| 3 | Effect of α-Ketoglutarate on Cyanide-induced Biochemical Alterations in Rat Brain and Liver显示文摘Objective To investigate the biochemical changes in rat brain and liver following acute exposure to a lethal dose of cyanide, and its response to treatment of α-ketoglutarate (α-KG) in the absence or presence of sodium thiosulfate (STS). Methods Female rats were administered 2.0 LD50 potassium cyanide (KCN; oral) in the absence or presence of pre-treatment (?10 min), simultaneous treatment (0 min) or post-treatment (+2-3 min) of α-KG (2.0 g/kg, oral) and/or STS (1.0 g/kg, intraperitoneal, ?15 min, 0 min or + 2-3 min). At the time of onset of signs and symptoms of KCN toxicity (2-4 min) and at the time of death (5-15 min), various parameters particularly akin to oxidative stress viz. cytochrome oxidase (CYTOX), superoxide dismutase (SOD), glutathione peroxidase (GPx), reduced glutathione (GSH) and oxidized glutathione (GSSG) in brain, and CYTOX, sorbitol dehydrogenase (SDH), alkaline phosphatase (ALP), GSH and GSSG in liver homogenate were measured. Results At both time intervals brain CYTOX, SOD, GPx, and GSH significantly reduced (percent inhibition compared to control) to 24%, 56%, 77%, and 65%, and 44%, 46%, 78%, and 57%, respectively. At the corresponding time points liver CYTOX and GSH reduced to 74% and 63%, and 44% and 68%, respectively. The levels of GSSG in the brain and liver, and hepatic ALP and SDH were unchanged. Pre-treatment and simultaneous treatment of α-KG alone or with STS conferred significant protection on above variables. Post-treatment was effective in restoring the changes in liver but failed to normalize the changes in the brain. Conclusions Oral treatment with α-KG alone or in combination with STS has protective effects on cyanide-induced biochemical alterations in rat brain and liver. | RAJKUMAR TULSAWANI R. | 4 | Impact of gout on in-hospital outcomes of acute coronary syndrome-related hospitalizations and revascularizations: Insights from the national inpatient sample显示文摘BACKGROUND Previous studies have established a role of gout in predicting risk and prognosis of cardiovascular diseases. However, large-scale data on the impact of gout on inpatient outcomes of acute coronary syndrome (ACS)-related hospitalizations and post-revascularization is inadequate. AIM To evaluate the impact of gout on in-hospital outcomes of ACS hospitalizations, subsequent healthcare burden and predictors of post-revascularization inpatient mortality. METHODS We used the national inpatient sample (2010-2014) to identify the ACS and goutrelated hospitalizations, relevant comorbidities, revascularization and postrevascularization outcomes using the ICD-9 CM codes. A multivariable analysis was performed to evaluate the predictors of post-revascularization in-hospital mortality. RESULTS We identified 3144744 ACS-related hospitalizations, of which 105198 (3.35%) also had gout. The ACS-gout cohort were more often older white males with a higher prevalence of comorbidities. Coronary artery bypass grafting was required more often in the ACS-gout cohort. Post-revascularization complications including cardiac (3.2% vs 2.9%), respiratory (3.5% vs 2.9%), and hemorrhage (3.1% vs 2.7%) were higher whereas all-cause mortality was lower (2.2% vs 3.0%) in the ACSgout cohort (P < 0.001). An older age (OR 15.63, CI: 5.51-44.39), non-elective admissions (OR 2.00, CI: 1.44-2.79), lower household income (OR 1.44, CI: 1.17- 1.78), and comorbid conditions predicted higher mortality in ACS-gout cohort undergoing revascularization (P < 0.001). Odds of post-revascularization inhospital mortality were lower in Hispanics (OR 0.45, CI: 0.31-0.67) and Asians (OR 0.65, CI: 0.45-0.94) as compared to white (P < 0.001). However, postoperative complications significantly raised mortality odds. Mean length of stay, transfer to other facilities, and hospital charges were higher in the ACS-gout cohort. | 8 | Seasonal variations of phytoplankton diversity in the Coleroon coastal waters,southeast coast of India显示文摘The results of an investigation carried out during June 2005 to May 2007 on physico-chemical parameters,species composition and community structure of phytoplankton including Chlorophyll a(Chl-a) at the Coleroon coastal waters(Southeast coast of India) are reported.Air and surface water temperatures(°C) varied from 25.1 to 30.1 and from 24.5 to 28.5 respectively.Salinity values varied from 6 to 28.5 and the pH ranged between 7.0 and 8.3.Variation in dissolved oxygen content was from 3.1 to 7.5 mg/dm-3 while the light extinction coefficient values(LEC) ranged between 3.1 and 10.1 cm.The ranges of inorganic nutrients(μmol/dm-3) viz.,nitrate,nitrite,phosphate and silicate were 10.1-23.4,1.2-8.9,0.2-3.1 and 55-125,respectively.The ranges of Chlorophyll a(μg/dm-3) values was 2.0-7.5.Presently,124 phytoplankton species representing different classes viz:Bacillariophyceae(77),Dinophyceae(19),Cyanophyceae(15),Chlorophyceae(10) and Chrysophyceae(3) were recorded.The phytoplankton cell abundance varied from 0.290 to 111.662 cells/cm-3,with peak diversity(3.38 bits/ind.) during summer season.The maximum abundance was found during summer season coinciding with the stable hydrographical conditions.The seasonal distribution and abundance of phytoplankton are discussed in relation to hydrographical parameters.Canonical Correspondence Analysis(CCA) was applied in this paper for discriminating environmental factors having effect on phytoplankton community at species level.Coleroon coastal water is subjected to long term fluctuations in physico-chemical parameters depending upon the seasonal tidal amplitude and freshwater influx resulting in a continuous exchange of organic,inorganic,plant and animal matters. | Thillai Rajasekar K Rajkumar M SUN Jun Ashok Prabu V Perumal P | 2010 | Acta Oceanologica Sinica2010,29,5: | 3 |

| 11 | Post-appendectomy pelvic abscess with extended-spectrum beta-lactamase producing Escherichia coli : A case report and review of literature显示文摘BACKGROUND Appendicitis, the inflammation of the appendix, is the most common abdominal surgical emergency requiring expedient surgical intervention. Extendedspectrum beta-lactamases(ESBLs) are bacterial enzymes that catalyse the degradation of the betalactam ring of penicillins and cephalosporins(but without carbapenemase activity), leading to resistance of these bacteria to beta-lactam antibiotics. Recent increases in incidence of ESBL-producing bacteria have caused alarm worldwide. Proportion estimates of ESBLEnterobacteriaceae hover around 46% in China, 42% in East Africa, 12% in Germany, and 8% in the United States.CASE SUMMARY The impact of ESBL-producing bacteria on appendiceal abscesses and consequent pelvic abscesses are yet to be examined in depth. A literature review using the search words 'appendiceal abscesses' and 'ESBL Escherichia coli(E. coli)' revealed very few cases involving ESBL E. coli in any capacity in the context of appendiceal abscesses. This report describes the clinical aspects of a patient with appendicitis whodeveloped a postoperative pelvic abscess infected with ESBL-producing E. coli. In this report, we discuss the risk factors for contracting ESBL E. coli infection in appendicitis and post-appendectomy pelvis abscesses. We also discuss our management approach for postappendectomy ESBL E. coli pelvic abscesses, including drainage, pathogen identification, and pathogen characterisation. When ESBL E. coli is confirmed, carbapenem antibiotics should be promptly administered, as was done efficaciously with this patient. | 12 | Effect of Pre-treatment of α-Ketoglutarate on Cyanide-induced Toxicity and Alterations in Various Physiological Variables in Rodents显示文摘Objective To investigate the effects of pre-treatment of α-ketoglutarate (α-KG) on cyanide-induced lethality and changes in various physiological parameters in rodents. Methods The LD50 of potassium cyanide (KCN) given orally (po), intraperitoneally (ip), subcutaneously (sc) or intravenously (iv) was determined in male mice, in the presence or absence α-KG given po, ip or iv. α-KG was administered 10, 20 or 40 min prior to KCN at 0.50, 1.0 or 2.0 g/kg by po or ip route, and at 0.10, 0.20 or 0.40 g/kg by iv route. Protection index (PI) was calculated as the ratio of LD50 of KCN in the presence of α-KG (protected animals) and LD50 of KCN in the absence of α-KG (unprotected animals). In a separate experiment, several physiological variables viz. mean arterial pressure (MAP), heart rate (HR), respiratory rate (RR), neuromuscular transmission (NMT) and rectal temperature (RT) were measured in anesthetized female rats pre-treated (-10 min) with po (2.0 g/kg) or iv (0.125 g/kg) α-KG and then administered sub-lethal (0.75 LD50) or lethal (2.0, 4.0 or 8.0 LD50) doses of KCN (po). Results PI of 4.52, 6.40 and 7.60 at -10 min, 3.20, 5.40 and 6.40 at -20 min, and 1.40, 3.20 and 5.40 at -40 min of po administration with α-KG was observed for 0.50, 1.0 and 2.0 g/kg doses, respectively, against KCN given by po route. When KCN was given ip, a PI of 3.38, 4.79 and 5.70 was observed for 0.50, 1.0 and 2.0 g/kg α-KG given ip (-10 min), respectively. A lower PI of 3.37, 2.83 and 2.38 was observed when KCN given sc was challenged by 2.0 g/kg α-KG given ip at -10, -20 or -40 min, respectively. Similarly, a PI of 3.37, 2.83 and 2.0 was noted when KCN given sc was antagonized by 2.0 g/kg α-KG given po at -10, -20 or -40 min, respectively. No appreciable protection was observed when lower doses of α-KG (ip or po) challenged KCN given by sc route. Pre-treatment of iv or po administration of α-KG did not afford any protection against KCN given po or iv route. Oral treatment of 0.75 LD50 KCN caused significant decrease in MAP and HR after 15 min, RR after 30 min and NMT after 60 min. There was no effect on RT. No reduction in MAP, HR, RR and RT was observed when rats received 2.0 or 4.0 LD50 KCN after pre-treatment of α-KG (po; 2.0 g/kg). However, no protection was observed on NMT. Protective efficacy of α-KG was not observed on MAP, HR, RR, and NMT decreased by 8.0 LD50 KCN. Decrease in MAP and NMT caused by 2.0 LD50 KCN (po) was resolved by iv administration of α-KG. Conclusions Cyanide antagonism by α-KG is best exhibited when both α-KG and KCN are given by po route. | 13 | Identification of SNP and InDel variations in the promoter and 5′ untranslated regions of γ-tocopherol methyl transferase(ZmVTE4) affecting higher accumulation of α-tocopherol in maize kernel显示文摘Deficiency of vitamin-E or tocopherol causes neurological and cardiovascular disorders in humans. Though maize kernel is rich in total tocopherol, the level of α-tocopherol possessing the highest vitamin-E activity is low. Mutant allele of ZmVTE4 with deletion of 7 bp and 118 bp (0/0: most favorable haplotype) in 5′UTR and promoter region, respectively significantly enhances α-tocopherol in maize kernel than wild type haplotype (7/118).α- tocopherol estimation in 15 diverse inbreds revealed that mean α-/γ-tocopherol and α-/ total-tocopherol was much higher in genotypes with favorable haplotype (1.51 and 0.41) than unfavorable class (0.19 and 0.13), respectively. However even within favorable class,α- tocopherol ranged from 4.76 to 30.07 μg g 1. Sequence analysis of part of 5′UTR and promoter of ZmVTE4 among the genotypes with favorable haplotype revealed 14 SNPs (SNP1 to SNP14) and eight InDels (InDel1 to InDel8). SNP7 at 606 bp (G to A), and InDels viz., InDel1 (27 bp), InDel4 (27 bp) and InDel8 (14 bp) differentiated low and high α-tocopherol accumulating inbreds with favorable haplotype. | 15 | Influence of Rice Genotypes on Folding and Spinning Behaviour of Leaffolder(Cnaphalocrocis medinalis) and Its Interaction with Leaf Damage显示文摘Folding and spinning behavior of Cnaphalocrocis medinalis(Guenee)(Lepidoptera: Pyralidae) in different categories of rice genotypes viz., resistant, susceptible, hybrid, scented, popular and wild rice genotypes were significantly different. Longer leaf selection time and folding time per primary fold; shorter primary fold and whole leaf fold; lower number of binds per primary fold and whole leaf fold were recorded in resistant and wild rice genotypes. In the correlation analysis, it was found that the leaf folding parameters were positively correlated to leaf folder damage whereas the leaf spinning parameters were negatively correlated. Similarly, the morphological characters differed significantly among the chosen genotypes and were related to leaffolder damage. The leaf width and total productive tiller number were positively correlated to leaffolder infestation. Results also indicated that the trichome density and length, leaf length and plant height might contribute to resistance whereas total number of green leaves had no effect on leaffolder infestation. In the scatter plot analysis between leaf folding and spinning characters and leaffolder damage, the genotypes were separated into four groups viz., resistant(TKM6, Ptb 33, LFR831311, Oryza rhizomatis and O. minuta), moderately resistant(ASD16 and CORH1), moderately susceptible(ADT36, Pusa Basmati and CB200290) and susceptible(IR36 and TN1).
