shu Xiaobo Yang Jian-Xin Song Mingliang Zhang Chengye Yao Wen Liu Muhan Huang Yuan Yu Qingyu Yang Tingju Zhu Jiqian Xu Jingfang Mu Yaxin Wang Hong Wang Tang Tang Yujie Ren Yongran Wu Shu-Hai Lin Yang Qiu Ding-Yu Zhang You Shang Xi Zhou 2020National Science Review2020,7,7:9
19Anomaly Models of Spatial Structures for Copper-Molybdenum Ore Deposits and their Application显示文摘This paper discusses the enrichment and depletion regularities for porphyry copper-molybdenum ore deposits in different regions and varied deposit genetic types in the same area, taking three porphyry copper-molybdenum ore deposits (i.e., the Chengmenshan in Jiangxi, Wunugetushan in Inner Mongolia, Baishantang in Gansu) and two copper deposits in Gansu Province (the Huitongshan skarn deposit and Gongpoquan composite deposit) as case studies. The results show that porphyry Cu-Mo deposits or skarn copper deposits include both enrichment of the ore-forming elements and associated elements, and depletion of some lithophile dispersed elements, rare earth elements (REE) and some major elements. And the depleted elements vary with deposits, having generality and their own features. On a deposit scale, the positive anomalies of enriched elements and negative anomalies of depleted elements follow in a sequence to comprise regular anomaly models of spatial structures. The exploration in the Tongchang deposit in Jiangxi and Huitongshan deposit in Gansu suggests that anomaly models play a key role in the identification of mineral occurrences and deposits compared to one single enriched element anomaly.
