| 1 | Establishment of a novel corneal endothelial cell line from domestic rabbit,Oryctolagus curiculus显示文摘To develop a rabbit corneal endothelial (RCE) cell line, in vitro culture of RCE cells was initiated from Oryctolagus curiculus corneas and a novel RCE cell line was established in this study. To initiate the primary culture of RCE cells, corneas from rabbit eyes were sliced and attached into glutin-coated wells with endothelial cell surface down. After being cultured at a time-gradient interval from 48 to 6 h, the corneal slices were detached and reattached into new wells, respectively. Cells in the wells containing only a pure population of RCE cells were collected and cultured in 20% FBS-DMEM/F12 medium con- taining chondroitin sulfate, ocular extract, epidermal growth factor (EGF), basic fibroblast growth factor (bFGF), carboxymethyl-chitosan, N-acetylglucosamine hydrochloride, glucosamine hydrochloride, culture medium of rabbit corneal stromal cells and oxidation-degradation products of chondroitin sul- fate at 37℃, 5% CO2. The cultured RCE cells, in quadrangle and polygonal shapes, proliferated to con- fluence 3 weeks later. During the subsequent subculture, the shape of RCE cells changed gradually from polygonal to more fibroblastic. A novel RCE cell line, growing at a steady rate, with a population doubling time of 53.8 h, has been established and subcultured to passage 67. Chromosome analysis showed that the RCE cells exhibited chromosomal aneuploidy with the modal chromosome number of 44. The results of immuno-cytochemical staining with neuron specific enolase (NSE) confirmed that the RCE cells were in neuroectodermal origin. Combined with the results of vascular endothelial growth factor (VEGF) treatment and endothelial cell morphology recovery, it can be concluded that the cell line established here is an RCE cell line. | 3 | Major advances in studies of the physical geography and living environment of China during the past 70 years and future prospects显示文摘The natural environment provides material essentials for human survival and development. The characteristics,processes, regional differentiation and forcing mechanisms of the elements of the natural environment(e.g. geomorphology,climate, hydrology, soil, etc.) are the main objects of research in physical geography. China has a complex natural environment and huge regional differentiation and therefore it provides outstanding reserach opportunities in physical geography. This review summarizes the most important developments and the main contributions of research in the physical geography and human living environment in China during the past 70 years. The major topics addressed are the uplift of the Tibetan Plateau and the evolution of its cryosphere, the development of fluvial systems, the acidification of the vast arid region of the Asian interior, variations in the monsoon and westerly climate systems on multiple timescales, the development of lakes and wetlands, the watershed system model, soil erosion, past human-environment interactions, biogeography, and physical geographic zonality. After briefly introducing international research developments, we review the history of research in physical geography in China, focusing on the major achievements and major academic debates, and finally we summarize the status of current research and the future prospects. | 4 | MicroRNA-324-3p Plays A Protective Role Against Coxsackievirus B3-Induced Viral Myocarditis显示文摘Viral myocarditis(VM) is an inflammatory disease of the myocardium associated with heart failure, which is caused by common viral infections. A majority of the infections are initiated by coxsackievirus B3(CVB3). Micro RNAs(mi RNAs)have a major role in various biological processes, including gene expression, cell growth, proliferation, and apoptosis, as well as viral infection and antiviral immune responses. Although, mi RNAs have been found to regulate viral infections,their role in CVB3 infection remains poorly understood. In the previous study, mi RNA microarray results showed that mi R-324-3 p expression levels were significantly increased when cells and mice were infected with CVB3. It was also found that miR-324-3p downregulated TRIM27 and decreased CVB3 replication in vitro and in vivo. In vitro, analysis of downstream signaling of TRIM27 revealed that, miR-324-3p inhibited CVB3 infection, and reduced cytopathic effect and viral plaque formation by reducing the expression of TRIM27. In vivo, miR-324-3p decreased the expression of TRIM27,reduced cardiac viral replication and load, thereby strongly attenuating cardiac injury and inflammation. | 5 | Analyses on the air and snow temperatures near ground with observations of an AWS at Dome A,the summit of Antarctic Plateau显示文摘As the summit of the Antarctic Plateau, Dome A has been received international attentions.In this paper, observational data of an automatic weather station (AWS) at Dome A in 2005–2007 were used to analyze the seasonal variations of air temperatures near the ground and snow temperatures within a depth of 10 m. Analyses on the air temperatures show a typical feature of the coreless winter, and strong inversion maintains during the long winter. Accordingly the stratification near the ground is dominated by the near-neutral stable states. Seasonal fluctuations of the snow temperature decrease in amplitude and lag in phase with depth increasing, which leads to distinct seasonal temperature profiles within the depth of 10 m. Measurements show the mean annual air temperature near ground is about 5°C higher than the 10 m firn temperature due to the strong inversion near the ground. However, our estimation of the annual mean of air temperature at the ground based on the boundary layer theory is close to the mean 10 m firn temperature. The lowest air temperature (–82.7°C) currently measured at the Dome A is not the lowest one ever recorded in Antarctica, but the extremely low mean 10 m firn temperature (–58.2°C) indicates very low ground temperature. | 6 | Effects of purified zearalenone on selected immunological and histopathologic measurements of spleen in post-weanling gilts显示文摘The present study was aimed at investigating the adverse effects of dietary zearalenone(ZEA) on the lymphocyte proliferation rate(LPR), interleukin-2(IL-2), mRNA expressions of pro-inflammatory cytokines, and histopathologic changes of spleen in post-weanling gilts. A total of 20 crossbred piglets(Yorkshire × Landrace × Duroc) with an initial BW of 10.36 ± 1.21 kg(21 d of age) were used in the study.Piglets were fed a basal diet with an addition of 0.1.1,2.0, or 3.2 mg/kg purified ZEA for 18 d ad libitum.The results showed that LPR and IL-2 production of spleen decreased linearly(P < 0.05) as dietary ZEA increased. Splenic mRNA expressions of interleukin-1β(IL-1β) and interleukin-6(IL-6) were linearly upregulated(P < 0.05) as dietary ZEA increased. On the contrary, linear down-regulation(P<0.05) of mRNA expression of interferon-γ(IFN-γ) was observed as dietary ZEA increased. Swelling splenocyte in1.1 mg/kg ZEA treatments, atrophy of white pulp and swelling of red pulp in 2.0 and 3.2 mg/kg ZEA treatments were observed. | 7 | Fabrication of a nano-sized ZSM-5 zeolite with intercrystalline mesopores for conversion of methanol to gasoline显示文摘Carbon deposition during methanol to hydrocarbons leads to the quick deactivation of ZSM-5 catalyst and it is one of the major problems for this technology. Decreasing the crystal size or introducing mesopores into ZSM-5 zeolites can improve its diffusion property and decrease the coke formation. In this paper, nano-sized ZSM-5 zeolite with intercrystalline mesopores combining the mesoporous and nanosized structure was fabricated. For comparison, the mesoporous ZSM-5 and nano-sized ZSM-5 were also prepared. These catalyst samples were characterized by XRD, BET, NH3-TPD, TEM, Py-IR and TG techniques and used on the conversion of methanol to gasoline in a fixed-bed reactor at T = 405 °C, WHSV = 4.74 h-1and P = 1.0 MPa. It was found that the external surface area of the nano-sized ZSM-5 zeolite with intercrystalline mesopores reached 104 m2/g, larger than that of mesoporous ZSM-5(66 m2/g) and nanosized ZSM-5(76 m2/g). Catalytic lifetime of the nano-sized ZSM-5 zeolite with intercrystalline mesopores was 93 h, which was only longer than that of mesoporous ZSM-5(86 h), but shorter than that of nanosized ZSM-5(104 h). Strong acidity promoted the coke formation and thus decreased the catalytic lifetime of the nano-sized ZSM-5 zeolite with intercrystalline mesopores though it presented large external surface that could improve the diffusion property. The special zeolite catalyst was further dealuminated to decrease the strong acidity. After this, its coke formation rate was slowed and catalytic lifetime was prolonged to 106 h because of the large external surface area and decreased weak acidity. | 8 | Development of an Inactivated Iridovirus Vaccine Against Turbot Viral Reddish Body Syndrome显示文摘Turbot(Scophthalmus maximus L.) reddish body iridovirus(TRBIV) was propagated in turbot fin cells(TF cells) and inactivated as the TRBIV vaccine with its protection efficiency evaluated in this study.TF cells were cultured in 10% bovine calf serum(BCS)-containing MEM medium(pH7.0) at 22℃,in which TRBIV propagated to a titer as high as 105.6 TCID50 mL-1.The TRBIV was inactivated with 0.1% formalin and formulated with 0.5% aluminum hydroxide.The inactivated vaccine caused neither cytopathogenic effect(CPE) on TF cells nor pathogenic effect on turbots.After being administered with the vaccine twice via muscle injection,the turbot developed high-tittered TRBIV neutralizing antibodies in a dose-dependent manner.The vaccine protected the turbot from dying with an immunoprotection rate of 83.3% as was determined via subcutaneous vaccination in the laboratory and 90.5% via bath vaccination in turbot farms,respectively.The inactivated vaccine was very immunogenic,efficiently preventing tur-bot from death.It holds the potential of being applied in aquaculture. | FAN Tingjun HU Xiuzhong WANG Liyan GENG Xiaofen JIANG Guojian YANG Xiuxia YU Miaomiao | 2012 | Journal of Ocean University of China2012,11,1: | 5 |
| 9 | Metformin activates chaperone-mediated autophagy and improves disease pathologies in an Alzheimer disease mouse model显示文摘Chaperone-mediated autophagy(CMA)is a lysosome-dependent selective degradation pathway implicated in the pathogenesis of cancer and neurodegenerative diseases.However,the mechanisms that regulate CMA are not fully understood.Here,using unbiased drug screening approaches,we discover Metformin,a drug that is commonly the first medication prescribed for type 2 diabetes,can induce CMA.We delineate the mechanism of CMA induction by Metformin to be via activation of TAK1-IKKα/β signaling that leads to phosphorylation of Ser85 of the key mediator of CMA,Hsc70,and its activation.Notably,we find that amyloid-beta precursor protein(APP)is a CMA substrate and that it binds to Hsc70 in an IKKα/β-dependent manner.The inhibition of CMA-mediated degradation of APP enhances its cytotoxicity.Importantly,we find that in the APP/PS1 mouse model of Alzheimer's disease(AD),activation of CMA by Hsc70 overexpression or Metformin potently reduces the accumulated brain Aβplaque levels and reverses the molecular and behavioral AD phenotypes.Our study elucidates a novel mechanism of CMA regulation via Metformin-TAK1-IKKα/β-Hsc70 signaling and suggests Metformin as a new activator of CMA for diseases,such as AD,where such therapeutic intervention could be beneficial. | Xiaoyan Xu Yaqin Sun Xufeng Cen Bing Shan Qingwei Zhao Tingxue Xie Zhe Wang Tingjun Hou Yu Xue Mengmeng Zhang Di Peng Qiming Sun Cong Yi Ayaz Najafov Hongguang Xia | 2021 | Protein & Cell2021,12,10: | 5 |

| 11 | Engineering osteoarthritic cartilage model through differentiating senescent human mesenchymal stem cells for testing disease-modifying drugs显示文摘Significant cellular senescence has been observed in cartilage harvested from patients with osteoarthritis(OA).In this study,we aim to develop a senescence-relevant OA-like cartilage model for developing disease-modifying OA drugs(DMOADs).Spe-cifically,human bone marrow-derived mesenchymal stromal cells(MSCs)were expanded in vitro up to passage 10(P10-MSCs).Following their senescent phenotype formation,P10-MSCs were subjected to pellet culture in chondrogenic medium.Results from qRT-PCR,histology,and immunostaining indicated that cartilage generated from P10-MSCs displayed both senescent and OA-like phenotypes without using other OA-inducing agents,when compared to that from normal passage 4(P4)-MSCs.Interestingly,the same gene expression differences observed between P4-MSCs and P10-MSC-derived cartilage tissues were also observed between the preserved and damaged OA cartilage regions taken from human samples,as demonstrated by RNA sequencing data and other analysis methods.Lastly,the utility of this senescence-initiated OA-like cartilage model in drug development was assessed by testing several potential DMOADs and senolytics.The results suggest that pre-existing cellular senescence can induce the generation of OA-like changes in cartilage.The P4-and P10-MSCs derived cartilage models also represent a novel platform for predicting the efficacy and toxicity of potential DMOADs on both preserved and damaged cartilage in humans. | Ning Wang Yuchen He Silvia Liu Meagan J.Makarcyzk Guanghua Lei Alexander Chang Peter G Alexander Tingjun Hao Anne-Marie Padget Nuria de Pedro Tsapekos Menelaos Hang Lin | 2022 | Science China(Life Sciences)2022,65,2: | 5 |

| 14 | A zirconium modified Co/SiO_2 Fischer-Tropsch catalyst prepared by dielectric-barrier discharge plasma显示文摘Co/SiO2 and zirconium promoted Co/Zr/SiO2 catalysts were prepared using dielectric-barrier discharge(DBD) plasma instead of the conventional thermal calcination method.Fischer-Tropsch Synthesis(FTS) performances of the catalyst were evaluated in a fixed bed reactor.The results indicated that the catalyst treated by DBD plasma shows the higher FTS activity and yield of heavy hydrocarbons as compared with that treated by the conventional thermal calcination method.Increase in CO conversion was unnoticeable on the Co/SiO2 catalyst,but significant on the Co/Zr/SiO2 catalyst,both prepared by DBD plasma.On the other hand,heavy hydrocarbon selectivity and chain growth probability (a value) were enhanced on all the catalysts prepared by the DBD plasma.In order to study the effect of the DBD plasma treatment on the FTS performance,the catalysts were characterized by N2-physisorption,H2-temperature programed reduction(H2-TPR),H2-temperature-programmed desorption(H2-TPD) and oxygen titration,transmission electron microscope(TEM) and X-ray diffraction(XRD).It was proved that,compared with the traditional calcination method,DBD plasma not only could shorten the precursor decomposition time,but also could achieve better cobalt dispersion,smaller Co3O4 cluster size and more uniform cobalt distribution.However,cobalt reducibility was hindered to some extent in the Co/SiO2 catalyst prepared by DBD plasma,while the zirconium additive prevented significantly the decrease in cobalt reducibility and increased cobalt dispersion as well as the FTS performance. | Yunhui Jiang Tingjun Fu Jing Lü Zhenhua Li | 2013 | Journal of Energy Chemistry2013,22,3: | 4 |
| 15 | Discovery of Novel Androgen Receptor Ligands by Structure-based Virtual Screening and Bioassays显示文摘Androgen receptor(AR) is a ligand-activated transcription factor that plays a pivotal role in the development and progression of many severe diseases such as prostate cancer, muscle atrophy, and osteoporosis. Binding of ligands to AR triggers the conformational changes in AR that may affect the recruitment of coactivators and downstream response of AR signaling pathway.Therefore, AR ligands have great potential to treat these diseases. In this study, we searched for novel AR ligands by performing a docking-based virtual screening(VS) on the basis of the crystal structure of the AR ligand binding domain(LBD) in complex with its agonist. A total of 58 structurally diverse compounds were selected and subjected to LBD affinity assay, with five of them(HBP1-3, HBP1-17, HBP1-38, HBP1-51, and HBP1-58) exhibiting strong binding to AR-LBD. The IC50 values of HBP1-51 and HBP1-58 are 3.96 m M and 4.92 m M, respectively, which are even lower than that of enzalutamide(Enz, IC50= 13.87 m M), a marketed second-generation AR antagonist. Further bioactivity assays suggest that HBP1-51 is an AR agonist, whereas HBP1-58 is an AR antagonist. In addition, molecular dynamics(MD) simulations and principal components analysis(PCA) were carried out to reveal the binding principle of the newlyidentified AR ligands toward AR. Our modeling results indicate that the conformational changes of helix 12 induced by the bindings of antagonist and agonist are visibly different. | 16 | Permafrost dynamics and their hydrologic impacts over the Russian Arctic drainage basin显示文摘Permafrost is an important component in hydrological processes because changes in runoff over the Arctic drainage basin cannot be well explained by changes in precipitation-related variables.However,current understanding of the influences of permafrost on hydrological dy-namics is insufficient.This study investigated historical variations in permafrost conditions and their potential hydrologic effects over the Russian Arctic drainage basin.The results show that soil temperature(at 0.40 m below surface)has increased about 1.4℃over the Ob,1.5℃over the Yenisei,and 1.8℃over the Lena River basin from 1936 through 2013,possibly resulted in a significant thawing of permafrost.Rapid active layer changes have occurred since the 1970s.The volume of the active layer increased by 28,142,and 228 km3 over the Ob,Yenisei,and Lena basins,respectively,since the 1970s.Melting ground ice caused by deepening active layer may be a limited contribution to annual runoff.Runoff during freeze season(October--April)showed significant positive correlations(p<0.05)to active layer thickness in the Yenisei and Lena basins while negative correlation(p>0.05)in the Ob basin.These results imply that,in basins with high permafrost coverage,a deeper active layer increased soil water storage capacity and perhaps contribute to an increase in winter runoff. | WANG Kang ZHANG Tingjun Daqing YANG | 2021 | Advances in Climate Change Research2021,12,4: | 3 |

| 18 | Modeled response of talik development under thermokarst lakes to permafrost thickness on the Qinghai-Tibet Plateau显示文摘Permafrost thickness under identical climates in cold regions can vary significantly because it is severely affected by climate change, topography, soil physical and thermal properties, and geothermal conditions. This study numerically investigates the response of ground thermal regime and talik development processes to permafrost with different thicknesses under a thermokarst lake on the Qinghai-Tibet Plateau. On the basis of observed data and information from a representative monitored lake in the Beiluhe Basin, we used a heat transfer model with phase change under a cylindrical coordinate system to conduct three simulation cases with permafrost thicknesses of 45 m, 60 m, and 75 m, respectively. The simulated results indicate that increases in permafrost thickness not only strongly retarded the open talik formation time, but also delayed the permafrost lateral thaw process after the formation of open talik. | 19 | Impacts of landscape and climatic factors on snow cover in the Altai Mountains,China显示文摘Snow properties and their changes are crucial to better understanding of hydrological processes,soil thermal regimes,and surface energy balances.Reliable data and information on snow depth and snow water equivalent(SWE)are also crucial for water resource assessments and socio-economic development at local and regional scales.However,these data are extremely limited and unreliable in northern Xinjiang,China.This study thus aims to investigate spatial variations of snow depth,SWE,and snow density based on winter snowfield surveys during 2015 through 2017 in the Altai Mountains,northwestern China.The results indicated that snow depth(25-114 cm)and SWE(40-290 mm)were greater in the alpine Kanas-Hemu region,and shallow snow accumulated(9-42 cm for snow depth,26-106 mm for SWE)on the piedmont sloping plain.While there was no remarkable regional difference in the distribution of snow density.Snow property distributions were strongly controlled by topography and vegetation.Elevation and latitude were the most important factors affecting snow depth and SWE,while snow density was strongly affected by longitude across the Altai Mountains in China.The influence of topography on snow property distributions was spatially heterogenous.Mean snow depth increased from 13.7 to 31.2 cm and SWE from 28.5 to 79.9 mm,respectively,with elevation increased from 400 to 1000 m a.s.l.on the piedmont sloping plain.Snow depth decreased to about 15.1 cm and SWE to about 28.5 mm from 1000 to 1800 m a.s.l.,then again increased to about 98.1 cm and 271.7 mm on peaks(-2000 m a.s.l.)in the alpine Kanas-Hemu.Leeward slopes were easier to accumulate snow cover,especially on north-,east-,and southeast-facing slopes.Canopy interception was also the cause of the difference in snow distribution.Snow depth,SWE,and snow density in forests were reduced by 8%-53%,2%-67%and-4%to+48%,respectively,compared with surrounding open areas.Especially when snow depth was less than 40 cm,snow depth and SWE differences in forests were more exaggerated.This study provides a basic data set of spatial distributions and variations of snow depth,SWE and snow density in the Altai Mountains,which can be used as an input parameter in climate or hydrological models.These first-hand observations will help to better understand the relationship between snow,topography and climate in mountainous regions across northern China and other high-mountain Asian regions. | ZHONG Xin-Yue ZHANG Tingjun SU Hang XIAO Xiong-Xin WANG Shu-Fa HU Yuan-Tao WANG Hui-Juan ZHENG Lei ZHANG Wei XU Min WANG Jian | 2021 | Advances in Climate Change Research2021,12,1: | 3 |
| 20 | Establishment of a Spleen Cell Line from Large Yellow Croaker Pseudosciaena crocea and its Primitive Application in Foreign Gene Transfection显示文摘A large yellow croaker,Pseudosciaena crocea,spleen(LYCS)cell line was established and the feasibility of using it for foreign gene transfection was evaluaed in this study.Primary culture of LYCS cells was initiated from spleen tissue pieces,which were cultured at 25℃ in Dulbecco's modiced Eagle medium/F12 medium(DMEM/F12,1:1)(pH7.2),supplemented with 20% fetal bovine serum,carboxymethyl chitosan,chondroitin sulfate,basic fibroblast growth factor(bFGF)and insulin-like growth factor-I(IGF-I).The cultured LYCS cells,in fibroblast shape,proliferated to 100% confluency 20 days later.Chromosome analyses indicated that the LYCS cells exhibited chromosomal aneuploidy with a modal chromosome number of 48 which displayed the normal diploid karyotype of P.crocea(6m+6sm+36t,NF=60).A LYCS cell line,with a population doubling time of 48.7 h at passage 60,has been established and subcultured to passage 70.Transgenic feasibility test demonstrated that positive green fluorescence protein(GFP)expression was observed in LYCS cells after pcDNA3.1-GFP plasmid transfection.In conclusion,a continuous foreign gene trans-fection feasible LYCS cell line has been established successfully.The cell line might serve as a valuable tool for studies of transgenic breeding and has potential applications for different kinds of cytotechnological studies. | SUN Ai FAN Tingjun YANG Xiuxia XU Bin XU Xiaohui GUO Xueyang | 2011 | Journal of Ocean University of China2011,10,4: | 3 |
