1Anti-inflammatory activity of curcumin-loaded tetrahedral framework nucleic acids on acute gouty arthritis显示文摘Gouty arthritis is a very familiar inflammatory arthritis.Controlling inflammation is the key to preventing gouty arthritis.However,colchicine,the most highly represented drug used in clinical practice,has strict contraindications owing to some severe side effects.Curcumin(Cur),a natural anti-inflammatory drug,has demonstrated good safety and efficacy.However,the rapid degradation,poor aqueous solubility,and low bioavailability of Cur limit its therapeutic effect.To strengthen the effectiveness and bioavailability of Cur.Cur loaded tetrahedral framework nucleic acids(Cur-TFNAs)were synthesized to deliver Cur.Compared with free Cur,Cur-TFNAs exhibit a preferable drug stability,good biocompatibility(CCK-8 assay),ease of uptake(immunofluorescence),and higher tissue utilization(in vivo biodistribution).Most importantly,Cur-TFNAs present better anti-inflammatory effect than free Cur both in vivo and in vitro experiments through the determination of inflammation-related cytokines expression.Therefore,we believe that Cur-TFNAs have great prospects for the prevention of gout and similar inflammatory diseases.Mei Zhang Xiaolin Zhang Taoran Tian Qi Zhang Yuting Wen Junyao Zhu Dexuan Xiao Weitong Cui Yunfeng Lin 2022Bioactive Materials2022,7,2:10
2Effect of tetrahedral DNA nanostructures on LPS-induced neuroinflammation in mice显示文摘Neuroinflammation plays a significant role in inducing depression-like behavior. Tetrahedral DNA nanostructures(TDNs) are molecules that exhibit anti-inflammatory properties and can effectively penetrate the blood-brain barrier. Thus, researchers have hypothesized that TDNs regulate the secretion of proinflammatory cytokines and consequently alleviate depression-like behavior. To test this hypothesis, we investigated the effect of TDNs on the depression-like behavior of C57 mice induced by lipopolysaccharide(LPS). We performed open-field, tail suspension, and sucrose preference tests on LPS-and LPS/TDNtreated mice. The results indicated that the injection of TDNs into LPS-treated mice resulted in increased velocity, center zone duration, frequency to the center zone, and sucrose preference, and decreased immobility time. Immunofluorescence results indicated that peripheral administration of LPS in the mice activated inflammation, which culminated in distinct depression-like behavior. However, TDNs effectively alleviated the inflammation and depression-like behavior through the reduction of the expression levels of proinflammatory cytokines, such as interleukin-1β and tumor necrosis factor-α in the brain. Additionally, TDNs normalized the expression level of microglia cell activation markers, such as ionized calcium binding adaptor molecule 1, in the hippocampus of mice. 3Tetrahedral framework nucleic acids promote scarless healing of cutaneous wounds via the AKT-signaling pathway显示文摘While the skin is considered the first line of defense in the human body,there are some vulnerabilities that render it susceptible to certain threats,which is an issue that is recognized by both patients and doctors.Cutaneous wound healing is a series of complex processes that involve many types of cells,such as fibroblasts and keratinocytes.This study showed that tetrahedral framework nucleic acids(tFNAs),a type of self-assembled nucleic-acid material,have the ability to promote keratinocyte(HaCaT cell line)and fibroblast(HSF cell line)proliferation and migration in vitro.In addition,tFNAs increased the secretion of vascular endothelial growth factor(VEGF)and basic fibroblast growth factor(bFGF)in HSF cells and reduced the production of tumor necrosis factor-alpha(TNF-α)and interleukin-1 beta(IL-1β)in HaCaT cells by activating the AKT-signaling pathway.During in vivo experiments,tFNA treatments accelerated the healing process in skin wounds and decreased the development of scars,compared with the control treatment that did not use tFNAs.This is the first study to demonstrate that nanophase materials with the biological features of nucleic acids accelerate the healing of cutaneous wounds and reduce scarring,which indicates the potential application of tFNAs in skin tissue regeneration.Junyao Zhu Mei Zhang Yang Gao Xin Qin Tianxu Zhang Weitong Cui Chenchen Mao Dexuan Xiao Yunfeng Lin 2020Signal Transduction and Targeted Therapy2020,5,1:1
4GCR-Net:3D Graph convolution-based residual network for robust reconstruction in cerenkov luminescence tomography显示文摘Cerenkov Luminescence Tomography(CLT)is a novel and potential imaging modality which can display the three-dimensional distribution of radioactive probes.However,due to severe ill-posed inverse problem,obtaining accurate reconstruction results is still a challenge for traditional model-based methods.The recently emerged deep learning-based methods can directly learn the mapping relation between the surface photon intensity and the distribution of the radioactive source,which effectively improves the performance of CLT reconstruction.However,the previously proposed deep learning-based methods cannot work well when the order of input is disarranged.In this paper,a novel 3D graph convolution-based residual network,GCR-Net,is proposed,which can obtain a robust and accurate reconstruction result from the photon intensity of the surface.Additionally,it is proved that the network is insensitive to the order of input.The performance of this method was evaluated with numerical simulations and in vivo experiments.The results demonstrated that compared with the existing methods,the proposed method can achieve efficient and accurate reconstruction in localization and shape recovery by utilizing threedimensional information.Weitong Li Mengfei Du Yi Chen Haolin Wang Linzhi Su Huangjian Yi Fengjun Zhao Kang Li Lin Wang Xin Cao 2023Journal of Innovative Optical Health Sciences2023,16,1:0
5Enhanced tensile ductility of tungsten microwires via high-density dislocations and reduced grain boundaries显示文摘Despite being strong with many outstanding physical properties,tungsten is inherently brittle at room temperature,restricting its structural and functional applications at small scales.Here,a facile strategy has been adopted,to introduce high-density dislocations while reducing grain boundaries,through electron backscatter diffraction(EBSD)-guided microfabrication of cold-drawn bulk tungsten wires.The designed tungsten microwire attains an ultralarge uniform tensile elongation of~10.6%,while retains a high yield strength of~2.4 GPa.in situ TEM tensile testing reveals that the large uniform elongation of tungsten microwires originates from the motion of pre-existing high-density dislocations,while the subsequent ductile fracture is attributed to crack-tip plasticity and the inhibition of grain boundary cracking.This work demonstrates the application potential of tungsten microcomponents with superior ductility and workability for micro/nanoscale mechanical,electronic,and energy systems.Chaoqun Dang Weitong Lin Fanling Meng Hongti Zhang Sufeng Fan Xiaocui Li Ke Cao Haokun Yang Wenzhao Zhou Zhengjie Fan Ji-jung Kai Yang Lu 2021Journal of Materials Science & Technology2021,,36:0
6Oxide formation mechanism of a corrosion-resistant CZ1 zirconium alloy显示文摘Tailoring microstructure and microchemistry by altering elemental compositions and thermomechanical treatment parameters enables superior corrosion performance in zirconium alloys for nuclear applications.However,our understanding of the relationship between various defects and the corrosion process remains limited in the newly developed zirconium alloys.Here we report the oxide formation mechanism of a CZ1 zirconium alloy with corrosion resistance surpassing many other zirconium alloy systems,such as Zircaloy-4 and Zr-1Nb-1Sn alloys.Autoclave experiments of CZ1 alloy and Zr1Nb-1Sn model alloy were performed in 360°C water for up to 820 d.We quantitively determined oxide phases by transmission Kikuchi diffraction(TKD)and examined lateral cracks,nano-porosity,and second-phase particles in oxide scales by transmission electron microscopy(TEM).Compared to the Zr-1Nb-1Sn model alloy,CZ1 alloy with lower Nb and Sn concentrations has shown smaller and lower-density lateral cracks but slightly larger oxide grains,reducing the diffusion route for oxidating species.Using analytical scanning and transmission electron microscopy,we demonstrate that due to the lower content of Sn(∼0.9 wt.%),there is less tetragonal ZrO_(2) phase formed in the oxide,and the level of tetragonal to the monoclinic phase transition is reduced.Although the Nb content(0.1 wt.%–0.3 wt.%)is lower than the solid solution limit of Nb in Zr,by introducing minor elements such as Fe,Cr,and Cu,there are still a reasonable number of second-phase particles to relieve the high stress associated with the metal-to-oxide transformation.These mechanisms have substantially changed the density and distribution of lateral cracks in the oxide,thus reducing the corrosion rate of zirconium alloys.Jing Hu Weitong Lin Qingyang Lv Changyuan Gao Jun Tan 2023Journal of Materials Science & Technology2023,,16:0
7Applications of tetrahedral DNA nanostructures in wound repair and tissue regeneration显示文摘Tetrahedral DNA nanostructures(TDNs)are molecules with a pyramidal structure formed by folding four single strands of DNA based on the principle of base pairing.Although DNA has polyanionic properties,the special spatial structure of TDNs allows them to penetrate the cell membrane without the aid of transfection agents in a caveolin-dependent manner and enables them to participate in the regulation of cellular processes without obvious toxic side effects.Because of their stable spatial structure,TDNs resist the limitations imposed by nuclease activity and innate immune responses to DNA.In addition,TDNs have good editability and biocompatibility,giving them great advantages for biomedical applications.Previous studies have found that TDNs have a variety of biological properties,including promoting cell migration,proliferation and differentiation,as well as having anti-inflammatory,antioxidant,anti-infective and immune regulation capabilities.Moreover,we confirmed that TDNs can promote the regeneration and repair of skin,blood vessels,muscles and bone tissues.Based on these findings,we believe that TDNs have broad prospects for application in wound repair and regeneration.This article reviews recent progress in TDN research and its applications.Yikai Dou Weitong Cui Xiao Yang Yunfeng Lin Xiaohong Ma Xiaoxiao Cai 2022Burns & Trauma2022,10,1:0
8Enhancing thermodynamic stability of single-crystal Ni-rich cathode material via a synergistic dual-substitution strategy显示文摘Nickel(Ni)-rich cathode materials have become promising candidates for the next-generation electrical vehicles due to their high specific capacity.However,the poor thermodynamic stability(including cyclic performance and safety performance or thermal stability)will restrain their wide commercial application.Herein,a single-crystal Ni-rich Li Ni_(0.83)Co_(0.12)Mn_(0.05)O_(2) cathode material is synthesized and modified by a dual-substitution strategy in which the high-valence doping element improves the structural stability by forming strong metal–oxygen binding forces,while the low-valence doping element eliminates high Li^(+)/Ni^(2+)mixing.As a result,this synergistic dual substitution can effectively suppress H2-H3 phase transition and generation of microcracks,thereby ultimately improving the thermodynamic stability of Ni-rich cathode material.Notably,the dual-doped Ni-rich cathode delivers an extremely high capacity retention of 81%after 250 cycles(vs.Li/Li+)in coin-type half cells and 87%after 1000 cycles(vs.graphite/Li^(+))in pouch-type full cells at a high temperature of 55℃.More impressively,the dual-doped sample exhibits excellent thermal stability,which demonstrates a higher thermal runaway temperature and a lower calorific value.The synergetic effects of this dual-substitution strategy pave a new pathway for addressing the critical challenges of Ni-rich cathode at high temperatures,which will significantly advance the high-energy-density and high-safety cathodes to the subsequent commercialization.Jixue Shen Hui Li Haoyu Qi Zhan Lin Zeheng Li Chuanbo Zheng Weitong Du Hao Chen Shanqing Zhang 2024Journal of Energy Chemistry2024,88,1:0
9Pre-training in Medical Data:A Survey显示文摘Medical data refers to health-related information associated with regular patient care or as part of a clinical trial program.There are many categories of such data,such as clinical imaging data,bio-signal data,electronic health records(EHR),and multi-modality medical data.With the development of deep neural networks in the last decade,the emerging pre-training paradigm has become dominant in that it has significantly improved machine learning methods′performance in a data-limited scenario.In recent years,studies of pre-training in the medical domain have achieved significant progress.To summarize these technology advancements,this work provides a comprehensive survey of recent advances for pre-training on several major types of medical data.In this survey,we summarize a large number of related publications and the existing benchmarking in the medical domain.Especially,the survey briefly describes how some pre-training methods are applied to or developed for medical data.From a data-driven perspective,we examine the extensive use of pre-training in many medical scenarios.Moreover,based on the summary of recent pre-training studies,we identify several challenges in this field to provide insights for future studies.Yixuan Qiu Feng Lin Weitong Chen Miao Xu 2023Machine Intelligence Research2023,20,2:0
10Dynamic Multi-Attribute Decision-Making Method with Double Reference Points and Its Application显示文摘To better reflect the psychological behavior characteristics of loss aversion,this paper builds a double reference point decision making method for dynamic multi-attribute decision-making(DMADM)problem,taking bottom-line and target as reference pints.First,the gain/loss function is given,and the state is divided according to the relationship between the gain/loss value and the reference point.Second,the attitude function is constructed based on the results of state division to establish the utility function.Third,the comprehensive utility value is calculated as the basis for alternatives classification and ranking.Finally,the new method is used to evaluate the development level of smart cities.The results show that the new method can judge the degree to which the alternatives meet the requirements of the decision-maker.While the new method can effectively screen out the unsatisfactory alternatives,the ranking results of other alternatives are consistent with those of traditional methods.Haoran Huang Qinyong Lin Weitong Chen Kai Fang Huazhou Chen Ken Cai 2021Computers, Materials & Continua2021,,7:0
