8GPR54 deficiency reduces the Treg population and aggravates experimental autoimmune encephalomyelitis in mice显示文摘GPR54 is highly expressed in the central nervous system and plays a crucial role in pubertal development. However, GRP54 is also expressed in the immune system, implying possible immunoregulatory functions. Here we investigated the role of GPR54 in T cell and immune tolerance. GPR54 deficiency led to an enlarged thymus, an increased number of thymocytes, and altered thymic micro-architecture starting around puberty, indicating GPR54 function in T-cell development through its regulatory effect on the gonadal system. However, flow cytometry revealed a significant reduction in the peripheral regulatory T cell population and a moderate decrease in CD4 single-positive thymocytes in prepubertal Gpr54^(-/-) mice. These phenotypes were confirmed in chimeric mice with GPR54 deficient bone marrow-derived cells. In addition, we found elevated T cell activation in peripheral and thymic T cells in Gpr54^(-/-) mice. When intact mice were immunized with myelin oligodendrocyte glycoprotein, a more severe experimental autoimmune encephalomyelitis(EAE) developed in the Gpr54^(-/-) mice. Interestingly, aggravated EAE disease was also manifested in castrated and bone marrow chimeric Gpr54^(-/-) mice compared to the respective wild-type control,suggesting a defect in self-tolerance resulting from GPR54 deletion through a mechanism that bypassed sex hormones. 12PRIMPOL competes with RAD51 to resolve G-quadruplexinduced replication stress via its interaction with RPA显示文摘PRIMPOL(primase-polymerase)is a recently discovered DNA primase-polymerase involved in DNA damage tolerance and replication stress response in eukaryotic cells.However,the detailed mechanism of the PRIMPOL response to replication stress remains elusive.Here,we demonstrate that replication-related factors,including replication protein A(RPA),regulate the accumulation of PRIMPOL in subnuclear foci in response to replication stress induced by replication inhibitors.Moreover,PRIMPOL works at G-quadruplexes(G4s)in human cells to resolve the replication stress induced by G4s.The formation of PRIMPOL foci persists throughout the cell cycle.We further demonstrate that PRIMPOL competes with RAD51 to resolve G4-induced replication stress.In conclusion,our results provide novel insight into the mechanism of PRIMPOL in G4s to resolve replication stress and competition between PRIMPOL(repriming)-and RAD51(fork reversal)-mediated pathways,which indicates a new strategy to improve the tumor response to DNA-damaging chemotherapy by targeting the PRIMPOL pathway.Tingfang Li Lu Tang Haomeng Kou Feng Wang 2023Acta Biochimica et Biophysica Sinica2023,55,3:0
13Mechanism Analysis of ' 7·21' Extremely Heavy Rainstorm Process in 2012 in Beijing显示文摘Extremely heavy rainstorm occurred in Beijing on July 21,2012,which was the most serious since 1961.Based on analyzing the precipitation characteristics,formation mechanism of the rainstorm process was analyzed.Results showed that when the precipitation process occurred,it was stable east-high and west-low situation at 500 hPa,and there was a steady stream of water vapor transportation at middle and low layers and strong vertical ascending motion at 700 hPa.The distribution of physical quantity field(relative humidity,vorticity and divergence)showed that they were all benefited to the formation of rainstorm.Then,falling zone of rainstorm and the movement of rain belt,generation,development and weakening of precipitation were analyzed.Finally,according to circulation situation and the distribution of physical quantity at each layer,vertical distribution of physical quantity and distribution of water vapor and jet stream, '7·21' rainstorm model was summarized.Mao Weiping Yan Yan Wang Tingfang Zhang Youshu 2015Meteorological and Environmental Research2015,6,1:0
