1Molecular identification of bloom-forming species Phaeocystis globosa(Prymnesiophyta)and its dispersal based on rDNA ITS sequence analysis显示文摘The colony-forming Phaeocystis species are causative agents of dense bloom occurrences in coastal waters worldwide. It is difficult to separate them because of the different morphologies associated with their colonial stages. In this study we applied molecular approaches to analyze the genetic variation of Phaeocystis globosa and Phaeocystis pouchetii from several geographic regions, and to assist in tracing the dispersal of bloom-forming Phaeocystis species in coastal waters of China. The sequences of the internal transcribed spacers ( ITS1 and ITS2) of rDNA and the 5.8S ribosomal RNA gene of Phaeocystis strains were determined. Sequence comparison shows that P.globosa was the most divergent to P. pouchetii, exhibiting sequence divergence higher than 0.08. However, lower genetic divergences existed between strains of P. globosa. The sequence comparison of the Phaeocystis rDNA ITS clearly shows that the species isolated from the southeast coast of China is identified as P. globosa rather than P. cf. pouchetii or other species. Furthermore, the significance of rDNA variation in distinct global populations of P. globosa suggested it might have had sufficient time to accumulate detectable mutations at the rDNA locus, supporting the hypothesis of ancient dispersal of P.globosa to many areas, meaning that P. globosa blooms in the coastal waters of China are endemic rather than a newly introduced species or a foreign source. Finally, based on the high divergent region of rDNA ITS, a pair of species-specific primers for P. globosa were designed, they could be useful to detect the presence of this species in mixed plankton assemblages or flagellate stages that are recognized with diffculties by means of conventional microscopy.ChenYueqin ShaoPeng WangNing ZhouHui QuLianghu LindaK.Medlin 2003Acta Oceanologica Sinica2003,22,2:9

3Injectable collagen scaffold promotes swine myocardial infarction recovery by long-term local retention of transplanted human umbilical cord mesenchymal stem cells显示文摘Stem cell therapy is an attractive approach for recovery from myocardial infarction(MI)but faces the challenges of rapid diffusion and poor survival after transplantation.Here we developed an injectable collagen scaffold to promote the long-term retention of transplanted cells in chronic MI.Forty-five minipigs underwent left anterior descending artery(LAD)ligation and were equally divided into three groups 2 months later(collagen scaffold loading with human umbilical mesenchymal stem cell(hUMSC)group,hUMSC group,and placebo group(only phosphate-buffered saline(PBS)injection)).Immunofluorescence staining indicated that the retention of transplanted cells was promoted by the collagen scaffold.Echocardiography and cardiac magnetic resonance imaging(CMR)showed much higher left ventricular ejection fraction(LVEF)and lower infarct size percentage in the collagen/hUMSC group than in the hUMSC and placebo groups at 12 months after treatment.There were also higher densities of vWf-,α-sma-,and cTnT-positive cells in the infarct border zone in the collagen/cell group,as revealed by immunohistochemical analysis,suggesting better angiogenesis and more cardiomyocyte survival after MI.Thus,the injectable collagen scaffold was safe and effective on a large animal myocardial model,which is beneficial for constructing a favorable microenvironment for applying stem cells in clinical MI.Qiang Wangn Xiaojun He Bin Wang Jun Pan Chunying Shi Jie Li Liudi Wang Yannan Zhao Jianwu Dai Dongjin Wang 2021Science China(Life Sciences)2021,64,2:5
4SPATIAL ANALYSIS OF SOIL EROSION AND NON-POINT SOURCE POLLUTION BASED ON GIS IN ERLONG LAKE WATERSHED, JILIN PROVINCE显示文摘Data collection, factor composition, nappe analysis and integrative simulation of natural geographical factors in Erlong Lake watershed have been carried out based on GIS. The risk areas where non-point source pollution may occur were compartmentalized and assessed, and the total soil erosion and the runoffs of N and P with rainfall in this valley were worked out by experiment and GIS mapping. The study indicated that the main type of soil erosion was moderate (erosion modulus is 1000-2500t/(km2·a)) at present , and the intense erosion areas are located in dry land with variable slope east of the lake and the midded ied ied ied ied ied ied ied iuntainous region (erosion modulus is more than 5000t/(km2·a)). Though the area is small, it should be paid attention to. The trend of non-point source pollution (NSP) of nitrogen and phosphorus loss was corresponded with the soil erosion. Spatial distribution and the reasons of the distribution difference have been presented and it was emphasized that the human activities among the influence factors was the most important.
