| 1 | Detection and characterization of diverse alpha-and betacoronaviruses from bats in China显示文摘Bats have been implicated as important reservoir hosts of alpha- and betacoronaviruses. In this study, diverse coronaviruses(CoVs) were detected in 50 of 951(positive rate 5.3%) intestinal specimens of eight bat species collected in four provinces and the Tibet Autonomous Region of China by pan-coronavirus RT-PCR screening. Based on 400-nt RNA-dependent RNA polymerase(Rd RP) sequence analysis, eight belonged to genus Alphacoronavirus and 42 to Betacoronavirus.Among the 50 positive specimens, thirteen gave rise to CoV full-length Rd RP gene amplification for further sequence comparison, of which three divergent sequences(two from a unreported province) were subjected to full genome sequencing. Two complete genomes of betacoronaviruses(JTMC15 and JPDB144) and one nearly-complete genome of alphacoronavirus(JTAC2) were sequenced and their genomic organization predicted. The present study has identified additional numbers of genetically diverse bat-borne coronaviruses with a wide distribution in China. | 2 | Coexistence of two sympatric and morphologically similar bat species Rhinolophus affinis and Rhinolophus pearsoni显示文摘Ecologists have long focused on the coexistence of sympatric species. Here, we investigated two horseshoe bat species, Rhinolophus a nis and Rhinolophus pearsoni inhabited in the same cave, for their foraging strategies, niche di erentiation, prey selection, and their coexistence status. These two species of horseshoe bats were di erent in the dominant frequency of their echolocation calls, but similar in their morphology. We found evidence for prey selectivity although there was a high degree of overlap in prey categories and sizes. R. a nis and R. pearsoni foraged on 16 and 7 categories insects, respectively, with Pyralidae, Geometridae, Melolonthidae dominating their diets. The degree of trophic niche overlap was 0.69. Pairwise comparisons suggested that there was no obvious di erentiation in prey categories and size. However, high prey availability in the environment (Simpson diversity index = 0.79 and Margalef richness index = 4.12) contributed to their coexistence by dampening the interspeci c competition. Since there are one or more mechanisms facil- itating species coexistence in a community, our results suggest that the spatial niche di erentiation in foraging microhabitats and in for- aging habitats at landscape scale may promote the coexistence of the two bat species. | 3 | Expression of Human Papillomavirus Type 16 L1 Protein in Transgenic Tobacco Plants显示文摘To develop a plant expression system for the production of the human papillomavirus type 16 (HPV16) vaccine, we investigated whether the HPV16 L1 protein can be expressed in tobacco plants and whether it can be used as the cheapest form of edible vaccine. The HPV16 L1 coding sequence was amplified by PCR using specific primers from the plasmid pGEM-T-HPV16 containing the template sequence, and subcloned into the intermediate vector pUCmT and binary vector pBI121 consecutively to obtain the plant expression plasmid pBI-L1. The T-DNA regions of the pBI-L1 binary vector contained the constitutive Cauliflower mosaic virus (CaMV) 35S promoter and the neomycin phosphotransferase npt Ⅱ gene, which allowed the selection of transformed plants using kanamycin. The tobacco plants were transformed by cocultivating them, using the leaf disc method, with Agrobacterium tumefaciens LBA4404, which harbored the plant expression plasmid. The regenerated transgenic tobacco plants were selected using kanamycin, and confirmed by PCR. The results of the Southern blot assay also showed that the HPV16 L1 gene was integrated stably into the genome of the transformed tobacco plants. The Western blot analysis showed that the transformed tobacco leaves could express the HPV 16 L1 protein. Furthermore, it was demonstrated by ELISA assay that the expressed protein accounted for 0.034%-0.076% of the total soluble leaf protein, was able to form 55nm virus-like particles compatible with HPV virus-like particle (VLP), and induced mouse erythrocyte hemagglutination in vitro. | 4 | BURST-LDA: A NEW TOPIC MODEL FOR DETECTING BURSTY TOPICS FROM STREAM TEXT显示文摘Topic models such as Latent Dirichlet Allocation(LDA) have been successfully applied to many text mining tasks for extracting topics embedded in corpora. However, existing topic models generally cannot discover bursty topics that experience a sudden increase during a period of time. In this paper, we propose a new topic model named Burst-LDA, which simultaneously discovers topics and reveals their burstiness through explicitly modeling each topic's burst states with a first order Markov chain and using the chain to generate the topic proportion of documents in a Logistic Normal fashion. A Gibbs sampling algorithm is developed for the posterior inference of the proposed model. | 5 | Patterns and causes of geographic variation in bat echolocation pulses显示文摘Evolutionary biologists have a long-standing interest in how acoustic signals in animals vary geographically,because divergent ecology and sensory perception play an important role in speciation.Geographic comparisons are valuable in determining the factors that influence divergence of acoustic signals.Bats are social mammals and they depend mainly on echolocation pulses to locate prey,to navigate and to communicate.Mounting evidence shows that geographic variation of bat echolocation pulses is common,with a mean 5-10 kHz differences in peak frequency,and a high level of individual variation may be nested in this geographical variation.However,understanding the geographic variation of echolocation pulses in bats is very difficult,because of differences in sample and statistical analysis techniques as well as the variety of factors shaping the vocal geographic evolution.Geographic differences in echolocation pulses of bats generally lack latitudinal,longitudinal and elevational patterns,and little is known about vocal dialects.Evidence is accumulating to support the fact that geographic variation in echolocation pulses of bats may be caused by genetic drift,cultural drift,ecological selection,sexual selection and social selection.Future studies could relate geographic differences in echolocation pulses to social adaptation,vocal learning strategies and patterns of dispersal.In addition,new statistical techniques and acoustic playback experiments may help to illustrate the causes and consequences of the geographic evolution of echolocation pulse in bats. | Tinglei JIANG Hui WU Jiang FENG | 2015 | Integrative Zoology2015,10,3: | 2 |

This unified ideal orbital resonance model is valid for all the commensurabilities between the rotational angular velocity of the Earth and the angular velocities of mean orbital motion of artificial satellites with arbitrary inclination and small eccentricity, and can be also transformed into Garfinkel's general expression of ideal resonance problem. Then 1/1 resonance of the 24-hour satellite with arbitrary inclination and small eccentricity is analyzed under the effect of harmonics of J2 and J 22 of the geopotential, based on the unified ideal model of mean motion resonance.
