1An aptamer-based shear horizontal surface acoustic wave biosensor with a CVD-grown single- layered graphene film for high-sensitivity detection of a label-free endotoxin显示文摘The thickness of the sensitive layer has an important influence on the sensitivity of a shear horizontal surface acoustic wave(SH-SAW)biosensor with a delay-line structure and lower layer numbers of graphene produce better sensitivity for biological detection.Therefore,a label-free and highly sensitive SH-SAW biosensor with chemical vapor deposition(CVD-)-grown single-layered graphene(SLG)for endotoxin detection was developed in this study.With this methodology,SH-SAW biosensors were fabricated on a 36°Y-90°X quartz substrate with a base frequency of 246.2MHz,and an effective detection cell was fabricated using acrylic material.To increase the surface hydrophilicity,chitosan was applied to the surface of the SLG film.Additionally,the aptamer was immobilized on the surface of the SLG film by cross-linking with glutaraldehyde.Finally,the sensitivity was verified by endotoxin detection with a linear detection ranging from 0 to 100 ng/mL,and the detection limit(LOD)was as low as 3.53 ng/mL.In addition,the stability of this type of SH-SAW biosensor from the air phase to the liquid phase proved to be excellent and the specificity was tested and verified by detecting the endotoxin obtained from Escherichia coli(E.coli),the endotoxin obtained from Pseudomonas aeruginosa(P.aeruginosa),and aflatoxin.Therefore,this type of SH-SAW biosensor with a CVD-grown SLG film may offer a promising approach to endotoxin detection,and it may have great potential in clinical applications.Junwang Ji Yiquan Pang Dongxiao Li Zheng Huang Zuwei Zhang Ning Xue Yi Xu Xiaojing Mu 2020Microsystems & Nanoengineering2020,6,1:3
2A newly defined dioxygenase system from Mycobacterium vanbaalenii PYR-1 endowed with an enhanced activity of dihydroxylation of high-molecular-weight polyaromatic hydrocarbons显示文摘NidA3B3 is a terminal dioxygenase whose favorable substrates are high-molecular-weight polyaromatic hydrocarbons(PAHs)from Mycobacterium vanbaalenii PYR-1,a powerftil PAHs degradation strain.NidA3B3 was reported to incorporate a dioxygen into the benzene ring of PAHs when equipped with an exogenous electron transport chain components PhdCD from Nocarciioides sp.strain KP7 by biotransformation,but this enzyme system was not particularly efficient.In this study,strain PYR-1 was confirmed to utilize four different PAHs at different growth rates.When PhtAcAd,an endogenous electron transport chain of a phthalate dioxygenase system,was substituted for PhdCD to couple with NidA3B3,the specific activity to convert phenanthrene by strain BL21(DE3)[pNidA3B3-PhAcAd]was 0.15±0.03 U/mg,but the specific activity of strain BL21(DE3)[pNidA3B3-PhdCD]was only 0.025±0.006U/mg.In addition,FNidA3,encoded by a newly defined ORF,has a prolonged 19-amino acid sequence at the N-terminus compared with NidA3.FNidA3B3 increased the activity by 50%approximately than NidA3B3 when using PhtAcAd.Components of the electron transport chain PhtAc and PhtAd were purified and characterized.The Km,kcal,kcat/Km values of the PhtAd were 123±26.9 pM,503±49.9 min^-1,4.1μM^-1·min^-1,respectively.And the,Km,Kcat,Kcat/Km values of the ferredoxin PhtAc were (52.5±9.7)μM,3.8±0.19min^-1 and 0.07μM^-1 min1,respectively.Basing on the phylogenetic analysis,NidA3/FNidA3 were far from its isoenzyme NidA from the same strain.Combining their primary differences of transcriptional pattern in vivo,it indicated that the functionally similar Rieske dioxygenases NidA3B3/FNidA3B3 and NidAB might originate from different ancestors.Yiquan Wu Ying Xu Ningyi Zhou 2020Frontiers of Environmental Science & Engineering2020,14,1:2
3The impact of mean dynamic topography on a sea-level anomaly assimilation in the South China Sea based on an eddy-resolving model显示文摘The sea-level anomaly (SLA) from a satellite altimeter has a high accuracy and can be used to improve ocean state estimation by assimilation techniques. However, the lack of an accurate mean dynamic topography (MDT) is still a bothersome issue in an ocean data assimilation. The previous studies showed that the errors in MDT have significant impacts on assimilation results, especially on the time-mean components of ocean states and on the time variant parts of states via nonlinear ocean dynamics. The temporal-spatial differences of three MDTs and their impacts on the SLA analysis are focused on in the South China Sea (SCS). The theoretical analysis shows that even for linear models, the errors in MDT have impacts on the SLA analysis using a sequential data assimilation scheme. Assimilation experiments, based on EnOI scheme and HYCOM, with three MDTs from July 2003 to June 2004 also show that the SLA assimilation is very sensitive to the choice of different MDTs in the SCS with obvious differences between the experimental results and observations in the centre of the SCS and in the vicinity of the Philippine Islands. A new MDT for assimilation of SLA data in the SCS was proposed. The results from the assimilation experiment with this new MDT show a marked reduction (increase) in the RMSEs (correlation coefficient) between the experimental and observed SLA. 10MORPHOLOGY OF BLACK CARBON AEROSOLS AND UBIQUITY OF 50-NANOMETER BLACK CARBON AEROSOLS IN THE ATMOSPHERE显示文摘Different-sized aerosols were collected by an Andersen air sampler to observe the detailed morphology of the black carbon (BC) aerosols which were separated chemically from the other accompanying aerosols, using a Scan-ning Electron Microscope equipped with an Energy Dispersive X-ray Spectrometer (SEM-EDX). The results indicate that most BC aerosols are spherical particles of about 50 nm in diameter and with a homogeneous surface. Results also show that these particles aggregate with other aerosols or with themselves to form larger agglomerates in the micrometer range. The shape of these 50-nm BC spherical particles was found to be very similar to that of BC particles released from petroleum-powered vehicular internal combustion engines. 12Interface construction of NiCo LDH/NiCoS based on the 2D ultrathin nanosheet towards oxygen evolution reaction显示文摘Carbon-free hydrogen as a promising clean energy source can be produced with electrocatalysts via water electrolysis.Oxygen evolution reaction(OER)as anodic reaction determines the overall efficiency of water electrolysis due to sluggish OER kinetics.Thus,it’s much desirable to explore the efficient and earth-abundant transition-metal-based OER electrocatalysts with high current density and superior stability for industrial alkaline electrolyzers.Herein,we demonstrate a significant enhancement of OER kinetics with the hybrid electrocatalyst arrays in alkaline via judiciously combining earth-abundant and ultrathin NiCo-based layered double hydroxide(NiCo LDH)nanosheets with nickel cobalt sulfides(NiCoS)with a facile metal-organic framework(MOF)-template-involved surface sulfidation process.The obtained NiCo LDH/NiCoS hybrid arrays exhibits an extremely low OER overpotential of 308 mV at 100 mA·cm^(−2),378 mV at 200 mA·cm^(−2)and 472 mV at 400 mA·cm^(−2)in 1 M KOH solution,respectively.A much low Tafel slope of 48 mV·dec^(−1)can be achieved.Meanwhile,with the current density from 50 to 250 mA·cm^(−2),the NiCo-LDH/NiCoS hybrid arrays can run for 25 h without any degradation.Our results demonstrate that the construction of hybrid arrays with abundant interfaces of NiCo LDH/NiCoS can facilitate OER kinetics via possible modulation of binding energy of O-containing intermediates in alkaline media.The present work would pave the way for the development of lowcost and efficient OER catalysts and industrial application of water alkaline electrolyzers.Jiahui Li Lili Wang Haojia He Yiquan Chen Zirui Gao Na Ma Bing Wang Linlin Zheng Rulin Li Yujia Wei Junqing Xu Yao Xu Bowen Cheng Zhen Yin Ding Ma 2022Nano Research2022,15,6:0

14Analytical Solution for an Elastic Sphere Indenting a Layered Half-Space显示文摘The contact problem for the elastic sphere indenting a layered half-space is considered. Analytical methods for solving this problem have been developed on the basis of the 3-D fundamental solution of a half space with a single coating layer under a normal concentrated force on the surface. The normal pressure distribution within the contact zone is assumed as Hertzian type. The solutions are constructed using superposition principle in the form of infinite series. Through comparing with the numerical results of FEM,it can be verified that the exact solutions have a rapid convergence rate and the stresses and displacements are mainly determined by the first term,which is corresponding to the solution of homogeneous half-space under Hertzian loading. 15Thermally Conductive Study of Polyethylene/Al2O3 Composite Networks Cross-linked by Electron Beam Irradiation显示文摘The surface-modifled Al2O3 particles were introduced into polyethylene(PE)to enhance the thermal conductivity,and PE/Al2O3 cross-linked networks with improved thermal and mechanical properties were prepared through electron beam(EB)irradiation technology.The incorporation of reactive irradiation sensitizer was useful in fabricating a high degree of cross-linking(DC)PE networks under a low irradiation dose.In the PE sample containing 2%sensitizer,DC ca.67.1%could be obtained under 60 kGy(1 kGy=1000 J/kg).EB-irradiation greatly improved thetensile stress of PE-based samples,and the tensile stresses of the samples with 0.2—5%TMPTA(trimethylolpr pane triacrvlate)under 60 kGy were 24.61—27.77 MPa.All the EB-irradiated samples had higher Vicat softening temperatures than the samples without irradiation.After treatment at 120 kGy,the Vicat softening temperatures of PE-Al2O3-44/TMPTA-2 increased from 127℃to 130.4℃.SEM images revealed that PE-Al2O3-5O samples with increased amount of Al2O3 particles showed a conduction'pathway,'and thennal conductivity reached 0.67 W/(m·K).Thus,high-performance pipes were extruded,which could satisfy the static hydraulic blasting test and exhibit improved thennal conduction capability.SHI Chengying YANG Jiayu LIU Yang WANG Yanmiao XU Wenge XU Yiquan HU Wei LIU Baijun 2020Chemical Research in Chinese Universities2020,36,5:0
16Transient axonal glycoprotein-1 induces apoptosisrelated gene expression without triggering apoptosis in U251 glioma cells显示文摘Previous studies show that transient axonal glycoprotein-1, a ligand of amyloid precursor protein, increases the secretion of amyloid precursor protein intracellular domain and is involved in apoptosis in Alzheimer's disease. In this study, we examined the effects of transient axonal glycoprotein-1 on U251 glioma cells. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay showed that transient axonal glycoprotein-1 did not inhibit the proliferation of U251 cells, but promoted cell viability. The terminal deoxynucleotidyl transferase dUTP nick end labeling assay showed that transient axonal glycoprotein-1 did not induce U251 cell apoptosis. Real-time PCR revealed that transient axonal glycoprotein-1 substantially upregulated levels of amyloid precursor protein intracellular C-terminal domain, and p53 and epidermal growth factor receptor mRNA expression. Thus, transient axonal glycoprotein-1 increased apoptosis-related gene expression in U251 cells without inducing apoptosis.
