1饲料中大豆油替代鱼油对大西洋鲑肝细胞脂肪酸代谢的影响显示文摘为探讨大豆油替代鱼油对大西洋鲑(Salmo salar L.)肝细胞脂肪酸代谢的影响,本试验分别以鱼油和大豆油为脂肪源配制2种试验饲料,并分别饲喂大西洋鲑幼鲑5个月。饲养结束后,分离和培养大西洋鲑肝细胞,一方面将细胞与碳14标记油酸([1-14C]OA)共孵育,然后测定细胞和细胞内各脂质成分以及脂肪酸β-氧化产物的放射活性;另一方面提取肝细胞总RNA,采用实时荧光定量聚合酶链式反应(RT-qPCR)测定脂肪酸结合蛋白3(FABP3)、脂肪酸结合蛋白10(FABP10)、Δ5去饱和酶(D5D)、Δ6去饱和酶(D6D)及肉碱棕榈酰转移酶Ⅰ(CPT-Ⅰ)基因的相对表达量。结果表明:1)鱼油组与大豆油组肝细胞内游离脂肪酸(FFA)放射活性百分率分别为(62.7±13.0)%和(55.9±9.1)%,2组间无显著差异(P>0.05);鱼油组与大豆油组肝细胞内磷脂(PLs)放射活性百分率分别为(20.9±8.7)%和(17.9±5.4)%,2组间无显著差异(P>0.05);鱼油组肝细胞内三酰甘油(TAG)放射活性百分率为(2.0±0.9)%,显著高于大豆油组的(0.8±0.4)%(P<0.05);鱼油组与大豆油组肝细胞内β-氧化产物放射活性百分率分别为(1.0±0.3)%和(1.0±0.2)%,2组间无显著差异(P>0.05)。2)大豆油组肝细胞中FABP3、D6D、D5D基因的相对表达量均显著高于鱼油组(P<0.05),而FABP10和CPT-Ⅰ基因的相对表达量在2组间无显著差异(P>0.05)。由此可见,大豆油替代饲料中的鱼油对大西洋鲑肝细胞内FFA、PLs合成及脂肪酸β-氧化等脂肪酸代谢途径无显著影响,但显著降低了肝细胞内TAG的合成,促进了肝细胞内脂肪酸的去饱和,因此大豆油替代鱼油在一定程度上影响了肝细胞内脂肪酸代谢。周继术 TORSTENSEN Bente Elisabeth STUBHAUG Ingunn 吉红 2013动物营养学报2013,25,11:4
