| 1 | Knockdown of OsHox33, a member of the class III homeodomain-leucine zipper gene family, accelerates leaf senescence in rice显示文摘The class III homeodomain-leucine zipper(HD-Zip III)gene family plays important roles in plant growth and development,including regulation of apical embryo patterning,embryonic shoot meristem formation,leaf polarity,vascular development,and meristem function,with a particularly crucial function in leaf development.Although HD-Zip III members are highly conserved in land plants,previous studies,such as genetic analyses based on multiple mutants in Arabidopsis and other plants,suggest that various HD-Zip III family genes have evolved with distinct functions and pleiotropic effects on plant growth and development.In this study,we analyzed a HD-Zip III member,OsHox33,and demonstrated that it plays an important role in age-dependent leaf senescence in rice.We constructed two specific RNAi vectors derived from the 5′-end region and 3′-UTR of OsHox33 to knockdown its expression.Transgenic plants harboring either RNAi construct displayed similar phenotypes of precocious leaf senescence symptoms,suggesting that knockdown of OsHox33 accelerates leaf senescence in rice.pOsHox33::GUS fusion expression and RT-PCR revealed that OsHox33 is highly expressed in young organs,especially in young meristems such as shoot apical meristems,intercalary meristems,and young callus.In addition,real-time PCR indicated that OsHox33 was more highly expressed in young leaves than in old leaves.To further investigate OsHox33 function,we analyzed chloroplast ultrastructure in different-aged leaves of RNAi plants,and found that OsHox33 knockdown accelerated chloroplast degradation,which is consistent with RNAi phenotypes.Finally,real-time PCR studies showed that OsHox33 can regulate the expression of GS1 and GS2,two senescence-associated genes.Taken together,the work presented here provides new insights into the function of HD-Zip III members in plants. | LUAN WeiJiang SHEN Ao JIN ZhiPing SONG SuSheng LI ZhengLong SHA AiHua | 2013 | Science China(Life Sciences)2013,56,12: | 13 |
| 2 | Xuefuzhuyu decoction for hyperlipidemia: a systematic review and Meta-analysis of randomized clinical trails显示文摘OBJECTIVE:To evaluate the efficacy and safety of Xuefuzhuyu decoction for hyperlipidemia.METHODS:Randomized clinical trials on hyperlipidemia treated by Xuefuzhuyu decoction,either alone or with Western Medicine,were searched in electronic databases.Databases searched were:MEDLINE,Allied and Complementary Medicine Database,EMBASE,The Cochrane Library 2013(Issue4),China National Knowledge Infrastructure Database,Chinese Biomedical Literature Database,and Wanfang Database up to 2 May,2013.Study selection,data extraction,quality assessment,and data analysis were conducted according to the Cochranestandards.RESULTS:Six randomized clinical trials involving748 patients(373 patients in the treatment group,375 patients in the control group)were included in the analysis.The studies were of low methodological quality.Meta-analysis indicated that the effect of Xuefuzhuyu decoction on hyperlipidemia was better than that in the control group[n=748,OR=5.07,95%CI(3.40,7.58),P<0.01].Weighted mean differencesin total cholesterol,low-densitylipoprotein cholesterol,triglycerides,and high-density lipoprotein cholesterol were﹣0.79,﹣0.74,﹣0.44,0.16,respectively,and Meta-analysis revealed that the treatment group was better than the control group with 95%CI(﹣1.21,﹣0.36),(﹣0.94,﹣0.55),(﹣0.77,﹣0.11),(0.04,0.27),respectively(all P<0.05).Some adverse events in evaluated studies wererecorded.CONCLUSION:Xuefuzhuyu decoction may be effective for treating hyperlipidemia.The studies we analyzed were of low methodological quality,which indicates that the above findings should be considered cautiously.Therefore,more strictly designed large-scale randomized clinical trials are needed to evaluate the efficacy of Xuefuzhuyu decoction in hyperlipidemia. | Jiangquan Liao Jiaxing Tian Tengfei Li Weijiang Song Weihan Zhao Jinhang Du | 2014 | Journal of Traditional Chinese Medicine2014,34,4: | 5 |
| 3 | Interaction between temperature and photoperiod in regulation of flowering time in rice显示文摘Photoperiod and temperature are two pivotal regulatory factors of plant flowering.The floral transition of plants depends on accurate measurement of changes in photoperiod and temperature.The flowering time of rice(Oryza sativa) as a facultative short-day(SD) plant is delayed under long-day(LD) and/or low temperature conditions.To elucidate the regulatory functions of photoperiod and temperature on flowering time in rice,we systematically analyzed the expression and regulation of several key genes(Hd3a,RFT1,Ehd1,Ghd7,RID1/Ehd2/OsId1,Se5) involved in the photoperiodic flowering regulatory pathway under different temperature and photoperiod treatments using a photoperiod-insensitive mutant and wild type plants.Our results indicate that the Ehd1-Hd3a/RFT1 pathway is common to and conserved in both the photoperiodic and temperature flowering regulatory pathways.Expression of Ehd1,Hd3a and RFT1 is dramatically reduced at low temperature(23°C),suggesting that suppression of Ehd1,Hd3a and RFT1 transcription is an essential cause of delayed flowering under low temperature condition.Under LD condition,Ghd7 mRNA levels are promoted at low temperature(23°C) compared with normal temperature condition(28°C),suggesting low temperature and LD treatment have a synergistic role in the expression of Ghd7.Therefore,upregulation of Ghd7 might be a crucial cause of delayed flowering under low temperature condition.We also analyzed Hd1 regulatory relationships in the photoperiodic flowering pathway,and found that Hd1 can negatively regulate Ehd1 transcription under LD condition.In addition,Hd1 can also positively regulate Ghd7 transcription under LD condition,suggesting that the heading-date of rice under LD condition is also regulated by the Hd1-Ghd7-Ehd1-RFT1 pathway. | SONG YuanLi GAO ZhiChao LUAN WeiJiang | 2012 | Science China(Life Sciences)2012,55,3: | 5 |

| 7 | A dual-modal probe for NIR fluorogenic and ratiometric photoacoustic imaging of Cys/Hcy in vivo显示文摘Biothiols, such as cysteine(Cys) and homocysteine(Hcy), play vital roles in biological homeostasis and are closely related to various pathological and physiological processes in the living systems. Therefore, the in vivo detection of biothiols is of great importance for early diagnosis of diseases and assessment of disease progression. In this work, we developed a near-infrared(NIR) fluorescence and photoacoustic dual-modal molecular probe(NIR-S) that can be specifically activated by Cys or Hcy. The aryl-thioether substituted cyanine probe can undergo nucleophilic substitution and Smiles rearrangement reaction, resulting in specific turn-on NIR fluorescence and ratiometric photoacoustic responses for Hcy/Cys. Thus, NIR-S not only realizes the specific NIR fluorescence and photoacoustic dual mode imaging to detect Hcy/Cys in solution, but also can be applied to living cells and mice to detect Hcy/Cys. | 9 | Photoinduced Carbene for Effective Photodynamic Therapy Against Hypoxic Cancer Cells显示文摘Photodynamic therapy(PDT)has attracted much attention because of its advantages over chemotherapy,such as broad spectrum,high selectivity,and low drug resistance.However,most photosensitizers(PSs)used in PDT are O_(2)-dependent and show limited therapeutic efficacy toward hypoxic solid tumors.Therefore,developing PSs that produce reactive oxygen species(ROS)in an O_(2)-independent manner is highly demanded.Herein,we constructed a novel O_(2)-independent PS(TPA-N)based onα-diazo-aryl acetate,which could generate reactive carbene under visible light irradiation.Photoinduced carbene could react with biosubstrates and cause significant carbene stress and ROS,effectively killing tumor cells even under hypoxic conditions.TPA-N showed much less O_(2)dependence compared with traditional PDT reagent protoporphyrin IX.A mechanistic study suggested that TPAN could induce mitochondrial membrane potential collapse and upregulate apoptosis-related proteins upon light irradiation.This work provided a reliable strategy for developing O_(2)-independent PSs against hypoxic tumor cells through photoinduced carbene. | Zhanghua Sun Yuncong Chen Shankun Yao Hao Yuan Dongfan Song Zijian Guo Weijiang He | 2023 | CCS Chemistry2023,5,9: | 0 |
