| 1 | Proteomic analysis of gastric cancer and immunoblot validation of potential biomarkers显示文摘AIM:To search or and validate di erentially expressed proteins in patients with gastric adenocarcinoma.METHODS:We used two-dimensional gel electrophoresis and mass spectrometry to search or di erentially expressed proteins in patients with gastric adenocarcinoma.A set o proteins was validated with immunoblotting.RESULTS:We identified 30 di erent proteins involved in various biological processes:metabolism,development,death,response to stress,cell cycle,cell communication,transport,and cell motility.Eight proteins were chosen or urther validation by immunoblotting.Our results show that gastrokine-1,39S ribosomal protein L12(mitochondrial precursor),plasma cell-induced resident endoplasmic reticulum protein,and glutathione S-trans erase mu 3 were signi icantly underexpressed in gastric adenocarcinoma relative to adjacent non-tumor tissue samples.On the other hand,septin-2,ubiquitin-conjugating enzyme E2 N,and transaldolase were significantly overexpressed.Translationally controlled tumor protein was shown to be di erentially expressed only in patients with cancer o the gastric cardia/esophageal border.CONCLUSION:This work presents a set o possible diagnostic biomarkers,validated or the first time.It might contribute to the e orts o understanding gastric cancer carcinogenesis. | Nina Kocevar Federico Odreman Alessandro Vindigni Snjezana Frkovic Grazio Radovan Komel | 2012 | World Journal of Gastroenterology2012，18，11： | 3 |
| 2 | Recurrence in node-negative advanced gastric cancer:Novel findings from an in-depth pathological analysis of prognostic factors from a multicentric series显示文摘AIM To analyze the clinicopathological characteristics of patients with both node-negative gastric carcinoma and diagnosis of recurrence during follow-up. METHODS We enrolled 41 patients treated with curative gastrectomy for p T2-4 a N0 gastric carcinoma between 1992 and 2010,who developed recurrence(Group 1). We retrospectively selected this group from the prospectively collected database of 4 centers belonging to the Italian Research Group for Gastric Cancer,and compared them with 437 p T2-4 a N0 patients without recurrence(Group 2). We analyzed lymphatic embolization,microvascular infiltration,perineural infiltration,and immunohistochemical determination of p53,Ki67,and HER2 in Group 1 and in a subgroup of Group 2(Group 2 bis) of 41 cases matched with Group 1 according to demographic and pathological characteristics. RESULTS T4 a stage and diffuse histotype were associated with recurrence in the group of p N0 patients.
