1Helicobacter pylori may be an initiating factor in newly diagnosed ulcerative colitis patients: A pilot study显示文摘AIM To directly visualize Helicobacter pylori(H. pylori) by the highly sensitive and specific technique of immunohistochemical staining in colonic tissue from patients newly diagnosed with ulcerative colitis(UC).METHODS Colonoscopic biopsies from thirty patients with newly diagnosed UC and thirty controls were stained with Giemsa stain and immunohistochemical stain for detection of H. pylori in the colonic tissue. Results were confirmed by testing H. pylori Ag in the stool then infected patients were randomized to receive either anti H. pylori treatment or placebo.RESULTS Twelve/30(40%) of the UC patients were positive for H. pylori by Giemsa, and 17/30(56.6%) by immunohistochemistry stain. Among the control group 4/30(13.3%) and 6/30(20 %) were positive for H. pylori by Giemsa and immunohistochemistry staining respectively. H. pylori was significantly higher in UC than in controls(P = 0.04 and 0.007). All Giemsa positive patients and controls were positive by immunohistochemical stain. Four cases of the control group positive for H. pylori also showed microscopic features consistent with early UC.CONCLUSION H. pylori can be detected in colonic mucosa of patients with UC and patients with histological superficial ulcerations and mild infiltration consistent with early UC. There seems to be an association between UC and presence of H. pylori in the colonic tissue. 2Evaluation of DNA damage profile in obese women and its association to risk of metabolic syndrome,polycystic ovary syndrome and recurrent preeclampsia显示文摘Metabolic syndrome(MS)is a cluster of metabolic abnormalities.Obesity and MS are always accompanied by elevated oxidative stress which might affect cellular biomolecules such as DNA.The aim of the present study is to investigate DNA damage profile in obese premenopausal women and its relation to the risk of MS,polycystic ovary syndrome(PCOS)and history of recurrent pre-eclampsia.The study included 90 obese women included cases with MS(nZ30),PCOS(n Z30)and previous history of recurrent preeclampsia(nZ30)and,age-matched healthy non-obese control women(n Z 50).The assessment of leukocyte DNA damage was done by comet assay for all cases and controls.Anthropometry and biochemical parameters have been measured.Results showed that mean percent of DNA damage was significantly higher in MS,PCOS as well as in women with the recurrent preeclampsia as compared to healthy controls.The high level of mean DNA damage frequency in obese women was significantly associated with the increased number of metabolic syndrome components.Cases with 2,3 and 3e5 components showed significantly higher levels of DNA damage than controls.Moreover,cases with 3e5 MS components showed significant higher DNA compared to those with the two components.Regarding PCOS,significant positive association between the mean frequency of DNA damage and waist circumference was observed.The study suggests that metabolic abnormalities,PCOS and recurrent pre-eclampsia might be contributed in development of DNA damage in obese women.DNA damage can serve as an early marker for obesity complications in premenopausal women.Moushira Zaki Walaa Basha Hala T.El-Bassyouni Safinaz El-Toukhy Tamer Hussein 2018Genes & Diseases2018,5,4:4
3Association of vitamin D receptor gene polymorphism(VDR)with vitamin D deficiency,metabolic and inflammatory markers in Egyptian obese women显示文摘Vitamin D deficiency might contribute to the pathogenesis of metabolic syndrome and could cause immune disturbance.The aim of this study is to analyze the associations between Vitamin D receptor(VDR)gene polymorphism,serum 25-hydroxy vitamin D,metabolic and inflammatory biomarkers in Egyptian obese women.The study included 201 obese women with vitamin D deficiency and 249 obese matched age healthy controls with sufficient vitamin D levels.Their age ranged between 25 and 35 years.Inflammatory biomarkers(interleukin-6 and C-reactive protein)and serum 25(OH)D were measured by enzyme-linked immunosorbent assay.Insulin resistance(IR)was determined by the homeostasis model assessment of insulin resistance(HOMA-IR).Vitamin D receptor(VDR)gene polymorphisms of FokI,ApaI,and TaqI were studied by PCR using the restriction fragment length polymorphism(RFLP)technique.Obese women with vitamin D deficiency had significant higher values of inflammatory and metabolic parameters compared to controls.Multivariable-logistic regression showed associations between 25(OH)D deficiency and metabolic components when comparing cases with controls.Moreover,cases carrying polymorphic alleles showed significant lower levels of serum 25(OH)D and higher HOMA-IR,blood pressure levels and lipid parameters compared to those with the wild type homozygote in obese cases with vitamin D deficiency.Vitamin D deficiency in Egyptian obese women with vitamin D deficiency is associated with abnormal metabolic components and abnormal inflammatory biomarkers.Moreover,VDR polymorphisms play important role in immune and inflammation status.Moushira Zaki Sanaa Kamal Walaa ABasha Eman Youness Wafaa Ezzat Hala El-Bassyouni Khalda Amr 2017Genes & Diseases2017,4,3:3
4Rosmarinic acid attenuates hepatic fibrogenesis via suppression of hepatic stellate cell activation/proliferation and induction of apoptosis显示文摘Objective:To investigate the antifibrotic role of rosmarinic acid(RA),a natural polyphenolic compound,on HSCs activation/proliferation and apoptosis in vitro and in vivo. Methods:The impact of RA on stellate cell line(HSC-T6) proliferation,activation and apoptosis was assessed along with its safety on primary hepatocytes. In vivo,rats were divided into:(i) normal;(ii) thioacetamide(TAA)-intoxicated rats for 12 weeks;(iii) TAA+silymarin or(iv) TAA+RA. At the end of experiment,liver functions,oxidative stress,inflammatory and profibrogenic markers,tissue inhibitor metalloproteinases type-1(TIMP-1) and hydroxyproline(HP) levels were evaluated. Additionally,liver histopathology and immunohistochemical examinations of alpha-smooth muscle actin(α-SMA),caspase-3 and proliferation cellular nuclear antigen(PCNA) were determined. Results:RA exhibited anti-proliferative effects on cultured HSCs in a time and concentration dependent manner showing an IC50 of 276 μg/mL and 171 μg/mL for 24 h and 48 h,respectively,with morphological reversion of activated stellate cell morphology to quiescent form. It significantly improved ALT,AST,oxidative stress markers and reduced TIMP-1,HP levels,inflammatory markers and fibrosis score(S1 vs S4). Furthermore,reduction in α-SMA plus elevation in caspase-3 expressions of HSCs in vitro and in vivo associated with an inhibition in proliferation of damaged hepatocytes were recorded. 5Anticancer activity of Aloe vera and Calligonum comosum extracts separetely on hepatocellular carcinoma cells显示文摘Objective: To investigate the in vitro anticancer effect of Aloe vera(A. vera) and Calligonum comosum(C. comosum) extracts against hepatocellular carcinoma(HepG 2) cells. Methods: Hep G2 cells were tested against different doses of A. vera and C. comosum. Viability of the cells was assessed by MTT assay. Evaluation of apoptosis and DNA damage in HepG 2 cells were performed using annexin V apoptosis detection kit. The expression of p53 and anti-apoptotic(Bcl-2) were tested by real time-PCR and flow cytometer analyser. Hematoxylin and eosin stained sections from untreated and treated HepG 2 cells were observed using light microscopy.Results: The IC50 values of A. vera and C. comosum extracts were(10.45 ± 0.31) and(9.60 ± 0.01) μg/mL respectively. The extracts separately increased cytotoxicity against HepG 2 cells in a time and dose dependent manners. 6Pre-treatment role of inosine triphosphate pyrophosphatase polymorphism for predicting anemia in Egyptian hepatitis C virus patients显示文摘AIM: To investigate and clarify, for the first time, the role of inosine triphosphate pyrophosphatase (ITPA ) polymorphism in Egyptian chronic hepatitis C virus (HCV) patients.METHODS:The human genomic DNA of all patients was extracted from peripheral blood cells in order to determine the single nucleotide polymorphism (SNP) of ITPA (rs1127354). SNP genotyping was performed by real time polymerase chain reaction (PCR, ABI TaqMan allelic discrimination kit) for 102 treatment-naive Egyptian patients with chronic HCV. All patients had no evidence of cardiovascular or renal diseases. They received a combination treatment of pegylated interferon α (PEG-IFNα) as a weekly subcutaneous dose plus an oral weight-adjusted dose of ribavirin (RBV). The majority received PEG-IFNα2a (70.6%) while 29.4% received PEG-IFNα2b. The planned duration of treatment was 24-48 wk according to the viral kinetics throughout the course of treatment. Pre-treatment liver biopsy was done for each patient for evaluation of fibrosis stage and liver disease activity. The basal viral load level was detected quantitatively by real time PCR while viral load throughout the treatment course was performed qualitatively by COBAS TaqMan assay. RESULTS: Ninety-three patients (91.2%) had ITPA SNP CC genotype and 9 (8.8%) had non-CC genotype (CA and AA). The percentage of hemoglobin (Hb) decline was higher for CC patients than for non-CC patients, particularly at weeks 4 and 8 (P=0.047 and 0.034, respectively). During the first 12 wk of treatment, CC patients had significantly more Hb decline > 3 g/dL than non-CC patients: 64.5% vs 22.2% at weeks 8 and 12, respectively, (P=0.024 and 0.038). Reduction of the amount of the planned RBV dose was significantly higher for CC patients than non-CC patients during the first 12 wk (18% ± 12.1% vs 8.5% ± 10.2%, P=0.021). The percentage of CC patients with RBV dose reduction was significantly greater than that of non-CC patients (77.4% vs 44.4%, P=0.044). Multivariate analysis identified only the percentage of RBV dose as a predictor for Hb decline. Platelet decline was significantly higher in non-CC patients than CC patients at weeks 12, 24 and 48 (P=0.018, 0.009 and 0.026, respectively). 9静脉注射草酸钾对犬肾功能和肾组织结构的影响显示文摘为研究草酸钙尿结石期间犬肾功能和肾脏组织结构变化,将10只比格犬随机分为2组,每组5只。按体质量(0.13mL/kg)经头静脉对处理组犬每天注射草酸钾溶液(0.5mol/L),对照组犬注射等剂量的生理盐水,每天3次,连续注射7d。观察犬的临床表现,每天采集的血清用于生化检查;在最后1次注射8h后,用手术方法采取肾脏,通过HE&Pizzolatos染色和过碘酸—雪夫氏染色(PAS)以及扫描电镜(SEM)与透射电镜(TEM)观察其病理组织结构和超微结构的变化。结果显示,处理组犬肾有点状出血灶,肾小球呈不同程度的破坏,肾小管中有大量结晶沉积,间质内有炎性细胞浸润;足细胞核皱缩、足突排列紊乱,有明显融合现象,线粒体脊断裂、双层膜结构消失;血清生化检查结果表明肾脏虑过功能下降。因此,草酸钾可导致犬肾小球功能下降,损伤肾小管上皮细胞,草酸钙结晶可在肾小管内大量沉积,并进一步导致肾功能衰退。关华伟 王亨 Walaa Mohamaden 李建基 2014中国兽医学报2014,34,4:1

13紫花苜蓿种带阪崎克罗诺杆菌的分离鉴定及其致病性初步研究显示文摘采用稀释平板法从6种表面消毒的紫花苜蓿种子中分离和纯化出3株阪崎克罗诺杆菌疑似分离物。选取代表分离物1gF3室内条件下通过菌悬液(≈109 cfu·mL-1)浸种紫花苜蓿种子,皿内发芽;腹腔注射洁净级昆明小白鼠试验;测定阪崎克罗诺杆菌疑似分离物对紫花苜蓿和小鼠的致病性。结果表明,阪崎克罗诺杆菌疑似分离物对紫花苜蓿不致病;小鼠在接种分离物1gF3 12h后病理特征显示化脓性脑炎、心肌水肿扩张,局部心肌纤维有断裂、肝脏局部坏死、急性脾炎、肺部急性弥散性出血以及肾脏弥散性血管内凝血等症状,1gF3对小鼠具有致病性。发病小鼠血液涂板分离到阪崎克罗诺杆菌疑似分离物TM2。结合表型特征和16SrDNA鉴定,确定分离物1gF3和TM2为阪崎克罗诺杆菌。在此基础上,测定了菌株1gF3的生物学特性,该菌对数生长期为24~36h,之后衰退;干旱胁迫加重其生长量线性下降;菌株1gF3能在较宽温度(10~41℃)和pH(3~11)范围内生长;该菌不需盐生长,NaCl浓度上升该菌的生长量受到抑制;黑暗交替的光照有利其生长。研究材料为具有动物和植物跨域寄主特点的一类病原菌资源,研究结果对隐藏于植物或动物材料中跨域病原菌侵染传播特征及相关检疫防控提供了理论基础。张振粉 苏静 杨成德 师尚礼 花立民 WALAA Mohamaden 2018草业学报2018,27,8:1
