| 1 | Effects of Flower Buds Extract of Tussilago farfara on Focal Cerebral Ischemia in Rats and Inflammatory Response in BV2 Microglia显示文摘Objective: To investigate the effects of the flower buds extract of Tussilago farfara Linné(Farfarae Flos; FF) on focal cerebral ischemia through regulation of inflammatory responses in activated microglia. Methods: Brain ischemia was induced in Sprague-Dawley rats by a transient middle cerebral artery occlusion(t MCAO) for 90 min and reperfusion for 24 h. Twenty rats were randomly divided into 4 groups(n=5 per group): normal, t MCAO-induced ischemic control, t MCAO plus FF extract 300 mg/kg-treated, and t MCAO plus MK-801 1 mg/kg-treated as reference drug. FF extract(300 mg/kg, p.o.) or MK-801(1 mg/kg, i.p.) was administered after reperfusion. Brain infarction was measured by 2,3,5,-triphenyltetrazolium chloride staining. Neuronal damage was observed by haematoxylin eosin, Nissl staining and immunohistochemistry using anti-neuronal nuclei(Neu N), anti-glial fibril ary acidic protein(GFAP), and antiCD11 b/c(OX42) antibodies in ischemic brain. The expressions of inducible nitric oxide synthase(i NOS), tumor necrosis factor(TNF-α), and hypoxia-inducible factor-1 a(HIF-1α) were determined by Western blot. BV2 microglial cel s were treated with FF extract or its main bioactive compound, tussilagone with or without lipopolysaccharide(LPS). Nitric oxide(NO) production was measured in culture medium by Griess assay. The expressions of i NOS, COX-2 and pro-inflammatory cytokines mRNA were analyzed by reverse transcription-polymerase chain reaction. The expression of i NOS, and COX-2 proteins, the phosphorylation of ERK1/2, JNK, and p38 MAPK and the nuclear expression of NF-κB p65 in BV2 cells were determined by Western blot. Results: FF extract significantly decreased brain infarctions in ischemic rats(P<0.01). The neuronal death and the microglia/astrocytes activation in ischemic brains were inhibited by FF extract. FF extract also suppressed i NOS, TNF-α, and HIF-1α expression in ischemic brains. FF extract(0.2 and 0.5 mg/m L, P<0.01) and tussilagone 20 and 50 μmol/L, P<0.01) significantly decreased LPS-induced NO production in BV2 microglia through downregulation of i NOS m RNA and protein expression. FF extract and tussilagone significantly inhibited LPS-induced expression of TNF-α, IL-1β, and IL-6 m RNA, and also suppressed the phosphorylation of ERK1/2, JNK and p38 MAPK and the nuclear expression of NF-κB in a dosedependent manner. | 2 | Applying real time seismic monitoring technology for slope stability assessment—An Indian opencast coal mine perspective显示文摘This paper outlines the results obtained from real time microseismic monitoring of an opencast coal mine in South India.The objective of the study is to investigate the stress changes within the rockmass along the slope due to underground mine development operation and their impact on the stability of the highwall slope.The installed microseismic systems recorded the seismic triggerings down toà2 moment magnitude.In general,most of the events recorded during the monitoring period are weak in seismic energy.The study adopts a simple and more reliable tool to characterize the seismically active zone for assessing the stability of the highwall in real time.The impact of underground working on the slope is studied on the basis of the seismic event impact contours and seismic clusters.During the monitoring period,it is observed that the intensity of the overall microseismic activity along the slope due to the mine development operations did not cause any adverse impact on the highwall stability. | Vinoth S. | 3 | Antimicrobial potential of Actinomycetes species isolated from marine environment显示文摘Objective:To evaluate the antimicrobial activity of Actinomycetes species isolated from marine environment.Methods:Twenty one strains of Actinomyceteswere isolated from samples of Royapuram, Muttukadu, Mahabalipuram sea shores and Adyar estuary. Preliminary screening was done using cross-streak method against two gram-positive and eight gram-negative bacteria.The most potent strains C11 and C12 were selected from which antibacterial substances were extracted. The antibacterial activities of the extracts were performed using Kirby-Bauer disc diffusion method. Molecular identification of those isolates was done.Results:All those twenty one isolates were active against at least one of the test organisms. Morphological characters were recorded. C11 showed activity againstStaphylococcusspecies(13.0暲0.5 mm),Vibrio harveyi(11.0暲0.2 mm),Pseudomonas species(12.0暲0.3 mm). | 4 | A comparative study of sequential vs.simultaneous type Ⅰ tympanoplasty in patients with bilateral chronic otitis media-Mucosal type显示文摘Background:Patients with bilateral chronic otitis media-mucosal type have been conventionally treated with sequential tympanoplasty.Bilateral simultaneous tympanoplasty is usually not preferred because of the theoretical risk of iatrogenic sensorineural hearing loss.With the advent of modern surgical instruments and surgical techniques,the risk is expected to be lower.This study compares the clinical outcomes in typeⅠtympanoplasty performed simultaneously and sequentially.Materials&methods:This randomized prospective study was carried out in a tertiary care hospital between August 2015 and July 2017.A total of 30 patients were divided into two groups of 15 each.This study analyzed the graft uptake,pure tone audiogram findings pre-and post-operatively,duration of surgery and number of hospital visit for each patient and the outco mes were co mpared between both the groups.Result:Patients undergoing bilateral simultaneous tympanoplasty had significantly lesser mean duration of surgery and number of hospital visits than the patients undergoing sequential tympanoplasty.Graft uptake and postoperative wound infections were similar in both the groups.Postoperative hearing improvement was significantly better in the bilateral simultaneous tympanoplasty group.However,further studies are needed to authenticate this observation.None of the patients had a postoperative deterioration of hearing or sensorineural hearing loss.Conclusion:Bilateral simultaneous tympanoplasty is not only feasible but also better than sequential tympanoplasty,especially in terms of operating time,follow-up and overall financial implications on the patient. | Preethi Umamaheswaran Sanjeev Mohanty Vinoth Manimaran Sathishkumar Jayagandhi Shiva Priya Jeyabalakrishnan | 2020 | Journal of Otology2020,15,2: | 3 |
| 5 | p-Coumaric acid ameliorates ethanol-induced kidney injury by inhibiting inflammatory cytokine production and NF-κB signaling in rats显示文摘Objective: To examine the effects of p-coumaric acid on ethanol-induced kidney injury in Swiss Wistar rats.Methods: Ethanol(25% v/v) was used to induce nephrotoxicity in rats. p-Coumaric acid was orally administered at 50, 100, or 200 mg/kg body weight. The levels of oxidative parameters were determined; pro-inflammatory biomarkers were analyzed by Western blotting and apoptotic protein was analyzed by immunohistochemistry. Results: Ethanol treated rats showed decreased levels of antioxidants and aberrant production of pro-inflammatory cytokines(IL-6, IL1β, TNF-α), NF-κB activation and imbalance of proand anti-apoptotic proteins(Bcl-2, Bax, caspase 3). Meanwhile, p-coumaric acid restored antioxidant levels and decreased the levels of inflammatory cytokines, NF-κB, and proapoptotic proteins and increased Bcl-2 expression. | 13 | Rare earth elements in flotation products of coals from East Kootenay coalfields,British Columbia显示文摘With the identification of coal deposits that are enriched in rare earth elements(REEs)across the world,coal and coal by-products are considered as an alternative source for REEs due to the combined supply and demand restrictions for these elements.Studies are being conducted to recover these elements from various coal sources.In this study,coal samples from the East Kootenay coalfields in British Columbia,Canada were examined for the presence of REEs in run-of-mine coal samples and flotation products.Results reveal that on an ash basis,the concentration of REEs varies from 174 to 1038 mg/kg in the samples.To understand the REE as sociation,which is essential for extraction purposes,a three-step sequential extraction process along with a mineralogical evaluation using scanning electron microscopy(SEM)with energy dispersive X-ray spectroscopy(EDX)and quantitative X-ray diffraction(XRD)data was used.It is found that a significant portion of REEs in the feed samples were reported to middling and tailings streams.The mineralogical analyses reveal REE association with aluminosilicate mineral in the samples. | Vinoth Kumar Kuppusamy Maria Holuszko | 2019 | Journal of Rare Earths2019,37,12: | 1 |
