| 1 | Antibodies Against Annexin V and Prothrombin,Their Correlation with Other Anti-phospholipid Antibodies in Recurrent Pregnancy Loss显示文摘Objective To study the findings of serum antibodies against annexin V, prothrombin,ph-inositol, ph-acid, ph-ethanolamine, ph-serine, ph-glycerol, cardiolipin, and beta2-glycoprotein I and analyze the trophoblast annexin V receptorsMethods Sera from 156 patients aged 26-41 years with recurrent pregnancy loss (3-7 times) were investigated. Eighty-four fertile healthy women aged 24-38 yearswere included in a control group. ELISA methods were used for detecting a panel of seraanti-phospholipid antibodies. Immunolocalization of annexin V receptors in 143trophoblast specimens of 156 patients was investigated by the immunofluorescencetechnique using Annexin V-FITC, Apoptosis and AnnexinV-CY3 commercial kits.Results Positivity for anti-phospholipid antibodies mainly against ph-serine, ph-ethanolamine, and ph-inositol was found together in 80.8% (126 out of 156 patients),anti-prothrombin antibodies in 12% (18), and anti-annexin V antibodies in 13. 5%(21) women. No significant levels of anti-phospholipid antibodies were found in 6controls. Placenta immunohistopathology also exhibited some changes manifested bythe presence of apoptotic and necrotic cells in trophoblast, and very fewmicrothrombotization in some intervillous spaces.Conclusion Our detailed study demonstrated the prevalence of majority of anti-phospholipid antibodies as a high risk factor for repeated reproductive failure. | 12 | Zinc phosphate-based nanoparticles as a novel antibacterial agent: in vivo study on rats after dietary exposure显示文摘Background: Development of new nanomaterials that inhibit or kil bacteria is an important and timely research topic. For example, financial losses due to infectious diseases, such as diarrhea, are a major concern in livestock productions around the world. Antimicrobial nanoparticles(NPs) represent a promising alternative to antibiotics and may lower antibiotic use and consequently spread of antibiotic resistance traits among bacteria, including pathogens.Results: Four formulations of zinc nanoparticles(Zn A, Zn B, Zn C, and Zn D) based on phosphates with spherical(Zn A, Zn B)or irregular(Zn C, Zn D) morphology were prepared. The highest in vitro inhibitory effect of our NPs was observed against Staphylococcus aureus(inhibitory concentration values, IC50, ranged from 0.5 to 1.6 mmol/L), fol owed by Escherichia coli(IC500.8–1.5 mmol/L). In contrast, methicil in resistant S. aureus(IC501.2–4.7 mmol/L) was least affected and this was similar to inhibitory patterns of commercial Zn O-based NPs and Zn O. After the successful in vitro testing, the in vivo study with rats based on dietary supplementation with zinc NPs was conducted. Four groups of rats were treated by 2,000 mg Zn/kg diet of Zn A, Zn B, Zn C, and Zn D, for comparison two groups were supplemented by 2,000 mg Zn/kg diet of Zn O-N and Zn O, and one group(control) was fed only by basal diet. The significantly higher(P < 0.05) Zn level in liver and kidney of al treated groups was found, nevertheless Zn NPs did not greatly influence antioxidant status of rats. However,the total aerobic and coliform bacterial population in rat feces significantly decreased(P < 0.05) in al zinc groups after 30 d of the treatment. | 18 | ETR1 Integrates Response to Ethylene and Cytokinins into a Single Multistep Phosphorelay Pathway to Control Root Growth显示文摘Cytokinins and ethylene control plant development via sensors from the histidine kinase(HK)family.However,downstream signaling pathways for the key phytohormones are distinct.Here we report that not only cytokinin but also ethylene is able to control root apical meristem(RAM)size through activation of the multistep phosphorelay(MSP)pathway.We found that both cytokinin and ethylene-dependent RAM shortening requires ethylene binding to ETR1 and the HK activity of ETR1.The receiver domain of ETR1 interacts with MSP signaling intermediates acting downstream of cytokinin receptors,further substantiating the role of ETR1 in MSP signaling.We revealed that both cytokinin and ethylene induce the MSP in similar and distinct cell types with ETR1-mediated ethylene signaling controlling MSP output specifically in the root transition zone.We identified members of the MSP pathway specific and common to both hormones and showed that ETR1-regulated ARR3 controls RAM size.ETR1-mediated MSP spatially differs from canonical CTR1/EIN2/EIN3 ethylene signaling and is independent of EIN2,indicating that both pathways can be spatially and functionally separated.Furthermore,we demonstrated that canonical ethylene signaling controls MSP responsiveness to cytokinin specifically in the root transition zone,presumably via regulation of ARR10,one of the positive regulators of MSP signaling in Arabidopsis. | Marketa Zdarska Abigail Rubiato Cuyacot Paul TTarr Amel Yamoune Agnieszka Szmitkowska Vendula Hrdinová Zuzana Gelová Elliot MMeyerowitz Jan Hejátko | 2019 | Molecular Plant2019,12,10: | 0 |
