2Extracellular CIRP dysregulates macrophage bacterial phagocytosis in sepsis显示文摘In sepsis, macrophage bacterial phagocytosis is impaired, but the mechanism is not well elucidated. Extracellular cold-inducible RNA-binding protein (eCIRP) is a damage-associated molecular pattern that causes inflammation. However, whether eCIRP regulates macrophage bacterial phagocytosis is unknown. Here, we reported that the bacterial loads in the blood and peritoneal fluid were decreased in CIRP^(−/−) mice and anti-eCIRP Ab-treated mice after sepsis. Increased eCIRP levels were correlated with decreased bacterial clearance in septic mice. CIRP−/− mice showed a marked increase in survival after sepsis. Recombinant murine CIRP (rmCIRP) significantly decreased the phagocytosis of bacteria by macrophages in vivo and in vitro. rmCIRP decreased the protein expression of actin-binding proteins, ARP2, and p-cofilin in macrophages. rmCIRP significantly downregulated the protein expression of βPIX, a Rac1 activator. We further demonstrated that STAT3 and βPIX formed a complex following rmCIRP treatment, preventing βPIX from activating Rac1. We also found that eCIRP-induced STAT3 phosphorylation was required for eCIRP’s action in actin remodeling. Inhibition of STAT3 phosphorylation prevented the formation of the STAT3-βPIX complex, restoring ARP2 and p-cofilin expression and membrane protrusion in rmCIRP-treated macrophages. The STAT3 inhibitor stattic rescued the macrophage phagocytic dysfunction induced by rmCIRP. 3Swelling Behaviors and Pervaporation Properties of CA/PAN Composite Membranes for Separation of Caprolactam/Water Mixtures显示文摘The composite membranes with cellulose acetate(CA) as the separating layer material and polyacrylonitrile(PAN) as the supporting layer material were prepared for separating caprolactam(CPL) from CPL/water mixtures by pervaporation technique.The swelling experiments were carried out to investigate the effects of swelling time and CPL concentration on the degree of swelling.The results showed that the CA membrane reached swelling equilibrium within 24 h,and the degree of swelling first slightly decreased then increased with the increase of CPL concentration in the feed under the experimental conditions.Respectively comparing flux with permeance,and separation factor with selectivity,we found out that the separation performance of the CA/PAN composite membrane is more strongly dependent on its hydrophilic/hydrophobic nature as well as on the effects of operating parameters,such as feed composition concentration and feed temperature.ZHANG Lei LUO Yunbai XUE Gaifeng YU Ping 2010Wuhan University Journal of Natural Sciences2010,15,5:0
4Cognitive function correlations with apolipoprotein Eε4 single nucleotide polymorphism in 1000 elderly patients following general anesthesia A randomized controlled study显示文摘BACKGROUND:Cognitive dysfunction occurs in elderly patients following general anesthesia, and this might be associated with genetics.Studies have shown that theε4 allele gene is closely associated with senile dementia. OBJECTIVE:To compare and analyze the correlations between cognitive dysfunction and single nucleotide polymorphism of apolipoprotein E(ApoE) following inhalation or intravenous anesthesia. DESIGN,TIME AND SETTING:A randomized,controlled study was performed.The patients were recruited from the Department of Anesthesia,Second Affiliated Hospital,Medical College, Xi'an Jiaotong University,China between May 2005 and December 2008.Genetic analyses were conducted at the Departments of Neuroanatomy and Forensic Medicine,Medical College,Xi'an Jiaotong University,China. PARTICIPANTS:A total of 1 000 patients of ASAⅠ-Ⅱgrade,without genetic connection,were enrolled in this study,comprising 520 males and 480 females,aged(70.1±4.6) years and weighing(57.3±7.5) kg.No patients suffered from cognitive dysfunction. METHODS:The patients were equally and randomly divided into intravenous anesthesia and gas anesthesia groups.Total intravenous anesthesia and inhaled anesthesia were used.Genomic DNA from whole blood was extracted.The ApoE gene was amplified by PCR.Restriction fragment length polymorphism of ApoE gene was analyzed.Cognitive function was evaluated by Mini-Mental State Examination(MMSE).Patients scoring<25 points were diagnosed with cognitive dysfunction. MAIN OUTCOME MEASURES:Correlation of ApoE gene frequency and ApoEε4 allele to MMSE scores was measured. RESULTS:MMSE scores in patients from the gas anesthesia group significantly decreased 3 days after surgery,compared with the intravenous anesthesia group.The proportion of patients that scored<25 points was significantly greater in the gas anesthesia group compared with the intravenous anesthesia group 3 days after surgery.Reduced MMSE scores closely correlated with expression of the ApoEε4 allele in the gas anesthesia group(odds ratio = 2.83;95%confidence interval was 1.25-6.39,P<0.05).However,decreased MMSE scores did not closely correlated with expression of the ApoEε4 allele in the intravenous anesthesia group(odds ratio = 0.96;95% confidence interval was 0.37-2.39,P>0.05). CONCLUSION:Results demonstrated a correlation between cognitive dysfunction and ApoE single nucleotide polymorphism in elderly patients after gas anesthesia.However,no relationship between cognitive dysfunction and ApoE single nucleotide polymorphism was determined in elderly patients following intravenous anesthesia.Therefore,elderly patients,especially those expressing the ApoEε4 gene,should be cautiously exposed to gas anesthesia.Yingmin Cai Haitao Hu Rongliang Xue Pengbin Liu Gaifeng Feng Weijiang Dong Bin Yu Minggang Zhao 2009Neural Regeneration Research2009,4,4:0
