| 1 | Design, synthesis and biological evaluation of chalcone analogues with novel dual antioxidant mechanisms as potential anti-ischemic stroke agents显示文摘Scavenging reactive oxygen species(ROS) by antioxidants is the important therapy to cerebral ischemia-reperfusion injury(CIRI) in stroke. The antioxidant with novel dual-antioxidant mechanism of directly scavenging ROS and indirectly through antioxidant pathway activation may be a promising CIRI therapeutic strategy. In our study, a series of chalcone analogues were designed and synthesized, and multiple potential chalcone analogues with dual antioxidant mechanisms were screened. Among these compounds, the most active 33 not only conferred cytoprotection of H2 O2-induced oxidative damage in PC12 cells through scavenging free radicals directly and activating NRF2/ARE antioxidant pathway at the same time, but also played an important role against ischemia/reperfusion-related brain injury in animals. More importantly, in comparison with mono-antioxidant mechanism compounds, 33 exhibited higher cytoprotective and neuroprotective potential in vitro and in vivo. | 4 | 中国-印尼积雪草药材化学成分对比研究显示文摘目的比较研究中国和印尼积雪草药材化学成分的差异。方法以积雪草苷为对照品,采用薄层色谱法(TLC)、高效液相色谱法(HPLC)及高效液相色谱蒸发光散射(HPLC-ELSD)指纹图谱对来源于中国和印尼的积雪草药材进行比较研究。结果薄层色谱鉴别:来源于中国的积雪草药材365 nm处检视有一特有的蓝色荧光斑点,两个不同来源的积雪草药材其他斑点颜色、数目及比移值(Rf值)基本一致;含量测定:来源于中国的积雪草药材中积雪草苷含量较高,达到0.52%(n=3),本批次的印尼积雪草药材中积雪草苷含量较低,为0.35%(n=3);HPLC-ELSD指纹图谱:两种不同来源的积雪草药材化学成分存在较大差异,其中1、2、5号峰为中国积雪草特有的峰,而3号(积雪草苷)、4号峰为两者共有的峰。结论首次初步建立了积雪草药材的HPLC-ELSD指纹图谱方法,有利于中医药研究的国际交流合作。 | 张培培 郭力 许莉 Wulan Tri Wahyuni | 2018 | 河南医学高等专科学校学报2018，30，5： | 3 |

| 7 | Effect of sucrose and potassium nitrate on biomass and saponin content of Talinum paniculatum Gaertn. hairy root in balloon-type bubble bioreactor显示文摘Objective:To increase biomass and saponin production in hairy root culture of Talinum paniculatum Gaertn.(T.paniculatum)in balloon-type bubble bioreactor(BTBB).Methods:Hairy roots which were collected from leaf explants of T.paniculatum were infected by Agrobacterium rhizogenes strain LB510.The hairy roots were cultivated at400 m L Murashige and Skoog liquid medium without growth regulator(MS0)in1 000 m L BTBB.Each BTBB had 2 g hairy roots as initial inoculum and these cultures were treated with various concentrations of sucrose(3%,4%,5%,6%w/v)and potassium nitrate(0.5,1.0,1.5 and 2.0 strength of MS medium).Cultures were maintained for 14days.Fresh and dry weights of hairy roots at the end of culture were investigated.Results:Various concentrations of sucrose influenced the biomass accumulation of hairy roots.Maximum biomass was reached by MS medium supplemented with 6%sucrose and it was approximately threefold higher than control.Culture supplemented with potassium nitrate at 2.0 strength of MS0 could increase biomass accumulation of hairy roots until 0.14 g dry weight and it was almost threefold higher than control.However,the maximum saponin content was obtained by MS medium supplemented with 5%sucrose and 2.0 strength potassium nitrate of MS.Conclusions:Based on this research,those conditions can be used to produce biomass and saponin of hairy root of T.paniculatum in the large scale. | Yosephine Sri Wulan Manuhara Alfinda Novi Kristanti Edy Setiti Wida Utami Arif Yachya | 2015 | Asian Pacific Journal of Tropical Biomedicine2015，5，12： | 3 |
