| 2 | Genetic analysis and fine mapping of the pubescence gene GL6 in rice (Oryza sativa L.)显示文摘The pubescence of the leaf blade surface is an important agronomic characteristic for rice morphology and significantly influences rice growth as well as physiological characteristics. This characteristic was analyzed in F1 and F2 plants derived by crossing cultivar 75-1-127 with the indica cultivar Minghui 63, as well as the glabrous cultivar Lemont and indica cultivar 9311. Results indicated that the pubescence of the leaf blade surface was a dominant trait and controlled by a single gene. The GL6 gene was primarily mapped on rice chromosome 6 with recessive F2 population derived from 75-1-127/Minghui 63 by combining bulked segregation analysis and recessive class analysis using the Mapmaker3.0/MapDraw software. The genetic distances between the simple sequence repeat markers RM20491 and RM20547 were 7.2 and 2.2 cM, respectively. The GL6 gene was fine mapped in the interval between InDel-106 and InDel-115 at genetic distances of 0.3 and 0.1 cM, respectively. The large, recessive F2 population was derived from 75-1-127/Minghui 63. A high-resolution genetic and physical map of GL6 was constructed. Derived from the map-based sequences published by the International Rice Genome Sequencing Project, the GL6 gene was localized at an interval of 79 (japonica) and 116.82 kb (9311) bracketed by InDel-106 and InDel-115 within the BAC accession numbers AP008403 and AP005760. Seven annotated genes (japonica) and eight annotated genes (9311) were present. | 4 | Physicochemical bases for protein folding,dynamics,and protein-ligand binding显示文摘Proteins are essential parts of living organisms and participate in virtually every process within cells.As the genomic sequences for increasing number of organisms are completed,research into how proteins can perform such a variety of functions has become much more intensive because the value of the genomic sequences relies on the accuracy of understanding the encoded gene products.Although the static three-dimensional structures of many proteins are known,the functions of proteins are ultimately governed by their dynamic characteristics,including the folding process,conformational fluctuations,molecular motions,and protein-ligand interactions.In this review,the physicochemical principles underlying these dynamic processes are discussed in depth based on the free energy landscape(FEL)theory.Questions of why and how proteins fold into their native conformational states,why proteins are inherently dynamic,and how their dynamic personalities govern protein functions are answered.This paper will contribute to the understanding of structure-function relationship of proteins in the post-genome era of life science research. | LI HuiMin XIE YueHui LIU CiQuan LIU ShuQun | 2014 | Science China(Life Sciences)2014,57,3: | 2 |
| 5 | LncRNA H19 inhibits oxidative stress injury of cochlear hair cells by regulating miR-653-5p/SIRT1 axis显示文摘Oxidative stress is one of the important mechanisms of inner ear cell damage,which can lead to age-related hearing loss(ARHL).LncRNA H19 is significantly downregulated in the cochlea of old mouse,however,the role of H19 in the development of ARHL remains unclear.In this study,we aim to investigate the expression and function of H19 in oxidative stress injury of cochlear hair cells induced by H_(2)O_(2).RT-qPCR and western blot analysis confirms that HEI-OC1 cells stimulated with H_(2)O_(2) decreases the expressions of H19 and SIRT1,but increases the expression of miR-653-5p.Overexpression of H19 could increase cell viability,ATP level and mitochondrial membrane potential,but reduce mitochondrial ROS generation and cell apoptosis ratio in H2O2-stimulated HEI-OC1 cells.MiR-653-5p is a target of H19,which can bind to the 3'-UTR of SIRT1.H19 is found to regulate the expression of SIRT1 through miR-653-5p.Further experiments demonstrates that H19 regulates HEI-OC1 cell viability,ATP level,mitochondrial membrane potential,mitochondrial ROS generation,and cell apoptosis ratio via the miR-653-5p/SIRT1 axis.In conclusion,lncRNA H19 inhibits oxidative stress injury of cochlear hair cells via the miR-653-5p/SIRT1 axis. | Wen Xie Ting Shu Haisen Peng Jiali Liu Chunhua Li Meiqun Wang Ping Wu Yuehui Liu | 2022 | Acta Biochimica et Biophysica Sinica2022,54,3: | 2 |

| 8 | Tree Planting:How Fast Can It Accelerate Post-fire Forest Restoration?——A Case Study in Northern Da Hinggan Mountains,China显示文摘In 1987,a catastrophic fire burned over 1330000 ha in the densely forested area of the Da Hinggan Mountains in the northeastern China.After the fire,intensive management including burned trunk harvesting and coniferous tree planting had been conducted to accelerate forest restoration.To study the long term effect of these activities on forest recovery,we used a simulation modeling approach to study long-term(300 years) forest dynamics under current planting and natural regeneration scenarios.Results indicate that under tree planting scenario in the severely burned area,the dominant species Dahurian larch(Larix gmelinii) can reach pre-fire level(60% of the area) within 20 years and the maximum abundance can reach nearly 90% within 100 years.While under natural regeneration scenario,it needs about 250 years to reach its pre-fire level.From the perspective of timber production,tree planting can bring twice as much timber volume as that under natural regeneration within 300 years,which is the average longevity of L.gmelinii.It needs about 70 years to reach the timber volume of pre-fire level under the planting scenario,whereas it requires at least 250 years to reach the timber volume of pre-fire level under natural regeneration scenario.Another dominant species Asian White birch(Betula platyphylla) responded negatively to the planting of coniferous species.In general,tree planting of coniferous species after fire can greatly accelerate forest restoration in terms of species abundance and target timber volume,with desirable ecological and economic returns. | LI Xiuzhen HE Hong S WANG Xugao XIE Fuju HU Yuanman LI Yuehui | 2010 | Chinese Geographical Science2010,20,6: | 1 |
| 9 | Research and clinical translation of trilayer stent-graft of expanded polytetrafluoroethylene for interventional treatment of aortic dissection显示文摘The aortic dissection(AD)is a life-threatening disease.The transcatheter endovascular aortic repair(EVAR)affords a minimally invasive technique to save the lives of these critical patients,and an appropriate stent-graft gets to be the key medical device during an EVAR procedure.Herein,we report a trilayer stent-graft and corresponding delivery system used for the treatment of the AD disease.The stent-graft is made of nitinol stents with an asymmetric Z-wave design and two expanded polytetrafluoroethylene(ePTFE)membranes.Each of the inner and outer surfaces of the stent-graft was covered by an ePTFE membrane,and the two membranes were then sintered together.The biological studies of the sintered ePTFE membranes indicated that the stent-graft had excellent cytocompatibility and hemocompatibility in vitro.Both the stent-graft and the delivery system exhibited satisfactory mechanical properties and operability.The safety and efficacy of this stent-graft and the corresponding delivery system were demonstrated in vivo.In nine canine experiments,the blood vessels of the animals implanted with the stent-grafts were of good patency,and there were no thrombus and obvious stenosis by angiography after implantation for 6months.Furthermore,all of the nine clinical cases experienced successful implantation using the stent-graft and its postrelease delivery system,and the 1-year follow-ups indicated the preliminary safety and efficacy of the trilayer stent-graft with an asymmetric Z-wave design for interventional treatment. | Gang Wang Caiyun Gao Benhao Xiao Jie Zhang Xunyuan Jiang Qunsong Wang Jingzhen Guo Deyuan Zhang Jianxiong Liu Yuehui Xie Chang Shu Jiandong Ding | 2022 | Regenerative Biomaterials2022,9,1: | 0 |
