2Sintilimab versus docetaxel as second-line treatment in advanced or metastatic squamous non-small-cell lung cancer:an open-label,randomized controlled phase 3 trial(ORIENT-3)显示文摘Background:Treatment options for Chinese patients with locally advanced or metastatic squamous-cell non-small-cell lung cancer(sqNSCLC)after failure of first-line chemotherapy are limited.This study(ORIENT-3)aimed to evaluate the efficacy and safety of sintilimab versus docetaxel as second-line treatment in patients with locally advanced or metastatic sqNSCLC.Methods:ORIENT-3 was an open-label,multicenter,randomized controlled phase 3 trial that recruited patients with stage IIIB/IIIC/IV sqNSCLC after failure with first-line platinum-based chemotherapy.Patients were randomized in a 1:1 ratio to receive either 200 mg of sintilimab or 75 mg/m^(2) of docetaxel intravenously every 3 weeks,stratified by the Eastern Cooperative Oncology Group performance status.The primary endpoint was overall survival(OS)in the full analysis set(FAS).Secondary endpoints included progression-free survival(PFS),objective response rate(ORR),disease control rate(DCR),duration of response(DoR)and safety.Results:Between August 25,2017,and November 7,2018,290 patients were randomized.For FAS,10 patients fromthe docetaxel armwere excluded.Themedian OS was 11.79(n=145;95%confidence interval[CI],10.28-15.57)months with sintilimab versus 8.25(n=135;95%CI,6.47-9.82)months with docetaxel(hazard ratio[HR]:0.74;95%CI,0.56-0.96;P=0.025).Sintilimab treatment significantly prolonged PFS(median 4.30 vs.2.79 months;HR:0.52;95%CI,0.39-0.68;P<0.001)and showed higher ORR(25.50%vs.2.20%,P<0.001)and DCR(65.50%vs.37.80%,P<0.001)than the docetaxel arm.The median DoRwas 12.45(95%CI,4.86-25.33)months in the sintilimab arm and 4.14(95%CI,1.41-7.23)months in the docetaxel arm(P=0.045).Treatment-related adverse events of grade≥3were reported in 26(18.1%)patients in the sintilimab arm and 47(36.2%)patients in the docetaxel arm.Exploratory biomarker analysis showed potential predictive values of expression levels of two transcription factors,including OVOL2(HR:0.35;P<0.001)and CTCF(HR:3.50;P<0.001),for sintilimab treatment.Conclusions:Compared with docetaxel,sintilimab significantly improved the OS,PFS,and ORR of Chinese patients with previously treated locally advanced or metastatic sqNSCLC.Yuankai Shi Lin Wu Xinmin Yu Puyuan Xing Yan Wang Jianying Zhou Airong Wang Jianhua Shi Yi Hu Ziping Wang Guangyu An Yong Fang Sanyuan Sun Caicun Zhou Changli Wang Feng Ye Xingya Li Junye Wang Mengzhao Wang Yunpeng Liu Yanqiu Zhao Ying Yuan Jifeng Feng Zhendong Chen Jindong Shi Tao Sun Gang Wu Yongqian Shu Qisen Guo Yi Zhang Yong Song Shucai Zhang Yuan Chen Wei Li Hongrui Niu Wenwei Hu Lijun Wang Jianan Huang Yang Zhang Ying Cheng Zhengdong Wu Bo Peng Jiya Sun Christoph Mancao Yanqi Wang Luyao Sun 2022Cancer Communications2022,42,12:8
3Unsteady aerodynamic characteristics of a horizontal wind turbine under yaw and dynamic yawing显示文摘Horizontal axis wind turbine(HAWT)often works under yaw due to the stochastic variation of wind direction.Yaw also can be used as one of control methods for load reduction and wake redirection of HAWT.Thus,the aerodynamic performance under yaw is very important to the design of HAWT.For further insight into the highly unsteady characteristics aerodynamics of HAWT under yaw,this paper investigates the unsteady variations of the aerodynamic performance of a small wind turbine under static yawed and yawing process with sliding grid method,as well as the there-dimensional effect on the unsteady characteristics,using unsteady Reynolds-averaged Navier-Stokes(URANS)simulations.The simulation results are validated with experimental data and blade element momentum(BEM)results.The comparisons show that the CFD results have better agreement with the experimental data than both BEM results.The wind turbine power decreases according to a cosine law with the increase of yaw angle.The torque under yaw shows lower frequency fluctuations than the non-yawed condition due to velocity component of rotation and the influence of spinner.Dynamic yawing causes larger fluctuate than static yaw,and the reason is analyzed.The aerodynamic fluctuation becomes more prominent in the retreating side than that in the advancing side for dynamic yawing case.Variations of effective angle of attack and aerodynamic forces along the blade span are analyzed.The biggest loading position moves from middle span to outer span with the increase of yaw angle.Three-dimensional stall effect presents load fluctuations at the inner board of blade,and becomes stronger with the increase of yaw angle.Zhaoliang Ye Xiaodong Wang Ziwen Chen Luyao Wang 2020Acta Mechanica Sinica2020,36,2:8
4The calcium-dependent protein kinase ZmCDPK7 functions in heat-stress tolerance in maize显示文摘Global warming poses a serious threat to crops.Calcium-dependent protein kinases(CDPKs)/CPKs play vital roles in plant stress responses,but their exact roles in plant thermotolerance remains elusive.Here,we explored the roles of heat-induced ZmCDPK7 in thermotolerance in maize.ZmCDPK7-overexpressing maize plants displayed higher thermotolerance,photosynthetic rates,and antioxidant enzyme activity but lower H2 O2 and malondialdehyde(MDA)contents than wild-type plants under heat stress.ZmCDPK7-knockdown plants displayed the opposite patterns.ZmCDPK7 is attached to the plasma membrane but can translocate to the cytosol under heat stress.ZmCDPK7 interacts with the small heat shock protein sHSP17.4,phosphorylates sHSP17.4 at Ser-44 and the respiratory burst oxidase homolog RBOHB at Ser-99,and up regulates their expression.Site-directed mutagenesis of sHSP17.4 to generate a Ser-44-Ala substitution reduced ZmCDPK7’s enhancement of catalase activity but enhanced ZmCDPK7’s suppression of MDA accumulation in heat-stressed maize protoplasts.sHSP17.4,ZmCDPK7,and RBOHB were less strongly upregulated in response to heat stress in the abscisic acid-deficient mutant vp5 versus the wild type.Pretreatment with an RBOH inhibitor suppressed sHSP17.4 and ZmCDPK7 expression.Therefore,abscisic acid-induced ZmCDPK7 functions both upstream and downstream of RBOH and participates in thermotolerance in maize by mediating the phosphorylation of sHSP17.4,which might be essential for its chaperone function.Yulong Zhao Hanwei Du Yankai Wang Huali Wang Shaoyu Yang Chaohai Li Ning Chen Hao Yang Yihao Zhang Yulin Zhu Luyao Yang Xiuli Hu 2021Journal of Integrative Plant Biology2021,63,3:6
5Silicon lithium-ion battery anode with enhanced performance: Multiple effects of silver nanoparticles显示文摘Silicon has been regarded as one of the most promising next generation lithium-ion battery anode. However, the poor cyclic stability of the Si based anode has severely limited its practical applications, which is even worse with high mass loading density(>1 mg cm^(-2)). A new concept has been developed to enhance the electrochemical performance of the Si nanoparticle anode. Silver nanoparticles are composited with the silicon nanoparticles in a facile way for the first time. It is found that the mechanical properties of the Si electrode have been significantly improved by the incorporation of the silver nanoparticles, leading to enhanced cyclic performance. With the Si/Ag mass ratio of 4:1, the reversible specific discharge capacity is retained as 1156 m A h g^(-1) after 100 cycles at 200 mA g^(-1), which is more than three times higher than that of the bare silicon(318 mA h g^(-1)). 6Impacts of Park Landscape Structure on Thermal Environment Using QuickBird and Landsat Images显示文摘Urban parks composed mostly of vegetation and water bodies can effectively mitigate the urban heat island effect. Many studies have investigated the cooling effects of urban parks; however, little attention has been given to park landscape structure. Based on landscape metrics, this study has explored the influences of the park landscape structure on its inner thermal environment, taking heavily urbanized Beijing Municipality in China as the study area. Three indices, including the percentage of landscape(PLAND), landscape shape index(LSI) and aggregation index(AI), were used to measure the composition and configuration characteristics of the landscape components inside the parks. The indices were calculated for five landscape types being interpreted from Quickbird images. Urban thermal conditions were measured using the land surface temperature(LST) derived from Landsat TM images. The results showed that the park LST had a negative relationship with the park size, but no significant relationship was found with park shape. For the park's interior landscape, however, the configuration and composition characteristics of the landscape components inside the park explained 70% of the park LST variance. The area percentage of water bodies and the aggregation index of woodland were identified as the key influencing characteristics. In addition, when the composition and configuration characteristics of the park landscape components were separately considered, the configuration characteristics(LSI and AI) explained approximately 54% of the variance in park LST, which was comparable with that explained by the composition characteristics(PLAND). 7Chromosome doubling of Sedum alfredii Hance: A novel approach for improving phytoremediation efficiency显示文摘Sedum alfredii Hance is a cadmium(Cd)/zinc(Zn)hyperaccumulator native to China.However,its relatively low biomass restricted the large-scale application for heavy metal contamination remediation.The chromosome set doubling of S.alfredii in vitro was achieved by 0.1%–0.2%(W/V)colchicine treatment.The plant DNA ploidy was analyzed by flow cytometry and chromosome set doubling plants(CSD)were identified based on the obvious different sharp peak.A tissue culture experiment with different Cd treated levels and a field trial with natural polluted mined soil were conducted to study the effects of chromosome doubling on plant biomass and Cd accumulation in shoots.The results suggested that S.alfredii is a mixoploid.Compared with the wild type plants(WT),CSD exhibited typical'gigas'characteristics in morphology including stem thickness,root hair production,number of leaves and size of stoma guard cell.Fresh weight and dry weight of CSD were increased to 1.62–2.03-fold and 2.26–3.25-fold of WT.And Cd content of CSD showed a17.49%–42.82%increase and 59%increase under tissue culture and field condition,accordingly.In addition,the TF and in BCF of CSD were 2.37-and 1.59-fold of WT,respectively.These results proved that it is feasible to promote phytoextraction efficiency of S.alfredii in Cd contaminated soils through chromosomal engineering,which provides a novel approach for hyperaccumulator application in phytoremediation.Ying Feng Qiong Wang Qian Meng Yongjun Liu Fengshan Pan Sha Luo Yingjie Wu Luyao Ma Xiaoe Yang 2019Journal of Environmental Sciences2019,31,12:4
8Toxicity and mechanisms of action of titanium dioxide nanoparticles in living organisms显示文摘Titanium dioxide nanoparticles(TiO_2 NPs) are one of the most widely used nanomaterials in the consumer products, agriculture, and energy sectors. Their large demand and widespread applications will inevitably cause damage to organisms and ecosystems. A better understanding of TiO_2 NP toxicity in living organisms may promote risk assessment and safe use practices of these nanomaterials. This review summarizes the toxic effects of TiO_2 NPs on multiple taxa of microorganisms, algae, plants, invertebrates, and vertebrates. 10Engineering self-healing adhesive hydrogels with antioxidant properties for intrauterine adhesion prevention显示文摘Intrauterine adhesion (IUA) is the fibrosis within the uterine cavity. It is the second most common cause of female infertility, significantly affecting women’s physical and mental health. Current treatment strategies fail to provide a satisfactory therapeutic outcome for IUA patients, leaving an enormous challenge for reproductive science. A self-healing adhesive hydrogel with antioxidant properties will be highly helpful in IUA prevention. In this work, we prepare a series of self-healing hydrogels (P10G15, P10G20, and P10G25) with antioxidant and adhesive properties. Those hydrogels exhibit good self-healing properties and can adapt themselves to different structures. They possess good injectability and fit the shape of the human uterus. Moreover, the hydrogels exhibit good tissue adhesiveness, which is desirable for stable retention and therapeutic efficacy. The in vitro experiments using P10G20 show that the adhesive effectively scavenges ABTS+, DPPH, and hydroxyl radicals, rescuing cells from oxidative stress. In addition, P10G20 offers good hemocompatibility and in vitro and in vivo biocompatibility. Furthermore, P10G20 lowers down the in vivo oxidative stress and prevents IUA with less fibrotic tissue and better endometrial regeneration in the animal model. It can effectively downregulate fibrosis-related transforming growth factor beta 1 (TGF-β1) and vascular endothelial growth factor (VEGF). 12Astragaloside Ⅳ suppresses post-ischemic natural killer cell infiltration and activation in the brain:involvement of histone deacetylase inhibition显示文摘Natural killer(NK)cells,a type of cytotoxic lymphocytes,can infiltrate into ischemic brain and exacerbate neuronal cell death.Astragaloside Ⅳ(ASIV)is the major bioactive ingredient of Astragalus membranaceus,a Chinese herbal medicine,and possesses potent immunomodulatory and neuroprotective properties.This study investigated the effects of ASIV on post-ischemic brain infiltration and activation of NK cells.ASIV reduced brain infarction and alleviated functional deficits in MCAO rats,and these beneficial effects persisted for at least 7 days.Abundant NK cells infiltrated into the ischemic hemisphere on day 1 after brain ischemia,and this infiltration was suppressed by ASIV.Strikingly,ASIV reversed NK cell deficiency in the spleen and blood after brain ischemia.ASIV inhibited astrocyte-derived CCL2 upregulation and reduced CCR2+NK cell levels in the ischemic brain.Meanwhile,ASIV attenuated NK cell activating receptor NKG2D levels and reduced interferon-γproduction.ASIV restored acetylation of histone H3 and the p65 subunit of nuclear factor-κB in the ischemic brain,suggesting inhibition of histone deacetylase(HDAC).Simultaneously,ASIV prevented p65 nuclear translocation.The effects of ASIV on reducing CCL2 production,restoring acetylated p65 levels and preventing p65 nuclear translocation were mimicked by valproate,an HDAC inhibitor,in astrocytes subjected to oxygen-glucose deprivation.Our findings suggest that ASIV inhibits post-ischemic NK cell brain infiltration and activation and reverses NK cell deficiency in the periphery,which together contribute to the beneficial effects of ASIV against brain ischemia.Furthermore,ASIV’s effects on suppressing NK cell brain infiltration and activation may involve HDAC inhibition.Baokai Dou Shichun Li Luyao Wei Lixin Wang Shiguo Zhu Zhengtao Wang Zunji Ke Kaixian Chen Zhifei Wang 2021Frontiers of Medicine2021,15,1:3

14Modulation of Beta Oscillations for Implicit Motor Timing in Primate Sensorimotor Cortex during Movement Preparation显示文摘Motor timing is an important part of sensorimotor control. Previous studies have shown that beta oscillations embody the process of temporal perception in explicit timing tasks. In contrast, studies focusing on beta oscillations in implicit timing tasks are lacking. In this study, we set up an implicit motor timing task and found a modulation pattern of beta oscillations with temporal perception during movement preparation. We trained two macaques in a repetitive visually-guided reach-to-grasp task with different holding intervals. Spikes and local field potentials were recorded from microelectrode arrays in the primary motor cortex, primary somatosensory cortex, and posterior parietal cortex. We analyzed the association between beta oscillations and temporal interval in fixedduration experiments(500 ms as the Short Group and1500 ms as the Long Group) and random-duration experiments(500 ms to 1500 ms). The results showed that the peak beta frequencies in both experiments ranged from15 Hz to 25 Hz. The beta power was higher during the hold period than the movement(reach and grasp) period.Further, in the fixed-duration experiments, the mean poweras well as the maximum rate of change of beta power in the first 300 ms were higher in the Short Group than in the Long Group when aligned with the Center Hit event. In contrast, in the random-duration experiments, the corresponding values showed no statistical differences among groups. The peak latency of beta power was shorter in the Short Group than in the Long Group in the fixed-duration experiments, while no consistent modulation pattern was found in the random-duration experiments. These results indicate that beta oscillations can modulate with temporal interval in their power mode. 17How text sentiment moderates the impact of motivational cues on crowdfunding campaigns显示文摘Arousing funders’motives for crowdfunding participation is a challenge for every entrepreneur.According to self-determination theory,crowdfunding involvement is driven by two kinds of motivations:extrinsic and intrinsic motives.Campaign narratives with different sentimental orientations could act as cues triggering funders’motives of capital-giving differently.Thus,we focus on the moderating effect of text sentiment on the motivational cues in crowdfunding campaigns.We hypothesize that the sentiment of campaign narratives moderates the relationship between motivational cues and fundraising success.Data are selected from 270,000 campaigns of the crowdfunding website Indiegogo.Empirical analysis demonstrates that the sentiment strength positively moderates the effect of intrinsic motives on fundraising success but negatively moderates the impact of extrinsic motives on fundraising success.The findings provide managerial insights for practitioners to stimulate specified motives by using the narratives with proper sentiment strength.Xiang Yuan Luyao Wang Xicheng Yin Hongwei Wang 2021Financial Innovation2021,7,1:1
18Salt stress-induced changes in soil metabolites promote cadmium transport into wheat tissues显示文摘Soil salinity is known to improve cadmium(Cd)mobility,especially in arid soils.However,the mechanisms involved in how salt stress-associated metabolic profiles participate in mediating Cd transport in the soil-plant system remain poorly understood.This study was designed to investigate the effects of salinity-induced changes in soil metabolites on Cd bioavailability.Sodium salts in different combinations according to molar ratio(NaCl:Na_(2)SO_(4)=1:1;NaCl:Na_(2)SO_(4):NaHCO_(3)=1:2:1;NaCl:Na_(2)SO_(4):NaHCO_(3):Na_(2)CO_(3)=1:9:9:1;NaCl:Na_(2)SO_(4):NaHCO_(3):Na_(2)CO_(3)=1:1:1:1)were applied to the Cd-contaminated soils,which increased soil Cd availability by 22.36%and the Cd content in wheat grains by 36.61%,compared to the control.Salt stress resulted in soil metabolic reprogramming,which might explain the decreased growth of wheat plants and increased Cd transport from the soil into wheat tissues.For example,down-regulation of starch and sucrose metabolism reduced the production of sugars,which adversely affected growth;up-regulation of fatty acid metabolism allowed wheat plants to maintain a normal intracellular environment under saline conditions;up-regulation of the tricarboxylic acid(TCA)cycle was triggered,causing an increase in organic acid synthesis and the accumulation of organic acids,which facilitated the migration of soil Cd into wheat tissues.In summary,salt stress can facilitate Cd transport into wheat tissues by the direct effect of salt-based ions and the combined effect of altered soil physicochemical properties and soil metabolic profiles in Cd-contaminated soils.Lifu Wang Luyao Qin Xiaoyi Sun Shuwen Zhao Lei Yu Shibao Chen Meng Wang 2023Journal of Environmental Sciences2023,,5:1
19Low-Temperature Aging Provides 22% E cient Bromine-Free and Passivation Layer-Free Planar Perovskite Solar Cells显示文摘Previous reports of formamidinium/methylamine(FAMA)-mixed halide perovskite solar cells have focused mainly on controlling the morphology of the perovskite film and its interface—for example,through the inclusion of bromine and surface passivation.In this paper,we describe a new processing pathway for the growth of a high-quality bromine-free FAMAPbI3 halide perovskites via the control of intermediate phase.Through low-temperature aging growth(LTAG)of a freshly deposited perovskite film,α-phase perovskites can be seeded in the intermediate phase and,at the same time,prevent beta-phase perovskite to nucleate.After postannealing,large grain-size perovskites with significantly reduced PbI2 presence on the surface can be obtained,thereby eliminating the need of additional surface passivation step.Our pristine LTAG-treated solar cells could provide PCEs of greater than 22%without elaborate use of bromine or an additional passivation layer.More importantly,when using this LTAG process,the growth of the pure alpha-phase FAMAPbI3 was highly reproducible.Xin Wang Luyao Wang Tong Shan Shibing Leng Hongliang Zhong Qinye Bao Zheng-Hong Lu Lin-Long Deng Chun-Chao Chen 2020Nano-Micro Letters2020,12,7:1
20NSUN2-mediated mRNA m^(5)C Modification Regulates the Progression of Hepatocellular Carcinoma显示文摘RNA modifications affect many biological processes and physiological diseases.The 5-methylcytosine(m^(5)C)modification regulates the progression of multiple tumors.However,its characteristics and functions in hepatocellular carcinoma(HCC)remain largely unknown.Here,we found that HCC tissues had a higher m^(5)C methylation level than the adjacent normal tissues.Transcriptome analysis revealed that the hypermethylated genes mainly participated in the phosphokinase signaling pathways,such as the Ras and PI3K-Akt pathways.The m^(5)C methyltransferase NSUN2 was highly expressed in HCC tissues.Interestingly,the expression of many genes was positively correlated with the expression of NSUN2,including GRB2,RNF115,AATF,ADAM15,RTN3,and HDGF.Real-time PCR assays further revealed that the expression of the mRNAs of GRB2,RNF115,and AATF decreased significantly with the down-regulation of NSUN2 expression in HCC cells.Furthermore,NSUN2 could regulate the cellular sensitivity of HCC cells to sorafenib via modulating the Ras signaling pathway.Moreover,knocking down NSUN2 caused cell cycle arrest.Taken together,our study demonstrates the vital role of NSUN2 in the progression of HCC.Dan Song Ke An Wenlong Zhai Luyao Feng Yingjie Xu Ran Sun Yueqin Wang Yun-Gui Yang Quancheng Kan Xin Tian 2023Genomics, Proteomics & Bioinformatics2023,21,4:1
