| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | China's regional transport dominance: Density, proximity, and accessibility显示文摘Transport infrastructure plays an important role in shaping the configuration of spatial socio-economic structures and influences regional accessibility.This paper defines transport dominance from three aspects:quality,quantity,and advantage,measured by density,proximity,and accessibility indices.County is the basic unit for analysis.The results reveal:(1)Transport dominance statistically follows a partial normal distribution.A very few counties,1.4%of the total,have extremely high transport dominance which strongly supports the socio-economic development in these areas.In contrast,one eighth of all counties have poor transport dominance which impedes local socio-economic development to some extent. | 4 | Phylogenetic reconstruction of Chirita and allies (Gesneriaceae) with taxonomic treatments显示文摘Chirita D. Don, a large genus in the subfamily Cyrtandroideae of Gesneriaceae, has been the subject of much debate whether it is a natural group or not. In addition, the highly heterogeneous Chirita has also been very problematic with regard to delimitation and subdivision. Here we used the nrDNA internal transcribed spacer and cpDNA trnL-F for molecular phylogenetic analaysis, combined with morphological data. Our results suggest that Chirita is an artificial, polyphyletic genus. The most important character that defines Chirita, the dorso-ventrally oblique and bilamellar stigma, has evolved convergently in different clades of diandrous Cyrtandroideae. Chirita sensu stricto only includes the species of Chirita sect. Chirita, whereas Chirita sect. Microchirita is an independent clade located at the basal node of the phylogenetic tree. Chirita sect. Liebigia is closely related to Didymocarpus with an entire stigma unlike other species of Chirita. The species of Chirita sect. Gibbosaccus, Chiritopsis, Primulina, and Wentsaiboea form a monophyletic group that is sister to a strongly supported clade comprising four monotypic genera Paralagarosolen, Calcareoboea, Petrocodon, and Tengia. | 9 | In vitro differentiation of adipose-derived stem cells and bone marrow-derived stromal stem cells into neuronal-like cells显示文摘Adipose-derived stem cells and bone marrow-derived stromal stem cells were co-cultured with untreated or Aβ1-40-treated PC12 cells, or grown in supernatant derived from untreated or Aβ1-40-treated PC12 cells. Analysis by western blot and quantitative real-time PCR showed that protein levels of Nanog, Oct4, and Sox2, and mRNA levels of miR/125a/3p were decreased, while expression of insulin-like growth factor-2 and neuron specific enolase was increased. In comparison, the generation of neuron specific enolase-positive cells was most successful when adipose-derived stem cells were co-cultured with Aβ1-40-treated PC12 cells. Our results demonstrate that adipose-derived stem cells and bone marrow-derived stromal stem cells exhibit trends of neuronal-like cell differentiation after co-culture with Aβ1-40-treated PC12 cells. | 10 | Chimeric antigen receptor-engineered T-cell therapy for liver cancer显示文摘Background: Chimeric antigen receptor-engineered T-cell(CAR-T) therapy is a newly developed immunotherapy used in the treatment of cancers. Because CAR-T therapy has shown great success in treating CD19-positive hematological malignancies, its application has been explored in the treatment of solid tumors, such as liver cancer. In this review, we discuss the immune characteristics of liver cancer, the obstacles encountered during the application of CAR-T therapy, and preclinical and clinical progress in the use of CAR-T therapy in patients with liver cancer.Data sources: The data on CAR-T therapy related to liver cancers were collected by searching Pub Med and the Web of Science databases prior to December 2017 with the keywords 'chimeric antigen receptor','CAR-T', 'liver cancer', 'hepatocellular carcinoma', and 'solid tumor'. Additional articles were identified by manual search of references found in the primary articles. The data for clinical trials were collected by searching Clinical Trials.gov.Results: The liver has a tolerogenic nature in the intrahepatic milieu and its tumor microenvironment significantly affects tumor progression. The obstacles that reduce the efficacy of CAR-T therapy in solid tumors include a lack of specific tumor antigens, limited trafficking and penetration of CAR-T cells to tumor sites, and an immunosuppressive tumor microenvironment. To overcome these obstacles, several strategies have emerged. In addition, several strategies have been developed to manage the side effects of CAR-T, including enhancing the selectivity of CARs and controlling CAR-T activity. To date, no clinical trials of CAR-T therapy against HCC have been completed. However, preclinical studies in vitro and in vivo have shown potent antitumor efficacy. Glypican-3, mucin-1, epithelial cell adhesion molecule, carcinoembryonic antigen, and other targets are currently being studied.Conclusions: The application of CAR-T therapy for liver cancer is just beginning to be explored and more research is needed. | 13 | Atmospheric particulate pollution of Lanzhou using magnetic measurements显示文摘To differentiate between natural and anthropogenic particulate sources in the atmosphere in Lanzhou City, samples were collected in different sites. The dust flux was calculated and magnetic measurements were conducted. Results show a distinct pattern of variation of dust flux within a year and it agrees with the shifts of atmospheric circulation regime. The magnetic parameters indicate that natural sources are the major components of atmospheric particulate during late spring and early summer, while anthropogenic sources contribute much more during winter months. | 14 | Overview on current criteria for heavy metals and its hint for the revision of soil environmental quality standards in China显示文摘Following rapid social and economic development over the past several decades, soil pollution by heavy metals(HMs) has been both serious and widespread in China. The Soil Environmental Quality Standards(SEQSs) in China(GB 15618-1995) have been introduced to encourage and enforce sustainable soil HM management. However, in recent years, HM contents in soils have frequently been found to exceed their associated standard values, while the crops growing on them might still meet regulatory standards, and vice versa. There is thus growing awareness that GB 15618-1995 does not effectively regulate current soil HM pollution, as it has encountered bottlenecks, and disappointing outcomes caused by poor execution along with deficiencies and gaps in the policies. However, due to the deficiency of scientific research about relationships between soil HM concentrations and their ecological or human health effects, the development of SEQSs in China is still greatly restricted. This paper discusses international SEQSs of HMs as well their development in China over time, then examines current Chinese SEQSs to demonstrate their potential regulatory deficiencies by referring to international SEQSs. The corresponding legislative policies are described, and scientific information or responses are outlined for maintaining soil environmental quality. | 17 | Calcium signaling and T-type calcium channels in cancer cell cycling显示文摘Regulation of intracellular calcium is an important signaling mechanism for cell proliferation in both normal and cancerous cells. In normal epithelial cells, free calcium concentration is essential for cells to enter and accomplish the S phase and the M phase of the cell cycle. In contrast, cancerous cells can pass these phases of the cell cycle with much lower cytoplasmic free calcium concentrations, indicating an alternative mechanism has developed for fulfilling the intracellular calcium requirement for an increased rate of DNA synthesis and mitosis of fast replicating cancerous cells. The detailed mechanism underlying the altered calcium loading pathway remains unclear; however, there is a growing body of evidence that suggests the T-type Ca2+ channel is abnormally expressed in cancerous cells and that blockade of these channels may reduce cell proliferation in addition to inducing apoptosis. Recent studies also show that the expression of T-type Ca2+ channels in breast cancer cells is proliferation state dependent, i.e. the channels are expressed at higher levels during the fast-replication period, and once the cells are in a non-proliferation state, expression of this channel isminimal. Therefore, selectively blocking calcium entry into cancerous cells may be a valuable approach for preventing tumor growth. | 19 | Grassland degradation in Northern Tibet based on remote sensing data显示文摘This study selected vegetation cover as the main evaluation index, calculated the grassland degradation index (GDI) and established the remote sensing monitoring and evaluation system for grassland degradation in Northern Tibet, according to the National Standard (GB19377-2003), based on the remote sensing data such as NDVI data derived from NOAA/AVHRR with a spatial resolution of 8 km of 1981-2000, from SPOT/VGT with a spatial resolution of 1 km of 2001 and from MODIS with a spatial resolution of 0.25 km of 2002-2004 respectively in this area, in combination with the actual condition of grassland degradation. The grassland degradation processes and their responses to climate change during 1981-2004 were discussed and analyzed in this paper. The result indicated that grassland degradation in Northern Tibet is very serious, and the mean value of GDI in recent 20 years is 2.54 which belongs to the serious degradation grade. From 1981 to 2004, the GDI fluctuated distinctly with great interannual variations in the proportion of degradation degree and GDI but the general tendency turned to severe-grade during this period with the grassland degradation grade changed from light degraded to serious degraded in Northern Tibet. The extremely serious degraded and serious degraded grassland occupied 1.7% and 8.0% of the study area, the moderate and light degraded grassland accounted for 13.2% and 27.9% respectively, and un-degraded grassland occupied 49.2% of the total grassland area in 2004. The grassland degradation was serious, especially in the conjunctive area of Naqu, Biru and Jiali counties, the headstream of the Yangtze River lying in the Galadandong snow mountain and glaciers, the area along the Qinghai-Tibet highway and railway, and areas around the Tanggula and Nianqingtanggula snow mountains and glaciers.
