| 2 | Offline two-dimensional liquid chromatography coupled with ion mobility-quadrupole time-of-flight mass spectrometry enabling fourdimensional separation and characterization of the multicomponents from white ginseng and red ginseng显示文摘Inherent complexity of plant metabolites necessitates the use of multi-dimensional information to accomplish comprehensive profiling and confirmative identification.A dimension-enhanced strategy,by offline two-dimensional liquid chromatography/ion mobility-quadrupole time-of-flight mass spectrometry(2 D-LC/IM-QTOF-MS)enabling four-dimensional separations(2 D-LC,IM,and MS),is proposed.In combination with in-house database-driven automated peak annotation,this strategy was utilized to characterize ginsenosides simultaneously from white ginseng(WG)and red ginseng(RG).An offline 2 DLC system configuring an Xbridge Amide column and an HSS T3 column showed orthogonality 0.76 in the resolution of ginsenosides.Ginsenoside analysis was performed by data-independent high-definition MSE(HDMSE)in the negative ESI mode on a Vion?IMS-QTOF hybrid high-resolution mass spectrometer,which could better resolve ginsenosides than MSEand directly give the CCS information.An in-house ginsenoside database recording 504 known ginsenosides and 58 reference compounds,was established to assist the identification of ginsenosides.Streamlined workflows,by applying UNIFI?to automatedly annotate the HDMSEdata,were proposed.We could separate and characterize 323 ginsenosides(including 286 from WG and 306 from RG),and 125 thereof may have not been isolated from the Panax genus.The established 2 D-LC/IM-QTOF-HDMSEapproach could also act as a magnifier to probe differentiated components between WG and RG.Compared with conventional approaches,this dimensionenhanced strategy could better resolve coeluting herbal components and more efficiently,more reliably identify the multicomponents,which,we believe,offers more possibilities for the systematic exposure and confirmative identification of plant metabolites. | Tiantian Zuo Chunxia Zhang Weiwei Li Hongda Wang Ying Hu Wenzhi Yang Li Jia Xiaoyan Wang Xiumei Gao Dean Guo | 2020 | Journal of Pharmaceutical Analysis2020,10,6: | 9 |
| 3 | TGIF2 promotes the progression of lung adenocarcinoma by bridging EGFR/RAS/ERK signaling to cancer cell stemness显示文摘TGF-β-induced factor homeobox 2(TGIF2)is a transcription regulator that plays essential roles in the regulation of development and cell fate decisions.Aberrant expression of TGIF family proteins has been observed in several cancers,including ovarian,esophageal,and colorectal cancers.Here,we report that TGIF2 mediates the EGFR–RAS–ERK signaling pathway to enhance the stemness of lung adenocarcinoma(LUAD)cells and,therefore,promote the progression and metastasis of LUAD.We found that high TGIF2 expression was closely correlated with tumor growth,lymph node metastasis,and survival of patients with LUAD.Mice bearing TGIF2-silenced H1299 xenografts developed smaller tumors and fewer lung metastases.Importantly,silencing TGIF2 decreased the cancer stem cell(CSC)-like properties in A549 and H1299 cells.Furthermore,we identified that TGIF2 binding to the OCT4 promoter promotes its expression.In both LUAD cells and in vivo LUAD mouse models,we revealed that EGFR–RAS–ERK signaling phosphorylated TGIF2 and increased its stability,which was important for TGIF2-promoted LUAD stemness since phosphorylation-deficient TGIF2 mutants lost these functions.Thus,our study revealed that an important factor,TGIF2,bridges EGFR signaling to the CSC characteristics of LUAD cells,which can be utilized as an effective target for LUAD therapy. | Renle Du Wenzhi Shen Yi Liu Wenjuan Gao Wei Zhou Jun Li Shuangtao Zhao Chong Chen Yanan Chen Yanhua Liu Peiqing Sun Rong Xiang Yi Shi Yunping Luo | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 7 |
| 4 | FEM Analysis on Acoustic Performance of Wall Flow Diesel Particulate Filters显示文摘Diesel powered vehicles, in compliance with the more strict exhaust emission standards such as Euro V, is likely to require a diesel particulate filter (DPF). A DPF used on a vehicle will affect the acoustic emission of the diesel engine, so it is important to investigate the sound propagation rule in DPF and further to propose the optimum DPF design. However, due to the geometrical complexity of the DPF, the traditional analysis method, such as analytical method, can not assess the acoustic performance of DPF accurately in medium and high frequency band. In this paper, a combined approach of finite element analysis and viscosity correction is proposed to predict acoustic performance of DPF. A simplified model of the full DPF is established and is used to analyze the sound propagation characteristic of the DPF. The distribution of the sound pressure and velocity, the transmission matrix of the DPF are obtained using the finite element method. In addition, the method of the viscosity correction is used in the transmission matrix of the DPF to evaluate the acoustic performance of DPF. Based on the FEM computation and the viscosity correction, the transmission losses under the rated load and idle condition of a diesel engine are calculated. The calculation results show that DPF can effectively attenuate exhaust noise, and sound attenuation increase with the rise of the frequency. | 5 | A hybrid CFD/characteristics method for fast characterization of hypersonic blunt forebody/inlet flow显示文摘A hybrid CFD/characteristic method(CCM) was proposed for fast design and evaluation of hypersonic inlet flow with nose bluntness, which targets the combined advantages of CFD and method of characteristics. Both the accuracy and efficiency of the developed CCM were verified reliably, and it was well demonstrated for the external surfaces design of a hypersonic forebody/inlet with nose bluntness. With the help of CCM method, effects of nose bluntness on forebody shock shapes and the flowfield qualities which dominate inlet performance were examined and analyzed on the two-dimensional and axisymmetric configurations. The results showed that blunt effects of a wedge forebody are more substantial than that of related cone cases. For a conical forebody with a properly blunted nose, a recovery of the shock front back to that of corresponding sharp nose is exhibited, accompanied with a gradually fading out of entropy layer effects. Consequently a simplification is thought to be reasonable for an axisymmetric inlet with a proper compression angle, and a blunt nose of limited radius can be idealized as a sharp nose, as the spillage and flow variations at the entrance are negligible, even though the nose scale increases to 10% cowl lip radius. | 6 | Effects of trips on the oscillatory flow of an axisymmetric hypersonic inlet with downstream throttle显示文摘Experimental investigations are conducted on an axisymmetric hypersonic inlet to evaluate the effects of trips on oscillatory flows. The model exit is throttled with a fixed block to generate oscillatory flows at a freestream Mach number of 6 in a conventional wind tunnel and a shock tunnel. Schlieren imaging and pressure measurements are adopted to record unsteady flow features.Results indicate that trips with a 1 mm thickness prominently suppress external separations, shorten oscillatory cycles, and modify pressure magnitudes. Trips can reduce the upstream movement ranges of separated shocks from nose regions to locations axially 142 mm downstream. The oscillatory cycles are shortened from 3.75 ms to 3.25 ms and from 4 ms to 3.13 ms in two facilities.Tripped cases generally exhibit higher pressure magnitudes than those of untripped cases, of which the increment is up to 21 times the freestream static pressure for the farthest downstream transducer in the shock tunnel. The effects of trips are related to the streamwise vortexes in wake flows, in which interactions between external separations modify the separated flow patterns and enhance the sustainment of the forebody boundary layers to backpressure. | 14 | Novel neutrophil extracellular trap-related mechanisms in diabetic wounds inspire a promising treatment strategy with hypoxia-challenged small extracellular vesicles显示文摘Neutrophil extracellular traps(NETs)have been considered a significant unfavorable factor for wound healing in diabetes,but the mechanisms remain unclear.The therapeutic application of small extracellular vesicles(sEVs)derived from mesenchymal stem cells(MSCs)has received considerable attention for their properties.Hypoxic preconditioning is reported to enhance the therapeutic potential of MSC-derived sEVs in regenerative medicine.Therefore,the aim of this study is to illustrate the detailed mechanism of NETs in impairment of diabetic wound healing and develop a promising NET-targeting treatment based on hypoxic pretreated MSC-derived sEVs(Hypo-sEVs).Excessive NETs were found in diabetic wounds and in high glucose(HG)-induced neutrophils.Further research showed that high concentration of NETs impaired the function of fibroblasts through activating endoplasmic reticulum(ER)stress.Hypo-sEVs efficiently promoted diabetic wound healing and reduced the excessive NET formation by transferring miR-17-5p.Bioinformatic analysis and RNA interference experiment revealed that miR-17-5p in Hypo-sEVs obstructed the NET formation by targeting TLR4/ROS/MAPK pathway.Additionally,miR-17-5p overexpression decreased NET formation and overcame NET-induced impairment in fibroblasts,similar to the effects of Hypo-sEVs.Overall,we identify a previously unrecognized NET-related mechanism in diabetic wounds and provide a promising NET-targeting strategy for wound treatment. | Ziqiang Chu Qilin Huang Kui Ma Xi Liu Wenhua Zhang Shengnan Cui Qian Wei Huanhuan Gao Wenzhi Hu Zihao Wang Sheng Meng Lige Tian Haihong Li Xiaobing Fu Cuiping Zhang | 2023 | Bioactive Materials2023,,9: | 1 |

| 16 | Responses of nitrification performance,triclosan resistome and diversity of microbes to continuous triclosan stress in activated sludge system显示文摘Triclosan(TCS)is commonly found in wastewater treatment plants,which often affects biological treatment processes.The responses of nitrification,antibiotic resistome and microbial community under different TCS concentrations in activated sludge system were evaluated in this study.The experiment was conducted in a sequencing batch reactor(SBR)for 240 days.Quantitative PCR results demonstrated that the abundance of ammonium oxidizing bacteria could be temporarily inhibited by 1 mg/L TCS and then gradually recovered.And the abundances of nitrite oxidizing bacteria(NOB)under 2.5 and 4 mg/L TCS were three orders of magnitude lower than that of seed sludge,which accounted for partial nitrification.When the addition of TCS was stopped,the abundance of NOB increased.The mass balance experiments of TCS demonstrated that the primary removal pathway of TCS changed from adsorption to biodegradation as TCS was continuously added into the SBR system.Moreover,TCS increased the abundance of mexB,indicating the efflux pump might be the main TCS-resistance mechanism.As a response to TCS,bacteria could secrete more protein(PN)than polysaccharide.Three-dimensional excitation-emission matrix revealed that tryptophan PN-like substances might be the main component in PN to resist TCS.High-throughput sequencing found that the relative abundances of Paracoccus,Pseudoxanthomonas and Thauera increased,which could secrete extracellular polymeric substances(EPS).And Sphingopyxis might be the main TCS-degrading bacteria.Overall,TCS could cause partial nitrification and increase the relative abundances of EPS-secreting bacteria and TCS-degrading bacteria. | Da Zhang Jingfeng Gao Lifang Zhang Wenzhi Zhang Jingxin Jia Huihui Dai Zhiqi Wang | 2020 | Journal of Environmental Sciences2020,32,6: | 1 |

| 19 | LncRNA-m18as1 competitively binds with miR-18a-5p to regulate follicle-stimulating hormone secretion through the Smad2/3 pathway in rat primary pituitary cells显示文摘Long noncoding RNAs(lncRNAs)are expressed in different species and different tissues,and perform different functions,but little is known about their involvement in the synthesis or secretion of follicle-stimulating hormone(FSH).In general,we have revealed lnc RNA-micro RNA(mi RNA)-messenger RNA(m RNA)interactions that may play important roles in rat primary pituitary cells.In this study,a new lncRNA was identified for the first time.First,we analyzed the gene expression of lncRNA-m18as1 in different tissues and different stages by reverse transcription-quantitative polymerase chain reaction(RT-qPCR)and observed the localization of lncRNA-m18as1 with fluorescence in situ hybridization,which indicated that this lncRNA was distributed mainly in the cytoplasm.Next,we used RT-qPCR and enzyme-linked immunosorbent assay(ELISA)to analyze the regulation of FSH synthesis and secretion after overexpression or knockdown of lncRNA-m18as1 and found that lncRNA-m18as1 was positively correlated with FSH synthesis and secretion.In addition,mothers against decapentaplegic homolog 2(Smad2)was highly expressed in our sequencing results.We also screened miR-18a-5p from our sequencing results as a miRNA that may bind to lncRNA-m18as1 and Smad2.We used RNA immunoprecipitation-qPCR(RIP-qPCR)and/or dual luciferase assays to confirm that lncRNA-m18as1 interacted with miR-18a-5p and miR-18a-5p interacted with Smad2.Fluorescence in situ hybridization(FISH)showed that lncRNA-m18as1 and miR-18a-5p were localized mainly in the cytoplasm.Finally,we determined the relationship among lncRNA-m18as1,miR-18a-5p,and the Smad2/3 pathway.Overall,we found that lncRNA-m18as1 acts as a molecular sponge of miR-18a-5p to regulate the synthesis and secretion of FSH through the Smad2/3 pathway. | Weidi ZHANG Wenzhi REN Dongxu HAN Guokun ZHAO Haoqi WANG Haixiang GUO Yi ZHENG Zhonghao JI Wei GAO Bao YUAN | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,6: | 0 |
