| 1 | Integrative analysis of DNA methylation and gene expression identified cervical cancer-specific diagnostic biomarkers显示文摘Cervical cancer is the leading cause of death among women with cancer worldwide.Here,we performed an integrative analysis of Illumina HumanMethylation450K and RNA-seq data from TCGA to identify cervical cancer-specific DNA methylation markers.We first identified differentially methylated and expressed genes and examined the correlation between DNA methylation and gene expression.The DNA methylation profiles of 12 types of cancers,including cervical cancer,were used to generate a candidate set,and machine-learning techniques were adopted to define the final cervical cancer-specific markers in the candidate set.Then,we assessed the protein levels of marker genes by immunohistochemistry by using tissue arrays containing 93 human cervical squamous cell carcinoma samples and cancer-adjacent normal tissues.Promoter methylation was negatively correlated with the local regulation of gene expression.In the distant regulation of gene expression,the methylation of hypermethylated genes was more likely to be negatively correlated with gene expression,while the methylation of hypomethylated genes was more likely to be positively correlated with gene expression.Moreover,we identified four cervical cancer-specific methylation markers,cg07211381(RAB3C),cg12205729(GABRA2),cg20708961(ZNF257),and cg26490054(SLC5A8),with 96.2%sensitivity and 95.2%specificity by using the tenfold cross-validation of TCGA data.The four markers could distinguish tumors from normal tissues with a 94.2,100,100,and 100%AUC in four independent validation sets from the GEO database.Overall,our study demonstrates the potential use of methylation markers in cervical cancer diagnosis and may boost the development of new epigenetic therapies. | Wanxue Xu Mengyao Xu Longlong Wang Wei Zhou Rong Xiang Yi Shi Yunshan Zhang Yongjun Piao | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 4 |
| 2 | Using irrigation intervals to optimize water-use efficiency and maize yield in Xinjiang,northwest China显示文摘Worldwide, scarce water resources and substantial food demands require efficient water use and high yield.This study investigated whether irrigation frequency can be used to adjust soil moisture to increase grain yield and water use efficiency(WUE) of high-yield maize under conditions of mulching and drip irrigation.A field experiment was conducted using three irrigation intervals in 2016: 6, 9, and 12 days(labeled D6, D9, and D12) and five irrigation intervals in 2017: 3, 6, 9, 12, and 15 days(D3, D6, D9, D12, and D15).In Xinjiang, an optimal irrigation quota is 540 mm for high-yield maize.The D3, D6, D9, D12, and D15 irrigation intervals gave grain yields of 19.7, 19.1–21.0, 18.8–20.0, 18.2–19.2, and 17.2 Mg ha^-1 and a WUE of 2.48, 2.53–2.80, 2.47–2.63, 2.34–2.45, and 2.08 kg m-3, respectively.Treatment D6 led to the highest soil water storage, but evapotranspiration and soil-water evaporation were lower than other treatments.These results show that irrigation interval D6 can help maintain a favorable soil-moisture environment in the upper-60-cm soil layer, reduce soilwater evaporation and evapotranspiration, and produce the highest yield and WUE.In this arid region and in other regions with similar soil and climate conditions, a similar irrigation interval would thus be beneficial for adjusting soil moisture to increase maize yield and WUE under conditions of mulching and drip irrigation. | Guoqiang Zhang Dongping Shen Bo Ming Ruizhi Xie Xiuliang Jin Chaowei Liu Peng Hou Jun Xue Jianglu Chen Wanxu Zhang Wanmao Liu Keru Wang Shaokun Li | 2019 | The Crop Journal2019,7,3: | 4 |
| 3 | Dynamic mechanical loading facilitated chondrogenic differentiation of rabbit BMSCs in collagen scaffolds显示文摘Mechanical signals have been played close attention to regulate chondrogenic differentiation of bone marrow mesenchymal stem cells(BMSCs).In this study,dynamic mechanical loading simulation with natural frequencies and intensities were applied to the 3D cultured BMSCs–collagen scaffold constructs.We investigated the effects of dynamic mechanical loading on cell adhesion,uniform distribution,proliferation,secretion of extracellular matrix(ECM)and chondrogenic differentiation of BMSCs–collagen scaffold constructs.The results indicated that dynamic mechanical loading facilitated the BMSCs adhesion,uniform distribution,proliferation and secretion of ECM with a slight contraction,which significantly improved the mechanical strength of the BMSCs–collagen scaffold constructs for better mimicking the structure and function of a native cartilage.Gene expression results indicated that dynamic mechanical loading contributed to the chondrogenic differentiation of BMSCs with higher levels of AGG,COL2A1 and SOX9 genes,and prevented of hypertrophic process with lower levels of COL10A1,and reduced the possibility of fibrocartilage formation due to down-regulated COL1A2.In conclusion,this study emphasized the important role of dynamic mechanical loading on promoting BMSCs chondrogenic differentiation and maintaining the cartilage phenotype for in vitro reconstruction of tissue-engineered cartilage,which provided an attractive prospect and a feasibility strategy for cartilage repair. | Wanxu Cao Weimin Lin Hanxu Cai Yafang Chen Yi Man Jie Liang Qiguang Wang Yong Sun Yujiang Fan Xingdong Zhang | 2019 | Regenerative Biomaterials2019,6,2: | 3 |
