1Mitochondrial dysfunction,oxidative stress and apoptotic induction in microglial BV-2 cells treated with sodium arsenate显示文摘The treatment of microglial BV-2 cells with sodium arsenate(As(V):0.1-400 μmol/L — 48 hr)induces a dose-dependent response.The neurotoxic effects of high concentrations of As(V)(100,200 and 400 μmol/L) are characterized by increased levels of mitochondrial complexesⅠ,Ⅱ,and Ⅳ followed by increased superoxide anion generation.Moreover,As(V) triggers an apoptotic mode of cell death,demonstrated by an apoptotic SubG1 peak,associated with an alteration of plasma membrane integrity.There is also a decrease in transmembrane mitochondrial potential and mitochondrial adenosine triphosphate ATP.It is therefore tempting to speculate that As(V) triggers mitochondrial dysfunction,which may lead to defective oxidative phosphorylation subsequently causing mitochondrial oxidative damage,which in turn induces an apoptotic mode of cell death.Wafa Kharroubi Samia Hai Ahmed Thomas Nury Pierre Andreoletti Rachid Sakly Mohamed Hammami Gerard Lizard 2017Journal of Environmental Sciences2017,29,1:5

14Perioperative steroid administration reduces overall complications in patients undergoing liver resection:A meta-analysis显示文摘BACKGROUND Hepatic resection(HR)results in an inflammatory response that can be modified by perioperative steroid administration.However,it remains to be determined if this response's attenuation translates to a reduction in complications.AIM To evaluate if perioperative administration of steroids reduces complications following HR.METHODS A systematic review of randomized controlled trials(RCTs)was conducted on PubMed,Embase,and Cochrane Central Register of Controlled Trials to evaluate the effect of perioperative steroid(compared to placebo or no intervention)use in patients undergoing HR.Clinical outcomes were extracted,and meta-analysis was performed.RESULTS 8 RCTs including 590 patients were included.Perioperative steroid administration was associated with significant reduction in postoperative complications[odds ratios:0.58;95%confidence intervals(CI):0.35-0.97,P=0.04].There was also improvement in biochemical and inflammatory markers,including serum bilirubin on postoperative day 1[MD:-0.27;95%CI:(-0.47,-0.06),P=0.01],Creactive protein on postoperative day 3[MD:-4.89;95%CI:(-5.83,-3.95),P<0.001],and interleukin-6 on postoperative day 1[MD:-54.84;95%CI:(-63.91,-45.76),P<0.001].CONCLUSION Perioperative steroids administration in HR may reduce overall complications,postoperative bilirubin,and inflammation.Further studies are needed to determine the optimal dose and duration and patient selection.Hao-Han Hai Phoebe Aw Thomas Zheng Jie Teng Vishal G Shelat 2021World Journal of Gastrointestinal Surgery2021,13,9:0
15Cathepsin D overexpression in the nervous system rescues lethality and Aβ42 accumulation of cathepsin D systemic knockout in vivo显示文摘The lysosome is responsible for protein and organelle degradation and homeostasis and the cathepsins play a key role in maintaining protein quality control.Cathepsin D(CTSD),is one such lysosomal protease,which when deficient in humans lead to neurolipofuscinosis(NCL)and is important in removing toxic protein aggregates.Prior studies demonstrated that CTSD germ-line knockout-CtsdKO(CDKO)resulted in accumulation of protein aggregates,decreased proteasomal activities,and postnatal lethality on Day 26±1.Overexpression of wildtype CTSD,but not cathepsin B,L or mutant CTSD,decreased a-synuclein toxicity in worms and mammalian cells.In this study we generated a mouse line expressing human CTSD with a floxed STOP cassette between the ubiquitous CAG promoter and the cDNA.After crossing with Nestin-cre,the STOP cassette is deleted in NESTIN+cells to allow CTSD overexpression-CTSDtg(CDtg).The CDtg mice exhibited normal behavior and similar sensitivity to subchronic 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)induced neurodegeneration.By breeding CDtg mice with CDKO mice,we found that over-expression of CTSD extended the lifespan of the CDKO mice,partially rescued proteasomal deficits and the accumulation of Aβ42 in the CDKO.This new transgenic mouse provides supports for the key role of CTSD in protecting against proteotoxicity and offers a new model to study the role of CTSD enhancement in vivo.Xiaosen Ouyang Willayat Y.Wani Gloria A.Benavides Matthew J.Redmann Hai Vo Thomas van Groen Victor M.Darley-Usmar Jianhua Zhang 2023Acta Pharmaceutica Sinica B2023,13,10:0
16Rapid assessment of knowledge,attitudes,practices,and risk perception related to the prevention and control of Ebola virus disease in three communities of Sierra Leone显示文摘Background:The recent outbreak of the Ebola virus disease(EVD)in Sierra Leone has been characterized by the World Health Organization as one of the most challenging EVD outbreaks to date.The first confirmed case in Sierra Leone was a young woman who was admitted to a government hospital in Kenema following a miscarriage on 24 May 2014.On 5 January 2015,intensified training for an EVD response project was initiated at the medical university of Sierra Leone in Jui.To understand the knowledge,attitudes,practices,and perceived risk of EVD among the public,especially after this training,a rapid assessment was conducted from 10 to 16 March 2015.Methods:Interviews were conducted with 466 participants based on questionnaires that were distributed from 10 to 16 March 2015 by cluster sampling in three adjacent communities,namely Jui,Grafton,and Kossoh Town,in the Western Area Rural District of Sierra Leone.Results:It was found that knowledge about EVD was comprehensive and high.Positive attitude towards prevention was found to be satisfactory.Nearly all participants knew the reporting phone number 117 and had reported some change in behavior since learning about Ebola.More than half(62%)of the participants had a history of travelling to urban areas,which increases the risk of infection.The multivariable logistic regression analysis showed that community and occupation were variables associated with perceived risk of EVD.Conclusions:Our study showed that community level social mobilization and community engagement were an effective strategy in the special context.Hai Jiang Guo-Qing Shi Wen-Xiao Tu Can-Jun Zheng Xue-Hui Lai Xin-Xu Li Qiang Wei Mei Li Li-Quan Deng Xiang Huo Ming-Quan Chen Feng Xu Long-Jie Ye Xi-Chen Bai Tong-Nian Chen Shao-Hua Yin Thomas T.Samba Xiao-Feng Liang 2016Infectious Diseases of Poverty2016,5,1:0
17A practical community-based response strategy to interrupt Ebola transmission in sierra Leone, 2014–2015显示文摘Background:The Ebola virus disease spread rapidly in West Africa in 2014,leading to the loss of thousands of lives.Community engagement was one of the key strategies to interrupt Ebola transmission,and practical community level measures needed to be explored in the field and tailored to the specific context of communities.Methods:First,community-level education on Ebola virus disease(EVD)prevention was launched for the community’s social mobilizers in six districts in Sierra Leone beginning in November 2014.Then,from January to May of 2015,in three pilot communities,local trained community members were organized to engage in implementation of EVD prevention and transmission interruption measures,by involving them in alert case report,contact tracing,and social mobilization.The epidemiological indicators of transmission interruption in three study communities were evaluated.Results:A total of 6016 community social mobilizers from 185 wards were trained by holding 279 workshops in the six districts,and EVD message reached an estimated 631680 residents.In three pilot communities,72 EVD alert cases were reported,with 70.8%of them detected by trained local community members,and 14 EVD cases were finally identified.Contact tracing detected 64.3%of EVD cases.The median duration of community infectivity for the cases was 1 day.The secondary attack rate was 4.2%,and no third generation of infection was triggered.No health worker was infected,and no unsafe burial and noncompliance to EVD control measures were recorded.The community-based measures were modeled to reduce 77 EVD cases,and the EVD-free goal was achieved four months earlier in study communities than whole country of Sierra Leone.Conclusions: The community-based strategy of social mobilization and community engagement was effective in casedetection and reducing the extent of Ebola transmission in a country with weak health system. 18Resolving impact volatilization and condensation from target rock mixing and hydrothermal overprinting within the Chicxulub impact structure显示文摘This work presents isotopic data for the non-traditional isotope systems Fe,Cu,and Zn on a set of Chicxulub impactites and target lithologies with the aim of better documenting the dynamic processes taking place during hypervelocity impact events,as well as those affecting impact structures during the post-impact phase.The focus lies on material from the recent IODP-ICDP Expedition 364 Hole M0077A drill core obtained from the offshore Chicxulub peak ring.Two ejecta blanket samples from the UNAM 5 and 7 cores were used to compare the crater lithologies with those outside of the impact structure.The datasets of bulk Fe,Cu,and Zn isotope ratios are coupled with petrographic observations and bulk major and trace element compositions to disentangle equilibrium isotope fractionation effects from kinetic processes.The observed Fe and Cu isotopic signatures,with δ^(56/54)Fe ranging from0.95‰to 0.58‰and δ^(65/63)Cu from0.73‰to 0.14‰,mostly reflect felsic,mafic,and carbonate target lithology mixing and secondary sulfide mineral formation,the latter associated to the extensive and long-lived(>105 years)hydrothermal system within Chicxulub structure.On the other hand,the stable Zn isotope ratios provide evidence for volatility-governed isotopic fractionation.The heavier Zn isotopic compositions observed for the uppermost part of the impactite sequence and a metamorphic clast(δ^(66/64)Zn of up to 0.80‰and 0.87‰,respectively)relative to most basement lithologies and impact melt rock units indicate partial vaporization of Zn,comparable to what has been observed for Cretaceous-Paleogene boundary layer sediments around the world,as well as for tektites from various strewn fields.In contrast to previous work,our data indicate that an isotopically light Zn reservoir(δ^(66/64)Zn down to0.49‰),of which the existence has previously been suggested based on mass balance considerations,may reside within the upper impact melt rock(UIM)unit.This observation is restricted to a few UIM samples only and cannot be extended to other target or impact melt rock units.Light isotopic signatures of moderately volatile elements in tektites and microtektites have previously been linked to(back-)condensation under distinct kinetic regimes.Although some of the signatures observed may have been partially overprinted during post-impact processes,our bulk data confirm impact volatilization and condensation of Zn,which may be even more pronounced at the microscale,with variable degrees of mixing between isotopically distinct reservoirs,not only at proximal to distal ejecta sites,but also within the lithologies associated with the Chicxulub impact crater.Thomas Déhais Stepan M.Chernonozhkin Pim Kaskes Sietze J.de Graaff Vinciane Debaille Frank Vanhaecke Philippe Claeys Steven Goderis 2022Geoscience Frontiers2022,13,5:0
