| 1 | A BAC clone of MDV strain GX0101 with REV-LTR integration retained its pathogenicity显示文摘The complete genome of Marek's disease virus (MDV) strain GX0101, which was integrated with the LTR sequences of REV, was cloned in Escherichia coli as a bacterial artificial chromosome (BAC). BAC vector sequences were introduced into the US2 locus of the MDV genome by homologous recombination. The viral DNA containing the BAC vector was used to transform Escherichia coli strain of DH10B. Then the recombinant virus was successfully rescued by transfection of the recombinant BAC DNA into primary chicken embryo fibroblast (CEF). This BAC viral clone was named bac-GX0101. When the reconstituted virus was inoculated into 1-day-old birds, visceral tumors could be detected as early as 62 d post infection. There was no difference in growth ability and pathogenicity to birds between the BAC derived virus and its parental virus. The BAC derived virus maintained its oncogenicity and immunosuppressive effects. In conclusion, the complete genome of GX0101 strain was successfully cloned into BAC and the infectious clone was rescued. | 8 | Sequential disruption of ALV host receptor genes reveals no sharing of receptors between ALV subgroups A, B, and J显示文摘Background: Previously, we showed that targeted disruption of viral receptor genes in avian leukosis virus(ALV)subgroups using clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9))-based genome editing confers resistance to ALV subgroups B and J. Here, we used the same strategy to target the receptor expressed by ALV subgroup A(TVA) and generate chicken cells resistant to infection by this virus.Results: CRISPR/Cas9-based disruption of exon 2 within the tva gene of DF-1 fibroblasts conferred resistance to infection by ALV subgroup A regardless of whether frameshift mutations were introduced during editing. Conversely,overexpression of the wild-type TVA receptor(wtTVA) by tva-modified DF-1 clones restored susceptibility to ALV subgroup A. The results confirm that exon 2, which contains the low-density lipoprotein receptor class A domain of TVA, is critical for virus entry. Furthermore, we sequentially modified DF-1 cells by editing the tva, tvb, and Na^+/H^+ exchange 1(chNHE1) genes, which are the specific receptors for ALV subgroups A, B, and J, respectively.Conclusions: Simultaneous editing of multiple receptors to block infection by different subgroups of ALV confirmed that ALV subgroups A, B, and J do not share host receptors. | 9 | Microstructural Characterization and Mechanical Properties of Powder Metallurgy Dual Phase Steel Preforms显示文摘In order to improve the mechanical properties of powder metallurgy (P/M) ferrite-pearlite steel, a dual phase (DP) ferrite-martensite steel was produced through intercritical annealing of sintered P/M preforms. Mi-crostructures of the sintered and DP steels were examined with optical, scanning and transmission electron microscopes. Mechanical properties were evaluated through hardness measurements and compression tests. Microstructural studies revealed that sintered steel contained polygonal ferrite-pearlite while the DP steel contained polygonal, lath and acicular ferrite along with lath-type martensite as microstructural constituents. In DP steels, with increasing mean preform density, the microstructure contained fine and continuous network of martensite colonies with minimum porosity. The work hardening rate vs plastic strain plots (Jaoul-Crussard analysis) of both the steels revealed typical three stage deformation behaviour for low and high mean preform densities. Physico-chemical characteristics of fresh, reduced and used catalysts were evaluated by XRD, TPR and O2 pulse chemisorptions, TEM and BET-SA techniques. XRD studies showed phases due to hydrotalcite-like precursors in oven dried form produced dispersed NiO species upon calcination in static air above 450 C. Raman spectra of deactivated samples revealed the presence of both ordered and disordered forms of carbon. Ni-La-Al2O3catalyst with a mole ratio of Ni : La : Al = 2 : 0.1 : 0.9 exhibited tremendously high longevity with a hydrogen production rate of 1300 molH2 mol 1 Ni. | 14 | 纤维蛋白胶与无缝无胶自体球结膜瓣固定术治疗翼状胬肉的疗效比较(英文)显示文摘目的:比较两种大小纤维蛋白胶与无缝无胶自体球结膜瓣固定术在原发性翼状胬肉手术中的患者舒适度、移植物稳定性、移植物炎症、复发等术后并发症。方法:本研究对2014-12/2016-08共80例翼状胬肉患者(80眼)进行前瞻性介入对比研究。研究组随机抽取并分为两组,组1接受纤维蛋白胶疗法,组2采用无缝合无胶结膜自体移植固定术。每组再将等量的受试者随机平均分为两组(组1:组A+组B,组2:组C+组D)。组A(自体移植物的大小与裸眼巩膜相同)和组B(比裸巩膜大1 mm)实施纤维蛋白胶疗法;组C(自体移植物的大小与裸眼巩膜相同)和组D(比裸巩膜大1 mm)采用无缝线无胶结膜自体移植固定术。每一位患者都接受翼状胬肉切除术,并根据组别使用不同大小的自体球结膜瓣。结果:组1平均年龄为43.525,组2平均年龄为42.2。男性20例,女性60例。37例患者右眼有翼状胬肉。四组间平均手术时长相似。在术后随访期间,除结膜下出血(纤维蛋白胶组在术后1wk内明显增多)外,4组间患者的舒适度、移植物回缩、移植物炎症和移植物水肿等术后参数均相似(P<0.0001)。组2仅3例患者出现4级移植物回缩/移位。术后1wk,组A和组D各出现1例移植囊肿患者。随访6mo后,组D中2例患者复发。结论:两种方法均安全有效,术后不适及并发症少。与纤维蛋白胶疗法相比,无缝无胶疗法更加经济且效果相当。由于该手术成本低,且具有纤维蛋白胶疗法的所有优点,无缝无胶法可被认为是治疗患者非常有效的方法。 | Manipur Raviraj Sahana Chikkanayakanahalli Venugopal Kavitha Acharya Pavana Bomalapura Ramamurthy Lakshmi | 2019 | 国际眼科杂志2019,19,11: | 2 |
| 15 | Cardiogenic differentiation of mesenchymal stem cells on elastomeric poly (glycerol sebacate)/collagen core/shell fibers显示文摘AIM: To facilitate engineering of suitable biomaterials to meet the challenges associated with myocardial infarction. METHODS: Poly (glycerol sebacate)/collagen (PGS/ collagen) core/shell fibers were fabricated by core/ shell electrospinning technique, with core as PGS and shell as collagen polymer; and the scaffolds were characterized by scanning electron microscope (SEM), fourier transform infrared spectroscopy (FTIR), contact angle and tensile testing for cardiac tissue engineering. Collagen nanofibers were also fabricated by electrospinning for comparison with core/shell fibers. Studies on cell-scaffold interaction were carriedout using cardiac cells and mesenchymal stem cells (MSCs) co-culture system with cardiac cells and MSCs separately serving as positive and negative controls respectively. The co-culture system was characterized for cell proliferation and differentiation of MSCs into cardiomyogenic lineage in the co-culture environment using dual immunocytochemistry. The co-culture cells were stained with cardiac specific marker proteins like actinin and troponin and MSC specific marker protein CD 105 for proving the cardiogenic differentiation of MSCs. Further the morphology of cells was analyzed using SEM.RESULTS: PGS/collagen core/shell fibers, core is PGS polymer having an elastic modulus related to that of cardiac fibers and shell as collagen, providing natural environment for cellular activities like cell adhesion, proliferation and differentiation. SEM micrographs of electrospun fibrous scaffolds revealed porous, beadless, uniform fibers with a fiber diameter in the range of 380 ± 77 nm and 1192 ± 277 nm for collagen fibers and PGS/collagen core/shell fibers respectively. The obtained PGS/collagen core/shell fibrous scaffolds were hydrophilic having a water contact angle of 17.9 ± 4.6° compared to collagen nanofibers which had a contact angle value of 30 ± 3.2°. The PGS/collagen core/shell fibers had mechanical properties comparable to that of native heart muscle with a young's modulus of 4.24 ± 0.7 MPa, while that of collagen nanofibers was comparatively higher around 30.11 ± 1.68 MPa. FTIR spectrum was performed to confirm the functional groups present in the electrospun scaffolds. Amide Ⅰ and amide Ⅱ of collagen were detected at 1638.95 cm -1 and 1551.64 cm -1 in the electrospun collagen fibers and at 1646.22 cm -1 and 1540.73 cm -1 for PGS/collagen core/shell fibers respectively. Cell culture studies performed using MSCs and cardiac cells co-culture environment, indicated that the cellproliferation significantly increased on PGS/collagen core/shell scaffolds compared to collagen fibers and the cardiac marker proteins actinin and troponin were expressed more on PGS/collagen core/shell scaffolds compared to collagen fibers alone. Dual immunofluorescent staining was performed to further confirm the cardiogenic differentiation of MSCs by employing MSC specific marker protein, CD 105 and cardiac specific marker protein, actinin. SEM observations of cardiac cells showed normal morphology on PGS/collagen fibers and providing adequate tensile strength for the regeneration of myocardial infarction. | 19 | Influence of Rotational Speed on Microstructure and Mechanical Properties of Dissimilar Metal AISI 304-AISI 4140 Continuous Drive Friction Welds显示文摘Fundamental investigation of continuous drive friction welding of austenitic stainless steel (AISI 304) and low alloy steel (AISI 4140) is described. The emphasis is made on the influence of rotational speed on the microstructure and mechanical properties such as hardness, tensile strength, notch tensile strength and impact toughness of the dissimilar joints. Hardness profiles across the weld show the interface is harder than the respective parent metals. In general, maximum peak hardness is observed on the stainless steel side, while other peak hardness is on the low alloy steel side. A trough in hardness distribution in between the peaks is located on the low alloy steel side. Peak hardness on the stainless steel and low alloy steel side close to the interface increases with a decrease in rotational speed. All transverse tensile joints fractured on stainless steel side near the interface. Notch tensile strength and impact toughness increase with increase in rotational speed up to 1 500 r/min and decrease thereafter. | 20 | 氧化铝和氧化锆催化剂上2-取代苯并咪唑和1,5-二取代苯并二氮的合成(英文)显示文摘In this study, alumina, zirconia, manganese oxide/alumina, and manganese oxide/zirconia have been investigated for their catalytic activity in the condensation reaction between o-phenylenediamine and an aldehyde or a ketone to synthesise 2-substituted benzimidazoles and 1,5-disubstituted benzodiazepines respectively. Surface area, surface acidity, and morphology of the catalysts have been analysed and correlated with their catalytic activity. The isolated yields of 2-substituted benzimidazoles and 1,5-disubstituted benzodiazepines are in the range of 30% to 95%. A good correlation between the amount of surface acid sites as well as the surface morphology of the catalysts and the catalytic activity has been observed. This method has been found to be simple and economical. The solid supports could be regenerated and reused without much loss in their activity.
