1The ChinaMAP analytics of deep whole genome sequences in 10,588 individuals显示文摘Metabolic diseases are the most common and rapidly growing health issues worldwide.The massive population-based human genetics is crucial for the precise prevention and intervention of metabolic disorders.The China Metabolic Analytics Project(ChinaMAP)is based on cohort studies across diverse regions and ethnic groups with metabolic phenotypic data in China.Here,we describe the centralized analysis of the deep whole genome sequencing data and the genetic bases of metabolic traits in 10,588 individuals from the ChinaMAP.The frequency spectrum of variants,population structure,pathogenic variants and novel genomic characteristics were analyzed.The individual genetic evaluations of Mendelian diseases,nutrition and drug metabolism,and traits of blood glucose and BMI were integrated.Our study establishes a large-scale and deep resource for the genetics of East Asians and provides opportunities for novel genetic discoveries of metabolic characteristics and disorders.Yanan Cao Lin Li Min Xu Zhimin Feng Xiaohui Sun Jieli Lu Yu Xu Peina Du Tiange Wang Ruying Hu Zhen Ye Lixin Shi Xulei Tang Li Yan Zhengnan Gao Gang Chen Yinfei Zhang Lulu Chen Guang Ning Yufang Bi Weiqing Wang The ChinaMAP Consortium 2020Cell Research2020,30,9:20
2Urban Water Resource Utilization Efficiency in China显示文摘The efficient use of water resources directly affects environmental, social, and economic development; therefore, it has a significant impact on urban populations. A slacks-based measure for data envelopment analysis(SBM-DEA) has been widely used in energy efficiency and environmental efficiency analyses in recent years. Based on this model, data from 316 cities were examined and a category method was employed involving three different sorting techniques to empirically evaluate the efficiency of urban water resource utilization in China between 2000 and 2012. The overall efficiency(OE) of urban water resource utilization in China was initially low, but has improved over the past decade. The scale efficiency(SE) was higher than the pure technological efficiency(PTE); PTE is a major determining factor of OE, and has had an increasingly significant effect. The efficiency of water resource utilization varied according to the region, urban scale, and economic function. The OE score for the eastern China was higher than for the rest of the region, and the OE score for the western China was higher than for the central China. The OE score for urban water resource utilization has improved with urban expansion, except in the case of small cities. The SE showed an inverted ′U-shaped′ trend with increasing urban expansion. The OE of urban water utilization in comprehensive functional cities was greater than in economic specialization cities, and was greater in heavy industry specialization cities than in other specialization cities. 3Correlation analysis of Treg/Th17 cells and related cytokines in patients with psoriasis vulgaris显示文摘OBJECTIVE: To investigate potential differences in circulating levels of T regulatory (Treg)/T helper 17 (Th17) cells, related inflammatory cytokines and specific transcription factors in healthy individuals and patients with psoriasis conforming to one of three Traditional Chinese Medicine (TCM) syndromes: blood-heat syndrome (BHS), blood-stasis syndrome (BSS) and blood-dryness syndrome (BDS). METHODS: Sixty-seven patients with psoriasis were recruited and assigned to one of three corresponding TCM syndrome groups: BHS (n = 40), BSS (n = 14) and BDS (n = 13 patients). The control group comprised 21 healthy individuals. The circulating levels of Treg/Th17 cells in peripheral blood were assessed using flow cytometry;the levels of inflammatory cytokines interleukin (IL)-10 and tumor necrosis factor (TNF)-α by enzyme-linked immunosorbent assay;and the mRNA expression of T cell-specific transcription factors retinoic acid-related orphan receptor γt (RORγt) and forkhead box P3 (Foxp3) by quantitative real-time PCR. RESULTS: The ratio of Th17 cells and the levels of TNF-α and RORγt were all significantly higher in the BHS and BSS groups than the control group (P < 0.05), while the ratio of Treg cells and the levels of IL-10 and Foxp3 mRNA in the BHS group were significantly lower compared with the control group (P < 0.05). No significant differences were seen between the BSS group and the control group. The ratio of Th17 cells and the levels of TNF-α and RORγt in the BDS group were not significantly different from those of the control group;however, the ratio of Treg cells and the levels of IL-10 and Foxp3 were all lower than those in the healthy controls (P < 0.05). CONCLUSION: Compared with healthy individuals, the ratio of Th17 cells and the levels of related cytokines were higher, while the ratio of Treg cells and the levels of related cytokines were lower, in the peripheral blood of psoriasis/BHS patients;corresponding results for the BSS and BDS groups also showed differences. 4Recombinant rabies virus expressing IL-15 enhances immunogenicity through promoting the activation of dendritic cells in mice显示文摘Rabies remains a public health threat that kills approximately 59,000 people worldwide each year,most of which are from the developing countries of Africa and Asia where dog rabies are endemic.Therefore, developing an affordable and efficacious vaccine is crucial for rabies control in these countries. Interleukin(IL)-15, an immunoregulatory cytokine, is a pluripotent molecule with therapeutic potential, which targets many cell types and links the innate and adaptive immune system. In this study, IL-15 gene was cloned and inserted into the genome of a recombinant rabies virus(RABV) strain LBNSE(designated as LBNSE-IL15), and the effect of over-expression of IL-15 on the immunogenicity of RABV was investigated. It was found that mice vaccinated with LBNSEIL15 could induce significantly higher level of virus-neutralizing antibody(VNA) than those immunized with LBNSE, resulting in the higher protection after challenge. Further investigation was performed to find out the possible role of IL-15 plays in the process of antibody induction, and it was found that LBNSE-IL15 could enhance the maturation of dendritic cells(DCs) in immunized mice. Furthermore, the mice immunized with LBNSE-IL15 could promote the T_(FH) cells differentiation and the generation of germinal center B cells and plasma cells. 5A Deep Learning-Based Computational Algorithm for Identifying Damage Load Condition: An Artificial Intelligence Inverse Problem Solution for Failure Analysis显示文摘In this work,we have developed a novel machine(deep)learning computational framework to determine and identify damage loading parameters(conditions)for structures and materials based on the permanent or residual plastic deformation distribution or damage state of the structure.We have shown that the developed machine learning algorithm can accurately and(practically)uniquely identify both prior static as well as impact loading conditions in an inverse manner,based on the residual plastic strain and plastic deformation as forensic signatures.The paper presents the detailed machine learning algorithm,data acquisition and learning processes,and validation/verification examples.This development may have significant impacts on forensic material analysis and structure failure analysis,and it provides a powerful tool for material and structure forensic diagnosis,determination,and identification of damage loading conditions in accidental failure events,such as car crashes and infrastructure or building structure collapses.Shaofei Ren Guorong Chen Tiange Li Qijun Chen Shaofan Li 2018Computer Modeling in Engineering & Sciences2018,,12:3
