| 1 | Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. | 2 | Coal and coal byproducts:A large and developable unconventional resource for critical materials—Rare earth elements显示文摘Rare earth elements(REEs) are critical materials and provide significant values to national security,energy production, environmental protection and economic growth. The supply of REEs in U.S. solely relies on import as domestic production of REEs was ceased because of the environmental concerns during mining and lack of competitiveness. Nonetheless, unconventional REEs-containing resources,including produced water. acid mine drainage, and coal and coal byproducts(C&CBs) contain significant amounts of REEs. However, the concentrations of REEs in these resources are several orders of magnitude lower than that of REEs ores. Thus, extraction of REEs from these materials is challenging. Here we report REEs extraction with environmentally friendly method that successfully concentrated REEs from312 ppm in fly ash to 99.4% in the final product. Especially, the five critically important REEs(Dy, Eu, Nd.Tb. and Y) account for up to ~63% of the total weight of all REEs in the final 99.4%-purity product. Coal fly ash is one of the major solid coal utilization byproducts, representing great potential resources for REEs extraction. | 3 | Dietary seaweed-derived polysaccharides improve growth performance of weaned pigs through maintaining intestinal barrier function and modulating gut microbial populations显示文摘Background:Seaweed-derived polysaccharides(SDP)represent an attractive source of prebiotic nutraceuticals for the food and animal husbandry industry.However,the mechanism by which SDP from Enteromorpha mediates pig growth are not fully understood.This study aimed to investigate how SDP supplementation influences the growth performance and intestinal health in weaned pigs.Results:In Exp.1,240 weaned pigs were randomly assigned to four dietary treatments and fed with a basal diet or a basal diet containing 200,400 or 800 mg/kg SDP,respectively,in a 21-day trial.Pigs on the 400 or 800 mg/kg SDP-supplemented group had greater ADG and lower F/G ratio than those on the control group(P<0.05).In Exp.2,20 male weaned pigs were randomly assigned to two treatments and fed with a basal diet(CON group)or a basal diet supplemented with 400 mg/kg SDP(the optimum does from Exp.1),in a 21-day trial.Pigs fed the SDP diet had greater ADG,the concentrations of serum IL-6 and TNF-αand the activities of glutathione peroxidase,superoxide dismutase and catalase(P<0.05),and lower F/G,diarrhea rate,as well as serum D-lactate concentrations and diamine oxidase activity(P<0.05).Moreover,dietary SDP supplementation enhanced secretory immunoglobulin A content,villus height and villous height:crypt depth ratio in small intestine,as well as the lactase and maltase activities in jejunum mucosa(P<0.05).SDP supplementation elevated the mRNA levels of inflammatory responserelated genes(IL-6,TNF-α,TLR4,TLR6 and MyD88),and the mRNA and protein levels of ZO-1,claudin-1 and occludin in jejunum mucosa(P<0.05).Importantly,SDP not only increased the Lactobacillus population but also reduced the Escherichia coli population in cecum(P<0.05).Furthermore,SDP increased acetic acid and butyric acid concentrations in cecum(P<0.05).Conclusions:These results not only suggest a beneficial effect of SDP on growth performance and intestinal barrier functions,but also offer potential mechanisms behind SDP-facilitated intestinal health in weaned pigs. | Tiande Zou Jin Yang Xiaobo Guo Qin He Zirui Wang Jinming You | 2021 | Journal of Animal Science and Biotechnology2021,12,3: | 10 |
| 4 | Regional difference of annual precipitation and discharge variation over west China during the last 50 years显示文摘Using annual precipitation and discharge data measured in the past five decades,this paper analyzed the regional differences over west China in terms of climate and discharge variations,and investigated the relationship between the regional characteristics and the activities of South and East Asian sum-mer monsoon. Results revealed that the precipitation and discharge in the upper reaches of the Yellow River (Central West China) have a negative correlation with those in Xinjiang (northwest China) and the Yarlung Zangbo River (the upper reaches of the Brahmaputra Rive,southwest China) regions. The geographical patterns of precipitation and discharge variations are different over west China,i.e. the regional climate displays the alteration of dry-wet-dry or wet-dry-wet from north to south in west China. The negative correlation of annual discharges between Xinjiang and the upper reaches of the Yellow River is found statistically significant in the decadal scale,and that between the Yarlung Zangbo River and the upper reaches of the Yellow River is found active in the interannual scale. | 8 | Hydrological effects of alpine permafrost in the headwaters of the Urumqi River,Tianshan Mountains显示文摘Against the background of climate change, alpine permafrost active layers have shown a gradual thickening trend and the hydrothermal conditions have undergone significant changes in the Tianshan Mountains and the Qinghai-Tibet Plateau, China. At the ice-free cirque basins in the headwaters of the Urumqi River(hereafter referred to as the Ice-Free Cirque) in eastern Tianshan, China, the hydrological effects of the alpine permafrost active layers appear to have also exhibited significant changes recently. The increasing trend of local precipitation is clear in May and June. The onset of winter and spring snowmelt runoff clearly lags behind increases of air temperature, and the runoff peak appears near the beginning of the melting season, which results in the spring runoff increasing. In summer, runoff decreases strongly and the maximum runoff occurs earlier. In our analysis of meteorological and hydrologic data from 1959 to 2010, the runoff and precipitation changes are significantly correlated. In the initial stage of runoff, the runoff-producing process is mainly under the control of the soil water content and soil temperature in the 0–30 cm active layers. Spring precipitation and snowmelt water are mainly involved in the processes of infiltration and evaporation while some melt water infiltrates into the seasonal thawed layer and stays above the frozen layers. During the strong ablation period in summer, the runoff-generating process is mainly controlled by soil water content in the active layers deeper than 60 cm.
