1Rapid isolation and immune profiling of SARS-CoV-2 specific memory B cell in convalescent COVID-19 patients via LIBRA-seq显示文摘B cell response plays a critical role against SARS-CoV-2 infection.However,little is known about the diversity and frequency of the paired SARS-CoV-2 antigen-specific BCR repertoire after SARS-CoV-2 infection.Here,we performed single-cell RNA sequencing and VDJ sequencing using the memory and plasma B cells isolated from five convalescent COVID-19 patients,and analyzed the spectrum and transcriptional heterogeneity of antibody immune responses.Via linking BCR to antigen specificity through sequencing(LIBRA-seq),we identified a distinct activated memory B cell subgroup(CD11c^(high) CD95^(high))had a higher proportion of SARS-CoV-2 antigen-labeled cells compared with memory B cells.Our results revealed the diversity of paired BCR repertoire and the non-stochastic pairing of SARS-CoV-2 antigen-specific immunoglobulin heavy and light chains after SARS-CoV-2 infection.The public antibody clonotypes were shared by distinct convalescent individuals.Moreover,several antibodies isolated by LIBRA-seq showed high binding affinity against SARS-CoV-2 receptor-binding domain(RBD)or nucleoprotein(NP)via ELISA assay.Two RBD-reactive antibodies C14646P3S and C2767P3S isolated by LIBRA-seq exhibited high neutralizing activities against both pseudotyped and authentic SARS-CoV-2 viruses in vitro.Our study provides fundamental insights into B cell response following SARS-CoV-2 infection at the single-cell level.Bing He Shuning Liu Yuanyuan Wang Mengxin Xu Wei Cai Jia Liu Wendi Bai Shupei Ye Yong Ma Hengrui Hu Huicui Meng Tao Sun Yanling Li Huanle Luo Mang Shi Xiangjun Du Wenjing Zhao Shoudeng Chen Jingyi Yang Haipeng Zhu Yusheng Jie Yuedong Yang Deyin Guo Qiao Wang Yuwen Liu Huimin Yan Manli Wang Yao-Qing Chen 2021Signal Transduction and Targeted Therapy2021,6,6:2

3A Novel Hybrid Intelligent Prediction Model for Valley Deformation: A Case Study in Xiluodu Reservoir Region, China显示文摘The narrowing deformation of reservoir valley during the initial operation period threatens the long-term safety of the dam,and an accurate prediction of valley deformation(VD)remains a challenging part of risk mitigation.In order to enhance the accuracy of VD prediction,a novel hybrid model combining Ensemble empirical mode decomposition based interval threshold denoising(EEMD-ITD),Differential evolutions—Shuffled frog leaping algorithm(DE-SFLA)and Least squares support vector machine(LSSVM)is proposed.The non-stationary VD series is firstly decomposed into several stationary subseries by EEMD;then,ITD is applied for redundant information denoising on special sub-series,and the denoised deformation is divided into the trend and periodic deformation components.Meanwhile,several relevant triggering factors affecting the VD are considered,from which the input features are extracted by Grey relational analysis(GRA).After that,DE-SFLA-LSSVM is separately performed to predict the trend and periodic deformation with the optimal inputs.Ultimately,the two individual forecast components are reconstructed to obtain the final predicted values.Two VD series monitored in Xiluodu reservoir region are utilized to verify the proposed model.The results demonstrate that:(1)Compared with Discrete wavelet transform(DWT),better denoising performance can be achieved by EEMD-ITD;(2)Using GRA to screen the optimal input features can effectively quantify the deformation response relationship to the triggering factors,and reduce the model complexity;(3)The proposed hybrid model in this study displays superior performance on some compared models(e.g.,LSSVM,Backward Propagation neural network(BPNN),and DE-SFLA-BPNN)in terms of forecast accuracy.Mengcheng Sun Weiya Xu Huanling Wang Qingxiang Meng Long Yan Wei-Chau Xie 2021Computers, Materials & Continua2021,,1:1

7Preparation and Photocatalytic Properties of Ag2CO3/Graphene Composite Photocatalyst显示文摘Silver-based photocatalytic semiconducting materials have drawn the attention of reseachers for their high visible photocatalytic activity. However, the silver-based photocatalytic semiconducting material exhibits light corrosion during the photocatalytic reaction, and the photocatalytic stability is poor. Therefore, improvingthe photocatalytic stability and inhibition of light corrosion of silver-based photocatalytic semiconductor materials have been the focus of attention. In this paper, according to the principle of photocatalysis and the principle of photo-corrosion, it is proposed to improve the photogenerated electrons and hole separation of photocatalytic semiconductor materials, to rapidly transfer photogenerated electrons, to inhibit photogenerated electrons and Ag + to prevent light corrosion, Stability of the catalyst. Ag2CO3/GO composite photocatalytic materials were synthesized by precipitation method using polystyrene as photocatalyst. The characterization and photocatalytic performance tests showed that the graphene has a good auxiliary effect, which can promote the separation of photogenerated electrons and holes of Ag2CO3 and transfer the photogenerated electrons into O2 in H2O in time, thus suppressing the light of Ag2CO3 / GO photocatalytic materials Corrosion phenomenon, improve the photocatalytic performance. Ag2CO3/GO-1.0 has the best catalytic activity for the catalytic activity and stability of Ag2CO3/GO in the photocatalytic decomposition of methyl orange. 9Dendritic cell-derived IL-27 p28 regulates T cell program in pathogenicity and alleviates acute graft-versus-host disease显示文摘Interleukin 27(IL-27),a heterodimeric cytokine composed of Epstein-Barr virus-induced 3 and p28,is a pleiotropic cytokine with both pro-and anti-inflammatory properties.However,the precise role of IL-27 in acute graft-versus-host disease is not yet fully understood.In this study,utilizing mice with IL-27 p28 deficiency in dendritic cells(DCs),we demonstrated that IL-27 p28 deficiency resulted in impaired Treg cell function and enhanced effector T cell responses,corresponding to aggravated aGVHD in mice.In addition,using single-cell RNA sequencing,we found that loss of IL-27 p28 impaired Treg cell generation and promoted IL1R2^(+)TIGIT^(+)pathogenic CD4+T cells in the thymus at a steady state.Mechanistically,IL-27 p28 deficiency promoted STAT1 phosphorylation and Th1 cell responses,leading to the inhibition of Treg cell differentiation and function.Finally,patients with high levels of IL-27 p28 in serum showed a substantially decreased occurrence of grade II-IV aGVHD and more favorable overall survival than those with low levels of IL-27 p28.Thus,our results suggest a protective role of DC-derived IL-27 p28 in the pathogenesis of aGVHD through modulation of the Treg/Teff cell balance during thymic development.IL-27 p28 may be a valuable marker for predicting aGVHD development after transplantation in humans.Huanle Gong Shoubao Ma Jia Chen Bingyu Yang Shuangzhu Liu Xin Liu Jingjing Han Xiaojin Wu Lei Lei Zhinan Yin Hongjian Sun Di Yu Haiyan Liu Yang Xu Depei Wu 2022Signal Transduction and Targeted Therapy2022,7,10:0
